1A 10-miRNA risk score-based prediction model for pathological complete response to neoadjuvant chemotherapy in hormone receptor-positive breast cancer显示文摘Patients with hormone receptor(HR)-positive tumors breast cancer usually experience a relatively low pathological complete response(p CR)to neoadjuvant chemotherapy(NAC).Here,we derived a 10-micro RNA risk score(10-mi RNA RS)-based model with better performance in the prediction of p CR and validated its relation with the disease-free survival(DFS)in 755 HRpositive breast cancer patients(273,265,and 217 in the training,internal,and external validation sets,respectively).This model,presented as a nomogram,included four parameters:the 10-mi RNA RS found in our previous study,progesterone receptor(PR),human epidermal growth factor receptor 2(HER2)status,and volume transfer constant(K).Favorable calibration and discrimination of 10-mi RNA RS-based model with areas under the curve(AUC)of 0.865,0.811,and 0.804 were shown in the training,internal,and external validation sets,respectively.Patients who have higher nomogram score(>92.2)with NAC treatment would have longer DFS(hazard ratio=0.57;95%CI:0.39–0.83;P=0.004).In summary,our data showed the 10-mi RNA RS-based model could precisely identify more patients who can attain p CR to NAC,which may help clinicians formulate the personalized initial treatment strategy and consequently achieves better clinical prognosis for patients with HRpositive breast cancer.Chang Gong Ziliang Cheng Yaping Yang Jun Shen Yingying Zhu Li Ling Wanyi Lin Zhigang Yu Zhihua Li Weige Tan Chushan Zheng Wenbo Zheng Jiajie Zhong Xiang Zhang Yunjie Zeng Qiang Liu RStephanie Huang Andrzej LKomorowski Eddy SYang François Bertucci Francesco Ricci Armando Orlandi Gianluca Franceschini Kazuaki Takabe Suzanne Klimberg Naohiro Ishii Angela Toss Mona PTan Mathew A Cherian Erwei Song 2022Science China(Life Sciences)2022,65,11:3
2Nanoscale monitoring of mitochondria and lysosome interactions for drug screening and discover显示文摘Technology advances in genomics,proteomics,and metabolomics largely expanded the pool of potential therapeutic targets.Compared with the in vitro setting,cell-based screening assays have been playing a key role in the processes of drug discovery and development.Besides the commonly used strategies based on colorimetric and cell viability,we reason that methods that capture the dynamic cellular events will facilitate optimal hit identification with high sensitivity and specificity.Herein,we propose a live-cell screening strategy using structured illumination microscopy (SIM) combined with an automated cell colocalization analysis software,CellprofilerTM,to screen and discover drugs for mitochondria and lysosomes interaction at a nanoscale resolution in living cells.This strategy quantitatively benchmarks the mitochondria-lysosome interactions such as mitochondria and lysosomes contact (MLC) and mitophagy.The automatic quantitative analysis also resolves fine changes of the mitochondria-lysosome interaction in response to genetic and pharmacological interventions.Super-resolution live-cell imaging on the basis of quantitative analysis opens up new avenues for drug screening and development by targeting dynamic organelle interactions at the nanoscale resolution,which could facilitate optimal hit identification and potentially shorten the cycle of drug discovery.Qixin Chen Xintian Shao Zhiqi Tian Yang Chen Payel Mondal Fei Liu Fengshan Wang Peixue Ling Weijiang He Kai Zhang Zijian Guo Jiajie Diao 2019Nano Research2019,12,5:2
3Super-resolution quantification of nanoscale damage to mitochondria in live cells显示文摘Mitochondrial damage,characterized by altered morphological distribution and the damage of cristae,is closely associated with mitochondrial disease.However,imaging methods for capturing mitochondrial morphology at the nanoscale level in live samples remain unavailable,which seriously hinders the accurate evaluation and diagnosis of mitochondrial-related diseases.In response,we propose a super-resolution quantification strategy based on structured illumination microscopy(SIM)for the rapid,accurate evaluation of mitochondrial morphology.Using the strategy,we accurately captured the morphological distribution of mitochondria at the nanoscale