| 5 | FRUCTOKINASE-LIKE PROTEIN 1 interacts with TRXz to regulate chloroplast development in rice显示文摘Chloroplast genes are transcribed by the plastidencoded RNA polymerase(PEP) or nucleus-encoded RNA polymerase. FRUCTOKINASE-LIKE PROTEINS(FLNs) are phosphofructokinase-B(Pfk B)-type carbohydrate kinases that act as part of the PEP complex; however, the molecular mechanisms underlying FLN activity in rice remain elusive.Previously, we identified and characterized a heat-stress sensitive albino(hsa_1) mutant in rice. Map-based cloning revealed that HSA_1 encodes a putative OsFLN_2. Here, we further demonstrated that knockdown or knockout of the OsFLN_1, a close homolog of HSA_1/OsFLN_2, considerably inhibits chloroplast biogenesis and the fln_1 knockout mutants, created by clustered regularly interspaced short palindromic repeats(CRISPR) and CRISPR-associate protein_9, exhibit severe albino phenotype and seedling lethality. Moreover, OsFLN_1 localizes to the chloroplast.Yeast two-hybrid, pull-down and bimolecular fluorescencecomplementation experiments revealed that OsFLN_1 and HSA_1/OsFLN_2 interact with THIOREDOXINZ(OsTRXz) to regulate chloroplast development. In agreement with this,knockout of OsTRXz resulted in a similar albino and seedling lethality phenotype to that of the fln_1 mutants. Quantitative reverse transcription polymerase chain reaction and immunoblot analysis revealed that the transcription and translation of PEP-dependent genes were strongly inhibited in fln_1 and trxz mutants, indicating that loss of OsFLN_1, HSA_1/OsFLN_2, or OsTRXz function perturbs the stability of the transcriptionally active chromosome complex and PEP activity. | 8 | Insights into the enhanced structure stability and electrochemical performance of Ti^(4+)/F^(-) co-doped P2-Na_(0.67)Ni_(0.33)Mn_(0.67)O_(2) cathodes for sodium ion batteries at high voltage显示文摘P2-Na_(0.67)N_(i0.33)Mn_(0.67)O_(2)is considered as a promising cathode material for sodium-ion battery (SIBs)because of its high capacity and discharge potential.However,its practical use is limited by Na^(+)/vacancy ordering and P2-O2 phase transition.Herein,a Ti^(4+)/F^(-) co-doping strategy is developed to address these issues.The optimal P2-Na_(0.67)Ni_(0.33)Mn_(0.37)Ti_(0.3)O_(1.9)F_(0.1) exhibits much enhanced sodium storage performance in the high voltage range of 2.0–4.4 V,including a cycling stability of 77.2%over 300cycles at a rate of 2 C and a high-rate capability of 87.7 m Ah g^(-1) at 6 C.Moreover,the P2-Na_(0.67)Ni_(0.33)Mn_(0.37)Ti_(0.3)O_(1.9)F_(0.1) delivers reversible capacities of 82.7 and 128.1 m Ah g^(-1) at-10 and 50℃ at a rate of 2 C,respectively.The capacity retentions over 200 cycles at-10℃ is 94.2%,implying more opportunity for practical application.In-situ X-ray diffraction analysis reveals that both P2-O2 phase transitions and Na^(+)/vacancy ordering is suppressed by Ti^(4+)/F^(-) co-doping,which resulting in fast Na^(+) diffusion and stable phase structure.The hard carbon//P2-Na_(0.67)Ni_(0.33)Mn_(0.37)Ti_(0.3)O_(1.9)F_(0.1) full cell exhibits a high energy density of 310.2 Wh kg^(-1) and remarkable cyclability with 82.1%retention after 300 cycles at 1 C in the voltage range of 1.5–4.2 V.These results demonstrate that the co-doping Ti^(4+)/F^(-) is a promising strategy to improve the electrochemical properties of P2-Na_(0.67)Ni_(0.33)Mn_(0.67)O_(2),providing a facile tactic to develop high performance cathode materials for SIBs. | Pengfei Zhou Jing Zhang Zhennan Che Zuhao Quan Ju Duan Xiaozhong Wu Junying Weng Jinping Zhao Jin Zhou | 2022 | Journal of Energy Chemistry2022,31,4: | 3 |

| 14 | Triangular element partition method with consideration of crack tip显示文摘In fracture simulation,how to model the pre-existing cracks and simulate their propagation without remeshing is an important topic.The newly developed triangular element partition method(TEPM)provides an efficient approach to this problem.It firstly meshes the cracked body regardless of the geometry integrity of the interesting object with triangular elements.After the meshing procedure is completed,some elements are intersected by cracks.For the element intersected by a crack,the TEPM takes the element partition technique to incorporate the discontinuity into the numerical model without