2In situ AFM observation of BSA adsorption on HOPG with nanobubble显示文摘Nanobubbles have been proven existent at the liquid/solid interface, and become a focus of research on varied interfacial processes. In the present work, by observing in situ with atomic force microscope (AFM), we found that nanobubbles could influence the adsorption process of bovine serum albumin (BSA) on hydrophobic surface of highly ordered pyrolytic graphite (HOPG). BSA could adsorb evenly, and coexist with nanobubbles at water/HOPG interface. After removing nanobubbles by injecting ethanol, some hollows were found in the BSA layers at the same positions of nanobubbles existing previously. These hollows were about 8 nm in depth and dozens of nanometers in diameter. The correlation coefficient between the areas of nanobubbles and that of the corresponding hollows reached 0.88―0.94, which strongly supported the assumption that the hollows were indeed caused by the nanobubbles. 3Application of image alignment and time averaging methods in AFM detection for single DNA molecules显示文摘Enhancement of the SNR (signal to noise ratio) in single-molecule imaging is significantly important for improving image resolu-tion and distinguishing the fine structures of single molecules at a higher precision level.Image processing techniques have dem-onstrated the remarkable capability to improve the SNR and the resolution level by breaking through some inherent limitations unresolved by instrument hardware optimization.In this paper, we focus on single-biomolecule imaging using atomic force mi-croscopy (AFM), a unique method in separated single-biomolecule imaging, and there was few suitable image processing tech-niques reported to increase the SNR of a single molecule’s AFM image.With the typical samples of separately dispersed DNA molecules, we replaced the classified averaging method, which is usually used when the molecules’ structure can be easily and repeatedly prepared, with the time averaging method to improve the SNR in a single molecule’s AFM imaging.Combining the time averaging technique with the image alignment method for the series of time-lapse AFM images of a single DNA molecule, we were able to improve the image’s SNR and recover some buried signals from the background noises.Furthermore, the fine structures of single molecules can potentially be further resolved if other image processing techniques are applied collaboratively with some newly developed imaging methods like Vibrating Mode Scanning Polarization Force Microscopy (VSPFM), and such combination will also be advantageous for the manipulation of single-biomolecules by AFM.In addition, the proposed algorithms for the estimations of drift, distortion and SNR are applicable for quantitative status characterization of AFM imaging.WANG ChunMei SUN JieLin LONG Fei ZHAO Jun HU Jun 2010Chinese Science Bulletin2010,55,16:3

8GPR162 activates STING dependent DNA damage pathway as a novel tumor suppressor and radiation sensitizer显示文摘In the treatment of most malignancies,radiotherapy plays a significant role.However,the resistance of cancer cells to ionizing radiation(IR)is the main reason for the failure of radiotherapy,which causes tumor recurrence and metastasis.In this study,we confirmed that GPR162,an orphan receptor in the G-protein-coupled receptor family,acted as a novel radiotherapy sensitizer by interacting with the stimulator of interferon genes(STING),which targeted DNA damage responses,activated IRF3,accelerated the activation of type I interferon system,promoted the expression of chemokines including CXCL10 and CXCL4,and inhibited the occurrence and development of tumors.Interestingly,the activation of STING by overexpression of GPR162 was independent of the classical pathway of cGAS.STING inhibitors could resist the antitumor effect of overexpression of GPR162 in IR-induced mouse models.In addition,most solid tumors showed low expression of GPR162.And the higher expression of GPR162 indicated a better prognosis in patients with lung adenocarcinoma,liver cancer,breast cancer,etc.In summary,these results suggested that GPR162 may serve as a potential sensitizer of radiotherapy by promoting radiotherapy-induced STING-IFN production and increasing the expression of chemokines including CXCL10 and CXCL4 in DNA damage response,providing an alternative strategy for improving cancer radiotherapy.Yao Long Jiaxing Guo Jielin Chen Jingyue Sun Haiyan Wang Xin Peng Zuli Wang WeiWei Lai Na Liu Long Shu Ling Chen Ying Shi Desheng Xiao Shuang Liu Yongguang Tao 2023Signal Transduction and Targeted Therapy2023,8,3:0

12Comparison of viral propagation and drug response among SARS-CoV-2 VOCs using replicons capable of recapitulating virion assembly and release显示文摘Several variants of concern(VOCs)have emerged since the WIV04 strain of severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)was first isolated in January 2020.Due to mutations in the spike(S)protein,these VOCs have evolved to enhance viral infectivity and immune evasion.However,whether mutations of the other viral proteins lead to altered viral propagation and drug resistance remains obscure.The replicon is a noninfectious viral surrogate capable of recapitulating certain steps of the viral life cycle.Although several SARS-CoV-2 replicons have been developed,none of them were derived from emerging VOCs and could only recapitulate viral genome replication and subgenomic RNA(sgRNA)transcription.In this study,SARS-CoV-2 replicons derived from the WIV04 strain and two VOCs(the Beta and Delta variants)were prepared by removing the S gene from their genomes,while other structural genes remained untouched.These replicons not only recapitulate viral genome replication and sgRNA transcription but also support the assembly and release of viral-like particles,as manifested by electron microscopic assays.Thus,the S-deletion replicon could recapitulate virtually all the post-entry steps of the viral life cycle and provides a versatile tool for measuring viral intracellular propagation and screening novel antiviral drugs,including inhibitors of virion assembly and release.Through the quantification of replicon RNA released into the supernatant,we demonstrate that viral intracellular propagation and drug response to remdesivir have not yet substantially changed during the evolution of SARS-CoV-2 from the WIV04 strain to the Beta and Delta VOCs.Lingqian Tian Qiuhong Liu Rongjuan Pei Yingshan Chen Chonghui Xu Jielin Tang Hao Sun Kunpeng Liu Qi Yang Lei Yang Leshan Li Yongli Zhang Yuan Zhou Chao Shan Xue Hu Xinwen Chen Yun Wang 2022Virologica Sinica2022,37,5:0