level in a way generally applicable to checking various cell processes and identifying patients with mitochondrial disease who exhibit the SLC25A46 mutation.We also used algorithm-assisted super-resolution imaging to quantitatively analyze damage to mitochondrial cristae,which supports a novel drug screening strategy—high-resolution drug screening—for investigating drugs’pharmacodynamics on organelles in living cells.In short,our strategy improves the accurate examination of changes in mitochondrial morphology in living cells and indicates new ways in which SIM-imaging can assist in diagnosing mitochondrial disease at the single-cell level.Xintian Shao Qixin Chen Lianting Hu Zhiqi Tian Liuyi Liu Fei Liu Fengshan Wang Peixue Ling Zong-Wan Mao Jiajie Diao 2020Nano Research2020,13,8:1

5Feasibility and physics potential of detecting ^(8)B solar neutrinos at JUNO显示文摘The Jiangmen Underground Neutrino Observatory(JUNO)features a 20 kt multi-purpose underground liquid scintillator sphere as its main detector.Some of JUNO's features make it an excellent location for^8B solar neutrino measurements,such as its low-energy threshold,high energy resolution compared with water Cherenkov detectors,and much larger target mass compared with previous liquid scintillator detectors.In this paper,we present a comprehensive assessment of JUNO's potential for detecting^8B solar neutrinos via the neutrino-electron elastic scattering process.A reduced 2 MeV threshold for the recoil electron energy is found to be achievable,assuming that the intrinsic radioactive background^(238)U and^(232)Th in the liquid scintillator can be controlled to 10^(-17)g/g.With ten years of data acquisition,approximately 60,000 signal and 30,000 background events are expected.This large sample will enable an examination of the distortion of the recoil electron spectrum that is dominated by the neutrino flavor transformation in the dense solar matter,which will shed new light on the inconsistency between the measured electron spectra and the predictions of the standard three-flavor neutrino oscillation framework.IfDelta m^(2)_(21)=4.8times10^(-5);(7.5times10^(-5))eV^(2),JUNO can provide evidence of neutrino oscillation in the Earth at approximately the 3sigma(2sigma)level by measuring the non-zero signal rate variation with respect to the solar zenith angle.Moreover,JUNO can simultaneously measureDelta m^2_(21)using^8B solar neutrinos to a precision of 20% or better,depending on the central value,and to sub-percent precision using reactor antineutrinos.A comparison of these two measurements from the same detector will help understand the current mild inconsistency between the value of Delta m^2_(21)reported by solar neutrino experiments and the KamLAND 9Thermal reliability analysis of the central detector of JUNO显示文摘Introduction The Jiangmen Underground Neutrino Observatory(JUNO)is a multipurpose neutrino experiment designed to determine neutrino mass hierarchy,precisely measure oscillation parameters and study solar neutrinos,supernova neutrinos and geo-neutrinos,etc.The central detector(CD)of JUNO has 20,000 tons liquid scintillator as target mass,which contains inside a huge acrylic sphere with inner diameter of 35.4 m,supported by a stainless steel structure.The whole structure of CD will be installed inside a cylindrical water pool,and the acrylic sphere will be submerged in the center of water pool.The operating temperature of CD is designed to be 21℃ as long as over 20 years,which is determined by the mechanical requirement of the structure and physics consideration.Methods For this operating temperature,a special cooling system will be used to maintain the temperature inside the water pool.The main structure of CD is composed of acrylic and stainless steel,and they have much different thermal expansion coefficients,strengths and life times.Change in temperature may affect the safety of CD.As part of reliability analysis,the effect of cooling system failure on the CD is considered,and finite element method is used in our thermal calculation.In this article,the temperature fields before and after cooling system failure are calculated and analyzed,and the temperatures of different locations of water pool after cooling system failure are compared and discussed in detail.Xiaoyu Yang Yuekun Heng Xiaoyan Ma Wei He Huafeng Li Kaixi Huang Tao Song Jiajie Ling Zhi Wu Xiao Tang Xiaolan Luo Xiaohui Qian Yatian Pei Nan Li Fengjiao Luo Zhiyan Cai Mengzhao Li 2019Radiation Detection Technology and Methods2019,3,4:0

