1HER2 heterogeneity is a poor prognosticator for HER2-positive gastric cancer显示文摘BACKGROUND The clinical significance of intratumoral human epidermal growth factor receptor 2(HER2)heterogeneity is unclear for HER2-positive gastric cancer,although it has been reported to be a significant prognosticator for HER2-positive breast cancer,which has received trastuzumab-based chemotherapy.AIM To clarify the clinical significance of intratumoral HER2 heterogeneity for HER2-positive gastric cancer,which has received trastuzumab-based chemotherapy.METHODS Patients with HER2-positive unresectable or metastatic gastric cancer who received trastuzumab-based chemotherapy as a first line treatment were included.The patients were classified into two groups according to their intratumoral HER2 heterogeneity status examined by immunohistochemistry(IHC)on endoscopic biopsy specimens before treatment,and their clinical response to chemotherapy and survival were compared.RESULTS A total of 88 patients were included in this study,and HER2 heterogeneity was observed in 23(26%)patients(Hetero group).The overall response rate was significantly better in patients without HER2 heterogeneity(Homo group)(Homo vs Hetero:79.5%vs 35.7%,P=0.002).Progression-free survival of trastuzumab-based chemotherapy was significantly better in the Homo group(median,7.9 vs 2.5 mo,HR:1.905,95%CI:1.109-3.268).Overall survival was also significantly better in the Homo group(median survival time,25.7 vs 12.5 mo,HR:2.430,95%CI:1.389-4.273).Multivariate analysis revealed IHC HER2 heterogeneity as one of the independent poor prognostic factors(HR:3.115,95%CI:1.610-6.024).CONCLUSION IHC of HER2 heterogeneity is the pivotal predictor for trastuzumab-based chemotherapy.Thus,HER2 heterogeneity should be considered during the assessment of HER2 expression.Akio Kaito Takeshi Kuwata Masanori Tokunaga Kohei Shitara Reo Sato Tetsuo Akimoto Takahiro Kinoshita 2019World Journal of Clinical Cases2019,7,15:5
2Testing Potential Effect of Environmental Endocrine Disruptors in Cow Milk on Reproductive Index in Female Rats显示文摘Objective To study the effect of endocrine disruptor chemicals in cow milk on female reproductive system. Methods A two-generation reproduction was conducted according to U. S. FDA standard. Milk was fed in special bottle to Wistar rats of both sexes through two successive generations (F0 and F1) in the milk group while artificial milk was fed to rats in the control group. Twenty-four rats of each sex were mated in each group. Measurements were made according to this guideline. Results Reproductive parameters in the milk group such as fertility index, gestation index, weights of uterus and ovary, days of vaginal opening, estrous cycles, histological morphological changes were comparable to those in the control group. However, the means of body weight had some differences. The body weight gains increased significantly in the milk-treated group in F1 and F2 generation compared with those in the control group. The concentration of insulin-like growth factor-1 (IGF-1) in blood in the milk group was comparable to that in the control group, but the standard deviation changed greatly in the milk-treated rats. 7Production of the Blood Pressure Lowing Peptides from Brown Alga(Undaria pinnatifida)显示文摘Brown alga (Undaria pinnatifida) was treated with alginate lyase and hydrolyzed using 17 kinds of proteases andthe inhibitory activity of the hydrolysates for the angiotensin-I-converting enzyme (ACE) was measured. Four hydrolysateswith potent ACE-inhibitory activity were administered singly and orally to spontaneously hypertensive rats (SHRs). The sys-tolic blood pressure of SHRs decreases significantly after single oral administration of the brown alga hydrolysates by pro-tease S ’Amano’ (from Bacillus stearothermophilus) at the concentration of 10 (mg protein) (kg body weight)-1. In the 17weeks of feeding experiment, 7-week-old SHRs were fed standard diet supplemented with the brown alga hydrolysates for 10weeks. In SHRs fed 1.0 and 0.1% brown alga hydrolysates, elevating of systolic bloodpressure was significantly suppressedfor 7 weeks. To elucidate the active components, the brown alga hydrolysates were fractionated by 1-butanol extraction andHPLC on a reverse-phase column. Seven kinds of ACE-inhibitory peptides were isolated and identified by amino acid compo-sition analysis, sequence analysis, and LC-MS with the results Val-Tyr, Ile-Tyr, Ala-Trp, Phe-Tyr, Val-Trp, Ile-Trp, andLeu-Trp. Each peptide was determined to have an antihypertensive effect after a single oral administration in SHRs. 16牛奶中雌激素类化合物对前列腺和睾丸的影响显示文摘目的 :研究牛奶中雌激素样物质对男性生殖系统的影响。 方法 :利用世界五大洲肿瘤发病数据库和联合国粮农组织 (FAO)数据库 ,对 42个国家 (地区 )前列腺癌和睾丸癌发病率 ( 1988~ 1992 )与相应的食物消费量资料 ( 1961~ 1990 )进行相关分析 ;根据美国食品药品管理局 (FDA)标准 ,选用Wistar大鼠 ,进行两代繁殖实验 ,主要研究牛奶中雌激素化合物对生殖器官的影响。 结果 :在所分析的食物中 ,前列腺癌与牛奶消费量的关系最密切(r =0 .711) ,其次是肉类与咖啡 ;奶酪与 2 0~ 3 9岁的睾丸癌标化发病率关系最密切 (r =0 .80 4) ,其次是动物性脂肪和牛奶 ;动物实验研究发现 ,大鼠经长期、大量地饮用牛奶后 ,未发现生殖器官明显减轻的现象。 结论李湘鸣 Davaasambuu Ganmaa 秦立强 刘秀梵 Akio Sato 2003中华男科学杂志2003,9,3:1
