2Jianpijiedu Fang improves survival of hepatocarcinoma mice by affecting phosphatase and tensin homolog, phosphoinositide 3-kinase, and focal adhesion kinase显示文摘OBJECTIVE: To investigate the effect of Jianpijiedu Fang (JPJDF) on phosphatase and tensin homolog (PTEN), phosphoinositide 3-kinase (PI3K), and focal adhesion kinase (FAK), and on the survival of hepatocellular carcinoma (HCC) nude mice. METHODS: Forty male nude mice were randomly divided into 4 groups. Human HCC tissue was implanted in the livers of three groups. After 24 h, the three groups were treated respectively with JPJDF (37.5 g/kg), saline (20 mL/kg) and Tegafur (FT-207, 160 mg/kg) once a day for 10 weeks. The control group without implanting the tissue was concurrently treated with saline (20 mL/kg). The survival data and body weight of all mice were recorded, and expression levels of PTEN, PI3K and FAK in normal tissue and cancer tissue of the livers were eval-uated with immunohistochemical method. RESULTS: The cumulative survival rate of the mice in the JPJDF group was higher than those of the other groups. The rate of weight loss was the lowest in JPJDF group. The survivability and weight loss rate in FT-207 group were the poorest in all groups. The expression intensity of PTEN was higher in normal tissues than in cancer tissues, and lower in the normal tissues of HCC models than in that of mice without HCC. The PTEN expression intensity in normal tissue and cancer tissue from mice treated with FT-207 were lower than that from the mice treated with JPJDF or saline.The expression intensity of PI3K was higher in cancer tissue than in normal tissue. The PI3K expression intensity was the lowest in normal tissue and cancer tissue from mice treated with JPJDF, and the intensity from mice treated with FT-207 was the highest. In mice treated with JPJDF, the expression intensity of FAK was higher in the normal tissue and lower in the cancer tissue than those of the other treatment groups. 4Whole-Genome Resequencing of a Worldwide Collection of Rapeseed Accessions Reveals the Genetic Basis of Ecotype Divergence显示文摘Rapeseed (Brassica napus),an important oilseed crop,has adapted to diverse climate zones and latitudes by forming three main ecotype groups,namely winter,semiwinter,and spring types. However,genetic variations underlying the divergence of these ecotypes are largely unknown. Here,we report the global pattern of genetic polymorphisms in rapeseed determined by resequencing a worldwide collection of 991 germplasm accessions.A total of 5.56 and 5.53 million singlenucleotide polymorphisms (SNPs)as Well as 1.86 and 1.92 million InDels were identified by mapping reads to the reference genomes of 'Darmor-bzh'and 'Tapidor,'respectively.We generated a map of allelic drift paths that shows splits and mixtures of the main populations,and revealed an asymmetric evolution of the two subgenomes of B.napus by calculating the genetic diversity and linkage disequilibrium parameters.Selective-sweep analysis revealed genetic changes in genes orthologous to those regulating various aspects of plant development and response to stresses.A genome-wide association study identified SNPs in the promoter regions of FLOWERING LOCUS T and FLOWERING LOCUS C orthologs that corresponded to the different rapeseed ecotype groups. 5Diagnostic value of 5 serum biomarkers for hepatocellular carcinoma with different epidemiological backgrounds:A large-scale,retrospective study显示文摘Objective:Hepatocellular carcinoma(HCC)is a lethal global disease that requires an accurate diagnosis.We assessed the potential of 5 serum biomarkers(AFP,AFU,GGT-II,GPC3,and HGF)in the diagnosis of HCC.Methods:In this retrospective study,we measured the serum levels of each biomarker using ELISAs in 921 participants,including 298 patients with HCC,154 patients with chronic hepatitis(CH),122 patients with liver cirrhosis(LC),and 347 healthy controls from 3 hospitals.Patients negative for hepatitis B surface antigen and hepatitis C antibody(called'NBNC-HCC')and patients positive for the above indices(called'HBV-HCC and HCV-HCC')were enrolled.The selected diagnostic model was constructed using a training cohort(n=468),and a validation cohort(n=453)was used to validate our results.Receiver operating characteristic analysis was used to evaluate the diagnostic accuracy.Results:Theα-L-fucosidase(AFU)/α-fetoprotein(AFP)combination was best able to distinguish NBNC-HCC[area under the curve:0.986(95%confidence interval:0.958–0.997),sensitivity:92.6%,specificity:98.9%]from healthy controls