| 3 | Fully-coupled simulations of thermally-induced cracking in pegmatite due to microwave irradiation显示文摘Fully-coupled thermo-mechanical simulations are implemented in COMSOL Multiphysics to investigate micro-scale stress-strain variability in pegmatite specimens subjected to thermal loading using microwaves. Thermally-induced compressive and tensile stresses increase as the microwave irradiation duration increases. The dielectric constant, coefficient of expansion, and type and size of mineralogical boundary have significant impacts on the responses of the rock to microwave irradiation. The maximum principal stress of the chlorite is the smallest, indicating that the chlorite experiences the most damage under microwave irradiation, followed by the quartz. The maximum principal stress values of plagioclase and orthoclase are larger, indicating that they are likely to incur the least damage. Where quartz or chlorite is dominant, the resulting von Mises stresses are consistently higher after 120 s of microwave irradiation. The rate of generation of von Mises stresses increases most rapidly along the interface between quartz and plagioclase, and the interface between quartz and orthoclase, followed by the interface between quartz and chlorite, and finally the interface between plagioclase and orthoclase. | 6 | Reduced function and disassembled microtubules of cultured cardiomyocytes in spaceflight显示文摘Lack of gravity during spaceflight has profound effects on cardiovascular system, but little is known about how the cardiomyocytes respond to microgravity. In the present study, the effects of spaceflight on the structure and function of cultured cardiomyocytes were reported. The primary cultures of neo- natal rat cardiomyocytes were carried on Shenzhou-6 spacecraft and activated at 4 h in orbit. 8 samples were fixed respectively at 4, 48 and 96 h after launching for immunofluorescence of cytoskeleton, and 2 samples remained unfixed to analyze contractile and secretory functions of the cultures. Ground sam- ples were treated in our laboratory in parallel. After 115 h spaceflight, video recordings displayed that the number of spontaneous beating sites in flown samples decreased significantly, and the cells in the beating aggregate contracted in fast frequency without synchrony. Radioimmunoassay of the medium showed that the atrial natriuretic peptide secreted from flown cells reduced by 59.6%. Confocal images demonstrated the time-dependant disassembly of mirotubules versus unchanged distribution and or- ganization of microfilaments. | 8 | In situ AFM observation of BSA adsorption on HOPG with nanobubble显示文摘Nanobubbles have been proven existent at the liquid/solid interface, and become a focus of research on varied interfacial processes. In the present work, by observing in situ with atomic force microscope (AFM), we found that nanobubbles could influence the adsorption process of bovine serum albumin (BSA) on hydrophobic surface of highly ordered pyrolytic graphite (HOPG). BSA could adsorb evenly, and coexist with nanobubbles at water/HOPG interface. After removing nanobubbles by injecting ethanol, some hollows were found in the BSA layers at the same positions of nanobubbles existing previously. These hollows were about 8 nm in depth and dozens of nanometers in diameter. The correlation coefficient between the areas of nanobubbles and that of the corresponding hollows reached 0.88―0.94, which strongly supported the assumption that the hollows were indeed caused by the nanobubbles. A new n-best list rescoring framework, Prefix tree based n-best list re-scoring(PTNR), is proposed to hundred percent eliminate the repeated computations which makes n-best list re-scoring ineffective. At the same time, the bunch mode technique,widely-used in speeding up the training of Feed-forward neural network language model(FF-NNLM), is combined with PTNR and the speed is further improved. Experimental results show that our approach is much faster than basic n-best list re-scoring. | 11 | Host HDAC4 regulates the antiviral response by inhibiting the phosphorylation of IRF3显示文摘Class II HDACs, such as HDAC4, are critical regulators of the immune response in various immune cells;however, its role in innate immunity remains largely unknown.Here, we report that the overexpression of HDAC4 suppresses the production of type I interferons triggered by pattern-recognition receptors (PRRs). HDAC4 repressed the translocation of transcription factor IRF3 to the nucleus, thereby decreasing IRF3-mediated IFN-β