1Filtration of fine particles in atmospheric aerosol with electrospinning nanofibers and its size distribution显示文摘In this study,the size distribution of atmospheric aerosol in Beijing was monitored by the scanning mobility particle sizer spectrometer and the optical particle sizer.The size of particles in atmospheric aerosol was primarily distributed in the range of less than 1 m.It showed different changes with the mass concentrations of particulate matters with an aerodynamic diameter of 2.5 m(PM2.5)for different sizes of fine particles.The amount of ultrafine particles(less than about 60 nm)decreased while the larger ones(>60 nm)increased along with the mass concentration of PM2.5 in atmospheric aerosol.This was because of the formation of the secondary atmospheric aerosol.The polylactic acid(PLA)nanofibers were prepared for filtering the aerosol by electrospinning.PLA nanofiber mats were used as the middle layer to design the composite filter membranes.Atmospheric aerosol was used as dust source in the filtration test.The results showed that the filtration efficiency of the composite filter media increased along with the thickness of nanofiber mats,which was controlled by the collection time during electrospinning.Filtration efficiency can be improved obviously by compositing with a thin layer of nanofibers.LI Juan SHI XiaoFei GAO Feng LIU LuQi CHEN Rui CHEN ChunYing ZHANG Zhong 2014Science China(Technological Sciences)2014,57,2:7
2Injectable collagen scaffold promotes swine myocardial infarction recovery by long-term local retention of transplanted human umbilical cord mesenchymal stem cells显示文摘Stem cell therapy is an attractive approach for recovery from myocardial infarction(MI)but faces the challenges of rapid diffusion and poor survival after transplantation.Here we developed an injectable collagen scaffold to promote the long-term retention of transplanted cells in chronic MI.Forty-five minipigs underwent left anterior descending artery(LAD)ligation and were equally divided into three groups 2 months later(collagen scaffold loading with human umbilical mesenchymal stem cell(hUMSC)group,hUMSC group,and placebo group(only phosphate-buffered saline(PBS)injection)).Immunofluorescence staining indicated that the retention of transplanted cells was promoted by the collagen scaffold.Echocardiography and cardiac magnetic resonance imaging(CMR)showed much higher left ventricular ejection fraction(LVEF)and lower infarct size percentage in the collagen/hUMSC group than in the hUMSC and placebo groups at 12 months after treatment.There were also higher densities of vWf-,α-sma-,and cTnT-positive cells in the infarct border zone in the collagen/cell group,as revealed by immunohistochemical analysis,suggesting better angiogenesis and more cardiomyocyte survival after MI.Thus,the injectable collagen scaffold was safe and effective on a large animal myocardial model,which is beneficial for constructing a favorable microenvironment for applying stem cells in clinical MI.Qiang Wangn Xiaojun He Bin Wang Jun Pan Chunying Shi Jie Li Liudi Wang Yannan Zhao Jianwu Dai Dongjin Wang 2021Science China(Life Sciences)2021,64,2:5
3SIRT7 orchestrates melanoma progression by simultaneously promoting cell survival and immune evasion via UPR activation显示文摘Melanoma is the most lethal type of skin cancer,originating from the malignant transformation of melanocyte.While the development of targeted therapy and immunotherapy has gained revolutionary advances in potentiating the therapeutic effect,the prognosis of patients with melanoma is still suboptimal.During tumor progression,melanoma frequently encounters stress from both endogenous and exogenous sources in tumor microenvironment.SIRT7 is a nuclear-localized deacetylase of which the activity is highly dependent on intracellular nicotinamide adenine dinucleotide(NAD+),with versatile biological functions in maintaining cell homeostasis.Nevertheless,whether SIRT7 regulates tumor cell biology and tumor immunology in melanoma under stressful tumor microenvironment remains elusive.Herein,we reported that SIRT7 orchestrates melanoma progression by simultaneously promoting tumor cell survival and immune evasion via the activation of unfolded protein response.We first identified that SIRT7 expression was the most significantly increased one in sirtuins family upon stress.Then,we proved that the deficiency of SIRT7 potentiated tumor cell death under stress in vitro and suppressed melanoma growth in vivo.Mechanistically,SIRT7 selectively activated the IRE1α-XBP1 axis to potentiate the pro-survival ERK signal pathway and the secretion of tumorpromoting cytokines.SIRT7 directly de-acetylated