any interpolation enrichment.By this approach,the TEPM can simulate fracture without mesh modification.In the TEPM,all the cracked elements are treated as the usual partitioned elements in which the crack runs through.The virtual node pairs(the intersection points of crack faces and elements)at the opposite faces of the crack move independently.Their displacements are respectively determined by their neighbor real nodes(nodes formatted in the original mesh scheme)at the same side of the crack.However,among these cracked elements,the element containing a crack tip,referred to as the crack tip element thereafter,behaves differently from those cut through by the crack.Its influence on the singular field at the vicinity of the fracture tip becomes increasingly significant with the element size increasing.In the crack tip element,the virtual node pair at the crack tip move consistently before fracture occurs while the virtual node pair separate and each virtual node moves independently after the fracture propagates.Accordingly,the crack tip element is automatically transformed into the usual partitioned element.In the present paper,the crack tip element is introduced into the TEPM to account for the effect of the crack tip.Validation examples indicate that the present method is almost free from the element size effect.It can reach the same precision as the conventional finite element method under the same meshing scheme.But the TEPM is much more efficient and convenient than the conventional finite element method because the TEPM avoids the troubles that the conventional finite element method suffers,e.g.,the meshing problem of cracked body,modification of mesh scheme,etc.Though the extended finite element method can also avoid these troubles,it introduces extra degrees of freedom due to node interpolation enrichment.Due to the simplicity of the present TEPM,it is believed that its perspective should be highly inspiring. | ZHANG ZhenNan ZHENG Hong GE XiuRun | 2013 | Science China(Technological Sciences)2013,56,8: | 1 |

| 17 | Glyphosate exposure deteriorates oocyte meiotic maturation via induction of organelle dysfunctions in pigs显示文摘Background:Recently,defects in mammalian oocytes maturation induced by environmental pollution results in the decreasing animal reproduction.Animal exposed to glyphosate is largely unavoidable because glyphosate is one of the most widely used herbicide worldwide due to its high-efficiency and broad-spectrum effects,which causes glyphosate an environmental contaminant found in soil,water and food.During the last few years,the growing and wider use of glyphosate has raised great concerns about its effects of reproductive toxicity.In this study,using porcine models,we investigated effects of glyphosate on organelle functions during oocyte meiosis.Results:The results showed glyphosate exposure disrupted porcine oocyte maturation.Expression levels of cumulus expansion-related genes were interfered,further indicating the meiotic defects.The damaging effects were mediated by destruction of mitochondrial distribution and functions,which induced ROS accumulation and oxidative stress,also indicated by the decreased mRNA expression of related antioxidant enzyme genes.We also found an interference of endoplasmic reticulum(ER)distribution,disturbance of Ca^(2+)homeostasis,as well as fluctuation of ER stress,showing with the reduced ER stress-related mRNA or protein expression,which could indicate the dysfunction of ER for protein processing and signal transduction in glyphosate-exposed oocytes.Moreover,glyphosate exposure induced the disruption of lysosome function for autophagy,showing with the decrease of LAMP2 expression and autophagy-related genes mRNA expression.Additionally,our data showed the distribution of Golgi apparatus and the functions of ribosome were disturbed after glyphosate exposure,which might affect protein synthesis and transport.Conclusions:Collectively,our study showed that exposed to glyphosate could affect animal reproduction by compromising the quality of oocytes through its wide toxic effects on organelle functions. | Chunhua Xing Shun Chen Yue Wang Zhennan Pan Yuanjing Zou Shaochen Sun Zili Ren Yu Zhang | 2022 | Journal of Animal Science and Biotechnology2022,13,6: | 1 |
