1Interleukin 17-Producing γδ T Cells Increased in Patients with Active Pulmonary Tuberculosis显示文摘Although it has been known that γδ T cells may play an important role in the immune response to infection of Mycobacterium tuberculosis (M. tb), the mechanisms by which the γδ T cells participate in the innate and/or acquired immunity to tuberculosis (TB) have not been full elucidated. In the present study, 27 patients with active pulmonary TB and 16 healthy donors (HD) were performed. We found that proportion of IL-17-producing cells among lymphocyte was similar between TB patients and HD, whereas the proportions of γδ T cells in IL-17-producing cells (59.2%) and IL-17-producing cells in γδ T cells (19.4%) in peripheral blood were markedly increased in TB patients when compared to those in HD (43.9% and 7.7%, respectively). In addition, the proportions of IFN-γ-producing γδ T cells in TB patients were obviously lower than that in HD. Upon re-stimulated with M. tb heat-treated antigen (M. tb-HAg) in vitro, fewer IL-17-producing γδ T cells were generated from HD and TB patients, whereas IFN-γ-producing γδ T cells were increased in TB patients compared to that in HD. 2Buyang Huanwu Decoction fraction protects against cerebral ischemia/reperfusion injury by attenuating the inflammatory response and cellular apoptosis显示文摘Buyang Huanwu Decoction fraction extracted from Buyang Huanwu Decoction contains saponins of Astragalus, total paeony glycoside and safflower flavones. The aim of this study was to demonstrate the neuroprotective effect and mechanism of Buyang Huanwu Decoction fraction on ischemic injury both in vivo and in vitro. In vivo experiments showed that 50-200 mg/kg Buyang Huanwu Decoction fraction reduced infarct volume and pathological injury in ischemia/reperfusion rats, markedly inhibited expression of nuclear factor-κB and tumor necrosis factor-α and promoted nestin protein expression in brain tissue. Buyang Huanwu Decoction fraction (200 mg/kg) exhibited significant effects, which were similar to those of 100 mg/kg Ginkgo biloba extract. In vitro experimental results demonstrated that 10-100 mg/L Buyang Huanwu Decoction fraction significantly improved cell viability, decreased the release of lactate dehydrogenase and malondialdehyde levels, and inhibited the rate of apoptosis in HT22 cells following oxygen-glucose deprivation. Buyang Huanwu Decoction fraction (100 mg/L) exhibited significant effects, which were similar to those of 100 mg/L Ginkgo biloba extract. 3Genetic Analysis of Two Weak Dormancy Mutants Derived from Strong Seed Dormancy Wild Type Rice N22 (Oryza sativa)显示文摘Two weak dormancy mutants,designated Q4359 and Q4646,were obtained from the rice cultivar N22 after treatment with 400 Gy 60Co gamma-radiation.Compared to the N22 cultivar,the dormancy of the mutant seeds was more readily broken when exposed to a period of room temperature storage.The mutants also showed a reduced level of sensitivity to abscisic acid compared to the N22 cultivar,although Q4359 was more insensitive than Q4646.A genetic analysis indicated that in both mutants,the reduced dormancy trait was caused by a single recessive allele of a nuclear gene,but that the mutated locus was different in each case.The results of quantitative trait locus(QTL)mapping,based on the F2 population from Q4359 x Nanjing35,suggested that Q4359 lacks the QTL qSdn-1 and carries a novel allele at QTL qSdn-9, while a similar analysis of the Q4646 x Nanjing35 F2 population suggested that Q4646 lacks QTL qSdn-5, both qSdn-1 and qSdn-5 are major effect seed dormancy QTL in N22.Therefore,these two mutants were helpful to understand the mechanism of seed dormancy in N22.Bingyue Lu KunXie Chunyan Yang Long Zhang Tao Wu Xi Liu Ling Jiang Jianmin Wan 2011Journal of Integrative Plant Biology2011,53,5:7
4Three-dimensional structure of a low salinity tongue in the southern Taiwan Strait observed in the summer of 2005显示文摘Cruise observations with CTD (conductivity-temperature-depth) profiler were carried out in the southern Taiwan Strait in the summer of 2005. Using the cruise data, two-dimensional maps of salinity and temperature distributions at depths of 5, 10, 15, 20, and 30 m were generated. The maps show a low salinity tongue sandwiched by low temperature and high salinity waters on the shallow water side and high temperature and high salinity waters on the deep water side. The further analysis indicates that the low salinity water has a nature of river-diluted water. A possible source of the diluted water is the Zhujiang (Pearl) Estuary. Meanwhile, the summer monsoon is judged as a possible driving force for this northeastward jet-like current. The coastal upwelling and the South China Sea Warm Current confine the low salinity water to flow along the central line of the strait. Previous investigations and a numerical model are used to verify that the upstream of the low salinity current is the Zhujiang Estuary. 