1MdGSTF6,activated by MdMYB1,plays an essential role in anthocyanin accumulation in apple显示文摘Anthocyanins are biosynthesized on the cytosolic surface of the endoplasmic reticulum and then transported into the vacuole for storage.Glutathione S-transferases(GSTs)are considered to be responsible for the transport of anthocyanins into the vacuole.However,the regulatory mechanisms of GSTs in plants are still unclear.Here,we performed a genome-wide analysis and identified 69 GST genes in apple.The expression of MdGSTF6 was positively correlated with the anthocyanin content(r=0.949)during‘Yanfu 8’fruit development.The overexpression of MdGSTF6 in the Arabidopsis thaliana tt19 mutant resulted in seedlings of 35S::MdGSTF6-GFP/tt19 that could accumulate anthocyanin and rescue its phenotype,suggesting that MdGSTF6 was an anthocyanin transporter.The silencing of MdGSTF6 affected anthocyanin accumulation in apple fruit.Moreover,the knockdown of MdGSTF6 by RNA interference in cultured‘Gala’seedlings inhibited anthocyanin accumulation.The interaction experiments showed that MdMYB1 could bind directly to the MdGSTF6 promoter to transcriptionally activate its expression.Collectively,our results demonstrate that MdGSTF6 encodes an important GST transporter of anthocyanins in apple fruit and provide evidence for the associated regulatory mechanisms.Therefore,MdMYB1 can not only regulate anthocyanin synthesis,but also control the transport of anthocyanin in apples.This information may be useful for further clarifying the regulation of anthocyanin transport in apple.Shenghui Jiang Min Chen Naibo He Xiaoliu Chen Nan Wang Qingguo Sun Tianliang Zhang Haifeng Xu Hongcheng Fang Yicheng Wang Zongying Zhang Shujing Wu Xuesen Chen 2019Horticulture Research2019,6,1:16
2Malus sieversii: the origin, flavonoid synthesis mechanism, and breeding of red-skinned and red-fleshed apples显示文摘Flavonoids play essential roles in human health.Apple(Malus domestica Borkh.),one of the most widely produced and economically important fruit crops in temperate regions,is a significant source of flavonoids in the human diet and is among the top nutritionally rated and most widely consumed fruits worldwide.Epidemiological studies have shown that the consumption of apples,which are rich in a variety of free and easily absorbable flavonoids,is associated with a decreased risk of various diseases.However,apple production is challenged by serious inbreeding problems.The narrowing of the hereditary base has resulted in apples with poor nutritional quality and low flavonoid contents.Recently,there have been advances in our understanding of the roles that Malus sieversii(Ledeb.)M.Roem has played in the process of apple domestication and breeding.In this study,we review the origin of cultivated apples and red-fleshed apples,and discuss the genetic diversity and construction of the core collections of M.sieversii.We also discuss current research progress and breeding programs on red-skinned and red-fleshed apples and summarize the exploitation and utilization of M.sieversii in the breeding of high-flavonoid,and red-fleshed apples.This study highlights a valuable pattern of horticultural crop breeding using wild germplasm resources.The future challenges and directions of research on the molecular mechanisms of flavonoid accumulation and high-flavonoid apple breeding are discussed.Nan Wang Shenghui Jiang Zongying Zhang Hongcheng Fang Haifeng Xu Yicheng Wang Xuesen Chen 2018Horticulture Research2018,5,1:15
3The acidic tumor microenvironment: a target for smart cancer nano-theranostics显示文摘The acidic tumor microenvironment(TME), which mainly results from the high glycolytic rate of tumor cells, has been characterized as a hallmark of solid tumors and found to be a pivotal factor participating in tumor progression. Recently, due to the increasing understanding of the acidic TME, it has been shown that the acidic TME could be utilized as a multifaceted target during the design of various pH-responsive nanoscale theranostic platforms for the precise diagnosis and effective treatment of cancers. In this article,we will give a focused overview on the latest progress in utilizing this characteristic acidic TME as the target of nano-theranostics to enable cancer-specific imaging and therapy. 