2Agonistic AT_1 Receptor Autoantibody Increases in Serum of Patients with Refractory Hypertension and Improves Ca^(2+) Mobilization in Cultured Rat Vascular Smooth Muscle Cells显示文摘Agonistic AT1 receptor autoantibodies (AT1-AAs) have been described in the patients with malignant hypertension or preeclampia. Furthermore, AT1-AAs were highly associated with refractory hypertension. Function of vascular smooth muscle cells (VSMCs) is important in the regulation of blood pressure. We investigated and compared the ability of angiotensin II (Ang II) and AT1-AAs to stimulate the intracellular calcium mobilization and cellular proliferation of rat VSMCs. Twenty-two patients with refractory hypertension, 24 patients with non-refractory hypertension and 37 normotensives were recruited. The serum of each patient was detected for the presence of AT1-AAs by ELISA. Ang II and the AT1-AAs from the sera of patients were used to stimulate rat VSMCs in vitro. AT1-AAs were detected in 10/22, 3/24 and 3/37 of patients with refractory hypertension, non-refractory hypertension and normotensives, respectively. AT1-AAs led the increase intracellular calcium mobilization in a dose-dependent manner and cellular proliferation of VSMCs just as Ang II. Both of these effects caused by AT1-AAs were blocked with losartan or a peptide corresponding to a part of the second extracellular loop of AT1 receptor. Since AT1-AAs exhibited pharmacological activity in rat VSMCs just as Ang II, they might play a role in the elevation of peripheral vascular resistance and in vascular remodeling. 9Surfactant assisted solvothermal synthesis of LiFePO4 nanorods for lithium-ion batteries显示文摘Well-shaped and uniformly dispersed LiFePO_4 nanorods with a length of 400–500 nm and a diameter of about 100 nm, are obtained with participation of a proper amount of anion surfactant sodium dodecyl sulfonate(SDS) without any further heating as a post-treatment. The surfactant acts as a self-assembling supermolecular template, which stimulated the crystallization of LiFePO_4 and directed the nanoparticles growing into nanorods between bilayers of surfactant(BOS). LiFePO_4 nanorods with the reducing crystal size along the b axis shorten the diffusion distance of Li^+ extraction/insertion, and thus improve the electrochemical properties of LiFePO_4 nanorods. Such prepared LiFePO_4 nanorods exhibited excellent specific capacity and high rate capability with discharge capacity of 151 mAh/g, 122 mAh/g and 95 mAh/g at 0.1C, 1 C and 5 C, respectively. Such excellent performance of LiFePO_4 nanorods is supposed to be ascribed to the fast Li^+ diffusion velocity from reduced crystal size along the b axis and the well electrochemical conductivity. 11ATF4/TXNIP/REDD1/mTOR signaling mediates the antitumor activities of liver X receptor in pancreatic cancers显示文摘Background:Limited by difficulties in early detection and availabilities of effective treatments,pancreatic cancer is a highly malignant disease with poor prognosis.Nuclear receptors are a family of ligand‐dependent transcription factors that are highly druggable therapeutic targets playing critical roles in human physiological and pathological development,including cancer.In this study,we explored the therapeutic potential as well as the molecular mechanisms of liver X receptor(LXR)agonist GW3965 in pancreatic cancer.Methods:Soft‐agar colony formation assay,xenograft tumors,Oligonucleotide microarray,Reverse transcription real‐time polymerase chain reaction,Western immunoblotting and Immunohistochemistry were used in this study.Results:We demonstrated pleotropic in vitro activities of GW3965 in pancreatic cell lines MIA PaCa‐2 and BXPC3 including reduction of cell viability,inhibition of cell proliferation,stimulation of cell death,and suppression of colony formation,which translated to significant inhibition of xenograft tumor growth in vitro.By mapping the gene expression profiles,we identified the up‐regulations of 188 and the down‐regulations of 92 genes common to both cell lines following GW3965 treatment.Genes responsive to GW3965 represent a variety of biological pathways vital for multiple cellular functions.Specifically,we identified that the activating transcription factor 4/thioredoxin‐interacting protein/regulated in development and DNA damage responses 1/mechanistic target of rapamycin(ATF4/TXNIP/REDD1/mTOR)signaling critically controls GW3965‐mediated regulation of cell proliferation/death.The significance of the ATF4/TXNIP/REDD1/mTOR pathway was further supported by associated expressions in xenograft tumors as well as human pancreatic cancer samples.Conclusions:This study provides the pre‐clinical evidence that LXR agonist is a promising therapy for pancreatic cancer.Zhikang Chen Xiaobo Lai Hui Ding Aijun Zhang Yufei Sun Jianhua Ling Paul J.Chiao Zihua Chen Xuefeng Xia 2022Cancer Innovation2022,1,1:1

