| 1 | GSDMB promotes non-canonical pyroptosis by enhancing caspase-4 activity显示文摘Gasdermin B (GSDMB) has been reported to be associated with immune diseases in humans, but the detailed molecular mechanisms remain unsolved. The N-terminus of GSDMB by itself, unlike other gasdermin family proteins, does not induce cell death. Here, we show that GSDMB is highly expressed in the leukocytes of septic shock patients, which is associated with increased release of the gasdermin D (GSDMD) N-terminus. GSDMB expression and the accumulation of the N-terminal fragment of GSDMD are induced by the activation of the non-canonical pyroptosis pathway in a human monocyte cell line. The downregulation of GSDMB alleviates the cleavage of GSDMD and cell death. Consistently, the overexpression of GSDMB promotes GSDMD cleavage, accompanied by increased LDH release. We further found that GSDMB promotes caspase-4 activity, which is required for the cleavage of GSDMD in non-canonical pyroptosis, by directly binding to the CARD domain of caspase-4. | 2 | Mitochondrial ROS promote macrophage pyroptosis by inducing GSDMD oxidation显示文摘Disrupted mitochondrial membrane potential (MMP) and reactive oxygen species (ROS) generation are often associated with macrophage pyroptosis. It remains unclear how these forms of mitochondrial dysfunction relate to inflammasome activation and gasdermin-D (Gsdmd) cleavage, two central steps of the pyroptotic process. Here, we also found MMP collapse and ROS generation induced by Nlrp3 inflammasome activation as previous studies reported. The elimination of ROS alleviated the cleavage of Gsdmd, suggesting that Gsdmd cleavage occurs downstream of ROS release. Consistent with this result, hydrogen peroxide treatment augmented the cleavage of Gsdmd by caspase-1. Indeed, four amino acid residues of Gsdmd were oxidized under oxidative stress in macrophages. The efficiency of Gsdmd cleavage by inflammatory caspase-1 was dramatically reduced when oxidative modification was blocked by mutation of these amino acid residues. | 8 | AntarcticaLC2000:The new Antarctic land cover database for the year 2000显示文摘Antarctica plays a key role in global energy balance and sea level change.It has been conventionally viewed as a whole ice body with high albedo in General Circulation Models or Regional Climate Models and the differences of land cover has usually been overlooked.Land cover in Antarctica is one of the most important drivers of changes in the Earth system.Detailed land cover information over the Antarctic region is necessary as spatial resolution improves in land process models.However,there is a lack of complete Antarctic land cover dataset derived from a consistent data source.To fill this data gap,we have produced a database named Antarctic Land Cover Database for the Year 2000(AntarcticaLC2000) using Landsat Enhanced Thematic Mapper Plus(ETM+) data acquired around 2000 and Moderate Resolution Imaging Spectrometer(MODIS) images acquired in the austral summer of 2003/2004 according to the criteria for the 1:100000-scale.Three land cover types were included in this map,separately,ice-free rocks,blue ice,and snow/firn.This classification legend was determined based on a review of the land cover systems in Antarctica(LCCSA) and an analysis of different land surface types and the potential of satellite data.Image classification was conducted through a combined usage of computer-aided and manual interpretation methods.A total of 4067 validation sample units were collected through visual interpretation in a stratified random sampling manner.An overall accuracy of 92.3%and the Kappa coefficient of 0.836 were achieved.Results show that the areas and percentages of ice-free rocks,blue ice,and snow/firn are 73268.81 km2(0.537%),225937.26 km2(1.656%),and 13345460.41 km2(97.807%),respectively.The comparisons with other different data proved a higher accuracy of our product and a more advantageous data quality.These indicate that AntarcticaLC2000,the new land cover dataset for Antarctica entirely derived from satellite data,is a reliable product for a broad spectrum of applications. | HUI FengMing KANG Jing LIU Yan CHENG Xiao GONG Peng WANG Fang LI Zhan YE YuFang GUO ZiQi | 2017 | Science China Earth Sciences2017,60,4: | 5 |
| 9 | Evaluation and scenario simulation for forest ecological security in China显示文摘Continuously growing populations and rapid economic development have led to the excessive use of forest resources,and the forest ecosystem is threatened.In response,forest ecological security(FES)has attracted attention.In this study,an integrated dynamic simulation model was constructed using the system dynamic method,and it was used to evaluate the FES in China from 1999 to 2014.A scenario analysis was then used to evaluate the changes in the FES under five forestry policy scenarios for the 2015–2050 period,including the baseline,afforestation policy,harvesting policies,management policy,investment policy,and a policy mix.The results showed that the evaluation values of the FES increased during the period from 1999 to 2002,the period from 2004 to 2010 and the year 2014,and they decreased in 2003 and during the period from 2011 to 2013.During the 2015–2050 simulation period,the FES improved continuously.In particular,China would enter a new stage when the economic