in the test cohort.For screening populations at risk of developing HCC(CH and LC),the AFP/AFU combination improved the diagnostic specificity for early-stage HCC[area under the curve:0.776(0.712–0.831),sensitivity:52.5%,specificity:91.6%in the test group].In all-stage HBV-HCC and HCV-HCC,AFU was also the best candidate biomarker combined with AFP[area under the curve:0.835(0.784–0.877),sensitivity 69.1%,specificity:87.4%in the test group].All results were verified in the validation group.Conclusions:The AFP/AFU combination could be used to identify NBNC-HCC from healthy controls and hepatitis-related HCC from at-risk patients.Dongming Liu Yi Luo Lu Chen Liwei Chen Duo Zuo Yueguo Li Xiaofang Zhang Jing Wu Qing Xi Guangtao Li Lisha Qi Xiaofen Yue Xiehua Zhang Zhuoyu Sun Ning Zhang Tianqiang Song Wei Lu Hua Guo 2021Cancer Biology & Medicine2021,18,1:10

7The biological effect of metal ions on the granulation of aerobic granular activated sludge显示文摘As a special biofilm structure,microbial attachment is believed to play an important role in the granulation of aerobic granular activated sludge(AGAS).This experiment was to investigate the biological effect of Ca^(2+),Mg^(2+),Cu^(2+),Fe^(2+),Zn^(2+),and K+which are the most common ions present in biological wastewater treatment systems,on the microbial attachment of AGAS and flocculent activated sludge(FAS),from which AGAS is always derived,in order to provide a new strategy for the rapid cultivation and stability control of AGAS.The result showed that attachment biomass of AGAS was about 300%higher than that of FAS without the addition of metal ions.Different metal ions had different effects on the process of microbial attachment.FAS and AGAS reacted differently to the metal ions as well,and in fact,AGAS was more sensitive to the metal ions.Specifically,Ca^(2+),Mg^(2+),and K+could increase the microbial attachment ability of both AGAS and FAS under appropriate concentrations,Cu^(2+),Fe^(2+),and Zn^(2+)were also beneficial to the microbial attachment of FAS at low concentrations,but Cu^(2+),Fe^(2+),and Zn^(2+)greatly inhibited the attachment process of AGAS even at extremely low concentrations.In addition,the acylated homoserine lactone(AHL)-based quorum sensing system,the content of extracellular polymeric substances and the relative hydrophobicity of the sludges were greatly influenced by metal ions.As all these parameters had close relationships with the microbial attachment process,the microbial attachment may be affected by changes of these parameters.Wen Hao Yaochen Li Junping Lv Lisha Chen Jianrong Zhu 2016Journal of Environmental Sciences2016,28,6:8
8Ranking effects of candidate drugs on biological process by integrating network analysis and Gene Ontology显示文摘There are high preclinical attrition rates in current drug discovery. The efficient assessment approach in the high throughout candidate drugs screening still needs great improvement. We propose two hypotheses. First, both drug action process and biological process can be converted to a common space of gene or gene product profiling. Second, the strength of drug action on biological process can be realized in the context of biological network. Based on the above hypotheses, we establish an algorithm termed Network-based Assessment for Drug Action (NADA) to assess the action strength of candidate drugs on certain biological processes. Then NADA is used to prioritize the effects of six compounds from traditional Chinese medicine on endothelial cell migration, a simple process defined by Gene Ontology, in the biological network specific for a given pathological process, angiogenesis. The computational results are subsequently tested by the experiment on the migration of Human Umbilical Vein Endothelial Cells in vitro. The experimental ranks for six compounds generally agree with the predicted output of NADA. NADA also outperforms the DAVID and meet/min methods in terms of the experimental orders, suggesting that the network topological features may have a key role in catching the mechanistic relationship between drug action and biological process. 10Neuroprotective effects of total saponins from Rubus parvifolius L. on cerebral ischemia/reperfusion injury in rats显示文摘This study examines the neuroprotective effects and mechanisms of action of total saponins from Rubus parvifolius L. (TSRP) on focal cerebral ischemia and reperfusion injury in rats. Focal cerebral ischemia and reperfusion injury was performed in rats using the suture method. The results indicate that intragastric injection of TSRP, at 5, 10 and 20 mg/kg, could decrease neurological impairment, reduce cerebral infarct volume, diminish pathological changes, and significantly inhibit the apoptosis of neurons surrounding the ischemic area. In addition, TSRP upregulated the expression of the anti-apoptotic factor Bcl-2, at the protein and mRNA levels, and it downregulated the expression of the pro-apoptotic factor Bax, at the protein and mRNA levels. 