2Impact of particle size and pH on protein corona formation of solid lipid nanoparticles:A proof-of-concept study显示文摘When nanoparticles were introduced into the biological media,the protein corona would be formed,which endowed the nanoparticles with new bio-identities.Thus,controlling protein corona formation is critical to in vivo therapeutic effect.Controlling the particle size is the most feasible method during design,and the infuence of media pH which varies with disease condition is quite important.The impact of particle size and pH on bovine serum albumin(BSA)corona formation of solid lipid nanoparticles(SLNs)was studied here.The BSA corona formation of SLNs with increasing particle size(120-480 nm)in pH 6.0 and 7.4 was investigated.Multiple techniques were employed for visualization study,conformational structure study and mechanism study,etc.'BSA corona-caused aggregation'of SLN2-3 was revealed in pH 6.0 while the dispersed state of SLNs was maintained in pH 7.4,which signifcantly affected the secondary structure of BSA and cell uptake of SLNs.The main interaction was driven by van der Waals force plus hydrogen bonding in p H 7.4,while by electrostatic attraction in pH 6.0,and size-dependent adsorption was confrmed.This study provides a systematic insight to the understanding of protein corona formation of SLNs.Wenhao Wang Zhengwei Huang Yanbei Li Wenhua Wang Jiayu Shi Fangqin Fu Ying Huang Xin Pan Chuanbin Wu 2021Acta Pharmaceutica Sinica B2021,11,4:3

4SOD1^(G93A) Induces a Unique PSAP-Dependent Mitochondrial Apoptosis Pathway via Bax-Bak Interaction显示文摘Amyotrophic lateral syndrome(ALS)is a progressive degenerative disorder characterized by motor neuron death and axon degeneration.Mitochondrial dysfunction plays a key role in the pathogenesis of ALS,the mechanism of which remains poorly understood.The B-cell lymphoma-2(Bcl-2)family of proteins that control and mediate mitochondrial function and apoptosis,including the pro-apoptotic members Bcl2-Associated X(Bax),are involved in ALS development.The death receptor 6(DR6)regulates motor neuron death in ALS,and DR6 antibodies can prevent axon degeneration and motor neuron damage by blocking DR6.Previous studies demonstrated that PSAP localized to mitochondria and was required for DR6-induced apoptosis.In this study,SOD1^(G93A) was transfected into the motor neuron cell line NSC-34 to serve as an ALS cell model in vitro.The data assessed the role of PSAP in SOD1^(G93A)induced apoptosis and demonstrated that the overexpression of SOD1^(G93A),but not wtSOD1,induced PARP cleavage,caspase-3 activation,cytochrome c release,and Bax translocation.PSAP,Bax,and Bak were necessary for SOD1^(G93A)induced apoptosis,as silencing PSAP inhibited SOD1^(G93A)-mediated cell death that was dependent on Bax-Bak interaction.HAN NIU XIN CHEN XUEQI FU JINGTIAN ZHANG GUODONG LI YUXIANG WANG JIAYUE SONG XUETING MA CHEN HU XUEMIN XU FUQIANG ZHANG LINLIN ZENG 2021BIOCELL2021,45,4:2
5Active oxygen species and oxidation mechanism over Ce-doped LaMn_(0.8)Ni_(0.2)O_(3)/hierarchical ZSM-5 in pentanal oxidation显示文摘Hierarchical ZSM-5(HZ)molecular sieves based on fly ash were synthesized using a method combining water heat treatment with step-by-step calcination.The coupling catalysts between La_(1-x)Ce_(x)Mn_(0.8)-Ni_(0.2)O_(3)(x≤0.5)perovskites and HZ were prepared through the impregnation method,which were characterized by X-ray diffraction(XRD),scanning electron microscopy(SEM),high-resolution transmission electron microscopy(HRTEM),N_(2)adsorption,X-ray photoelectron spectroscopy(XPS),NH3-temperature programmed desoprtion(NH3-TPD),H_(2)-temperature programmed reduction(H_(2)-TPR)and O_(2)-TPD techniques and investigated regarding pentanal oxidation at 120-390℃to explore the effects of Ce doping on the catalytic activity and the active oxygen species of the coupling catalysts,meanwhile,the reaction mechanism and pathway of pentanal oxidation were also studied.The results reveal that Ce substitution at La sites can change the electronic interactions between all the elements and promote the electronic transfer among La,Ce,Ni,Mn and HZ,influencing directly the physicochemical characteristics of the catalysts.Moreover,the amount and transfer ability of surface adsorbed oxygen(O_(2)-and O-)regarded as the reactive oxygen species and the low temperature reducibility are the main influence factors in pentanal oxidation.Additionally,La_(0.8)Ce_(0.2)Mn_(0.8)Ni_(0.2)O_(3)/HZ exhibits the best catalytic activity and deep oxidation capacity as well as a better water resistance