| 2 | Engineering Broad-Spectrum Bacterial Blight Resistance by Simultaneously Disrupting Variable TALE-Binding Elements of Multiple Susceptibility Genes in Rice显示文摘Xanthomonas oryzae pv.oryzae(Xoo),the causal agent of bacterial blight of rice,employs the transcription activator-like effectors(TALEs)to induce the expression of the OsSWEET family of putative sugar transporter genes,which function in conferring disease susceptibility(S)in rice plants.To engineer broadspectrum bacterial blight resistance,we used CRISPR/Cas9-mediated gene editing to disrupt the TALEbinding elements(EBEs)of two S genes,OsSWEETH and OsSWEET14,in rice cv.Kitaake,which harbors the recessive resistance allele of Xa25/OsSWEET13.The engineered rice line MS14K exhibited broadspectrum resistance to most Xoo strains with a few exceptions,suggesting that the compatible strains may contain new TALEs.We identified two PthXo2-like TALEs,Tal5LN18 and Tal7PX061,as major virulence factors in the compatible Xoo strains LN18 and PX061,respectively,and found that Xoo encodes at least five types of PthXo2-like effectors.Given that PthXo2/PthXo2.1 target OsSlVEETf3 for transcriptional activation,the genomes of 3000 rice varieties were analyzed for EBE variationsin the OsSWEET13 promoter,and 10Xa25-like haplotypes were identified.We found that Tal5LN18 and Tal7PX〇6i bind slightly different EBE sequences in the OsSWEET13 promoter to activate its expression.CRISPR/Cas9 technology was then used to generate InDels in the EBE of the OsSWEET13 promoter in MS14K to creat a new germplasm with three edited OsSWEET EBEs and broad-spectrum resistance against all Xoo strains tested.Collectively,our findings illustrate how to disarm TALE-S co-evolved loci to generate broad-spectrum resistance through the loss of effector-triggered susceptibility in plants. | Zhengyin Xu Xiameng Xu Qiang Gong Ziyang Li Ying Li Sai Wang Yangyang Yang Wenxiu Ma Longyu Liu Bo Zhu Lifang Zou Gongyou Chen | 2019 | Molecular Plant2019,12,11: | 29 |
| 3 | Fe2N nanoparticles boosting FeNx moieties for highly efficient oxygen reduction reaction in Fe-N-C porous catalyst显示文摘Replacing Pt-based electrocatalysts for the oxygen reduction reaction (ORR) with high performance and low-cost non-precious metal catalysts is crucial for the commercialization of fuel cells.Herein,we present a highly efficient Fe-N-C porous ORR electrocatalyst with FeNx moieties promoted by Fe2N nanoparticles derived from Fe-doped zeolitic imidazolate framework.The best-performing Fe-N-C/HPC-NH3 catalyst exhibits a superior ORR activity with an onset (E0) and half-wave (E1/2) potential of 0.945 and 0.803 V (RHE),respectively,which is comparable to those of the commercial Pt/C in acidic media.Probing and acid-leaching experiments prove that FeNx moieties play an important role in the ORR and the Fe2N can further improve the ORR activity.Density functional theory calculation reveals a synergistic effect that the existence of Fe2N weakens the adsorption of ORR intermediates on active sites and lowers the reaction free energy of the potential limiting step,thus facilitating the ORR.Both experimental evidence and theoretical analysis for the enhancement of ORR activity by Fe2N decoration in Fe-N-C catalyst might inspire a new strategy for rational design of high performance non-precious metal catalysts. | Xiao Liu Hong Liu Chi Chen Liangliang Zou Yuan Li Qing Zhang Bo Yang Zhiqing Zou Hui Yang | 2019 | Nano Research2019,12,7: | 14 |
| 4 | End-stage liver disease score and future liver remnant volume predict post-hepatectomy liver failure in hepatocellular carcinoma显示文摘BACKGROUND Hepatocellular carcinoma(HCC)is the world’s sixth most common malignant tumor and the third cause of cancer death.Although great progress has been made in hepatectomy,it is still associated with a certain degree of risk of posthepatectomy liver failure(PHLF),which extends the length of hospital stay and remains the leading cause of postoperative death.Studies have shown that assessment of hepatic functional reserve before hepatectomy is beneficial for reducing the incidence of PHLF.AIM To assess the value of model for end-stage liver disease(MELD)score combined with standardized future liver remnant(sFLR)volume in predicting PHLF in patients undergoing hepatectomy for HCC.METHODS This study was attended by 238 patients with HCC who underwent hepatectomy between January 2015 and January 2018.Discrimination of sFLR volume,MELD score,and sFLR/MELD ratio to predict PHLF was evaluated according to the area under the receiver operating characteristic curve.RESULTS The patients were divided into two