| 2 | Regulation of non-classical immune parameters in immune thrombocytopenic purpura mice by a spleen-invigorating,qi-replenishing and blood-containing formula显示文摘Objective:This study investigated the regulatory effect of non-classical immune parameters on immune thrombocytopenic purpura(ITP)mice by a spleen-invigorating,qi-replenishing and blood-containing formula(SQBF).Method:A total of 80 BALB/c mice were randomly divided into four equal groups(20 mice each):control group,model group,prednisone group and spleen-invigorating,qi-replenishing and blood-containing(SQBF)group.Mice in the model group,prednisone group,and SQBF group were administered anti-platelet serum to induce ITP.The dynamic variations of platelet counts in ITP mice were measured with an automatic blood analyzer before modeling and 48 h,and 8,12 and 15 days following APS injection.Levels of b-endorphin(b-EP),vasoactive intestinal peptide(VIP)and salivary IgA(SIgA)were detected by enzyme-linked immunosorbent assay(ELISA)on 15th day of experiment.Results:SQBF enhanced peripheral blood platelet counts in ITP mice similar to that of prednisone,and both groups showed a statistically significant response compared with the model group(P<.01).The SQBF significantly decreased b-EP levels compared with the model and prednisone intervention groups(P<.05),significantly increased the levels of VIP and SIgA in ITP mice compared with the model group(P<.05)and had significant protective effects on the thymus of ITP mice compared with the model group(P<.01).Conclusions:The SQBF had a similar effect to prednisone with regards to enhancing peripheral blood platelet counts in ITP mice.Furthermore,it decreased b-EP levels and increased VIP and SIgA,and protected the thymus.This shows that,on base of the brain-gut axis functions,some non-classical immune vascular active factors or neurotransmitters are also involved in immune responses,and also have relationship with the onset of ITP and bleeding and/or hemostasis.It needs further study to determine whether a change in these active factors is related to immediate hemostasis. | Tiantian Li Hao He Li Hou Yue Xu Xiaoyong Wu Yanping Sun Lei Zheng Zhixiong Chen Xinyi Chen Bei Qin | 2015 | Journal of Traditional Chinese Medical Sciences2015,2,2: | 6 |

| 4 | Clinical Predictors of Functional Cure in Children 1–6 Years-old with Chronic Hepatitis B显示文摘Background and Aims:Hepatitis B surface antigen(HBsAg)clearance is significantly more common in children with chronic hepatitis B(CHB)than in adults;however,the possible influencing factors related to HBsAg loss have yet to be found.This study aimed to explore the efficacy of long-term interferon(IFN)αtherapy in treating children with CHB and analyzed the factors influencing functional cure after treatment.Methods:A total of 236 children aged 1–6 years and diagnosed with CHB via liver biopsy were included in the study,all receiving IFNαtreatment(IFNα-2b monotherapy,IFNα-2b followed by lamivudine[LAM]or IFNα-2b combined with LAM)and followed up for 144 weeks.A comprehensive analysis was conducted on clinical data,including biochemical items,serum markers of hepatitis B virus(HBV)and immunological indexes,and logistic regression analysis was used to screen the influencing factors related to HBsAg loss.Results:The cumulative loss rates of HBsAg were 79.5%,62.1%and 42.1%at 144 weeks after the start of treatment in the 1–3 years-old group,3–5 years-old group and 5–7 years-old group,respectively(p<0.05).IFNα-2b combined with LAM treatment displayed the highest HBsAg loss rates compared with monotherapy and sequential treatment(p=0.011).Younger baseline age and lower HBsAg levels were independent factors for the prediction of HBsAg loss(p<0.05).The baseline PreS1 and hepatitis B core antibody levels in the HBsAg loss group were lower than those in the HBsAg non-loss group.In addition,the PreS1 level was positively corelated with the level of HBsAg,HBV DNA and liver inflammation.Conclusions:Long-term treatment with IFNαwas effective in achieving HBsAg loss in CHB children aged 1–6 years-old.Age less than 3 years-old and lower HBsAg levels are independent predictors of functional cure in children with CHB. | Jing Pan Haiyan Wang Tiantian Yao Xuejiao Liao Hao Cheng Suthat Liangpunsakul Yan Wang Min Zhang Zheng Zhang | 2022 | Journal of Clinical and Translational Hepatology2022,10,3: | 3 |