5Rat model of cholelithiasis with human gallstones implanted in cholestasis-induced virtual gallbladder显示文摘AIM:To facilitate translational research on cholelithiasis,we have developed a rat model of human gallstones by exploiting the unique biliopancreatic features of this species.METHODS:Under anesthesia,16 adult rats of equal genders underwent two times of abdominal surgery.First,their common bile duct(CBD)was ligated to cause cholestasis by total biliary obstruction(TBO).On day 0,1,3,7,14,21 and 28 after TBO,magnetic resonance imaging(MRI)was conducted to monitor the dilatation of the CBD,and blood was sampled to analyze total serum bilirubin(TSB).Secondly,on day 30,the abdomen was re-opened and gallstone(s)collected from human patients were implanted in the dilated CBD asa virtual gallbladder(VGB),which was closed by suture ligation.This rat cholelithiasis model was examined by MRI,clinical observation,microcholangiography and histology.RESULTS:All rats survived two laparotomies.After ligation,the CBD was dilated to a stable size of 4 to30 mm in diameter on day 21-28,which became a VGB.The rats initially showed signs of jaundice that diminished over time,which paralleled with the evolving TSB levels from 0.6±0.3 mg/d L before ligation,through a peak of 10.9±1.9 mg/dL on day 14,until a nearly normalized value after day 28.The dilated CBD with thickened wall allowed an incision for implantation of human gallstones of 1-10 mm in diameter.The rat cholelithiasis was proven by in vivo MRI and postmortem microcholangiography and histomorphology.CONCLUSION:A rat model cholelithiasis with human gallstones has been established,which proves feasible,safe,reliable,nontoxic and cost-effective.Given the gallstones of human origin,applications of this model may be of help in translational research such as optical detection and lysis of gallstones by systemic drug administration.Marlein Miranda Cona Yewei Liu Ting Yin Yuanbo Feng Feng Chen Stefaan Mulier Yue Li Jian Zhang Raymond Oyen Yicheng Ni 2016World Journal of Methodology2016,6,2:7
6MdMYB6 regulates anthocyanin formation in apple both through direct inhibition of the biosynthesis pathway and through substrate removal显示文摘Anthocyanin biosynthesis and sugar metabolism are important processes during plant growth,but the molecular interactions underlying these pathways are still unclear.In this work,we analyzed the anthocyanin and soluble sugar contents,as well as the transcript levels of transcription factors that are known to be related to the biosynthesis of anthocyanin in‘Hongcui 1’apple flesh during fruit development.Overexpression of MdMYB6 in red-fleshed calli was found to reduce anthocyanin content and result in downregulated expression of the MdANS and MdGSTF12 proteins.Yeast one-hybrid and electrophoretic mobility shift analyses showed that MdMYB6 could directly bind to the promoters of MdANS and MdGSTF12,indicating that MdMYB6 could inhibit anthocyanin biosynthesis by regulating MdANS and MdGSTF12.Overexpression of MdTMT1 in the Arabidopsis tmt1 mutant restored the glucose and fructose contents to the wild-type levels,while overexpression of MdTMT1 in red-fleshed calli increased the contents of glucose and fructose but reduced the contents of UDP-glucose,UDP-galactose,and anthocyanin.Using a GUS reporter system,yeast one-hybrid,chromatin immunoprecipitation-PCR and electrophoretic mobility shift analyses,we found that MdMYB6 could bind to the promoter of MdTMT1,resulting in increased promoter activity.Overexpression of MdMYB6 in calli overexpressing MdTMT1 increased the expression of MdTMT1,which led to reduced contents of UDP-glucose and UDP-galactose and decreased anthocyanin content compared to those of the calli that overexpressed MdTMT1.This finding suggested that MdMYB6 could also inhibit anthocyanin biosynthesis by regulating MdTMT1 to decrease the contents of UDP-glucose and UDP-galactose.Taken together,these results showed that MdMYB6 and MdTMT1 play key roles in both anthocyanin biosynthesis and sugar transport.Haifeng Xu Qi Zou Guanxian Yang Shenghui Jiang Hongcheng Fang Yicheng Wang Jing Zhang Zongying Zhang Nan Wang Xuesen Chen 2020Horticulture Research2020,7,1:6
7Effects of stress dependent electrochemical reaction on voltage hysteresis of lithium ion batteries显示文摘Intercalation of lithium ions into the electrodes of lithium ion batteries is affected by the stress of active materials, leading to energy dissipation and stress dependent voltage hysteresis. A reaction-diffusion-stress coupling model is established to investigate the stress effects under galvanostatic and potentiostatic operations. It is found from simulations that the stress hysteresis contributes to the voltage hysteresis and leads to the energy dissipation. In addition, the stress induced voltage hysteresis is small in low rate galvanostatic operations but extraordinarily significant in high rate cases. In potentiostatic operations, the stresses and stress induced overpotentials increase to a peak value very soon after the operation commences and decays all the left time. Therefore,a combined charge-discharge operation is suggested, i.e., first the galvanostatic one and then the potentiostatic one. 8Autophagy receptor CCDC50 tunes the STING-mediated interferon response in viral infections and autoimmune diseases显示文摘DNA sensing and timely activation of interferon(IFN)-mediated innate immunity are crucial for the defense against DNA virus infections and the