6An event-related potential study of working memory in children显示文摘To examine the neural mechanisms o working memory in children, event-related potentials (ERPs) were recorded from the 12―13 year-old while they performed a delayed match-to-sample task. The ERP results revealed that new and studied objects both evoked a late positive ERP componen peaking around 350 ms during the working memory process. New objects evoke a more positive ERP waveform than the studied objects. The scalp distri bution showed that the frontal-central electrode sites were associated with object working memory proc esses. When tracking new or studied targets among visual distracters, ERPs of targets and distracters revealed differential responses as early as 150 ms The visual targets evoked larger and more positive ERP responses than the distracters. The typica old-new effect was observed between ERPs of stud ied and new distracters. However, ERPs of new and studied targets differed at about 250 ms, in which new targets evoked more positive-going and slightly earlier ERP responses. 10Chronic intermittent hypoxia promotes the development of experimental non-alcoholic steatohepatitis by modulating Treg/Th17 differentiation显示文摘The present study aims to characterize the effect of chronic intermittent hypoxia and HIF1α on the non-alcoholic steatohepatitis (NASH)process in mice,and to explore the role of the Treg/Th17 balance in the formation of NASH inflammation and fibrosis.To achieve this purpose,simple steatosis was induced in mice by high-fat diet administration.Subsequently,chronic intermittent hypoxia was simulated by intraperitoneally injecting sodium nitrite.The changes of inflammation, fibrosis,and Treg/Th17 balance in the liver were.quantified under chronic intermittent hypoxia condition and after tail vein injection of HIF1α-siRNA.In addition,T cells were cultured in vitro, and HIF1α expression was either blocked or overexpressed under chronic intermittent hypoxia or normal conditions.Then,the changes of Treg/Th17 balance,inflammatory factors,and cell pathways were measured in each group.Our results demonstrated that chronic intermittent hypoxia accelerates the NASH process,while tail vein injection of HIF1α-siRNA improves liver histology and function.Chronic intermittent hypoxia alters the ratio of Th17 and Treg cells through HIF1α and roTOR signaling,and increases the expressions of NF-κB,IL-6,and IL-17,but decreases IL-10 expression.Inhibition of the mTOR-HIF1α-TLR4-IL-6 pathway increases the ratio of Treg/Th17. 11Precipitation Responses to Radiative Effects of Ice Clouds:A Cloud-Resolving Modeling Study of a Pre-Summer Torrential Precipitation Event显示文摘The precipitation responses to the radiative effects of ice clouds are investigated through analysis of five-day and horizontally averaged data from 2D cumulus ensemble model experiments of a pre-summer torrential precipitation event.The exclusion of the radiative effects of ice clouds lowered the precipitation rate through a substantial reduction in the decrease of hydrometeors when the radiative effects of water clouds were switched on,whereas it increased the precipitation rate through hydrometeor change from an increase to a decrease when the radiative effects of ice clouds were turned off.The weakened hydrometeor decrease was associated with the enhanced longwave radiative cooling mainly through the decreases in the melting of non-precipitating ice to non-precipitating water.The hydrometeor change from an increase to a decrease corresponded to the strengthened longwave radiative cooling in the upper troposphere through the weakened collection of non-precipitating water by precipitation water.Xinyong SHEN Wenyan HUANG Chunyan GUO Xiaocen JIANG 2016Advances in Atmospheric Sciences2016,33,10:3
12Carrier Cells for Delivery of Oncolytic Measles Virus into Tumors:Determinants of Efficient Loading显示文摘Oncolytic measles virus(OMV) is a promising antitumor agent. However, the presence of anti-measles neutralizing antibodies(NAbs) against the hemagglutinin(H) protein of OMV is a major barrier to the therapeutic application of OMV in clinical practice. In order to overcome this challenge, specific types of cells have been used as carriers for OMV.Differential loading strategies appear to result in different therapeutic outcomes; despite this, only few studies have reported practical ex vivo loading strategies required for effective treatment. To this end, we systematically evaluated the antitumor efficacy of OMV using different loading strategies; this involved varying the in vitro loading duration and loading dose of OMV. We found that improved oncolysis of carrier cells was achieved by a prolonged loading duration in the absence of NAbs. However, the enhanced oncolytic effect was abrogated in the presence of NAbs. Further, we found that the expression of H protein on the surface of carrier cells was predominantly determined by the loading duration rather than the loading dose. Finally, we showed that NAbs blocked viral transfer by targeting H protein prior to the occurrence of cell-to-cell interactions. 