systems,social systems and ecosystems were in harmony after 2040.To improve the FES and the current status of the FES,a scenario analysis showed the most suitable scenario to be Scenario 5 from 2015 to 2020 and Scenario 2 from 2021 to 2050.To relieve pressure,the most suitable scenario would be Scenario 5 from 2015 to 2040 and from 2046 to 2050,and the most suitable scenario would be Scenario 4 for 2041–2045.A policy mix(Scenario 5)would be most efficient under current conditions,while the effects of all the benefits of the forestry policies would weaken over the long term.The integrated method can be regarded as a decision support tool to help policy makers understand FES and promulgate a reasonable forestry policy. | Qin Zhang Guangyu Wang Feng Mi Xuanchang Zhang Lianzhen Xu Yufang Zhang Xiaoli Jiang | 2019 | Journal of Forestry Research2019,30,5: | 5 |

Two-line hybrids developed using the PGMS/TGMS system now account for a large proportion of rice production in China. In this paper, we summarize recent advances on molecular regulation mechanisms and genetics of PGMS/TGMS in rice. We suggest that temperature-sensitive splicing, an important posttranscriptional regulatory mechanism in modulating gene expression and eventually development and differentiation, is also an important molecular regulation mechanism of TGMS in rice. We review those factors involved in temperature-sensitive splicing like cis splice site, snRNA, trans premRNA splicing protein and SR proteins, and delineate that splicing could be regulated by a complex cell signaling pathway. | 13 | An improved Landsat Image Mosaic of Antarctica显示文摘A revised Landsat Image Mosaic of Antarctica (LIMA) is presented, using the 1073 multi-band scenes of the original Landsat-7 ETM+ LIMA image collection available at the United States Geological Survey (USGS: http://lima.usgs.gov/). Three improvements have been applied during the data processing: (1) DN saturation is adjusted by adopting a linear regression, which has a lower root mean square error than the ratio regression used by LIMA; (2) solar elevation angle is calculated using pixel-level latitude/longitude and the acquisition time and date of the central pixel of the scene, improving slightly upon the bilinear interpolation of the solar elevation angles of scene corners applied in LIMA; and (3) two additional image bands, Band 5 and Band 7, are sharpened using the panchromatic band (Band 8) and a Gram-Schmidt Spectral Sharpening algorithm to more easily distinguish snow, cloud and exposed rocks. The final planetary reflectance product is stored in 16-bit bands to preserve the full radiometric content of the scenes. | 14 | Coupled relationships among anammox,denitrification, and dissimilatory nitrate reduction to ammonium along salinity gradients in a Chinese estuarine wetland显示文摘Salinization in estuarine wetlands significantly alters the balance between their nitrogen(N)removal and retention abilities but these processes have not yet been characterized effectively.In the present study,the potential rates of sediment denitrification,anaerobic ammonium oxidation(anammox),and dissimilatory nitrate reduction to ammonium(DNRA)were mapped using N isotope tracing methods along salinity gradients across the Yellow River Delta wetland(YRDW)in China.The contribution of anammox to total dissimilatory N transformations in YRDW was merely 6.8%,whereas denitrification and DNRA contributed 52.3%and 40.9%,respectively.The potential rate of denitrification(5.82μmol/kg/h)decreased significantly along salinity gradients and markedly exceeded DNRA potential rate(2.7μmol/kg/h)in fresh wetlands,but was lower than that of DNRA in oligohaline wetlands(3.06 and 3.18μmol/kg/h,respectively).Moreover,a significantly positive relationship between salinity and DNRA/denitrification was obeserved,indicating that increased salinity may favor DNRA over denitrification.Furthermore,total sulfur(TS)content and ratio of total organic carbon to total nitrogen(C/N)increased with the salinity gradient and showed evident positive relationships with the DNRA/denitrification ratio.In this study,we proved that increased salinization resulted in the dominance of DNRA over denitrification,possible through the addition of S and alteration of the C/N in estuarine wetlands,leading to increased N retention in estuarine wetlands during salinization,which would enhance the eutrophication potential within wetlands and in downstream ecosystems. | Zijun Zhou Lei Ge Yufang Huang Yuqian Liu Siyang Wang | 2021 | Journal of Environmental Sciences2021,33,8: | 4 |
