2Efficacy and safety of a novel anti-HER2 therapeutic antibody RC48 in patients with HER2-overexpressing,locally advanced or metastatic gastric or gastroesophageal junction cancer:a single-arm phase II study显示文摘Background:Current treatment options for human epidermal growth factor receptor 2(HER2)-overexpressing gastric cancer at third-line have shown limited clinical benefit.Further,there is no specific treatment for HER2 immunohistochemistry(IHC)2+and fluorescence in-situ hybridization-negative patients.Here,we report the efficacy and safety of a novel anti-HER2 antibody RC48 for patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer.Methods:Patients with HER2-overexpressing(IHC 2+or 3+),locally advanced or metastatic gastric or gastroesophageal junction cancer who were under at least second-line therapy were eligible and received RC482.5 mg/kg alone every 2 weeks.The primary endpoint was the objective response rate(ORR)assessed by an independent review committee.Secondary endpoints included progressionfree survival(PFS),overall survival(OS),duration of response,time to progression,disease control rate,and safety.Results:Of 179 patients screened,125 were eligible and received RC48 treatment.The ORR was 24.8%(95%confidence interval[CI]:17.5%-33.3%).The median PFS and OS were 4.1 months(95%CI:3.7-4.9 months)and 7.9 months(95%CI:6.7-9.9 months),respectively.The most frequently reported adverse events were decreased white blood cell count(53.6%),asthenia(53.6%),hair loss(53.6%),decreased neutrophil count(52.0%),anemia(49.6%),and increased aspartate aminotransferase level(43.2%).Serious adverse events(SAEs)occurred in 45(36.0%)patients,and RC48-related SAEs were mainly decreased neutrophil count(3.2%).Seven patients had adverse events that led to death were not RC48-related.Conclusions:RC48 showed promising activity with manageable safety,suggesting potential application in patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer who have previously received at least two lines of chemotherapy.Zhi Peng Tianshu Liu Jia Wei Airong Wang Yifu He Liuzhong Yang Xizhi Zhang Nanfeng Fan Suxia Luo Zhen Li Kangsheng Gu Jianwei Lu Jianming Xu Qingxia Fan Ruihua Xu Liangming Zhang Enxiao Li Yuping Sun Guohua Yu Chunmei Bai Yong Liu Jiangzheng Zeng Jieer Ying Xinjun Liang Nong Xu Chao Gao Yongqian Shu Dong Ma Guanghai Dai Shengmian Li Ting Deng Yuehong Cui Jianmin Fang Yi Ba Lin Shen 2021Cancer Communications2021,41,11:29
3Discrimination of the rare medicinal plant Dendrobium officinale based on naringenin, bibenzyl, and polysaccharides显示文摘The aim of this study was to establish a method for discriminating Dendrobium officinale from four of its close relatives Dendrobium chrysanthum, Dendrobium crystallinum, Dendrobium aphyllum and Dendrobium devonianum based on chemical composition analysis. We analyzed 62 samples of 24 Dendrobium species. High performance liquid chromatography analysis confirmed that the four low molecular weight compounds 4′,5,7-trihydroxyflavanone (naringenin), 3,4-dihydroxy-4′,5-dime- thoxybibenzyl (DDB-2), 3′,4-dihydroxy-3,5′-dimethoxybibenzyl (gigantol), and 4,4′-dihydroxy-3,3′,5-trimethoxybibenzyl (moscatilin), were common in the genus. The phenol-sulfuric acid method was used to quantify polysaccharides, and the monosaccharide composition of the polysaccharides was determined by gas chromatography. Stepwise discriminant analysis was used to differentiate among the five closely related species based on the chemical composition analysis. This proved to be a simple and accurate approach for discriminating among these species. The results also showed that the polysaccharide content, the amounts of the four low molecular weight compounds, and the mannose to glucose ratio, were important factors for species discriminant. 