| 9 | An Adversarial Smart Contract Honeypot in Ethereum显示文摘A smart contract honeypot is a special type of smart contract.This type of contract seems to have obvious vulnerabilities in contract design.If a user transfers a certain amount of funds to the contract,then the user can withdraw the funds in the contract.However,once users try to take advantage of this seemingly obvious vulnerability,they will fall into a real trap.Consequently,the user’s investment in the contract cannot be retrieved.The honeypot induces other accounts to launch funds,which seriously threatens the security of property on the blockchain.Detection methods for honeypots are available.However,studying the manner by which to defend existing honeypots is insufficient to fight against honeypots.The new honeypots that may appear in the future from the perspective of an attacker must also be predicted.Therefore,we propose a type of adversarial honeypot.The code and behavioral features of honeypots are obtained through a comparative analysis of the 158,568 nonhoneypots and 352 honeypots.To build an adversarial honeypot,we try to separately hide these features and make the honeypot bypass the existing detection technology.We construct 18 instances on the basis of the proposed adversarial honeypot and use an open-source honeypot detection tool to detect these instances.The experimental result shows that the proposed honeypot can bypass the detection tool with a 100%ratio.Therefore,this type of honeypot should be given attention,and defensive measures should be proposed as soon as possible. | Yu Han Tiantian Ji Zhongru Wang Hao Liu Hai Jiang Wendi Wang Xiang Cui | 2021 | Computer Modeling in Engineering & Sciences2021,,7: | 1 |

| 14 | Detection and classification of breast lesions using multiple information on contrast-enhanced mammography by a multiprocess deep-learning system: A multicenter study显示文摘Objective: Accurate detection and classification of breast lesions in early stage is crucial to timely formulate effective treatments for patients. We aim to develop a fully automatic system to detect and classify breast lesions using multiple contrast-enhanced mammography(CEM) images.Methods: In this study, a total of 1,903 females who underwent CEM examination from three hospitals were enrolled as the training set, internal testing set, pooled external testing set and prospective testing set. Here we developed a CEM-based multiprocess detection and classification system(MDCS) to perform the task of detection and classification of breast lesions. In this system, we introduced an innovative auxiliary feature fusion(AFF)algorithm that could intelligently incorporates multiple types of information from CEM images. The average freeresponse receiver operating characteristic score(AFROC-Score) was presented to validate system’s detection performance, and the performance of classification was evaluated by area under the receiver operating characteristic curve(AUC). Furthermore, we assessed the diagnostic value of MDCS through visual analysis of disputed cases,comparing its performance and efficiency with that of radiologists and exploring whether it could augment radiologists’ performance.Results: On the pooled external and prospective testing sets, MDCS always maintained a high standalone performance, with AFROC-Scores of 0.953 and 0.963 for detection task, and AUCs for classification were 0.909[95% confidence interval(95% CI): 0.822-0.996] and 0.912(95% CI: 0.840-0.985), respectively. It also achieved higher sensitivity than all senior radiologists and higher specificity than all junior radiologists on pooled external and prospective testing sets. Moreover, MDCS performed superior diagnostic efficiency with an average reading time of 5 seconds, compared to the radiologists’ average reading time of 3.2 min. | 16 | Evaluation of an innovative pediatric isolation(PI)bed using fluid dynamics simulation and aerosol isolation efficacy显示文摘Airborne transmission is an important mechanism of spread for both viruses and bacteria in hospitals,with nosocomial infections putting a great burden on public health.In this study,we designed and manufactured a bed for pediatric clinic consultation rooms providing air isolation to protect patients and medical personnel from pathogen transmission.The pediatric isolation bed has several primary efficiency filters and a high-efficiency particulate air filter in the bedside unit.The air circulation between inlet and outlet forms negative pressure to remove the patient's exhaled air timeously and effectively.A computational fluid dynamics model was used to calculate the speed of the airflow and the angle of sampler.Following this,we conducted purification experiments using cigarette smoke,Staphylococcus albus(S.albus)and human adenovirus type 5(HAdV-5)to demonstrate the isolation efficacy.The results showed that the patient's head should be placed as close to the air inlet hood as possible,and an air intake wind speed of 0.86 m/s was effective.The isolation efficacy of the pediatric isolation bed was demonstrated by computational fluid dynamics technology.The isolation efficiency against cigarette smoke exceeded 91.8%,and against S.albus was greater than 99.8%,while the isolation efficiency against HAdV-5 was 100%.The pediatric isolation bed could be used where isolation wards are unavailable,such as in intensive care units and primary clinical settings,to control hospital acquired infections. | Tiantian Liu Yubing Guo Xiaotang Hao Mei Wang Shicong He Zhengshi Lin Rong Zhou | 2021 | Building Simulation2021,14,5: | 0 |