173-Hydroxybutyrate ameliorates insulin resistance by inhibiting PPARγSer273 phosphorylation in type 2 diabetic mice显示文摘3-Hydroxybutyrate(3HB)is a small ketone body molecule produced endogenously by the body in the liver.Previous studies have shown that 3HB can reduce blood glucose level in type 2 diabetic(T2D)patients.However,there is no systematic study and clear mechanism to evaluate and explain the hypoglycemic effect of 3HB.Here we demonstrate that 3HB reduces fasting blood glucose level,improves glucose tolerance,and ameliorates insulin resistance in type 2 diabetic mice through hydroxycarboxylic acid receptor 2(HCAR2).Mechanistically,3HB increases intracellular calcium ion(Ca^(2+))levels by activating HCAR2,thereby stimulating adenylate cyclase(AC)to increase cyclic adenosine monophosphate(cAMP)concentration,and then activating protein kinase A(PKA).Activated PKA inhibits Raf1 proto-oncogene serine/threonine-protein kinase(Raf1)activity,resulting in a decrease in extracellular signal-regulated kinases 1/2(ERK1/2)activity and ultimately inhibiting peroxisome proliferator-activated receptorγ(PPARγ)Ser273 phosphorylation in adipocytes.Inhibition of PPARγSer273 phosphorylation by 3HB altered the expression of PPARγregulated genes and reduced insulin resistance.Collectively,3HB ameliorates insulin resistance in type 2 diabetic mice through a pathway of HCAR2/Ca^(2+)/cAMP/PKA/Raf1/ERK1/2/PPARγ.Yudian Zhang Zihua Li Xinyi Liu Xinyu Chen Shujie Zhang Yuemeng Chen Jiangnan Chen Jin Chen Fuqing Wu Guo-Qiang Chen 2023Signal Transduction and Targeted Therapy2023,8,6:0

19Geological characteristics of the Nankai Trough subduction zone and their tectonic significances显示文摘The Nankai Trough subduction zone is a typical subduction system characterized by subduction of multiple geological units of the Philippine Sea Plate(the Kyushu-Palau Ridge,the Shikoku Basin,the Kinan Seamount Chain,and the Izu-Bonin Arc)beneath the Eurasian Plate in the southwest of Japan.This study presents a geophysical and geochemical analysis of the Nankai Trough subduction zone in order to determine the features and subduction effects of each geological unit.The results show that the Nankai Trough is characterized by lowgravity anomalies(–20 mGal to–40 mGal)and high heat flow(60–200 mW/m2)in the middle part and low heat flow(20–80 mW/m2)in the western and eastern parts.The crust of the subducting plate is 5–20 km thick.The mantle composition of the subducting plate is progressively depleted from west to east.Subduction of aseismic ridges(e.g.,the Kyushu-Palau Ridge,the Kinan Seamount Chain,and the Zenisu Ridge)is a common process that leads to a series of subduction effects at the Nankai Trough.Firstly,aseismic ridge or seamount chain subduction may deform the overriding plate,resulting in irregular concave topography along the front edge of the accretionary wedge.Secondly,it may have served as a seismic barrier inhibiting rupture propagation in the 1944 Mw 8.1 and 1946 Mw 8.3 earthquakes.In addition,subduction of the Kyushu-Palau Ridge and hot and young Shikoku Basin lithosphere may induce slab melting,resulting in adakitic magmatism and the provision of ore-forming metals for the formation of porphyry copper and gold deposits in the overriding Japan Arc.Based on comparisons of their geophysical and geochemical characteristics,we suggest that,although the Izu-Bonin Arc has already collided with the Japan Arc,the Kyushu-Palau Ridge,which represents a remnant arc of the Izu-Bonin Arc,is still at the subduction stage characterized by a single-vergence system and a topographic boundary with the Japan Arc.Jie Zhang Ling Chen Zihua Cheng Limei Tang 2020Acta Oceanologica Sinica2020,39,10:0