11The role of bone marrow-derived cells in the origin of liver cancer revealed by single-cell sequencing显示文摘Objective:Epithelial cancers often originate from progenitor cells,while the origin of hepatocellular carcinoma(HCC)is still controversial.HCC,one of the deadliest cancers,is closely linked with liver injuries and chronic inflammation,which trigger massive infiltration of bone marrow-derived cells(BMDCs)during liver repair.Methods:To address the possible roles of BMDCs in HCC origination,we established a diethylnitrosamine(DEN)-induced HCC model in bone marrow transplanted mice.Immunohistochemistry and frozen tissue immunofluorescence were used to verify DENinduced HCC in the pathology of the disease.The cellular origin of DEN-induced HCC was further studied by single cell sequencing,single-cell nested PCR,and immunofluorescence-fluorescence in situ hybridization.Results:Studies by using single cell sequencing and biochemical analysis revealed that HCC cells in these mice were coming from donor mice BMDCs,and not from recipient mice.Furthermore,the copy numbers of mouse orthologs of several HCC-related genes previously reported in human HCC were also altered in our mouse model.DEN-induced HCCs exhibited a similar histological phenotype and genomic profile as human HCCs.Conclusions:These results suggested that BMDCs are an important origin of HCC,which provide important clues to HCC prevention,detection,and treatments.Lu Chen Xianfu Yi Piao Guo Hua Guo Ziye Chen Chunyu Hou Lisha Qi Yongrong Wang Chengwen Li Peng Liu Yucun Liu Yuanfu Xu Ning Zhang 2020Cancer Biology & Medicine2020,17,1:4
12CPI-17-mediated contraction of vascular smooth muscle is essential for the development of hypertension in obese mice显示文摘Several factors have been implicated in obesity-related hypertension, but the genesis of the hypertension is largely unknown. In this study, we found a significantly upregulated expression of CPI-17(C-kinasepotentiated protein phosphatase 1 inhibitor of 17 kDa) and protein kinase C(PKC) isoforms in the vascular smooth muscles of high-fat diet(HFD)-fed obese mice. The obese wild-type mice showed a significant elevation of blood pressure and enhanced calcium-sensitized contraction of vascular smooth muscles. However, the obese CPI-17-deficient mice showed a normotensive blood pressure, and the calcium-sensitized contraction was consistently reduced. In addition, the mutant muscle displayed an abolished responsive force to a PKC activator and a 30%-50% reduction in both the initial peak force and sustained force in response to various G protein-coupled receptor(GPCR) agonists. Our observations showed that CPI-17-mediated calcium sensitization is mediated through a GPCR/PKC/CPI-17/MLCP/RLC signaling pathway. 13Pyridinic nitrogen enriched porous carbon derived from bimetal organic frameworks for high capacity zinc ion hybrid capacitors with remarkable rate capability显示文摘Aqueous zinc ion hybrid capacitors(ZIHCs)hold great potential for large-scale energy storage applications owing to their high safety and low cost,but suffer from low capacity and energy density.Herein,pyridinic nitrogen enriched porous carbon(nPC)was successfully synthesized via the growth,subsequent annealing and acid etching of bimetal organic frameworks for high capacity and safe ZIHCs with exceptional rate capability.Benefiting from the mesopores for easy ion diffusion,high electrical conductivity enabled by in-situ grown carbon nanotubes matrix and residual metal Co nanoparticles for fast electron transfer,sufficient micropores and high N content(8.9 at%)with dominated pyridinic N(54%)for enhanced zinc ion storage,the resulting nPC cathodes for ZIHCs achieved high capacities of 302 and137 m Ah g^(-1) at 1 and 18 A g^(-1),outperforming most reported carbon based cathodes.Theoretical results further disclosed that pyridinic N possessed larger binding energy of-4.99 eV to chemically coordinate with Zn2+than other N species.Moreover,quasi-solid-state ZIHCs with gelatin based gel electrolytes exhibited high energy density of 157.6 Wh kg^(-1) at 0.69 kW kg^(-1),high safety and mechanical flexibility to withstand mechanical deformation and drilling.This strategy of developing pyridinic nitrogen enriched porous carbon will pave a new avenue to construct safe ZIHCs with high energy densities.Yao Li Pengfei Lu Ping Shang Lisha Wu Xiao Wang Yanfeng Dong Ronghuan He Zhong-Shuai Wu 2021Journal of Energy Chemistry2021,30,5:3