| 18 | Autoinhibitory structure of preligand association state implicates a new strategy to attain effective DR5 receptor activation显示文摘Members of the tumor necrosis factor receptor superfamily(TNFRSF)are important therapeutic targets that can be activated to induce death of cancer cells or stimulate proliferation of immune cells.Although it has long been implicated that these receptors assemble preligand associated states that are required for dominant interference in human disease,such states have so far eluded structural characterization.Here,we find that the ectodomain of death receptor 5(DR5-ECD),a representative member of TNFRSF,can specifically self-associate when anchored to lipid bilayer,and we report this self-association structure determined by nuclear magnetic resonance(NMR).Unexpectedly,two non-overlapping interaction interfaces are identified that could propagate to higher-order clusters.Structure-guided mutagenesis indicates that the observed preligand association structure is represented on DR5-expressing cells.The DR5 preligand association serves an autoinhibitory role as single-domain antibodies(sdAbs)that partially dissociate the preligand cluster can sensitize the receptor to its ligand TRAIL and even induce substantial receptor signaling in the absence of TRAIL.Unlike most agonistic antibodies that require multivalent binding to aggregate receptors for activation,these agonistic sdAbs are monovalent and act specifically on an oligomeric,autoinhibitory configuration of the receptor.Our data indicate that receptors such as DR5 can form structurally defined preclusters incompatible with signaling and that true agonists should disrupt the preligand cluster while converting it to signaling-productive cluster.This mechanism enhances our understanding of a long-standing question in TNFRSF signaling and suggests a new opportunity for developing agonistic molecules by targeting receptor preligand clustering. | Gang Du Linlin Zhao Yumei Zheng Anissa Belfetmi Tiantian Cai Boying Xu Karen Heyninck Kim Van Den Heede Marie-Ange Buyse Pietro Fontana Michael Bowman Lih-Ling Lin Hao Wu James Jeiwen Chou | 2023 | Cell Research2023,33,2: | 0 |
| 19 | GRK2 mediated degradation of SAV1 initiates hyperplasia of fibroblast-like synoviocytes in rheumatoid arthritis显示文摘Hyperplasia and migration of fibroblast-like synoviocytes(FLSs)are the key drivers in the pathogenesis of rheumatoid arthritis(RA)and joint destruction.Abundant Yes-associated protein(YAP),which is a powerful transcription co-activator for proliferative genes,was observed in the nucleus of inflammatory FLSs with unknown upstream mechanisms.Using Gene Expression Omnibus database analysis,it was found that Salvador homolog-1(SAV1),the pivotal negative regulator of the Hippo-YAP pathway,was slightly downregulated in RA synovium.However,SAV1 protein expression is extremely reduced.Subsequently,it was revealed that SAV1 is phosphorylated,ubiquitinated,and degraded by interacting with an important serine-threonine kinase,G protein-coupled receptor(GPCR)kinase 2(GRK2),which was predominately upregulated by GPCR activation induced by ligands such as prostaglandin E2(PGE2)in RA.This process further contributes to the decreased phosphorylation,nuclear translocation,and transcriptional potency of YAP,and leads to aberrant FLSs proliferation.Genetic depletion of GRK2 or inhibition of GRK2 by paroxetine rescued SAV1 expression and restored YAP phosphorylation and finally inhibited RA FLSs proliferation and migration.Similarly,paroxetine treatment effectively reduced the abnormal proliferation of FLSs in a rat model of collagen-induced arthritis which was accompanied by a significant improvement in clinical manifestations.Collectively,these results elucidate the significance of GRK2 regulation of Hippo-YAP signaling in FLSs proliferation and migration and the potential application of GRK2 inhibition in the treatment of FLSs-driven joint destruction in RA. | Paipai Guo Ji Jiang Rui Chu Feng He Mingli Ge Ruhong Fang Qiuyun Guan Huijuan Cheng Chunru Jiang Tiantian Su Zhenduo Zhu Hao Liu Wei Wei Shihao Zhang Qingtong Wang | 2024 | Acta Pharmaceutica Sinica B2024,14,3: | 0 |
