Treatment of multiple sclerosis by transplantation of neural stem cells derived from induced pluripotent stem cells显示文摘Multiple sclerosis(MS) is an autoimmune disease of the central nervous system(CNS), with focal T lymphocytic infiltration and damage of myelin and axons. The underlying mechanism of pathogenesis remains unclear and there are currently no effective treatments. The development of neural stem cell(NSC) transplantation provides a promising strategy to treat neurodegenerative disease. However, the limited availability of NSCs prevents their application in neural disease therapy. In this study, we generated NSCs from induced pluripotent stem cells(iPSCs) and transplanted these cells into mice with experimental autoimmune encephalomyelitis(EAE), a model of MS. The results showed that transplantation of iPSC-derived NSCs dramatically reduced T cell infiltration and ameliorated white matter damage in the treated EAE mice. Correspondingly, the disease symptom score was greatly decreased, and motor ability was dramatically rescued in the iPSC-NSC-treated EAE mice, indicating the effectiveness of using iPSC-NSCs to treat MS. Our study provides pre-clinical evidence to support the feasibility of treating MS by transplantation of iPSC-derived NSCs. | Chao Zhang Jiani Cao Xiaoyan Li Haoyu Xu Weixu Wang Libin Wang Xiaoyang Zhao Wei Li Jianwei Jiao Baoyang Hu Qi Zhou Tongbiao Zhao | 2016 | Science China(Life Sciences)2016,59,9: | 9
Abnormal activation of the occipital lobes during emotion picture processing in major depressive disorder patients显示文摘A large number of studies have demonstrated that depression patients have cognitive dysfunction. With recently developed brain functional imaging, studies have focused on changes in brain function to investigate cognitive changes. However, there is still controversy regarding abnormalities in brain functions or correlation between cognitive impairment and brain function changes. Thus, it is important to design an emotion-related task for research into brain function changes. We selected positive, neutral, and negative pictures from the International Affective Picture System. Patients with major depressive disorder were asked to judge emotion pictures. In addition, functional MRI was performed to synchronously record behavior data and imaging data. Results showed that the total correct rate for recognizing pictures was lower in patients compared with normal controls. Moreover, the consistency for recognizing pictures for depressed patients was worse than normal controls, and they frequently recognized positive pictures as negative pictures. The consistency for recognizing pictures was negatively correlated with the Hamilton Depression Rating Scale. Functional MRI suggested that the activation of some areas in the frontal lobe, temporal lobe, parietal lobe, limbic lobe, and cerebellum was enhanced, but that the activation of some areas in the frontal lobe, parietal lobe and occipital lobe was weakened while the patients were watching positive and neutral pictures compared with normal controls. The activation of some areas in the frontal lobe, temporal lobe, parietal lobe, and limbic lobe was enhanced, but the activation of some areas in the occipital lobe were weakened while the patients were watching the negative pictures compared with normal controls. These findings indicate that patients with major depressive disorder have negative cognitive disorder and extensive brain dysfunction. Thus, reduced activation of the occipital lobe may be an initiating factor for cognitive disorder in depressed patients. | Jianying Li Cheng Xu Xiaohua Cao Qiang Gao Yan Wang Yanfang Wang Juyi Peng Kerang Zhang | 2013 | Neural Regeneration Research2013,8,18: | 7