9Nanoencapsulation of arsenate with nanoscale zero-valent iron(nZVI):A 3D perspective显示文摘The principal forces driving the efficient enrichment and encapsulation of arsenic(As) into nanoscale zero-valent iron(nZVI) are the disordered arrangement of the atoms and the gradient chemical potentials within the core-shell interface. The chemical compositions and the fine structure of nZVI are characterized with a combination of spherical aberration corrected scanning transmission electron microscopy(Cs-STEM), X-ray energy-dispersive spectroscopy(XEDS), electron energy loss spectroscopy(EELS), and high-resolution X-ray photoelectron spectroscopy(HR-XPS). Atomically resolved EELS at the oxygen K-edge unfolds that the Fe species in nZVI are well stratified from Fe(Ⅲ) oxides in the outermost periphery to a mixed Fe(Ⅲ)/Fe(Ⅱ) interlayer, then Fe(Ⅱ) oxide and the pure Fe(0) phase. Reactions between As(Ⅴ)and nZVI suggest that a well-structured local redox gradient exists within the shell layer, which serves as a thermodynamically favorable conduit for electron transfer from the iron core to the surface-bound As(Ⅴ). HR-XPS with ion sputtering shows that arsenic species shift from As(Ⅴ), As(Ⅲ)/As(Ⅴ) to As(Ⅴ)/As(Ⅲ)/As(0) from the iron oxide shell–water interface to the Fe(0) core. 10Optical properties of estuarine dissolved organic matter isolated using cross-flow ultrafiltration显示文摘Dissolved organic matter(DOM) from freshwater, mid-salinity, and seawater endmember samples in the Jiulong River Estuary, China were fractionated using cross-flow ultrafiltration with a 10-kDa membrane. The colloidal organic matter(COM; 10 kDa–0.22 μm) retentate, low molecular weight(LMW) DOM(<10 kDa) permeate, and bulk samples were analyzed using absorption spectroscopy and three-dimensional fluorescence excitation-emission-matrix spectroscopy. The UV-visible spectra of COM were very similar to those obtained for permeate and bulk samples, decreasing monotonically with increasing wavelength. Most of the chromophoric DOM(CDOM, expressed as the absorption coefficient a355) occurred in the LMW fraction, while the percentage of CDOM in the colloidal fraction was substantially higher in the freshwater endmember(13.4% of the total) than in the seawater endmember(6.8%). The bulk CDOM showed a conservative mixing behavior in the estuary, while there was removal of the COM fraction and a concurrent addition of the permeate fraction in the mid-salinity sample, implying that part of the colloidal CDOM was transformed into LMW CDOM. Two humic-like components(C1: <250, 325/402 nm; and C2: 265, 360/458 nm) and one protein-like component(C3: 275/334 nm) were identified using parallel factor analysis. The contributions of the C1, C2, and C3 components of the COM fraction to the bulk sample were 2.5%–8.7%, 4.8%–12.6%, and 7.4%–14.7%, respectively, revealing that fluorescent DOM occurred mainly in the LMW fraction in the Jiulong River Estuary. The C1 and C2 components in the retentate and permeate samples showed conservative mixing behavior, but the intensity ratio of C2/C1 was higher in the retentate than in the permeate fractions for all salinity samples, showing that the humic component was more enriched in the COM than the fulvic component. The intensity ratio of C3/(C1+C2) was much higher in the retentate than in the permeate fraction for mid-salinity and seawater samples, revealing that the protein-like component was relatively more enriched in COM than the humic-like component. 11Novel SNPs in IL-17F and IL-17A genes associated with somatic cell count in Chinese Holstein and Inner-Mongolia Sanhe cattle显示文摘Background: Bovine mastitis is the most common and costly disease of lactating cattle worldwide. Apart from milk somatic cell count(SCC) and somatic cell score(SCS), serum cytokines such as interleukin-17(IL-17) and interleukin-4(IL-4) may also be potential indicators for bovine mastitis. The present study was designed to investigate the effects of single nucleotide polymorphisms(SNPs) in bovine IL-17 F and IL-17 A genes on SCC, SCS and serum cytokines in Chinese Holstein and Inner-Mongolia Sanhe cattle, and to compare the m RNA expression variations of the cows with different genotypes.Results: A total of 464 lactating cows(337 Holstein and 127 Inner-Mongolia Sanhe cattle) were screened for SNPs identification and the data were analyzed using fixed effects of herd, parity, season and year of calving by general linear model procedure. The results revealed that SNP g.24392436 C > T in IL-17 F and SNP g.24345410 A > G in IL-17 A showed significant effects on SCC and IL-4 in Holstein(n = 337) and on IL-17 and IL-4 in Sanhe cattle(n = 127). 