| 15 | The metastable constituent effects on size-dependent deformation behavior of nanolaminated micropillars: Cu/FeCoCrNi vs Cu/CuZr显示文摘Metastable high entropy alloys(HEAs) and amorphous metallic glasses(MGs), with the chemical disordered character, are intensively studied due to their excellent performance. Here, we introduce Cu to separately constrain these two metastable materials and comparatively investigate their deformation behaviors and mechanical properties of Cu/HEA Fe Co Cr Ni and Cu/MG Cu Zr nanolaminated micropillars in terms of intrinsic layer thickness h and extrinsic pillar diameter D. The metastable HEA layers, as the hard phase in Cu/HEA micropillars, are stable and dominate the deformation, while transformation(crystallization) occurs in MG which plays a minor role in deformation of Cu/MG micropillars. The h-controlled deformation mode transits from the D-independent homogenous-like deformation at large h to the Ddependent shear banding at small h in both Cu/HEA and Cu/MG micropillars. Although both Cu/HEA and Cu/MG micropillars exhibit a maximum strain hardening capability controlled by h, the former manifests much lower hardening capability compared with the latter. The intrinsic size h and extrinsic size D have a strong coupling effect on the strength of Cu/HEA and Cu/MG micropillars. The strength of strength of Cu/HEA micropillars exhibits the D-dependent transition from 'smaller is stronger' to 'smaller is weaker'with increasing h. | 16 | N-acetyltransferase 10 promotes colon cancer progression by inhibiting ferroptosis through N4-acetylation and stabilization of ferroptosis suppressor protein 1 (FSP1) mRNA显示文摘Background:N-acetyltransferase 10(NAT10)is the only enzyme known tomediate the N4-acetylcytidine(ac4C)modification of mRNA and is crucial formRNA stability and translation efficiency.However,its role in cancer development and prognosis has not yet been explored.This study aimed to examine the possible role of NAT10 in colon cancer.Methods:The expression levels ofNAT10were evaluated by immunohistochemical analyses with a colon cancer tissue microarray,and its prognostic value in patients was further analyzed.Quantitative real-time polymerase chain reaction(qRT-PCR)and Western blotting were performed to analyze NAT10 expression in harvested colon cancer tissues and cell lines.Stable NAT10-knockdown and NAT10-overexpressing colon cancer cell lines were constructed using lentivirus.The biological functions of NAT10 in colon cancer cell lines were analyzed in vitro by Cell Counting Kit-8(CCK-8),wound healing,Transwell,cell cycle,and ferroptosis assays.Xenograft models were used to analyze the effect of NAT10 on the tumorigenesis and metastasis of colon cancer cells in vivo.Dot blotting,acetylated RNA immunoprecipitation-qPCR,and RNA stability analyses were performed to explore the mechanism by which NAT10 functions in colon cancer progression.Results:NAT10 was upregulated in colon cancer tissues and various colon cancer cell lines.This increased NAT10 expression was associated with shorter patient survival.Knockdown of NAT10 in two colon cancer cell lines(HT-29 and LoVo)impaired the proliferation,migration,invasion,tumor formation and metastasis of these cells,whereas overexpression of NAT10 promoted these abilities.Further analysis revealed that NAT10 exerted a strong effect on the mRNA stability and expression of ferroptosis suppressor protein 1(FSP1)in HT-29 and LoVo cells.In these cells,FSP1 mRNA was found to be modified by ac4C acetylation,and this epigenetic modification was associated with the inhibition of ferroptosis.Conclusions:Our study revealed that NAT10 plays a critical role in colon cancer development by affecting FSP1 mRNA stability and ferroptosis,suggesting that NAT10 could be a novel prognostic and therapeutic target in colon cancer. | Xiao Zheng Qi Wang You Zhou Dachuan Zhang Yiting Geng Wenwei Hu Changping Wu Yufang Shi Jingting Jiang | 2022 | Cancer Communications2022,42,12: | 3 |

| 20 | Late Quaternary Activity of the Huashan Piedmont Fault and Associated Hazards in the Southeastern Weihe Graben,Central China显示文摘The Weihe Graben is not only an important Cenozoic fault basin in China but also a significant active seismic zone. The Huashan piedmont fault is an important active fault on the southeast side of the Weihe Graben and has been highly active since the Cenozoic. The well–known Great Huaxian County Earthquake of 1556 occurred on the Huashan piedmont fault. This earthquake, which claimed the lives of approximately 830000 people, is one of the few large earthquakes known to have occurred on a high–angle normal fault. The Huashan piedmont fault is a typical active normal fault that can be used to study tectonic activity and the associated hazards. In this study, the types and characteristics of late Quaternary deformation along this fault are discussed from geological investigations, historical research and comprehensive analysis. On the basis of its characteristics and activity, the fault can be divided into three sections, namely eastern, central and western. The eastern and western sections display normal slip. Intense deformation has occurred along the two sections during the Quaternary; however, no deformation has occurred during the Holocene. The central section has experienced significant high–angle normal fault activity during the Quaternary, including the Holocene. Holocene alluvial fans and loess cut by the fault have been identified at the mouths of many stream valleys of the Huashan Mountains along the central section of the Huashan piedmont fault zone. Of the three sections of the Huashan piedmont fault, the central section is the most active and was very active during the late Quaternary. The rate of normal dip–slip was 1.67–2.71±0.11 mm/a in the Holocene and 0.61±0.15 mm/a during the Mid–Late Pleistocene. As is typical of normal faults, the late Quaternary activity of the Huashan piedmont fault has produced a set of disasters, which include frequent earthquakes, collapses, landslides, mudslides and ground fissures.
