| 1 | GSDMB promotes non-canonical pyroptosis by enhancing caspase-4 activity显示文摘Gasdermin B (GSDMB) has been reported to be associated with immune diseases in humans, but the detailed molecular mechanisms remain unsolved. The N-terminus of GSDMB by itself, unlike other gasdermin family proteins, does not induce cell death. Here, we show that GSDMB is highly expressed in the leukocytes of septic shock patients, which is associated with increased release of the gasdermin D (GSDMD) N-terminus. GSDMB expression and the accumulation of the N-terminal fragment of GSDMD are induced by the activation of the non-canonical pyroptosis pathway in a human monocyte cell line. The downregulation of GSDMB alleviates the cleavage of GSDMD and cell death. Consistently, the overexpression of GSDMB promotes GSDMD cleavage, accompanied by increased LDH release. We further found that GSDMB promotes caspase-4 activity, which is required for the cleavage of GSDMD in non-canonical pyroptosis, by directly binding to the CARD domain of caspase-4. | 2 | Early inoculation with caecal fermentation broth alters small intestine morphology,gene expression of tight junction proteins in the ileum,and the caecal metabolomic profiling of broilers显示文摘Background:The establishment of stable microbiota in early life is beneficial to the individual.Changes in the intestinal environment during early life play a crucial role in modulating the gut microbiota.Therefore,early intervention to change the intestinal environment can be regarded as a new regulation strategy for the growth and health of poultry.However,the effects of intestinal environmental changes on host physiology and metabolism are rarely reported.This study was conducted to investigate the effects of early inoculation with caecal fermentation broth on small intestine morphology,gene expression of tight junction proteins in the ileum,and cecum microbial metabolism of broilers.Results:Our data showed that early inoculation with caecal fermentation broth could improve intestine morphology.The small intestine villus height was significantly increased(P<0.05)in the intervened broilers compared to the control group,especially on day 28.A similar result was observed in the ratio of villus height to crypt depth(P<0.05).Meanwhile,we found early inoculation significantly increased(P<0.05)the expression levels of zonula occludens-1(ZO1)on days 14 and 28,claudin-1(CLDN1)on day 28,whereas the gene expression of claudin-2(CLDN2)was significantly decreased(P<0.05)on days 14 and 28.Gas chromatography time-of-flight/mass spectrometry(GC-TOF/MS)technology was further implemented to systematically evaluate the microbial metabolite profiles.Principal component analysis(PCA)and orthogonal partial least squares discriminant analysis(OPLS-DA)displayed a distinct trend towards separation between the fermentation broth group(F group)and the control group(C group).The differentially expressed metabolites were identified,and they were mainly functionally enriched in beta-alanine metabolism and biosynthesis of unsaturated fatty acids.In addition,1,3-diaminopropane was selected as a key biomarker that responded to early inoculation with caecal fermentation broth.Conclusions:These results provide insight into intestinal metabolomics and confirm that early inoculation with caecal fermentation broth can be used as a potential strategy to improve intestinal health of broilers. | Yujie Gong Wenrui Xia Xueting Wen Wentao Lyu Yingping Xiao Hua Yang Xiaoting Zou | 2020 | Journal of Animal Science and Biotechnology2020,11,2: | 3 |
| 3 | Reducing energy loss via tuning energy levels of polymer acceptors for efficient all-polymer solar cells显示文摘The open-circuit voltage(Voc) of all-polymer solar cells(all-PSCs) is typically lower than 0.9 V even for the most efficient ones.Large energy loss is the main reason for limiting Voc and efficiency of all-PSCs. Herein, through materials design using electron deficient building blocks based on bithiophene imides, the lowest unoccupied molecular orbital(LUMO) energy levels of polymer acceptors can be effectively tuned, which resulted in a reduced energy loss induced by charge generation and recombination loss due to the suppressed charge-transfer(CT) state absorption. Despite a negligible driving force, all-PSC based on the polymer donor and acceptor combination with well-aligned energy levels exhibited efficient charge transfer and achieved an external quantum efficiency over 70% while maintaining a large Voc of 1.02 V, leading to a 9.21% efficiency. | 4 | Optical and electrical properties of a spiral LED filament显示文摘This paper introduces a new type of spiral white light-emitting diodes(WLED) filament with high luminous efficiency and uniform optical performance. The optical and thermal