due to its larger amount of surface adsorbed oxygen species and higher low temperature reducibility.What’s more,appropriate Ce substitution can significantly enhance the amount of O_(2)-ions,which can distinctly enhance the catalytic activity of the catalyst,and moderate acid strength and appropriate acid amount can also facilitate the improvement of the pentanal oxidation activity.It is found that there is a synergic catalytic effect between surface acidity and redox ability of the catalyst.According to the in situ DRIFTS and GC/MS analyses,pentanal can be oxidized gradually to CO_(2)and H_(2)O by the surface oxygen species with the form of adsorption in air following the Langmuir-Hinshelwood(L-H)reaction mechanism.Two reaction pathways for the pentanal oxidation process are proposed,and the conversion of the formates to carbonates may be one of the main rate-determining steps.Jian Li Yingjie Shi Xiaoheng Fu Yun Shu Jiayu Huang Jinwei Zhu Gang Tian Jingnan Hu 2021Journal of Rare Earths2021,39,9:2
6The BZR1-EDS1 module regulates plant growth-defense coordination显示文摘Plants have developed sophisticated strategies to coordinate growth and immunity,but our understanding of the underlying mechanism remains limited.In this study,we identified a novel molecular module that reg-ulates plant growth and defense in both compatible and incompatible infections.This module consisted of BZR1,a key transcription factor in brassinosteroid(BR)signaling,and EDS1,an essential positive regulator of plant innate immunity.We found that EDS1 interacts with BZR1 and suppresses its transcriptional activ-ities.Consistently,upregulation of EDS1 function by a virulent Pseudomonas syringae strain or salicylic acid treatment inhibited BZR1-regulated expression of BR-responsive genes and BR-promoted growth.Furthermore,we showed that the cytoplasmic fraction of BZR1 positively regulates effector-triggered im-munity(ETI)controlled by the TIR-NB-LRR protein RPS4,which is attenuated by BZR1's nuclear transloca-tion.Mechanistically,cytoplasmic BZR1 facilitated AvrRps4-triggered dissociation of EDS1 and RPS4 by binding to EDS1,thus leading to efficient activation of RPS4-controlled ETI.Notably,transgenic expression of a mutant BZR1 that accumulates exclusively in the cytoplasm improved pathogen resistance without compromising plant growth.Collectively,these results shed new light on plant growth-defense coordina-tion and reveal a previously unknown function for the cytoplasmic fraction of BZR1.The BZR1-EDS1 mod-ule may be harnessed for the simultaneous improvement of crop productivity and pathogen resistance.Guang Qi Huan Chen Dian Wang Hongyuan Zheng Xianfeng Tang Zhengzheng Guo Jiayu Cheng Jian Chen Yiping Wang Ming-yi Bai Fengquan Liu Daowen Wang Zheng Qing Fu 2021Molecular Plant2021,14,12:1
7The genetic control of glabrous glume during African rice domestication显示文摘African cultivated rice,Oryza glaberrima,is characterized by its glabrous glumes.During domestication,the pubescent glumes of its wild ancestor,Oryza barthii,lost their trichomes,and in this study,we show that glabrous glume 5(GLAG5),a WUSCHEL-like homeobox transcription factor gene on chromosome 5,is required for trichome development.DNA methylation associated with an hATtransposable element inserted in the promoter region of GLAG5 is found to reduce its expression,leading to the formation of glabrous glumes and leaves in African cultivated rice.Among 82 African cultivated rice varieties investigated in this study,59(approximately 71%)lines exhibit glabrous glumes and harbor the hAT transposon;however,the other 23 varieties(approximately 29%),which exhibit pubescent glumes,lack the hAT transposon,indicating that glag5 had undergone strong artificial selection.Theπ;/π;ratios also show the hAT transposon insertions influence the genetic diversity of an approximately 150-kb interval encompassing the GLAG5 locus.The identification of the GLAG5 gene provides new insights into the domestication of cultivated rice in Africa.We speculate that the selection of varieties with mutations in their promoter regions is an important aspect of crop domestication.Leqin Chang Min Hu Jing Ning Wei He Jiayu Gao Marie-Noelle Ndjiondjop Yongcai Fu Fengxia Liu Hongying Sun Ping Gu Chuanqing Sun Zuofeng Zhu 2022Journal of Genetics and Genomics2022,49,5:1

