1Genome-wide CRISPR screen identifies synthetic lethality between DOCK1 inhibition and metformin in liver cancer显示文摘Metformin is currently a strong candidate anti-tumor agent in multiple cancers.However,its anti-tumor effectiveness varies among different cancers or sub-populations,potentially due to tumor heterogeneity.It thus remains unclear which hepatocellular carcinoma(HCC)patient subpopulation(s)can benefit from met-formin treatment.Here,through a genome-wide CRISPR-Cas9-based knockout screen,we find that DOCK1 levels determine the anti-tumor effects of met-formin and that DOCK1 is a synthetic lethal target of metformin in HCC.Mechanistically,metformin promotes DOCK1 phosphorylation,which activates RAC1 to facilitate cell survival,leading to metformin resistance.The DOCK1-selective inhibitor,TBOPP,potentiates anti-tumor activity by metformin in vitro in liver cancer cell lines and patient-derived HCC organoids,and in vivo in xenografted liver cancer cells and immunocompetent mouse liver cancer models.Notably,metformin improves overall survival of HCC patients with low DOCK1 levels but not among patients with high DOCK1 expression.This study shows that metformin effective-ness depends on DOCK1 levels and that combining metformin with DOCK1 inhibition may provide a promising personalized therapeutic strategy for met-formin-resistant HCC patients.Junru Feng Hui Lu Wenhao Ma Wenjing Tian Zhuan Lu Hongying Yang Yongping Cai Pengfei Cai Yuchen Sun Zilong Zhou Jiaqian Feng Jiazhong Deng Ying Shu Kun Qu Weidong Jia Ping Gao Huafeng Zhang 2022Protein & Cell2022,13,11:5

5China's foreign aid for global poverty alleviation:artemisinin-based combination therapies against malaria in Togo显示文摘From providing funds for the global fight against infectious diseases,to actively participating in global health security actions,to strengthening mutual cooperation in the field of health,and providing medical treatment,training and scholarships to countries in need,China’s foreign aid on global poverty alleviation is increasingly diversified and expanding in scale.Indeed,China is playing an increasingly important leading role in the global health agenda.It is worth mentioning that over the years,artemisinin compound have saved millions of lives all over the world,especially in poverty-stricken areas.China’s work mode of malaria elimination has also been written into WHO’s technical documents and recommended to other countries.Since 2007,Chinese medical staff has carried out the Artemisinin Compound Malaria Control Project in Comoros,bringing Chinese prevention and treatment programs to the local area.By 2014,Comoros had achieved zero deaths from malaria,and the number of cases had dropped by 98%.Now,this program is also extended to Togo,another African country.This article preliminarily summarizes the malaria profile in Togo and introduces China-Togo Cooperative Artemisinin Malaria Control Demonstration Project to provide a reference for better anti-malaria assistance in Togo,and also shows one of the substantive actions of China’s participation in global health governance,which contributes Chinese wisdom and offers Chinese solutions to global poverty alleviation.Qi Wang Chuyi Yu Hongying Zhang Shaoqin Zheng Jianping Song Changsheng Deng 2021Global Health Journal2021,5,3:3

13Cable-driven legged landing gear for unmanned helicopter:Prototype design,optimization and performance assessment显示文摘Unmanned helicopters equipped with adaptive landing gear will dramatically extend their application especially in dealing with challenging terrains.This study presents a novel cable-driven legged landing gear(CLG)with differential transmission for unmanned helicopters in complex landing environments.To obtain the preferred configuration of the legged mechanism,a multi-objective optimization framework for the CLG is constructed by concurrently considering terrain adaptability,landing stability and reasonable linkage of internal forces.The non-dominated sorting genetic algorithmⅡis employed to numerically acquire the optimal scale parameters that guide the mechanical design of the CLG.An unmanned helicopter prototype equipped with the devised CLG is developed with key performance assessment.Experimental results show that the devised CLG can provide energy-efficient support over uneven terrains(totally driven torque demand less than 0.1 N m)in quasi-static landing tests,and favorable terrain adaptability(posture fluctuation of the fuselage less than±1°)in unknown slope landing tests.These exhibited merits give the proposed CLG the potential to enhance the landing performance of future aircraft in extreme environments.TIAN BaoLin GAO HaiBo YU HaiTao SHAN HaoMin HOU JunChen YU HongYing DENG ZongQuan 2024Science China(Technological Sciences)2024,67,4:0