properties of the flexible filament were investigated at different stretching heights, namely 0, 1, 2, and 3 cm. The results indicated that the filament showed the best optical characteristics at the stretching height of 2 cm, because of good heat dissipation. In addition, the radiation temperature of the filament was inversely proportional to the output luminous flux. The reliability of the filament at a stretching height of 2 cm was also evaluated after 1000 h of use. The result demonstrated that the luminous flux decay of the bulb was only 0.85%. | 5 | SFRP5 inhibits melanin synthesis of melanocytes in vitiligo by suppressing the Wnt/b-catenin signaling显示文摘Secreted frizzled-related protein 5(SFRP5)plays a pivotal role in regulating the development of many tissues and organs,however,as an inhibitor of Wnt signaling,the role of SFRP5 in vitiligo remains unknown.Hence,we speculated that SFRP5 might be associated with melanogenesis in melanocytes by regulating Wnt signaling in vitiligo.In this study,we found that SFRP5 was overexpressed in the skin lesions of patients with vitiligo.Compared with that in normal epidermal melanocytes(PIG1),the expression of SFRP5 was increased in vitiligo melanocytes(PIG3V).To investigate the effect of SFRP5 on melanin synthesis,PIG1 cells were infected with recombinant SFRP5 adenovirus(AdSFRP5),and PIG3V cells were infected with recombinant siSFRP5 adenovirus(AdsiSFRP5).The results showed that SFRP5 overexpression inhibited melanin synthesis in PIG1 cells through downregulation of microphthalmia-associated transcription factor(MITF)and its target proteins via suppression of the Wnt/b-catenin signaling pathway.Accordingly,SFRP5 silencing increased melanin synthesis and activated the Wnt signaling pathway in PIG3V cells.Moreover,SFRP5 overexpression also downregulated the transcriptional activity of T cell factor/lymphoid enhancer factor(TCF/LEF)in PIG1 cells.Furthermore,this inhibitory effect of SFRP5 on melanin synthesis was reversed by treatment with the b-catenin agonist,SKL2001.The inhibitory action of SFRP5 in pigmentation was further confirmed in vivo using a nude mouse model.Hence,our results indicate that SFRP5 can inhibit melanogenesis in melanocytes.Additionally,our findings showed that SFRP5 plays a vital role in the development of vitiligo,and thus may serve as a potential therapeutic target for vitiligo. | Daopei Zou Yangmei Chen Lingzhao Zhang Xiaohui Yuan Yujie Zhang Adelina Inggawati Pham Thi Kieu Nguyet Tianwen Gao Jin Chen | 2021 | Genes & Diseases2021,8,5: | 2 |

| 7 | Folding Kinetics of HDV Ribozyme with C13A:G82U and A16U:U79A Mutations显示文摘Gene mutations influence the folding kinetics of hepatitis delta virus(HDV) ribozyme. In this work, we study the effect of the double mutation on the folding kinetics of HDV ribozyme. By using the master equation method combined with RNA folding free energy landscape, we predict the folding kinetics of C13A:G82U and A16U:U79A mutated HDV sequences. Their folding pathways are identified by recursively searching the states with high net flux-in(out) population starting from the native state. The results indicate that the folding kinetics of C13A:G82U mutation sequence is bi-phasic, which is similar to the wild type(wt HDV) sequence. While the folding kinetics of A16U:U79A mutation sequence is mono-phasic, it quickly folds to the native state in 30 s. | 18 | Landscape of genomic imprinting and its functions in the mouse mammary gland显示文摘Genomic imprinting is an epigenetic modification of DNA,whereby gene expression is restricted to either maternally or paternally inherited alleles.Imprinted genes(IGs)in the placenta and embryo are essential for growth regulation and nutrient supply.However,despite being an important nutrition delivery organ,studies on mammary gland genomic imprinting remain limited.In this study,we found that both the number of IGs and their expression levels decreased during development of the mouse mammary gland.IG expression was lineage-specific and related to mammary gland development and lactation.Meta-analysis of single-cell RNA sequencing data revealed that mammary gland IGs were co-expressed in a network that regulated cell sternness and differentiation,which was confirmed by our functional studies.Accordingly,our data indicated that IGs were essential for the self-renewal of mammary gland stem cells and IG decline was correlated with mammary gland maturity.Taken together,our findings revealed the importance of IGs in a poorly studied nutrition-related organ,i.e.the mammary gland,thus providing a reference for further studies on genomic imprinting. | Haibo Xu Lina Zhao Xu Feng Yujie Ma Wei Chen Li Zou Qin Yang Jihong Sun Hong Yu Baowei Jiao | 2020 | Journal of Molecular Cell Biology2020,12,11: | 0 |