11Development of a radiolabeled site-specific single-domain antibody positron emission tomography probe for monitoring PD-L1 expression in cancer显示文摘Despite advances in immunotherapy for the treatment of cancers,not all patients can benefit from programmed cell death ligand 1(PD-L1)immune checkpoint blockade therapy.Anti-PD-L1 therapeutic effects reportedly correlate with the PD-L1 expression level;hence,accurate detection of PD-L1 expression can guide immunotherapy to achieve better therapeutic effects.Therefore,based on the high affinity antibody Nb109,a new site-specifically radiolabeled tracer,^(68)Ga-NODA-cysteine,aspartic acid,and valine(CDV)-Nb109,was designed and synthesized to accurately monitor PD-L1 expression.The tracer ^(68)Ga-NODA-CDV-Nb109 was obtained using a site-specific conjugation strategy with a radiochemical yield of about 95%and radiochemical purity of 97%.It showed high affinity for PD-L1 with a dissociation constant of 12.34±1.65 nM.Both the cell uptake assay and positron emission tomography(PET)imaging revealed higher tracer uptake in PD-L1-positive A375-hPD-L1 and U87 tumor cells than in PD-L1-negative A375 tumor cells.Meanwhile,dynamic PET imaging of a NCI-H1299 xenograft indicated that doxorubicin could upregulate PD-L1 expression,allowing timely interventional immunotherapy.In conclusion,this tracer could sensitively and dynamically monitor changes in PD-L1 expression levels in different cancers and help screen patients who can benefit from anti-PD-L1 immunotherapy.Yinfei Chen Shiyu Zhu Jiayu Fu Jianguo Lin Yan Sun Gaochao Lv Minhao Xie Tao Xu Ling Qiu 2022Journal of Pharmaceutical Analysis2022,12,6:0

13Themis suppresses the effector function of CD8^(+)T cells in acute viral infection显示文摘CD8^(+)T cells play a central role in antiviral immune responses.Upon infection,naive CD8^(+)T cells differentiate into effector cells to eliminate virus-infected cells,and some of these effector cells further differentiate into memory cells to provide long-term protection after infection is resolved.Although extensively investigated,the underlying mechanisms of CD+T-cell differentiation remain incompletely understood.Themis is a T-cell-specific protein that plays critical roles in T-cell development.Recent studies using Themis T-cell conditional knockout mice also demonstrated that Themis is required to promote mature CD8^(+)T-cell homeostasis,cytokine responsiveness,and antibacterial responses.In this study,we used LCMV Armstrong infection as a probe to explore the role of Themis in viral infection.We found that preexisting CD8^(+)T-cell homeostasis defects and cytokine hyporesponsiveness do not impair viral clearance in Themis T-cell conditional knockout mice.Further analyses showed that in the primary immune response,Themis deficiency promoted the differentiation of CD8^(+)effector cells and increased their TNF and IFNy production.Moreover,Themis deficiency impaired memory precursor cell(MPEC)differentiation but promoted short-lived effector cell(SLEC)differentiation.Themis deficiency also enhanced effector cytokine production in memory CD8^(+)T cells while impairing central memory CD8^(+)T-cell formation.Mechanistically,we found that Themis mediates PD-1 expression and its signaling in effector CD8^(+)T cells,which explains the elevated cytokine production in these cells when Themis is disrupted.Jian Tang Xian Jia Jian Li Junchen Dong Jiayu Wang Wanyun Li Yuzhen Zhu Yanyan Hu Bowen Hou Chunjie Lin Yu Cong Tong Ren Changsheng Yan Hongying Yang Qian Lai Haiping Zheng Yuzhou Bao Namrata Gautam Hong-Rui Wang Bing Xu Xiao Lei Chen Qing Li Nicholas R.J.Gascoigne Guo Fu 2023Cellular & Molecular Immunology2023,20,5:0
14A Survey of the Regional Language Landscape of Uyghur Language Use-Take Urumqi Metro as an Example显示文摘Uyghur is generally used in the Xinjiang Uygur Autonomous Region,so the Uyghur language used in this topic refers to the Xinjiang Uygur Autonomous Region.Through the investigation of Metro Line 1,this paper analyzes from the perspectives of language code orientation,language harmony,consensus comparison,and questionnaires.It is found that Urumqi Metro Line 1 is mainly composed of Chinese(in this paper,when we talk about Chinese which means mandarin,the national language in China)and Uygur languages.Chinese occupies a dominant position in multi-language signage,Uygur language is relatively inferior,and English is relatively weak.And other national languages do not appear on all kinds of signs.Through the investigation and research on the language landscape of Line 1,this paper puts forward the urban language usage in Urumqi.Some suggestions contribute to the construction of beautiful Urumqi.FU Yi SHEN Jiayu 2022US-China Foreign Language2022,20,6:0