SMAD4 to antagonize the TGF-β-SMAD4 signal,which relieved the transcriptional repression on IRE1αand induced the activation of the IRE1α-XBP1 axis.Moreover,SIRT7 up-regulation eradicated anti-tumor immunity by promoting PD-L1 expression via the IRE1α-XBP1 axis.Additionally,the synergized therapeutic effect of SIRT7 suppression and anti-PD-1 immune checkpoint blockade was also investigated.Taken together,SIRT7 can be employed as a promising target to restrain tumor growth and increase the effect of melanoma immunotherapy.Xiuli Yi Huina Wang Yuqi Yang Hao Wang Hengxiang Zhang Sen Guo Jianru Chen Juan Du Yangzi Tian Jingjing Ma Baolu Zhang Lili Wu Qiong Shi Tianwen Gao Weinan Guo Chunying 2023Signal Transduction and Targeted Therapy2023,8,4:1

7Specific bFGF targeting of KIM-1 in ischemic kidneys protects against renal ischemia-reperfusion injury in rats显示文摘Renal ischemia-reperfusion(I/R)injury is one of the major causes of acute kidney injury.However,there is still no effective treatment for this disease.Basic fibroblast growth factor(bFGF)has been reported to be beneficial for recovery from ischemic diseases.It is vital to increase the local concentration and reduce the diffusion of bFGF in vivo for renal I/R injury therapy.A targeted growth factor delivery system that responds to specific biological signals in the regenerative environment to guide release has been highlighted in tissue repair.In the present study,a specific peptide was fused with bFGF and called bFGF-kidney injury targeting(KIT-bFGF),and this compound specifically targeted kidney injury molecule-1 both in hypoxic renal HK-2 cells in vitro and ischemic kidneys in vivo after intravenous injection.When administered to rat models of renal I/R injury,KIT-bFGF attenuated renal tubule damage and fibrosis,and promoted functional recovery compared to the effects of native bFGF and the control.We also investigated the mechanism by which KIT-bFGF activated the ERK1/2 and Akt signaling pathways to significantly reduce apoptosis and protect against ischemic injury in the kidney.These results demonstrated that targeted delivery of KIT-bFGF could be an effective strategy for the treatment of renal I/R injury.Siqi Song Xianglin Hou Weiwei Zhang Xinyu Liu Wei Wang Xiaoya Wang Wenxuan Cao Yujun Xia Wei Chen Chunying Shi 2022Regenerative Biomaterials2022,9,1:0
8Differential effects of recombinant fusion proteins TAT-OCT4 and TAT-NANOG on adult human fibroblasts显示文摘OCT4 and NANOG are two important tran-scription factors for maintaining the pluripotency and self-renewal abilities of embryonic stem(ES)cells.Meanwhile they play key roles in the induced pluripotent stem(iPS)cells.In this study,recombinant transcript factors TAT-NANOG and TAT-OCT4,which contained a fused powerful protein transduction domain(PTD)TAT from human immunodeficiency virus(HIV),were produced.Each fusion protein could be transported into human adultfibroblasts(HAF)successfully and activated the endogen-ous transcription of both nanog and oct4.Our study revealed the inter-regulation and autoregulation abilities of solo oct4 or nanog in the process of iPS cell reprogram-ming.Meanwhile the transduction of TAT-NANOG could accelerate the growth rate of HAF cells,and the key cell cycle regulator cdc25a was up-regulated.Thus cdc25a may be involved in the regulation of cell growth by NANOG.In addition,the TAT fusion protein technology provided a novel way to improve cell growth that is more controllable and safer.Jiani CAO Zhifeng XIAO Bing CHEN Yuan GAO Chunying SHI Jinhuan WANG Jianwu DAI 2010Frontiers in Biology2010,5,5:0
9Robust nanoporous Cu/TiO_(2) ceramic filter membrane with promoted bactericidal function显示文摘Filter membrane technology has been extensively used for water purification.However,due to their high brittleness and biological fouling,filter membranes will gradually deteriorate.Here we develop a robust Cu/TiO_(2) ceramic filter membrane for water treatment,which has excellent inherent antibacterial activities both in the dark and under visible light irradiation.Onedimensional nanobelt TiO_(2) can be sintered into a nanoporous membrane structure by overlapping and interweaving with each other,thereby achieving high flux simultaneously and solving the problem of high brittleness of inorganic ceramics.Meanwhile,such one-dimensional TiO_(2) nanostructures can provide more deposition sites for Cu nanoparticles with inherent antibacterial activity in the dark.To realise the surface plasmon resonance-enhanced antibacterial activity under visible light,Cu/TiO_(2) heterostructures are used.ZHOU Li SHI ChunYing XU XiaoHong JIA ChuanCheng 2022Science China(Technological Sciences)2022,65,11:0
