| 2 | The Prevalence of Nonalcoholic Fatty Liver Disease and its Association with Lifestyle/dietary Habits among University Faculty and Staff in Chengdu显示文摘Objective To investigate the prevalence of nonalcoholic fatty liver disease(NAFLD) in different university categories and its association with lifestyle/dietary habits.Methods A cross-sectional study was carried out on 9 378 faculty members and staff who participated in an annual health checkup at three universities selected by random cluster sampling.Demographic,anthropometric,biochemical indices and abdominal ultrasound measurements were collected.A nested case-control study was conducted with 200 NAFLD cases and 200 controls matched by gender,age(±3 years),and university.Results The overall prevalence of NAFLD was 10.3%(13.7% in males and 6.8% in females).The prevalence was significantly higher in the science and engineering university(22.1%) than in the comprehensive universities with(6.4%) and without(10.9%) medical colleges.Obesity/overweight,hyperlipidemia,diabetes mellitus,and family history of NAFLD were independently associated with higher risk of NAFLD,as were frequent consumption of desserts and salty/spicy foods.Using nutritional supplements was a protective factor against NAFLD.Intake of coarse cereals,potatoes,vegetables,fruits,and milk was significantly lower,and intake of red meat,viscera,candies and pastries,cooking oil,and total energy was significantly higher in participants with NAFLD than in controls.Conclusion Science and engineering university faculty and staff are key targets for NAFLD prevention.NAFLD is closely associated with age,gender,university type,metabolic diseases,and lifestyle/dietary habits. | SHI Lei LIU Zhong Wu LI Yun GONG Cai ZHANG Huan SONG Li Juan HUANG Cheng Yu LI Ming | 2012 | Biomedical and Environmental Sciences2012,25,4: | 16 |

| 5 | Purification and characterization of keratinase from a new Bacillus subtilis strain显示文摘The aim of this study was to purify and characterize a keratinase produced by a new isolated Bacillus subtilis KD-N2 strain. The keratinase produced by the isolate was purified using ammonium sulphate precipitation, Sephadex G-75 and DEAE (diethylaminoethyl)-Sepharose chromatographic techniques. The purified enzyme was shown to have a molecular mass of 30.5 kDa, as determined by sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) analysis. The optimum pH at 50 °C was 8.5 and the optimum temperature at pH 8.5 was 55 °C. The keratinase was partially inactivated by some metal ions, organic solvents and serine protease inhibitor phenylmethanesulfonyl fluoride (PMSF). Sodium dodecyl sulfate (SDS) and ethylene diamine tetraacetic acid (EDTA) had positive effect on the keratinase activity. Reducing agents including dithiothreitol (DTT), mercaptoethanol, L-cysteine, sodium sulphite, as well as chemicals of SDS, ammonium sulfamate and dimethylsulfoxide (DMSO) stimulated the enzyme activity upon a feather meal substrate. Besides feather keratin, the enzyme is active upon the soluble proteins ovalbumin, bovine serum albumin (BSA), casein and insoluble ones as sheep wool and human hair. | 6 | Pediatric reference intervals in China(PRINCE):design and rationale for a large,multicenter collaborative cross-sectional study显示文摘There is a lack of accurate pediatric reference intervals(RIs) in China, with most commonly used RIs established without consideration of the effect of age and gender. The Pediatric Reference Intervals in China(PRINCE) project aims to establish and verify pediatric RIs for 31 common laboratory measurands.The project will be a large, multicenter cross-sectional study:14,490 healthy children and adolescents aged up to 19 years will be surveyed by 10 children's hospitals and one pediatric department of a university hospital. To evaluate the feasibility and efficiency of the study methods, 602 children were surveyed in the pilot phase of the PRINCE study in April 2017: it found that some measurands were distinctly age dependent and that there were differences between values for males and females. The results of the pilot study affirmed the necessity of the PRINCE project for Chinese pediatrics. The pilot also indicated potential difficulties in the full survey, e.g., difficulties in recruiting children aged under 3 years and insufficient collection of blood samples from infants. | 8 | The adult human testis transcriptional cell atlas显示文摘Human adult spermatogenesis balances spermatogonial stem cell (SSC) self-renewal and differentiation,alongside complex germ cell-niche interactions,to ensure long-term fertility and faithful genome