12NF-κB and FosB mediate inflammation and oxidative stress in the blast lung injury of rats exposed to shock waves显示文摘Blast lung injury(BLI)is the major cause of death in explosion-derived shock waves;however,the mechanisms of BLI are not well understood.To identify the time-dependent manner of BLI,a model of lung injury of rats induced by shock waves was established by a fuel air explosive.The model was evaluated by hematoxylin and eosin staining and pathological score.The inflammation and oxidative stress of lung injury were also investigated.The pathological scores of rats’lung injury at 2 h,24 h,3 days,and 7 days post-blast were 9.75±2.96,13.00±1.85,8.50±1.51,and 4.00±1.41,respectively,which were significantly increased compared with those in the control group(1.13±0.64;P<0.05).The respiratory frequency and pause were increased significantly,while minute expiratory volume,inspiratory time,and inspiratory peak flow rate were decreased in a time-dependent manner at 2 and 24 h post-blast compared with those in the control group.In addition,the expressions of inflammatory factors such as interleukin(IL)-6,IL-8,FosB,and NF-κB were increased significantly at 2 h and peaked at 24 h,which gradually decreased after 3 days and returned to normal in 2 weeks.The levels of total antioxidant capacity,total superoxide dismutase,and glutathione peroxidase were significantly decreased 24 h after the shock wave blast.Conversely,the malondialdehyde level reached the peak at 24 h.These results indicated that inflammatory and oxidative stress induced by shock waves changed significantly in a time-dependent manner,which may be the important factors and novel therapeutic targets for the treatment of BLI.Hong Wang Wenjuan Zhang Jinren Liu Junhong Gao Le Fang Zhiyong Liu Baoqing Xia Xiaolin Fan Cunzhi Li Qing Lu Airong Qian 2021Acta Biochimica et Biophysica Sinica2021,53,3:4

14General formulas for drag coefficient and settling velocity of sphere based on theoretical law显示文摘The settlement of particles is of great importance in many areas.The accurate determination of drag coefficient and settling velocity in wide Reynolds number(Re)range remains a problem.In this paper,a series of new formulas for drag coefficient of spherical particles based on theoretical laws,such as the Stokes law,the Oseen law,and the Goldstein law,were developed and fitted using 480 groups of experimental data(Re<2 105).The results show that the 2nd approximation of a rational function containing only one parameter can describe CD–Re relationship accurately over the whole Re range of0–2 105.The new developed formulas containing five parameters show higher goodness over wide Re range than presently existing equations.The introduction of the Oseen law is helpful for improving the fitting goodness of the empirical formulas.On the basis of one of the Oseen-based CD–Re formulas giving the lowest sum of squared relative errors Q over the whole Re range(Re<2 105),a general formula for settling velocity utbased on dimensionless parameters was proposed showing high goodness.Yang Hongli Fan Minqiang Liu Airong Dong Lianping 2015International Journal of Mining Science and Technology2015,25,2:3
15N-P fertilization did not reduce AMF abundance or diversity but alter AMF composition in an alpine grassland infested by a root hemiparasitic plant显示文摘Fertilization has been shown to have suppressive effects on arbuscular mycorrhizal fungi(AMF) and root hemiparasites separately in numerous investigations, but its effects on AMF in the presence of root hemiparasites remain untested. In view of the contrasting nutritional effects of AMF and root hemiparasites on host plants, we tested the hypothesis that fertilization may not show strong suppressive effects on AMF when a plant community was infested by abundant hemiparasitic plants. Plants and