15Pomalidomide improves the function of CD133-or HER2-specific CAR T cells显示文摘Chimeric antigen receptor(CAR)T-cell therapy is mostly limited to hematological malignancies and has a poor effect on solid tumors.CAR T cells as a kind of immune cell may be affected by some immunomodulatory drugs such as pomalidomide,so the use of pomalidomide may improve the effect of CAR T cells on solid tumors.In this study,CD133-or HER2-specific CAR T cells were chosen to investigate whether pomalidomide can regulate the function of CAR T cells in vitro.We found that pomalidomide can significantly enhance the ability of CD133-CAR T cells and HER2-CAR T cells to kill tumor cells and increase the cytokine secretion of CD133-CAR T cells and HER2-CAR T cells.Also,pomalidomide was shown to induce down-regulation of protein levels of IL-2 transcriptional repressors Aiolos and Ikaros in CAR T cells.This study suggests that the combination of pomalidomide and CAR T cells may be a new strategy for the treatment of solid tumors.ZHIXIONG WANG NA RISU JIAYU FU HUI LIU GUOMIN ZHOU QIAN LIU YAN ZOU JIAXING TANG LONG LI XUEKAI ZHU 2021BIOCELL2021,45,1:0
16Genomic analysis of indigenous goats in Southwest Asia reveals evidence of ancient adaptive introgression related to desert climate显示文摘Understanding how evolutionary pressures related to climate change have shaped the current genetic background of domestic animals is a fundamental pursuit of biology. Here, we generated wholegenome sequencing data from native goat populations in Iraq and Pakistan. Combined with previously published data on modern, ancient(Late Neolithic to Medieval periods), and wild Capra species worldwide, we explored the genetic population structure, ancestry components, and signatures of natural positive selection in native goat populations in Southwest Asia(SWA). Results revealed that the genetic structure of SWA goats was deeply influenced by gene flow from the eastern Mediterranean during the Chalcolithic period, which may reflect adaptation to gradual warming and aridity in the region. Furthermore, comparative genomic analysis revealed adaptive introgression of the KITLG locus from the Nubian ibex(C. nubiana) into African and SWA goats. The frequency of the selected allele at this locus was significantly higher among goat populations located near northeastern Africa. 18Effects of Shenlian formula(参连方)on microbiota and inflammatory cytokines in adults with type 2 diabetes:a double-blind randomized clinical trial显示文摘OBJECTIVES:To observe the efficacy of Shenlian formula(SL formula,参连方),which consist of Huanglian(Rhizoma Coptidis)and Renshen(Radix Ginseng),in the treatment of type 2 diabetes mellitus(T2DM)and explore the effects on gut microbiota and serum inflammatory cytokines.METHODS:In a double-blind,randomized,placebocontrolled parallel-group clinical trial,31 adults with T2DM were randomly allocated to receive the SL formula or placebo for 12 weeks.Body mass index(BMI),blood lipid indices,glycemic biomarkers including glycated hemoglobin(HbA1C),fasting plasma glucose(FPG),postprandial blood glucose(PBG),fasting insulin levels(FIL),fasting C-peptide(C-P),homoeostasis model assessment for insulin resistance(HOMA-IR)and inflammatory cytokines were assessed at baseline and 12 weeks.The contents of gut microbiota were determined by pyrosequencing of the V3-V4 regions of 16S rRNA genes.RESULTS:Sixteen cases were allocated in the treatment group and 15 in the placebo group.Compared with the placebo,SL formula resulted in a higher significant reduction in PBG[(-1.318±0.772)vs(-0.008±1.404)mmol/L,P=0.003],BMI[(-0.611±0.524)vs(0.957±2.212)kg/m2,P=0.01],FIL[(-1.627±6.268)vs(3.976±6.85)μIU/mL,P=0.02],HOMA-IR[(-0.530±1.461)vs(1.511±2.288),P=0.006],and C-reactive protein(CRP)[(-1.307±0.684)vs(0.828±0.557)mg/L,P=0.04].In terms of gut microbiota,compared with the placebo,the SL formula resulted in a significant decrease in species richness and evenness.CONCLUSIONS:The SL formula showed the efficacy to improve postprandial blood glucose,insulin resistance,BMI and CRP levels.In addition,it could reduce the total number,richness and evenness of species,meanwhile increase the abundance of probiotics to modulate the structure of gut microbiota in patients with T2DM.However,further studies are required for exploring the deeper mechanism of TCM on gut microbiota.JIANG Li FU Qiang WANG Shidong ZHAO Jinxi CHEN Yu LI Jiayue XIAO Yonghua HUANG Weijun SUN Ruixi XIAO Yao SHEN Aijia WANG Junheng LIU Jiangteng FU Xiaozhe LI Yuanyuan ZHAO Yu XUE Taiqi 2023Journal of Traditional Chinese Medicine2023,43,4:0