11Anti-fatigue Effect of Water Extract from Centella asiatica(L.)Urban on Mice显示文摘[Objectives]To study the anti-fatigue effect of water extract from Centella asiatica(L.)Urban on mice.[Methods]After intragastric administration of low,medium and high concentration of water extract from Centella asiatica(L.)Urban for 14 d,the rotarod time and the content of serum lactic acid in mice were determined,respectively.[Results]Compared with the control group,the rotarod time of mice in the low and medium concentration groups was significantly prolonged(P<0.05),but there was no significant difference between the control group and the high concentration group;the content of serum lactic acid in the medium concentration group was significantly lower than that in the control group(P<0.01),but there was no significant difference between low concentration group and high concentration group and the control group.[Conclusions]A certain concentration of water extract from Centella asiatica(L.)Urban had a good anti-fatigue effect.Chunmei MO Jinye LIN Yanfang MA Jiajie ZHOU Ling LIANG Nian LIN Suoyi HUANG Gang XIAO 2023Medicinal Plant2023,14,1:0
12A systematic review of the efficacy of traditional Chinese medicine retention enema in the treatment of ulcerative colitis显示文摘The objective of this study is to summarize the efficacy and safety of traditional Chinese medicine(TCM)enema in the treatment of ulcerative colitis(UC).The randomized controlled trials on TCM enema intervention in the treatment of UC were searched in seven databases:PubMed,Embase,Web of Science,CBM,CNKI,Wanfang Database,and VIP Database from January 1,2013 to June 6,2022,and the data were analyzed using RevMan 5.3 software.A total of 18 studies involving 1514 UC patients were included.Meta analysis results showed that compared with conventional Western medicine,Chinese medicine enema had a significant effect on UC,and the clinical effective rate of the experimental group using Chinese medicine enema was 4.45 times that of the control group using conventional Western medicine(odds ratio=4.45,95%confidence interval[3.27,6.06]).Therefore,Chinese medicine enema is effective in the treatment of UC,and can significantly reduce related symptoms.Jiaji LI Ling TANG Ye LI Jingjin XU Jing ZHANG 2024Journal of Integrative Nursing2024,6,1:0
13Robust Joule-heating ceramic reactors for catalytic CO oxidation显示文摘Joule-heating reactors have the higher energy efficiency and product selectivity compared with the reactors based on radiative heating.Current Joule-heating reactors are constructed with electrically-conductive metals or carbon materials,and therefore suffer from stability issue due to the presence of corrosive or oxidizing gases during high-temperature reactions.In this study,chemicallystable and electrically-conductive(La_(0.80)Sr_(0.20))_(0.95)FeO_(3)(LSF)/Gd_(0.1)Ce_(0.9)O_(2)(GDC)ceramics have been used to construct Joule-heating reactors for the first time.Taking the advantage of the resistance decrease of the ceramic reactors with temperature increase,the ceramic reactors heated under current control mode achieved the automatic adjustment of heating to stabilize reactor temperatures.In addition,the electrical resistance of LSF/GDC reactors can be tuned by the content of the highconductive LSF in composite ceramics and ceramic density via sintering temperature,which offers flexibility to control reactor temperatures.The ceramic reactors with dendritic channels(less than 100μm in diameter)showed the catalytic activity for CO oxidation,which was further improved by coating efficient MnO_(2)nanocatalyst on reactor channel wall.The Joule-heating ceramic reactors achieved complete CO oxidation at a low temperature of 165℃.Therefore,robust ceramic reactors have successfully demonstrated effective Joule heating for CO oxidation,which are potentially applied in other high-temperature catalytic reactions.Fangsheng LIU Zhibo ZHAO Yuyao MA Yi GAO Jiajie LI Xun HU Zhengmao YE Yihan LING Dehua DONG 2022Journal of Advanced Ceramics2022,11,7:0
