| 1 | Strata movement controlling effect of waste and fly ash backfillings in fully mechanized coal mining with backfilling face显示文摘A fully mechanized coal mining with backfilling (FMCMB) provides advantages of safety and efficiency for coal mining under buildings, railways, and water bodies. According to the field geological conditions, we analyzed the controlling effect of strata movement by the waste and fly ash backfilling in FMCMB face. Based on the key strata theory, we established the equivalent mining thickness model, and analyzed the action of the bulk factor of backfilling body to the equivalent mining thickness. In addition, we numerically simulated the controlling function of the strata movement by backfilling bodies with differ- ent strength. And the numerical simulation result show that the deformation of stratum and the subsi- dence of surface can be controlled by FMCMB. | 2 | GSDMB promotes non-canonical pyroptosis by enhancing caspase-4 activity显示文摘Gasdermin B (GSDMB) has been reported to be associated with immune diseases in humans, but the detailed molecular mechanisms remain unsolved. The N-terminus of GSDMB by itself, unlike other gasdermin family proteins, does not induce cell death. Here, we show that GSDMB is highly expressed in the leukocytes of septic shock patients, which is associated with increased release of the gasdermin D (GSDMD) N-terminus. GSDMB expression and the accumulation of the N-terminal fragment of GSDMD are induced by the activation of the non-canonical pyroptosis pathway in a human monocyte cell line. The downregulation of GSDMB alleviates the cleavage of GSDMD and cell death. Consistently, the overexpression of GSDMB promotes GSDMD cleavage, accompanied by increased LDH release. We further found that GSDMB promotes caspase-4 activity, which is required for the cleavage of GSDMD in non-canonical pyroptosis, by directly binding to the CARD domain of caspase-4. | 3 | Mitochondrial ROS promote macrophage pyroptosis by inducing GSDMD oxidation显示文摘Disrupted mitochondrial membrane potential (MMP) and reactive oxygen species (ROS) generation are often associated with macrophage pyroptosis. It remains unclear how these forms of mitochondrial dysfunction relate to inflammasome activation and gasdermin-D (Gsdmd) cleavage, two central steps of the pyroptotic process. Here, we also found MMP collapse and ROS generation induced by Nlrp3 inflammasome activation as previous studies reported. The elimination of ROS alleviated the cleavage of Gsdmd, suggesting that Gsdmd cleavage occurs downstream of ROS release. Consistent with this result, hydrogen peroxide treatment augmented the cleavage of Gsdmd by caspase-1. Indeed, four amino acid residues of Gsdmd were oxidized under oxidative stress in macrophages. The efficiency of Gsdmd cleavage by inflammatory caspase-1 was dramatically reduced when oxidative modification was blocked by mutation of these amino acid residues. | 6 | Method of neural network modulation recognition based on clustering and Polak-Ribiere algorithm显示文摘To improve the recognition rate of signal modulation recognition methods based on the clustering algorithm under the low SNR, a modulation recognition method is proposed. The characteristic parameter of the signal is extracted by using a clustering algorithm, the neural network is trained by using the algorithm of variable gradient correction(Polak-Ribiere) so as to enhance the rate of convergence, improve the performance of recognition under the low SNR and realize modulation recognition of the signal based on the modulation system of the constellation diagram.Simulation results show that the recognition rate based on this algorithm is enhanced over 30% compared with the methods that adopt clustering algorithm or neural network based on the back propagation algorithm alone under the low SNR. | 7 | LyP-1 peptide-functionalized gold nanoprisms for SERRS imaging and tumor growth suppressing by PTT induced-hyperthermia显示文摘Gold-based nanomaterials with plasmonic properties exhibit various potentials for biomedical applications. In this work, gold nanoprisms (GNPs) was synthesized and then modified with LyP-1, a tumor-homing peptide, to improve the affinity of the GNPs to tumor cells, thus, to improve the efficacy of tumor-targeted photothermal therapy. The introduction of NIR dye IR780 not only enabled the GNPs-based nanosystem with the surface-enhanced resonant Raman scattering (SERRS) property, but also enhanced the plasmonic photothermal property which delivering therapeutic heating by 660 nm laser irradiation. The obtained GNPs/IR780-LyP-1 presented significantly increased of photothermal conversion in vitro and in vivo, which resulted in enhanced tumor-targeting photothermal therapeutic efficacy after laser irradiation. | 10 | High expression of human serum albumin in milk of trans-genie mice directed by the goat β-casein gene promoter region显示文摘We have constructed a mammary gland expression vector that contained the goat β-casein gene pro-moter, 5’upstream regulatory region, exons 1, 2, intron 1 as well as the human serum albumin (hALB) mini-gene (including the full-long sequences of hALB cDNA and its intron 1). Injection of the vector into mouse tail veins showed that the recombinant construct was expressed only in mammary glands. The vector was microinjected into the mouse fertilized eggs, followed by transferring the eggs into the foster mice. 33 F0 mice were obtained. Of the 33, 8 mice (5 , 3 ) were transgenic with hALB gene integration identified by PCR as well as Southern blot hybridization. The integration rate was 24.2% (8/33). Western blot analysis showed that 3 female transgenic mice had hALB expression in their milk. In this paper, we first analyse BSR system for different amplitudes of 4-ary PAM signals. The steadystate of the bistable system will be statistically distinct, and the feasibility of the proposed detection scheme is confirmed. On this basis, we present a detailed study on steady-state transitions of the BSR system, and an explicit expression of the bistable system parameters is derived. By setting the bistable system parameters, bistable system, 4-ary PAM signal, and noise reach the resonance state, and the BSR-based detection scheme is implemented. Moreover, we derive an analytical expression to calculate the symbol error rate(SER) of 4-ary PAM signals with the BSR-based detection under additive white Gaussian noise(AWGN).