FY-4A/GIIRS Temperature Validation in Winter and Application to Cold Wave Monitoring显示文摘In order to improve the operational application ability of the Fengyun-4A(FY-4A)new sounding dataset,in this paper, validation of the FY-4A Geosynchronous Interferometric Infrared Sounder(FY-4A/GIIRS)temperature was carried out using the balloon sounding temperature from meteorological sounding stations.More than 350,000 samples were obtained through time–space matching, and the results show that the FY-4A/GIIRS temperature mean bias(MB)is 0.07°C,the mean absolute error(MAE)is 1.80°C,the root-mean-square error(RMSE)is 2.546°C,and the correlation coefficient(RR)is 0.95.The FY-4A/GIIRS temperature error is relatively larger in the upper and lower troposphere, and relatively smaller in the middle troposphere;that is,the temperature at 500 hPa is better than that at 850 hPa.The temporal variation is smaller in the upper and middle troposphere than in the lower troposphere. The reconstruction of missing data of FY-4A/GIIRS temperature in cloudy areas is also carried out and the results are evaluated. The spatial distribution of reconstructed FY-4A/GIIRS temperature and the fifth generation ECMWF reanalysis(ERA5)data is consistent and completely retains the minimum temperature center with high precision of FY-4A/GIIRS.There are more detailed characteristics of intensity and position at the cold center than that of the reanalysis data. Therefore,an operational satellite retrieval temperature product with time–space continuity and high accuracy is formed. The reconstructed FY-4A/GIIRS temperature is used to monitor a strong cold wave event in November 2021.The results show that the product effectively monitors the movement and intensity of cold air activities, and it also has good indication for the phase transition of rain and snow triggered by cold wave. | Suling REN Jianying JIANG Xiang FANG Hui LIU Zhiqiang CAO | 2022 | Journal of Meteorological Research2022,36,4: | 3
Effect of Microsilica Addition on Properties of CMA72 Bonded Al2O3-MgO Castables显示文摘CMA72 bonded Al_2O_3-MgO castable is promising for application of steel ladle wall, because of unique combination of thermo-mechanical properties, slag corrosion resistance and cost benefit. In these castables, microsilica can be introduced to counterbalance the expansion generated by spinel formation. In this paper, the effect of microsilica dosage on properties of castables was evaluated. Expansion, expressed by the permanent linear change( PLC),is highly dependent on the dosage of microsilica. Unexpected expansion occurs when the dosage of microsilica is too low due to dominant effect of spinel and CA_6 formation. Too high dosage results in sintering shrinkage, which is related to amount of liquid phase generated by microsilica addition. In addition,HMOR declines dramatically with increasing microsilica dosage. Considering the balance between expansion control and hot property retention,1. 0 mass% of microsilica is recommended for the castable containing 4 mass%of magnesia. | GAO Jianying ZHANG Ziyi BRUNO Touzo CAO Biao | 2016 | China's Refractories2016,25,2: | 2