14Effect of Traditional Chinese Medicine plus narrow-band medium-wave ultraviolet B radiation on moderate-to-severe psoriasis vulgaris in a case series显示文摘OBJECTIVE: To investigate the efficacy and safety of Traditional Chinese Medicine (TCM) therapy combined with ultraviolet B light therapy in the treatment of moderate-to-severe psoriasis. METHODS: Patients with moderate-to-severe psoriasis (skin lesion area > 10% of the body surface area) for 2 consecutive years were treated with TCM (oral and external use of herbal medicines, acupuncture, and herbal bathing) and narrow-band medium-wave ultraviolet B light treatment for 12 weeks. The treatment effect was evaluated based on the Psoriasis Area Severity Index (PASI), the achievement of a 50% reduction in the PASI (PASI50), the achievement of a 75% reduction in the PASI (PASI75), pruritus score, Dermatology Life Quality Index, and safety. RESULTS: A total of 95 outpatients were enrolled, and 92 subjects (96.8%) completed the 12-week treatment course. At baseline, the average proportion of the body surface area covered by skin lesions was 12.4%, and the average PASI was 17.7. All patients had previously been treated with conventional medicine (89.1% of patients received ultraviolet light treatment, 50.0% received glucocorticoids, and 21.7% received acitretin). After the 12-week treatment course, 22 patients (23.9%) achieved PASI75, and 43 (46.7%) achieved PASI50. The post-treatment pruritus score and Dermatology Life Quality Index of all treated patients were significantly lower than the respective baseline values (P < 0.0001). 15Expression and prognostic value of plasminogen activator inhibitor type 1 in node-negative breast cancer显示文摘Objective:To investigate the expressions of plasminogen activator inhibitor type 1 (PAI-1), C-erbB-2, VEGF and Ki-67 by immunohistostaining and then to evaluate the prognostic value of PAI-1 in node-negative breast cancer. Methods:The study included a retrospective series of 62 female patients with axillary lymph node-negative breast cancer. Expressions of PAI-1, C-erbB-2, VEGF and Ki-67 were determined by immunohistostaining on formalin-fixed paraffin-embedded tissue sections from these patients after a median follow-up of 69 months (range 22–117 months). Correlations with well known clinicopathologic factors were assessed and multivariate survival analyses were performed. Results: High PAI-1 level was positively associated with high histologic grade of the tumors. Disease-free survival (DFS) was significantly shorter for the patients with moderate to intensive expression of PAI-1 than for those with negative (χ2 = 25.46, P < 0.001; χ2 = 23.07, P < 0.001) to mild expression (χ2 = 19.75, P < 0.001; χ2 = 17.40, P < 0.001). Although on univariate analysis of the prognostic factors, tumor size, location of primary tumor and age as well as expressions of PAI-1, VEGF and Ki-67 were all significantly prognostic factors for DFS (P < 0.05), PAI-1 was the only independent prognostic factor on multivariate analysis (P<0.0001; hazard ratio [HR], 4.041; 95% confidence interval [CI], 1.928–8.468). 16Biomedical polymers: synthesis, properties, and applications显示文摘Biomedical polymers have been extensively developed for promising applications in a lot of biomedical fields, such as therapeutic medicine delivery, disease detection and diagnosis, biosensing, regenerative medicine, and disease treatment. In this review, we summarize the most recent advances in the synthesis and application of biomedical polymers, and discuss the comprehensive understanding of their property-function relationship for corresponding biomedical applications. In particular, a few burgeoning bioactive polymers, such as peptide/biomembrane/microorganism/cell-based biomedical polymers, are also introduced and highlighted as the emerging biomaterials for cancer precision therapy. Furthermore, the foreseeable challenges and outlook of the development of more efficient, healthier and safer biomedical polymers are discussed. 19Improving the ammonia synthesis activity of Ru/CeO_(2) through enhancement of the metal–support interaction显示文摘The metal–support interactions induced by high-temperature hydrogen reduction have a strong influence on the catalytic performance of ceria-supported Ru catalysts. However, the appearance of the strong metal–support interaction leads to covering of the Ru species by Ce suboxides, which is detrimental to the ammonia synthesis reaction that requires metallic species as active sites. In the present work, the interaction between Ru and ceria in the Ru/CeO_(2) catalyst was induced by NaBH_(4) treatment. NaBH_(4) treatment enhanced the fraction of metallic Ru, proportion of Ce^(3+), content of exposed Ru species, and amount of surface oxygen species. As a result, a larger amount of hydrogen species would desorb by the H_(2)-formation pathway and the strength of hydrogen adsorption would be weaker, weakening the inhibition effect of the hydrogen species on ammonia synthesis. In addition, the strong electronic metal–support interaction aids in nitrogen dissociation. 20Composition engineering of Sb_2S_3 film enabling high performance solar cells显示文摘Sb_2S_3 is a kind of stable light absorption materials with suitable band gap, promising for practical applications. Here we demonstrate that the engineering on the composition ratio enables significant improvement in the device performance. We found that the co-evaporation of sulfur or antimony with Sb_2S_3 is able to generate sulfur-or antimony-rich Sb_2S_3. This composition does not generate essential influence on the crystal structure, optical band and film formability, while the carrier concentration and transport dynamics are considerably changed. The device investigations show that sulfur-rich Sb_2S_3 film is favorable for efficient energy conversion, while antimony-rich Sb_2S_3 leads to greatly decreased device performance.