propagation. Here,we performed single-cell RNA sequencing of -6500 testicular cells from young adults. We found five niche/somatic cell types (Leydig,myoid,Sertoli,endothelial,macrophage),and observed germline-niche interactions and key human-mouse differences. Spermatogenesis,including meiosis,was reconstructed computationally,revealing sequential coding,non-coding,and repeat-element transcriptional signatures. Interestingly,we identified five discrete transcriptional/developmental spermatogonial states,including a novel early SSC state,termed State 0. Epigenetic features and nascent transcription analyses suggested developmental plasticity within spermatogonial States. To understand the or丨gin of State 0,we profiled testicular cells from infants,and identified distinct similarities between adult State 0 and infant SSCs. | 10 | Breviscapine attenuates acute pancreatitis by inhibiting expression of PKCα and NF-κB in pancreas显示文摘AIM:To study the effect of breviscapine (Bre) on activity of protein kinase Cα (PKCα) and nuclear factor (NF)-κB in pancreas,and the mechanism of Bre attenuating acute pancreatitis (AP). METHODS:One hundred and eight rats were randomly divided into acute necrotizing pancreatitis (ANP) group,Bre group (ANP + Bre group) and sham operation (SO) group,36 rats in each group. ANP model was induced by a retrograde injection of 4% sodium deoxycholate into the bilio-pancreatic duct. Fifteen minutes after the ANP model was induced,the rats in Bre group were intraperitoneally injected with Bre (0.4 mg/100 g body weight or 0.1 mL/100 g body weight). Survival time and mortality of rats were calculated. Serum amylase and malondialdehyde levels were measured,volume of ascites was recorded and morphology of pancreas and lung was evaluated at 1,5 and 10 h,after the ANP model was induced,respectively. Expressions of PKCα and subunit p65 of NF-κB in pancreas were detected by immunohistochemistry and Western blotting. RESULTS:The life span of rats was longer and the mortality was lower in Bre group than in ANP group 13.51 ± 5.46 vs 25.36 ± 8.11 (P < 0.05). The amylase and MDA levels as well as the volume of ascites were lower and the pathological changes in pancreas and lung were less in Bre group than ANP group (P < 0.05),indicating that the pancreatitis is less severe in Bre group than ANP group. The activation of PKCα and NF-κB p65 in pancreas was induced rapidly and reached their peak at 1 h or 5 h after ANP,but their activity in Bre group was significantly inhibited. | 13 | Efficacy and safety of the long-acting fusion inhibitor albuvirtide in antiretroviral-experienced adults with human immunodeficiency virus-1:interim analysis of the randomized,controlled,phase 3,non-inferiority TALENT study显示文摘Background:Albuvirtide is a once-weekly injectable human immunodeficiency virus(HIV)-1 fusion inhibitor.We present interim data for a phase 3 trial assessing the safety and efficacy of albuvirtide plus lopinavir-ritonavir in HIV-1-infected adults already treated with antiretroviral drugs.Methods:We carried out a 48-week,randomized,controlled,open-label non-inferiority trial at 12 sites in China.Adults on the World Health Organization(WHO)-recommended first-line treatment for>6 months with a plasma viral load>1000 copies/mL were enrolled and randomly assigned(1:1)to receive albuvirtide(once weekly)plus ritonavir-boosted lopinavir(ABT group)or the WHO-recommended second-line treatment(NRTI group).The primary endpoint was the proportion of patients with a plasma viral load below 50 copies/mL at 48 weeks.Non-inferiority was prespecified with a margin of 12%.Results:At the time of analysis,week 24 data were available for 83 and 92 patients,and week 48 data were available for 46 and 50 patients in the albuvirtide and NRTI groups,respectively.At 48 weeks,80.4%of patients in the ABT group and 66.0%of those in the NRTI group had HIV-1 RNA levels below 50 copies/mL,meeting the criteria for non-inferiority.For the per-protocol population,the superiority of albuvirtide over NRTI was demonstrated.The frequency of grade 3 to 4 adverse events was similar in the two groups;the most common adverse events were diarrhea,upper respiratory tract infections,and grade 3 to 4 increases in triglyceride concentration.Renal function was significantly more impaired at 12 weeks in the patients of the NRTI group who received tenofovir disoproxil fumarate than in those of the ABT group.Conclusions:The TALENT study is the first phase 3 trial of an injectable long-acting HIV drug.This interim analysis indicates that once-weekly albuvirtide in combination with ritonavir-boosted lopinavir is well