Biomarkers of aging显示文摘Aging biomarkers are a combination of biological parameters to(i)assess age-related changes,(ii)track the physiological aging process, and(iii)predict the transition into a pathological status.Although a broad spectrum of aging biomarkers has been developed, their potential uses and limitations remain poorly characterized. An immediate goal of biomarkers is to help us answer the following three fundamental questions in aging research:How old are we?Why do we get old?And how can we age slower? This review aims to address this need. Here,we summarize our current knowledge of biomarkers developed for cellular, organ, and organismal levels of aging, comprising six pillars:physiological characteristics, medical imaging, histological features, cellular alterations, molecular changes, and secretory factors. To fulfill all these requisites,we propose that aging biomarkers should qualify for being specific, systemic, and clinically relevant. | Aging Biomarker Consortium Hainan Bao Jiani Cao Mengting Chen Min Chen Wei Chen Xiao Chen Yanhao Chen Yu Chen Yutian Chen Zhiyang Chen Jagadish K Chhetri Yingjie Ding Junlin Feng Jun Guo Mengmeng Guo Chuting He Yujuan Jia Haiping Jiang Ying Jing Dingfeng Li Jiaming Li Jingyi Li Qinhao Liang Rui Liang Feng Liu Xiaoqian Liu Zuojun Liu Oscar Junhong Luo Jianwei Lv Jingyi Ma Kehang Mao Jiawei Nie Xinhua Qiao Xinpei Sun Xiaoqiang Tang Jianfang Wang Qiaoran Wang Siyuan Wang Xuan Wang Yaning Wang Yuhan Wang Rimo Wu Kai Xia Fu-Hui Xiao Lingyan Xu Yingying Xu Haoteng Yan Liang Yang Ruici Yang Yuanxin Yang Yilin Ying Le Zhang Weiwei Zhang Wenwan Zhang Xing Zhang Zhuo Zhang Min Zhou Rui Zhou Qingchen Zhu Zhengmao Zhu Feng Cao Zhongwei Cao Piu Chan Chang Chen Guobing Chen Hou-Zao Chen Jun Chen Weimin Ci Bi-Sen Ding Qiurong Ding Feng Gao Jing-Dong JHan Kai Huang Zhenyu Ju Qing-Peng Kong Ji Li Jian Li Xin Li Baohua Liu Feng Liu Lin Liu Qiang Liu Qiang Liu Xingguo Liu Yong Liu Xianghang Luo Shuai Ma Xinran Ma Zhiyong Mao Jing Nie Yaojin Peng Jing Qu Jie Ren Ruibao Ren Moshi Song Zhou Songyang Yi Eve Sun Yu Sun Mei Tian Shusen Wang Si Wang Xia Wang Xiaoning Wang Yan-Jiang Wang Yunfang Wang Catherine CL Wong Andy Peng Xiang Yichuan Xiao Zhengwei Xie Daichao Xu Jing Ye Rui Yue Cuntai Zhang Hongbo Zhang Liang Zhang Weiqi Zhang Yong Zhang Yun-Wu Zhang Zhuohua Zhang Tongbiao Zhao Yuzheng Zhao Dahai Zhu Weiguo Zou Gang Pei Guang-Hui Liu | 2023 | Science China(Life Sciences)2023,66,5: | 2
Maternal heterozygous mutation in CHEK1 leads to mitotic arrest in human zygotes显示文摘Dear Editor,The first mitotic division in zygotes is crucial for the beginning of the life cycle for the human. After fertilization, zygotes reactivate cell cycle, both paternal and maternal genomes replicate and reprogram to become totipotent. In the meantime, the male and female pronucleus move to the center of the zygote and merge. Then zygotes enter the metaphase, and sister chromatids separate into two daughter cells(Eckersley-Maslin et al.,2018;Reichmann et al.,2018).This is a sensitive time window and many perturbances may cause the first mitosis to fail. | Beili Chen Jianying Guo Ting Wang Qianhui Lee Jia Ming Fangfang Ding Haitao Li Zhiguo Zhang Lin Li Yunxia Cao Jie Na | 2022 | Protein & Cell2022,13,2: | 1
Cellular metabolism and homeostasis in pluripotency regulation显示文摘Pluripotent stem cells(PSCs)can immortally self-renew in culture with a high proliferation rate, and they possess unique metabolic characteristics that facilitate pluripotency regulation. Here,we review recent progress in understanding the mechanisms that link cellular metabolism and homeostasis to pluripotency regulation, with particular emphasis on pathways involving amino acid metabolism, lipid metabolism, the ubiquitin-proteasome system and autophagy.Metabolism of amino acids and lipids is tightly coupled to epigenetic modification, organelle remodeling and cell signaling pathways for pluripotency regulation.PSCs harness enhanced proteasome and autophagy activity to meet the material and energy requirements for cellular homeostasis. These regulatory events reflect a fine balance between the intrinsic cellular requirements and the extrinsic environment.A more complete understanding of this balance will pave new ways to manipulate PSC fate. | Kun Liu Jiani Cao Xingxing Shi Liang Wang Tongbiao Zhao | 2020 | Protein & Cell2020,11,9: | 1