14Decadal variations in the community status of economically important invertebrates in the Bohai Sea显示文摘With the decline of fish stocks,the proportions of economically important invertebrates like crustaceans and cephalopods have increased in the Bohai Sea.The community structure and trophic level of economically important invertebrates were analyzed using the bottom trawl survey data collected by the Yellow Sea Fisheries Research Institute in the Bohai Sea in May and August of 1958–1959,1982,1992–1993,2004,2009 and 2015.A total of 37 species of economically important invertebrates,belonging to 5 orders,24 families,were captured.The biomass densities of economically important invertebrates in the Bohai Sea displayed an overall downward trend from 1982 to 2015.Oratosquilla oratoria and Loligo spp.were the most dominant species in the past 30 years,the biomass proportion of O.oratoria increased gradually in both May and August from 1982 to 2015.Moreover,biodiversity indices of economically important invertebrates in the Bohai Sea appeared to decline from 1982 to 2004 and then increased in 2015.Similarly,the mean trophic level of economically important invertebrates declined from 1982 to 2004 and increased slightly in 2015.Overall,although the proportions of invertebrates have increased,the biomass densities in the Bohai Sea have displayed an overall downward trend from 1982 to 2015.The increases in the biodiversity and trophic level of economically important invertebrates after the 2000s,possibly benefit from stock enhancement projects implemented by governments at different levels and national fishery management measures such as the“double-control”of the total number and engine power of fishing vessels and summer moratorium of fishing.Qiang Wu Lisha Guan Zhongyi Li Xiujuan Shan Xianshi Jin 2019Acta Oceanologica Sinica2019,38,10:3
15Imaging changes in neural circuits in patients with depression using 1 H-magnetic resonance spectroscopy and diffusion tensor imaging显示文摘1 H-magnetic resonance spectroscopy imaging and diffusion tensor imaging were performed in 19 patients with mild depression and in 13 controls.The mean age of the patients was 31 years.The mean Hamilton depression score of the patients was 22.5±13.2.N-acetylaspartate,choline and creatine concentrations and the average diffusion coefficient and fractional anisotropy values were measured in the bilateral hippocampus,striatum,thalamus and prefrontal deep white matter. Compared with the control group,the mild depressed patients had:(1)a higher choline/creatine ratio and a negative correlation between the choline/creatine ratio and the average diffusion coefficient in the hippocampus;(2)a lower choline/creatine ratio and a higher fractional anisotropy in the striatum;(3)a lower fractional anisotropy and a positive correlation between the fractional anisotropy and the choline/creatine ratio in the prefrontal deep white matter;and(4)a higher average diffusion coefficient and a positive correlation between the choline/creatine ratio and the N-acetylaspartate/creatine ratio in the thalamus,as well as positive correlation between the choline/creatine ratio and Hamilton depression scores.These data suggest evidence of abnormal connectivity in neurofibrotic microstructures and abnormal metabolic alterations in the limbic-cortical-striatal-pallidal-thalamic neural circuit in patients with mild depression.Shaoqiong Chen Lisha Lai Zhuang Kang Xiao Luo Jiansheng Zhang Jianfang Li 2012Neural Regeneration Research2012,7,24:2
16Improved Electrochemical Performance Based on Nanostructured SnS_2@CoS_2–rGO Composite Anode for Sodium-Ion Batteries显示文摘A promising anode material composed of SnS_2@CoS_2 flower-like spheres assembled from SnS_2 nanosheets and CoS_2 nanoparticles accompanied by reduced graphene oxide(rGO) was fabricated by a facile hydrothermal pathway. The presence of rGO and the combined merits of SnS_2 and CoS_2 endow the SnS_2@-CoS_2–rGO composite with high conductivity pathways and channels for electrons and with excellent properties as an anode material for sodium-ion batteries(SIBs). A high capacity of 514.0 mAh g^(-1) at a current density of200 m A g^(-1) after 100 cycles and a good rate capability can be delivered. 