expression. In particular, we also determined that HDAC4 can be phosphorylated and simultaneously block the phosphorylation of IRF3 at Ser386 and Ser396 by TBK1 and IKKε, respectively, by interacting with the kinase domain of TBK1 and IKKε. Furthermore, IFN-β may stimulate the expression of HDAC4. The region containing OOV words in the sentence to be decoded is detected with the help of the word-level LM and the probabilities of OOV words are derived from the character-level LM. The implementation of the proposed approach is based on a dynamic decoder. The proposed approach is evaluated in terms of Actual term weighted value(ATWV) on two Mandarin data sets. | 14 | Histone Deacetylase 3 Inhibitor Suppresses Hepatitis C Virus Replication by Regulating Apo-A1 and LEAP-1 Expression显示文摘Histone deacetylase (HDAC) inhibitors show clinical promise for the treatment of cancers, including hepatocellular carcinoma (HCC). In this study, we investigated the effect of HDAC inhibitor treatment on hepatitis C virus (HCV)replication in Huh7 human liver cells and in a mouse model of HCV infection. Viral replication was markedly suppressed by the HDAC3 inhibitor at concentrations below 1 mmol/L, with no cellular toxicity. This was accompanied by upregulation of liver-expressed antimicrobial peptide 1 (LEAP-1) and downregulation of apolipoprotein-A1 (Apo-A1), as determined by microarray and quantitative RT-PCR analyses. Moreover, HDAC3 was found to modulate the binding of CCAAT-enhancer-binding protein a (C/EBPa), hypoxia-inducible factor 1 a (HIF1 a), and signal transducer and activator of transcription 3 (STAT3) to the LEAP-1 promoter. HDAC3 inhibitor treatment also blocked HCV replication in a mouse model of HCV infection. | 15 | Analysis of risk factors associated with endoscopic retrograde cholangiopancreatography for patients with liver cirrhosis: a multicenter, retrospective, clinical study显示文摘Background:Endoscopic retrograde cholangiopancreatography(ERCP)is the endoscopic modality of choice for the treatment of biliary and pancreatic diseases.However,patients with cirrhosis,particularly those with decompensated cirrhosis,are believed to be at increased risk for complications associated with ERCP.There is a paucity of literature describing the outcomes of ERCP for patients with cirrhosis.This study aimed to investigate the outcomes of ERCP for cirrhosis patients,especially adverse events,and evaluated its safety and efficacy.Methods:We performed a multicenter,retrospective study of all patients at Beijing Friendship Hospital affiliated to Capital Medical University,Xijing Hospital affiliated to Air Force Military Medical University,Beijing Youan Hospital affiliated to Capital Medical University,and the Fifth Medical Center of the People’s Liberation Army General Hospital from June 2003 to August 2019.The adverse events of inpatient ERCP for patients with(n=182)and without liver cirrhosis(controls;n=385)were compared.Results:A total of 567 patients underwent ERCP between January 2003 and December 2019 were enrolled in this study.Compared to patients without cirrhosis,patients with cirrhosis were at higher risk for postoperative complications(odds ratio[OR],4.172;95%confidence interval[CI],1.232–7.031;P<0.001)such as postoperative pancreatitis(OR,2.026;95%CI,1.002–4.378;P=0.001)and cholangitis(OR,3.903;95%CI,1.001–10.038;P=0.036).The main indications for ERCP for patients with cirrhosis in this study included choledocholithiasis(101 cases;55.5%),benign bile duct strictures(46 cases;25.3%),and malignant bile duct strictures(28 cases;15.4%).Among them,23 patients(12.6%)underwent balloon dilation and 79 patients(43.4%)underwent sphincterotomy.Of the patients with cirrhosis,delayed bleeding occurred in ten patients(5.5%),postoperative pancreatitis occurred in 80 patients(44.0%),and postoperative cholangitis occurred in 25 patients(13.7%).An additional multivariate analysis showed that the total bilirubin(TBIL)level(OR,4.58;95%CI,2.37–6.70)and Child-Pugh score of C(OR,3.11;95%CI,1.04–5.37)were risk factors for postoperative complications in patients with cirrhosis.Conclusions:Compared with the general population of patients undergoing ERCP,patients with cirrhosis were more prone to postoperative pancreatitis and cholangitis.TBIL levels and Child-Pugh scores were risk factors for postoperative complications in patients with cirrhosis. | Jielin Li Jiexuan Hu Peng Li Yongdong Wu Yongjun Wang Ming Ji Haiyang Hua Wenbin Ran Yanglin Pan Shutian Zhang | 2022 | Chinese Medical Journal2022,135,19: | 2 |