clearance of abnormal cells.However,overactivation of immune responses may lead to tissue damage and autoimmune diseases;therefore,these processes must be intricately regulated.STING is the key adaptor protein,which is activated by cyclic GMP-AMP,the second messenger derived from cGAS-mediated DNA sensing.Here,we report that CCDC50,a newly identified autophagy receptor,tunes STING-directed type I IFN signaling activity by delivering K63-polyubiquitinated STING to autolysosomes for degradation.Knockout of CCDC50 significantly increases herpes simplex virus 1(HSV-1)-or DNA ligand-induced production of type I IFN and proinflammatory cytokines.Ccdc50-deficient mice show increased production of IFN,decreased viral replication,reduced cell infiltration,and improved survival rates compared with their wild-type littermates when challenged with HSV-1.Remarkably,the expression of CCDC50 is downregulated in systemic lupus erythematosus(SLE),a chronic autoimmune disease.CCDC50 levels are negatively correlated with IFN signaling pathway activation and disease severity in human SLE patients.CCDC50 deficiency potentiates the cGAS-STING-mediated immune response triggered by SLE serum.Thus,our findings reveal the critical role of CCDC50 in the immune regulation of viral infections and autoimmune diseases and provide insights into the therapeutic implications of CCDC50 manipulation.Panpan Hou Yuxin Lin Zibo Li Ruiqing Lu Yicheng Wang Tian Tian Penghui Jia Xi Zhang Liu Cao Zhongwei Zhou Chunmei Li Jieruo Gu Deyin Guo 2021Cellular & Molecular Immunology2021,18,10:3
9Updated developments on molecular imaging and therapeutic strategies directed against necrosis显示文摘Cell death plays important roles in living organisms and is a hallmark of numerous disorders such as cardiovascular diseases, sepsis and acute pancreatitis. Moreover, cell death also plays a pivotal role in the treatment of certain diseases, for example, cancer. Noninvasive visualization of cell death contributes to gained insight into diseases, development of individualized treatment plans, evaluation of treatment responses, and prediction of patient prognosis. On the other hand, cell death can also be targeted for the treatment of diseases. Although there are many ways for a cell to die, only apoptosis and necrosis have been extensively studied in terms of cell death related theranostics. This review mainly focuses on molecular imaging and therapeutic strategies directed against necrosis. Necrosis shares common morphological characteristics including the rupture of cell membrane integrity and release of cellular contents, which provide potential biomarkers for visualization of necrosis and necrosis targeted therapy. In the present review, we summarize the updated joint efforts to develop molecular imaging probes and therapeutic strategies targeting the biomarkers exposed by necrotic cells. 10A method of elevated temperatures coupled with magnetic stirring to predict real time release from long acting progesterone PLGA microspheres显示文摘The object of the study was to develop a quick and reproducible accelerated in vitro release method to predict and deduce the function of the real time(37 °C) release for long acting PLGA microspheres. The method could be described in several steps. First, the release of the microspheres were studied using the sample and separate method at 37 °C with normal orbital shaking and elevated temperatures with magnetic stirring to further accelerate the release. Second, the most similar profile at elevated temperatures with the real time release was chosen with the help of the n value in the fitted Korsmeyer-Peppas Function. Third,the Weibull function and conversion ratio were used to deduce the function of real time release according to the chosen profile at elevated temperatures. The key point in this study was to provide a quick and precise method to predict the real time release for long acting progesterone PLGA microspheres. So the elevated temperatures coupled with magnetic stirring were used to accelerate the release further, and when there have many similar release profiles with the real time release at elevated temperatures, releasing time at elevated temperatures and the R2 of the final deduced function will be used to help choosing the most similar release profile with the real time release. 