tolerated and non-inferior to the WHO-recommended second-line regimen in patients with first-line treatment failure.Trial registration:ClinicalTrials.gov Identifier:NCT02369965;https://www.clinicaltrials.gov.Chinese Clinical Trial Registry No.ChiCTR-TRC-14004276;http://www.chictr.org.cn/enindex.aspx. | Bin Su Cheng Yao Qing-Xia Zhao Wei-Ping Cai Min Wang Hong-Zhou Lu Yuan-Yuan Chen Li Liu Hui Wang Yun He Yu-Huang Zheng Ling-Hua Li Jin-Feng Chen Jian-Hua Yu Biao Zhu Min Zhao Yong-Tao Sun Wen-Hui Lun Wei Xia Li-Jun Sun Li-Li Dai Tai-Yi Jiang Mei-Xia Wang Qing-Shan Zheng Hai-Yan Peng Yao Wang Rong-Jian Lu Jian-Hua Hu Hui Xing Yi-Ming Shao Dong Xie Tong Zhang Fu-Jie Zhang Hao Wu TALENT Study Team | 2020 | Chinese Medical Journal2020,,24: | 10 |

| 16 | Psychological status and diabetes-related distress of Chinese type 1 diabetes patients in Jiangsu province, China显示文摘We sought to assess the psychological status and diabetes-related distress of Chinese type 1 diabetes patients in Jiangsu province,China.According to the World Health Organization criteria,42 patients with type 1 diabetes were enrolled in the study and assessed with the Self-Rating Anxiety Scale(SAS),Self-Rating Depression Scale(SDS)and Diabetes Distress Scale(DDS).All data were tabulated and statistical analyses were performed.The study summarized cases of 42 patients with type 1 diabetes,including 17 males and 25 females with a mean age of 23±12 years and a mean duration of diabetes of 7±6 years.Compared to the Chinese normative data,the SAS standard score was significantly higher,whereas SDS standard score had no statistical significance.The SAS standard score was most highly correlated with diabetes duration(γ=0.547,P=0.011).Additionally,19.5%of the patients had moderate or even severe diabetes-related distress and 21.4%had moderate or even severe emotional burden while 26.2%had regimen-related distress.Multiple stepwise regression analysis showed that the mean correlation between DDS and the four domains was high,particularly the emotional burden domain(estimatedβ=0.363,P<0.001)and regimen-related distress domain(estimatedβ=0.356,P<0.001).The correlation between SAS and DDS was positive(estimatedβ=0.039,P=0.027).In conclusion,the results showed the importance of psychological aspects in Chinese individuals with type 1 diabetes.Screening and treatment of psychological aspects may result in better adherence and increased quality of life for patients with diabetes. | Lulu Kong Yun Cai Gang Mei Rong Gu Xiaojiao Zhang Yao Qin Ya Cai Yan Li Hongwen Zhou Mei Zhang Tao Yang | 2013 | The Journal of Biomedical Research2013,27,5: | 9 |

| 18 | How do Chinese medicines that tonify the kidney inhibit dopaminergic neuron apoptosis?显示文摘Wistar rats were intragastrically perfused with Chinese medicines used for tonifying the kidney.These included 0.180 g/mL of Herba Epimedii(Epimedium), Semen Cuscutae(Dodder Seed), or Herba Cistanches(Desertliving Cistanche), 0.04 mg/mL monoamine oxidase-B inhibitor selegiline,or distilled water for 14 consecutive days to prepare drug-containing serum or blank serum.MES23.5 cells in the logarithmic phase were cultured in media supplemented with 15%drug-containing serum for 24 hours, followed by incubation in culture solution containing 100 μmol/L H2O2for 3 hours. 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MTT) and flow cytometry results showed that all drug-containing serums improved the survival rate of H2O2-injured MES23.5 cells, inhibited pro-apoptotic FasL and caspase-3 expression, promoted anti-apoptotic Bcl-2 expression. However, drug-containing serums had little influence on Fas expression in H2O2-injured MES23.5 cells. Enzyme-linked immunosorbent assay results showed that serum containing Herba Cistanches or Herba Epimedii increased the expression of nerve growth factor,brain-derived neurotrophic factor, and glial cell line-derived neurotrophic factor in injured MES23.5cells; serum containing Semen Cuscutae only increased brain-derived neurotrophic factor expression; while expression of the above neurotrophic factors remained the same in cells treated with serum containing selegiline. | 20 | Novel Ru - K/Carbon Nanotubes Catalyst for Ammonia Synthesis显示文摘A novel ammonia synthesis catalyst, potassium-promoted ruthenium supported on carbon nanotubes, was developed. It was found that the Ru-K/carbon nanotubes catalyst had higher activity for ammonia synthesis (20.85 ml NH3/h/g-cat ) than the Ru-K/fullerenes (13.3 ml NH3/h/g-cat ) at atmospheric pressure and 623 K. The catalyst had activity even at 473 K. and had the highest activity (23.46 ml NH3/h/g-cat ) at 643 K.