Multiplexed imaging of tumor immune microenvironmental markers in locally advanced or metastatic non-small-cell lung cancer characterizes the features of response to PD-1 blockade plus chemotherapy显示文摘Background:Although programmed cell death 1(PD-1)blockade plus chemotherapy can significantly prolong the progression-free survival(PFS)and overall survival(OS)in first-line settings in patients with driver-negative advanced non-small-cell lung cancer(NSCLC),the predictive biomarkers remain undetermined. Here,we investigated the predictive value of tumor immune microenvironmental marker expression to characterize the response features to PD-1 blockade plus chemotherapy. Methods:Tumor tissue samples at baseline were prospectively collected from 144 locally advanced or metastatic NSCLC patients without driver gene alterations who received camrelizumab plus chemotherapy or chemotherapy alone.Tumor immune microenvironmental markers, including PD-1 ligand(PDL1),CD8,CD68,CD4 and forkhead box P3,were assessed using multiplex immunofluorescence(mIF)assays.Kaplan-Meier curveswere used to determine treatment outcome differences according to their expression status.Mutational profiles were compared between tumors with distinct expression levels of these markers and their combinations. Results:Responders had significantly higher CD8/PD-L1(P=0.015)or CD68/PD-L1 co-expression levels(P=0.021)than non-responders in the camrelizumab plus chemotherapy group, while no difference was observed in the chemotherapy group.Patients with high CD8/PD-L1 or CD68/PD-L1 co-expression level was associated with significantly longer PFS(P=0.002,P=0.024;respectively)and OS(P=0.006,P=0.026;respectively)than those with low co-expression in camrelizumab plus chemotherapy group. When comparing survival in the camrelizumab plus chemotherapy with chemotherapy by CD8/PD-L1 co-expression stratification, significantly better PFS(P=0.003)and OS(P=0.032)were observed in high co-expression subgroups. The predictive value of CD8/PD-L1 and CD68/PD-L1 co-expression remained statistically significant for PFS and OS when adjusting clinicopathological features.Although the prevalence of TP53 or KRAS mutations was similar between patients with and without CD8/PD-L1 or CD68/PD-L1 co-expression,the positive groups had a significantly higher proportion of TP53/KRAS co-mutations than the negative groups(both 13.0%vs.0.0%,P=0.023).Notably,enriched PI3K(P=0.012)and cell cycle pathway(P=0.021)were found in the CD8/PD-L1 co-expression group. Conclusion:Tumor immune microenvironmental marker expression, especially CD8/PD-L1 or CD68/PD-L1 co-expression,was associated with the efficacy of PD-1 blockade plus chemotherapy as first-line treatment in patients with advanced NSCLC. | Fengying Wu Tao Jiang Gongyan Chen Yunchao Huang Jianying Zhou Lizhu Lin Jifeng Feng Zhehai Wang Yongqian Shu Jianhua Shi Yi Hu Qiming Wang Ying Cheng Jianhua Chen Xiaoyan Lin Yongsheng Wang Jianan Huang Jiuwei Cui Lejie Cao Yunpeng Liu Yiping Zhang Yueyin Pan Jun Zhao LiPing Wang Jianhua Chang Qun Chen Xiubao Ren Wei Zhang Yun Fan Zhiyong He Jian Fang Kangsheng Gu Xiaorong Dong Tao Zhang Wei Shi Jianjun Zou Xuejuan Bai Shengxiang Ren Caicun Zhou | 2022 | Cancer Communications2022,42,12: | 1