groups according to whether PHLF occurred after hepatectomy.The incidence of PHLF was 8.4%in our research.The incidence of PHLF increased with the decrease in sFLR volume and the increase in MELD score.Both sFLR volume and MELD score were considered independent predictive factors for PHLF.Moreover,the cut-off value of the sFLR/MELD score to predict PHLF was 0.078(P<0.001).This suggests that an sFLR/MELD≥0.078 indicates a higher incidence of PHLF than an sFLR/MELD<0.078.CONCLUSION MELD combined with sFLR is a reliable and effective PHLF predictor,which is superior to MELD score or sFLR volume alone. | Fan-Hua Kong Xiong-Ying Miao Heng Zou Li Xiong Yu Wen Bo Chen Xi Liu Jiang-Jiao Zhou | 2019 | World Journal of Clinical Cases2019,7,22: | 14 |

| 6 | Ciliary parathyroid hormone signaling activates transforming growth factor-βto maintain intervertebral disc homeostasis during aging显示文摘Degenerative disc disease(DDD) is associated with intervertebral disc degeneration of spinal instability. Here, we report that the cilia of nucleus pulposus(NP) cells mediate mechanotransduction to maintain anabolic activity in the discs. We found that mechanical stress promotes transport of parathyroid hormone 1 receptor(PTH1 R) to the cilia and enhances parathyroid hormone(PTH) signaling in NP cells. PTH induces transcription of integrin α_vβ_6 to activate the transforming growth factor(TGF)-β-connective tissue growth factor(CCN2)-matrix proteins signaling cascade. Intermittent injection of PTH(iPTH) effectively attenuates disc degeneration of aged mice by direct signaling through NP cells, specifically improving intervertebral disc height and volume by increasing levels of TGF-β activity, CCN2, and aggrecan. PTH1 R is expressed in both mouse and human NP cells. Importantly,knockout PTH1 R or cilia in the NP cells results in significant disc degeneration and blunts the effect of PTH on attenuation of aged discs. | 7 | Insight-HXMT observations of the first binary neutron star merger GW170817显示文摘Finding the electromagnetic(EM) counterpart of binary compact star merger, especially the binary neutron star(BNS) merger,is critically important for gravitational wave(GW) astronomy, cosmology and fundamental physics. On Aug. 17, 2017,Advanced LIGO and Fermi/GBM independently triggered the first BNS merger, GW170817, and its high energy EM counterpart,GRB 170817 A, respectively, resulting in a global observation campaign covering gamma-ray, X-ray, UV, optical, IR, radio as well as neutrinos. The High Energy X-ray telescope(HE) onboard Insight-HXMT(Hard X-ray Modulation Telescope) is the unique high-energy gamma-ray telescope that monitored the entire GW localization area and especially the optical counterpart(SSS17 a/AT2017 gfo) with very large collection area(~1000 cm^2) and microsecond time resolution in 0.2-5 MeV. In addition,Insight-HXMT quickly implemented a Target of Opportunity(ToO) observation to scan the GW localization area for potential X-ray emission from the GW source. Although Insight-HXMT did not detect any significant high energy(0.2-5 MeV) radiation from GW170817, its observation helped to confirm the unexpected weak and soft nature of GRB 170817 A. Meanwhile,Insight-HXMT/HE provides one of the most stringent constraints(~10^(-7) to 10^(-6) erg/cm^2/s) for both GRB170817 A and any other possible precursor or extended emissions in 0.2-5 MeV, which help us to better understand the properties of EM radiation from this BNS merger. | 9 | R-CHOP regimen can significantly decrease the risk of disease relapse and progression in patients with non-germinal centerB-cell subtype diffuse large B-cell lymphoma显示文摘To further explore the role of rituximab when added to the CHOP-like regimen in the treatment of immunohistochemically defined non-germinal center B-cell subtype (non-GCB) diffuse large B-celllymphoma (DLBCL), 159 newly diagnosed DLBCL patients were studied retrospectively based on the immunohistochemical evaluation of CD10, Bcl-6, MUM-1, and Bcl-2. Altogether, 110 patients underwent the CHOP-like regimen, and rituximab was added for the other 49 patients. Cox regression analysis showed that compared with the CHOP-like regimen, the rituximab-based regimen (R-CHOP regimen)significantly decreased the risk of disease relapse and progression in CD10-negative patients (P=0.001),Bcl-6-negative patients (P=0.01), and MUM-1-positive patients (P=0.003). The risk of disease relapse in patients with non-GCB subtype (P=0.002) also decreased. In contrast, patients with the opposite immunohistochemical marker expression profile and GCB subtype did not benefit from treatment with