soil samples were collected from experimental field plots in Bayanbulak Grassland, where N and P fertilizers had been applied for three continuous years for control against a spreading root hemiparasite, Pedicularis kansuensis. Shoot and root biomass of each plant functional group were determined. Root AMF colonization levels, soil spore abundance, and extraradical hyphae length density were measured for three soil depths(0 e10 cm, 10 e20 cm, 20 e30 cm). Partial 18 S r RNA gene sequencing was used to detect AMF diversity and community composition. In addition, we analyzed the relationship between relative abundance of different AMF genera and environmental factors using Spearman's correlation method. In contrast to suppressive effects reported by many previous studies, fertilization showed no significant effects on AMF root colonization or AMF species diversity in the soil. Instead, a marked increase in soil spore abundance and extraradical hyphae length density were observed. However, fertilization altered relative abundance and AMF composition in the soil. 16Study of colloidal phosphorus variation in estuary with salinity显示文摘The variation of colloidal phosphorus with salinity in estuary was studied in this paper,which was compared with those of particle and truly dissolved phosphorus with the purpose of finding out the similarities and differences between their behaviors in the estuary. Distribution patterns of phosphorus in particle,colloidal and truly dissolved phase at different salinities and their relationships with suspended particulate matter were also studied to understand the transformation and transportation of colloidal phosphorus in estuarine area. The result showed that the concentrations of total colloidal phosphorus,organic colloidal phosphorus and inorganic colloidall phosphate all descended from river-end to sea-end,illustrating their terrigenous source. Ratios of organic to total colloidal phosphorus decreased with salinity's increment,indicating that organic phosphorus was influenced by scavenging process of colloid more significantly as compared with inorganic phosphate. 17On estimation of attraction domain for port-controlled Hamiltonian systems subject to actuator saturation显示文摘This paper investigates the estimation of domain of attraction for nonlinear port-controlled Hamiltonian (PCH) systems with actuator saturation (AS). Several conditions are established under which an ellipsoid is contractively invariant, and thus can be employed to find the biggest ellipsoid contained in the domain of attraction. It is shown that the proposed conditions can be expressed in the form of the linear matrix inequalities (LMIs) optimization problem with constraints. 18Macroporous 3D carbon-nitrogen(CN)confined MoO_(x) catalyst for enhanced oxidative desulfurization of dibenzothiophene显示文摘Macroporous 3D carbon doped with nitrogen confined Mo catalyst(MoO_(x)@CN)had been prepared by a facile one-step pyrolysis technique using silica as a template and was employed for oxidative desulfurization(ODS)of dibenzothiophene(DBT)in model fuel with H2O_(2) as oxidant.The effect of different ope rating conditions(i.e.,reaction te mperature and time,catalyst dosage,H2O_(2)/DBT(O/S)molar ratio)were also systematic investigated.Under the optimal reaction condition,MoO_(x)@CN catalyst exhibited highly excellent ODS performance toward DBT,the highest sulfur removal efficiency can be up to 99.9%and sulfur content was wiped out from 800 ppm to 10 ppm.Due to the robust 3D structure promoting rapid transfer,in addition to the increased number of active sites induced by the Mo vacancies,the catalyst,prepared using chitosan and ammonium heptamolybdate in a mass ratio of 1:0.5,displayed rapid kinetics and low activation energy in the oxidation of dibenzothiophene.Moreover,it exhibited excellent recyclability after five cycles without any obvious decrease in catalytic activity for the oxidative desulfurization reaction.Jinmei Ye Jie Wen Dongdong Zhao Peilun Zhang Airong Li Lianhong Zhang Hui Zhang Mengqi Wu 2020Chinese Chemical Letters2020,31,10:3