Oxidative phosphorylation safeguards pluripotency via UDP-N-acetylglucosamine显示文摘Dear Editor,Embryonic stem cells(ESCs)have been assumed to possess immature mitochondria and to favor anaerobic glycolysis over oxidative phosphorylation(OXPHOS)for energy production. This proposition is largely based on the findings that ESCs possess globular mitochondria with blurred cristae, and the facts that ESCs have higher glycolysis activity and lower mitochondrial respiration capacity than somatic cells(Kondoh et al.,2007;Folmes et al.,2011,2012;Zhang et al.,2012;Xu et al.,2013;Ito and Suda,2014;Gu et al.,2016).However,recent studies have shown that mitochondrial autophagy and mitochondrial dynamics are pivotal for ESC self-renewal and pluripotency(Todd et al.,2010;Liu et al.,2016,2020;Wang et al.,2019;Zhong et al.,2019).These studies have raised a fundamental question:what is the contribution and functional mechanism of mitochondrial respiration in pluripotency regulation? | Jiani Cao Meng Li Kun Liu Xingxing Shi Ning Sui Yuchen Yao Xiaojing Wang Shiyu Li Yuchang Tian Shaojing Tan Qian Zhao Liang Wang Xiahua Chai Lin Zhang Chong Liu Xing Li Zhijie Chang Dong Li Tongbiao Zhao | 2023 | Protein & Cell2023,14,5: | 1
Envonalkib versus crizotinib for treatment-naive ALK-positive non-small cell lung cancer: a randomized, multicenter, open-label, phase III trial显示文摘Anaplastic lymphoma kinase(ALK)rearrangements are present in about 5–6%of non-small cell lung cancer(NSCLC)cases and associated with increased risks of central nervous system(CNS)involvement.Envonalkib,a novel ALK inhibitor, demonstrated promising anti-tumor activity and safety in advanced ALK-positive NSCLC in the first-in-human phase I study. This phase III trial(ClinicalTrials.gov NCT04009317)investigated the efficacy and safety of first-line envonalkib in advanced ALK-positive NSCLC cases.Totally 264 participants were randomized 1:1 to receive envonalkib(n=131)or crizotinib(n=133).Median independent review committee(IRC)-assessed progression-free survival(PFS)times were 24.87(95%confidence interval[CI]:15.64–30.36)and 11.60(95%CI:8.28–13.73)months in the envonalkib and crizotinib groups, respectively(hazard ratio[HR]=0.47,95%CI:0.34–0.64,p<0.0001).IRC-assessed confirmed objective response rate(ORR)was higher(81.68%vs.70.68%,p=0.056)and duration of response was longer(median,25.79[95%CI,16.53–29.47]vs.11.14[95%CI,9.23–16.59]months,p=0.0003)in the envonalkib group compared with the crizotinib group. In participants with baseline brain target lesions,IRC-assessed CNS-ORR was improved with envonalkib compared with crizotinib(78.95%vs.23.81%).Overall survival(OS)data were immature, and median OS was not reached in either group(HR=0.84,95%CI:0.48–1.47,p=0.5741).The 12-month OS rates were 90.6%(95%CI,84.0%–94.5%)and 89.4%(95%CI,82.8%–93.6%)in the envonalkib and crizotinib groups,respectively.Grade≥3 treatment-related adverse events were observed in 55.73%and 42.86%of participants in the envonalkib and crizotinib groups, respectively.Envonalkib significantly improved PFS and delayed brain metastasis progression in advanced ALK-positive NSCLC. | Yunpeng Yang Jie Min Nong Yang Qitao Yu Ying Cheng Yanqiu Zhao Manxiang Li Hong Chen Shou’an Ren Jianying Zhou Wu Zhuang Xintian Qin Lejie Cao Yan Yu Jian Zhang Jianxing He Jifeng Feng Hao Yu Li Zhang Wenfeng Fang | 2023 | Signal Transduction and Targeted Therapy2023,8,9: | 0