17Improvement in the risk assessment of oral leukoplakia through morphology-related copy number analysis显示文摘Oral leukoplakia is the most common type of oral potentially malignant disorders and considered a precursor lesion to oral squamous cell carcinoma.However,a predictor of oral leukoplakia prognosis has not yet been identified.We investigated whether copy number alteration patterns may effectively predict the prognostic outcomes of oral leukoplakia using routinely processed paraffin sections.Comparison of copy number alteration patterns between oral leukoplakia with hyperplasia(HOL,n=22)and dysplasia(DOL,n=21)showed that oral leukoplakia with dysplasia had a higher copy number alteration rate(86%)than oral leukoplakia with hyperplasia(46%).Oral leukoplakia with dysplasia exhibited a wider range of genomic variations across all chromosomes compared with oral leukoplakia with hyperplasia.We also examined a retrospective cohort of 477 patients with oral leukoplakia with hyperplasia with detailed follow-up information.The malignant transformation(MT,n=19)and leukoplakia recurrence(LR,n=253)groups had higher frequencies of aneuploidy events and copy number loss rate than the free of disease(FD,n=205)group.Together,our results revealed the association between the degree of copy number alterations and the histological grade of oral leukoplakia and demonstrated that copy number alteration may be effective for prognosis prediction in oral leukoplakia patients with hyperplasia.Xiaotian Li Lu Liu Jianyun Zhang Ming Ma Lisha Sun Xuefen Li Heyu Zhang Jianbin Wang Yanyi Huang Tiejun Li 2021Science China(Life Sciences)2021,64,9:2

To evaluate the effect of FGFR2-IIIb in pig lung development, we employed somatic cell nuclear transfer(SCNT) technology to generate transgenic pig fetuses overexpressing the transmembrane(dn FGFR2-IIIb-Tm) and soluble(dn FGFR2-IIIb-HFc) forms of the dominant-negative human FGFR2-IIIb driven by the human surfactant protein C(SP-C) promoter,which was specifically expressed in lung epithelia. Eight dn FGFR2-IIIb-Tm transgenic and twelve dn FGFR2-IIIb-HFc transgenic pig fetuses were collected from three and two recipient sows, respectively.Repression of FGFR2-IIIb in lung epithelia resulted in smaller lobes and retardation of alveolarization in both forms of dn FGFR2-IIIb transgenic fetuses. Moreover, the dn FGFR2-IIIb-HFc transgenic ones showed more deterioration in lung development. Our results demonstrate that disruption of FGFR2-IIIb signaling in the epithelium impedes normal branching and alveolarization in pig lungs, which is less severe than the results observed in transgenic mice. 19The dichotomous and incomplete adaptive immunity in COVID-19 patients with different disease severity显示文摘The adaptive immunity that protects patients from coronavirus disease 2019(COVID-19),caused by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2),is not well characterized.In particular,the asymptomatic patients have been found to induce weak and transient SARS-CoV-2 antibody responses,but the underlying mechanisms remain unknown;meanwhile,the protective immunity that guide the recovery of these asymptomatic patients is elusive.Here,we characterized SARS-CoV-2-specific B-cell and T-cell responses in 10 asymptomatic patients and 64 patients with other disease severity(mild,n=10,moderate,n=32,severe,n=12)and found that asymptomatic or mild symptomatic patients failed to mount virus-specific germinal center(GC)B cell responses that result in robust and prolonged humoral immunity,assessed by GC response indicators including follicular helper T(TFH)cell and memory B cell responses as well as serum CXCL13 levels.Alternatively,these patients mounted potent virus-specific TH1 and CD8+T cell responses.In sharp contrast,patients of moderate or severe disease induced vigorous virus-specific GC B cell responses and associated TFH responses;however,the virus-specific TH1 and CD8+T cells were minimally induced in these patients.These results,therefore,uncovered the protective immunity in asymptomatic patients and also revealed the strikingly dichotomous and incomplete humoral and cellular immune responses in COVID-19 patients with different disease severity,providing important insights into rational design of effective COVID-19 vaccines.Leiqiong Gao Jing Zhou Sen Yang Lisha Wang Xiangyu Chen Yang Yang Ren Li Zhiwei Pan Jing Zhao Zhirong Li Qizhao Huang Jianfang Tang Li Hu Pinghuang Liu Guozhong Zhang Yaokai Chen Lilin Ye 2021Signal Transduction and Targeted Therapy2021,6,4:2