11Growth inhibition and oxidative damage of Microcystis aeruginosa induced by crude extract of Sagittaria trifolia tubers显示文摘Aquatic macrophytes are considered to be promising in controlling harmful cyanobacterial blooms. In this research, an aqueous extract of Sagittaria trifolia tubers was prepared to study its inhibitory effect on Microcystis aeruginosa in the laboratory. Several physiological indices of M. aeruginosa, in response to the environmental stress, were analyzed. Results showed that S. trifolia tuber aqueous extract significantly inhibited the growth of M. aeruginosa in a concentration-dependent way. The highest inhibition rate reached 90% after 6 day treatment. The Chlorophyll-a concentration of M. aeruginosa cells decreased from 343.1 to314.2 μg/L in the treatment group. The activities of superoxide dismutase and peroxidase and the content of reduced glutathione in M. aeruginosa cells initially increased as a response to the oxidative stress posed by S. trifolia tuber aqueous extract, but then decreased as time prolonged. The lipid peroxidation damage of the cyanobacterial cell membranes was reflected by the malondialdehyde level, which was notably higher in the treatment group compared with the controls. 12Neutrophil-derived heparin binding protein triggers vascular leakage and synergizes with myeloperoxidase at the early stage of severe burns(With video)显示文摘Background:Burn shock caused by vascular leakage is one of the main causes of high mortality in severe burn injury.However,the pathophysiological mechanism of vascular leakage is still unclear.The purpose of this study was to explore the molecular mechanism of vascular leakage in the early stage of severe burn and provide a new target for the treatment of severe burns.Methods:Neutrophils were isolated from human peripheral blood by magnetic beads sorting.ELISA was used to detect neutrophil-derived granule proteins and glycocalyx injury products in plasma.The vascular leakage and neutrophil movement were assessed by in vivo laser confocal imaging in mice,and high-quality video were provided.Adhesion-related molecules were investigated by qRT-PCR.The damage to glycocalyx of mice vascular endothelial cellswas observed by transmission electron microscope and scanning electron microscope.Proteomic analysis,flow cytometry and immunofluorescence were used to further study the relationship between human peripheral blood neutrophil-derived hypochlorite(HOCl)and CD44 of human vascular endothelial cells.Results:In this study,we found that rapidly increasing activated neutrophils secrete heparin binding protein(HBP)andmyeloperoxidase(MPO)after severe burn injury.Increased HBP triggers vascular leakage with synergy of MPO,results in systemic edema and burn shock.Furthermore,we found that the MPO catalytic product HOCl but not MPO triggers CD44 extracellular domain shedding from vascular endothelial cells to damage the glycocalyx.Damage to the glycocalyx results in firm adhesion of neutrophils and increases vascular leakage.However,MPO inhibitors partially protect the glycocalyx of vascular endothelial cells.The combination of HBP and MPO inhibitors markedly reduces vascular leakage and systemic edema in the early stage of severe burns.Conclusions:Taken together,these data reveal that neutrophil-derived HBP and MPO play an important synergies role in triggering vascular leakage at the early stage of severe burns.Targeted intervention in these two biomolecules may introduce new strategies for helping to reduce large amount of fluid loss and subsequent burn shock.Lu Liu Yiming Shao Yixuan Zhang Yunxi Yang Jiamin Huang Linbin Li Ran Sun Yuying Zhou Yicheng Su Bingwei Sun 2021Burns & Trauma2021,9,1:3

14Two-way coupled analysis of lithium diffusion and diffusion induced finite elastoplastic bending of bilayer electrodes in lithium-ion batteries显示文摘A fully coupling model for the diffusion induced finite elastoplastic bending of bilayer electrodes in lithium-ion batteries is proposed. The effect of the mechanical stress on the lithium diffusion is accounted for by the mechanical part of the chemical potential derived from the Gibbs free energy along with the logarithmic stress and strain. Eight dimensionless parameters, governing the stress-assisted diffusion and the diffusion induced elastoplastic bending, are identified. It is found that the finite plasticity starting from the interface of the bilayer increases the chemical potential gradient and thereby facilitates the lithium diffusion. The full plastic flow makes the abnormal lithium concentration distribution possible, i.e., the concentration at the lithium inlet can be lower than the concentration at the interface(downstream). 15Two-step Optimal Allocation of Stationary and Mobile Energy Storage Systems in Resilient Distribution Networks显示文摘Energy storage systems (ESSs) are acknowledged to be a promising option to cope with issues in high penetration of renewable energy and guarantee a highly reliable power supply. In this paper, a two-step optimal allocation model is proposed to obtain the optimal allocation (location and size) of stationary ESSs (SESSs) and mobile ESSs (MESSs) in the resilient distribution networks (DNs). In the first step, a mixed-integer linear programming (MILP) problem is formulated to obtain the preselected location of ESSs with consideration of different scenarios under normal operation conditions. In the second step, a two-stage robust optimization model is established to get the optimal allocation results of ESSs under failure operation conditions which are solved by column-and-constraint generation (C&CG) algorithm. A hybrid ESS allocation strategy based on the subjective and objective weight analysis is proposed to give the final allocation scheme of SESSs and MESSs. 