VEGF-B prevents excessive angiogenesis by inhibiting FGF2/FGFR1 pathway显示文摘Although VEGF-B was discovered as a VEGF-A homolog a long time ago,the angiogenic effect of VEGF-B remains poorly understood with limited and diverse findings from different groups.Notwithstanding, drugs that inhibit VEGF-B together with other VEGF family members are being used to treat patients with various neovascular diseases. It is therefore critical to have a better understanding of the angiogenic effect of VEGF-B and the underlying mechanisms.Using comprehensive in vitro and in vivo methods and models,we reveal here for the first time an unexpected and surprising function of VEGF-B as an endogenous inhibitor of angiogenesis by inhibiting the FGF2/FGFR1 pathway when the latter is abundantly expressed.Mechanistically,we unveil that VEGF-B binds to FGFR1,induces FGFR1/VEGFR1 complex formation, and suppresses FGF2-induced Erk activation, and inhibits FGF2-driven angiogenesis and tumor growth. Our work uncovers a previously unrecognized novel function of VEGF-B in tethering the FGF2/FGFR1 pathway.Given the anti-angiogenic nature of VEGF-B under conditions of high FGF2/FGFR1 levels, caution is warranted when modulating VEGF-B activity to treat neovascular diseases. | Chunsik Lee Rongyuan Chen Guangli Sun Xialin Liu Xianchai Lin Chang He Liying Xing Lixian Liu Lasse DJensen Anil Kumar Harald FLanger Xiangrong Ren Jianing Zhang Lijuan Huang Xiangke Yin JongKyong Kim Juanhua Zhu Guanqun Huang Jiani Li Weiwei Lu Wei Chen Juanxi Liu Jiaxin Hu Qihang Sun Weisi Lu Lekun Fang Shasha Wang Haiqing Kuang Yihan Zhang Geng Tian Jia Mi Bi-Ang Kang Masashi Narazaki Aaron Prodeus Luc Schoonjans David MOrnitz Jean Gariepy Guy Eelen Mieke Dewerchin Yunlong Yang Jing-Song Ou Antonio Mora Jin Yao Chen Zhao Yizhi Liu Peter Carmeliet Yihai Cao Xuri Li | 2023 | Signal Transduction and Targeted Therapy2023,8,9: | 0
PIM2 regulates stemness through phosphorylation of 4E-BP1显示文摘Embryonic stem cells(ESCs) can undergo unlimited self-renewal and maintain pluripotency to differentiate into any cell type of the three germ layers. Extensive studies have shown ESC identity is regulated by transcription factors, epigenetic regulators and multiple signal transduction pathways. However, the kinase regulation of pluripotency is not well understood. Here we show that the serine/threonine kinase PIM2, which is highly expressed in ESCs but not in somatic cells, functions as a crucial stemness regulator in ESCs. Knockout of Pim2 inhibits the self-renewal and differentiation capability of ESCs. Mechanistic studies identified that PIM2 can directly phosphorylate 4E-BP1, leading to release of e IF4E which facilitates the translation of pluripotent genes in ESCs. Our study highlights a novel kinase cascade pathway for ESC identity maintenance. | Hongyan Sun Jiani Cao Lin Zhao Shaohua Zhu Shenghui Chen Yaqiong Li Bin Zhao Tongbiao Zhao | 2017 | Science Bulletin2017,62,10: | 0

Differential effects of recombinant fusion proteins TAT-OCT4 and TAT-NANOG on adult human fibroblasts显示文摘OCT4 and NANOG are two important tran-scription factors for maintaining the pluripotency and self-renewal abilities of embryonic stem(ES)cells.Meanwhile they play key roles in the induced pluripotent stem(iPS)cells. In this study, recombinant transcript factors TAT-NANOG and TAT-OCT4,which contained a fused powerful protein transduction domain(PTD)TAT from human immunodeficiency virus(HIV),were produced. Each fusion protein could be transported into human adultfibroblasts(HAF)successfully and activated the endogen-ous transcription of both nanog and oct4.Our study revealed the inter-regulation and autoregulation abilities of solo oct4 or nanog in the process of iPS cell reprogram-ming.Meanwhile the transduction of TAT-NANOG could accelerate the growth rate of HAF cells, and the key cell cycle regulator cdc25a was up-regulated.Thus cdc25a may be involved in the regulation of cell growth by NANOG.In addition, the TAT fusion protein technology provided a novel way to improve cell growth that is more controllable and safer. | Jiani CAO Zhifeng XIAO Bing CHEN Yuan GAO Chunying SHI Jinhuan WANG Jianwu DAI | 2010 | Frontiers in Biology2010,5,5: | 0