16Genome-wide analysis of expression quantitative trait loci(eQTLs)reveals the regulatory architecture of gene expression variation in the storage roots of sweet potato显示文摘Dissecting the genetic regulation of gene expression is critical for understanding phenotypic variation and species evolution.However,our understanding of the transcriptional variability in sweet potato remains limited.Here,we analyzed two publicly available datasets to explore the landscape of transcriptomic variations and its genetic basis in the storage roots of sweet potato.The comprehensive analysis identified a total of 724,438 high-confidence single nucleotide polymorphisms(SNPs)and 26,026 expressed genes.Expression quantitative trait locus(eQTL)analysis revealed 4408 eQTLs regulating the expression of 3646 genes,including 2261 local eQTLs and 2147 distant eQTLs.Two distant eQTL hotspots were found with target genes significantly enriched in specific functional classifications.By combining the information from regulatory network analyses,eQTLs and association mapping,we found that IbMYB1-2 acts as a master regulator and is the major gene responsible for the activation of anthocyanin biosynthesis in the storage roots of sweet potato.Our study provides the first insight into the genetic architecture of genome-wide expression variation in sweet potato and can be used to investigate the potential effects of genetic variants on key agronomic traits in sweet potato.Lei Zhang Yicheng Yu Tianye Shi Meng Kou Jian Sun Tao Xu Qiang Li Shaoyuan Wu Qinghe Cao Wenqian Hou Zongyun Li 2020Horticulture Research2020,7,1:2

18Overexpression of phosphatidylserine synthase IbPSS1 affords cellular Na+homeostasis and salt tolerance by activating plasma membrane Na^(+)/H^(+)antiport activity in sweet potato roots显示文摘Phosphatidylserine synthase(PSS)-mediated phosphatidylserine(PS)synthesis is crucial for plant development.However,little is known about the contribution of PSS to Na^(+)homeostasis regulation and salt tolerance in plants.Here,we cloned the IbPSS1 gene,which encodes an ortholog of Arabidopsis AtPSS1,from sweet potato(Ipomoea batatas(L.)Lam.).The transient expression of IbPSS1 in Nicotiana benthamiana leaves increased PS abundance.We then established an efficient Agrobacterium rhizogenes-mediated in vivo root transgenic system for sweet potato.Overexpression of IbPSS1 through this system markedly decreased cellular Na^(+)accumulation in salinized transgenic roots(TRs)compared with adventitious roots.The overexpression of IbPSS1 enhanced salt-induced Na^(+)/H^(+)antiport activity and increased plasma membrane(PM)Ca^(2+)-permeable channel sensitivity to NaCl and H2O2 in the TRs.We confirmed the important role of IbPSS1 in improving salt tolerance in transgenic sweet potato lines obtained from an Agrobacterium tumefaciens-mediated transformation system.Similarly,compared with the wild-type(WT)plants,the transgenic lines presented decreased Na^(+)accumulation,enhanced Na^(+)exclusion,and increased PM Ca^(2+)-permeable channel sensitivity to NaCl and H2O2 in the roots.Exogenous application of lysophosphatidylserine triggered similar shifts in Na^(+)accumulation and Na^(+)and Ca^(2+)fluxes in the salinized roots of WT.Overall,this study provides an efficient and reliable transgenic method for functional genomic studies of sweet potato.Our results revealed that IbPSS1 contributes to the salt tolerance of sweet potato by enabling Na^(+)homeostasis and Na^(+)exclusion in the roots,and the latter process is possibly controlled by PS reinforcing Ca^(2+)signaling in the roots.Yicheng Yu Ying Xuan Xiaofeng Bian Lei Zhang Zhiyuan Pan Meng Kou Qinghe Cao Zhonghou Tang Qiang Li Daifu Ma Zongyun Li Jian Sun 2020Horticulture Research2020,7,1:2

20Imaging algorithm of multi-ship motion target based on compressed sensing显示文摘An imaging algorithm based on compressed sensing(CS) for the multi-ship motion target is presented. In order to reduce the quantity of data transmission in searching the ships on a large sea area, both range and azimuth of the moving ship targets are converted into sparse representation under certain signal basis. The signal reconstruction algorithm based on CS at a distant calculation station, and the Keystone and fractional Fourier transform(FRFT) algorithm are used to compensate range migration and obtain Doppler frequency. When the sea ships satisfy the sparsity, the algorithm can obtain higher resolution in both range and azimuth than the conventional imaging algorithm.