the R-CHOP regimen. In addition, non-GCB subtype patients had a significantly higher expression rate of Bcl-2 than GCB subtype patients (P=0.042). Although univariate analysis found that both Bcl-2-positive and-negative patients had significantly higher event-free survival rates with the R-CHOP regimen, only Bcl-2 positivity (P=0.004) maintained significance in the Cox regression analysis. | 10 | Effects of targeting magnetic drug nanoparticles on human cholangiocarcinoma xenografts in nude mice显示文摘BACKGROUND:Targeting is a new therapeutic tool for malignant tumor as a result of combining nanotechnology with chemotherapeutics.The aim of our study was to investigate the effects of magnetic nanoparticles enveloping a chemotherapeutic drug on human cholangiocarcinoma xenografts in nude mice. METHODS:The human cholangiocarcinoma xenograft model was established in nude mice with the QBC939 cell line.The nude mice were randomly assigned to 7 groups. 0.9%saline or magnetic nanoparticles,including high (group 2),medium(group 4)and low(group 5)dosages, were given to nude mice through the tail vein 20 days after the QBC939 cell line was implanted.Calculations were made 35 days after treatment in order to compare the volumes,inhibition ratios and growth curves of the tumors in each group.Mice in each group were sacrificed randomly to collect tumor tissues and other organs for electron microscopy and pathological examination. RESULTS:The high and medium dosage groups were significantly different from the control group(P<0.05). The tumor inhibition ratios for the high,medium and low dosage groups were 39.6%,14.6%and 7.9%,respectively. The morphology, chemical structure and dispersity of the modified graphene were characterized with SEM, FTIR, Raman, and XPS methods.In addition, the heat-resistance behavior, mechanical properties, thermal conductivity, and electrical conductivity of the RTV-2 silicone rubber were also studied systematically. The results showed that the SiO_2 nanoparticles were coated on graphene oxide successfully, and HM-SiO_2-G was uniformly dispersed in RTV-2 silicone rubber. | 12 | The FGOALS climate system model as a modeling tool for supporting climate sciences:An overview显示文摘Climate system models are useful tools for understanding the interactions among the components of the climate system and predicting/projecting future climate change. The development of climate models has been a central focus of the State Key Laboratory of Numerical Modeling for Atmospheric Sciences and Geophysical Fluid Dynamics, Institute of Atmospheric Physics, Chinese Academy of Sciences(LASG/IAP) since the establishment of the laboratory in 1985. In China, many pioneering component models and fully coupled models of the climate system have been developed by LASG/IAP. The fully coupled climate system developed in the recent decade is named FGOALS(Flexible Global Ocean-Atmosphere-Land System Model). In this paper, an application-oriented review of the LASG/IAP FGOALS model is presented. The improved model performances are demonstrated in the context of cloud-radiation processes, Asian monsoon, ENSO phenomena, Atlantic Meridional Overturning Circulation(AMOC) and sea ice. The FGOALS model has contributed to both CMIP5(Coupled Model Intercomparison Project-phase 5) and IPCC(Intergovernmental Panel on Climate Change) AR5(the Fifth Assessment Report). The release of FGOALS data has supported the publication of nearly 500 papers around the world. The results of FGOALS are cited ~106 times in the IPCC WG1(Working Group 1) AR5. In addition to the traditional long-term simulations and projections, near-term decadal climate prediction is a new set of CMIP experiment, progress of LAGS/IAP in the development of nearterm decadal prediction system is reviewed. The FGOALS model has supported many Chinese national-level research projects and contributed to the national climate change assessment report. | 13 | Ruminal fermentation and microbial community differently influenced by four typical subtropical forages in vitro显示文摘The present study evaluated the effects of 4 typical subtropical forages on ruminal microbial community composition to formulate a better diet for buffalo.Corn straw silage,elephant grass,cassava residues and sugarcane tail silage were used as substrates for in vitro fermentation.Eight replicates were set up for every substrate,and fermentation was carried out in a 100-m L glass syringe,using buffalo rumen inoculum.Every replicate was anaerobically dispensed with 10 m L of rumen inoculum,20 m L of Mc Dougall's buffer and 200 mg of dried substrate,and placed in a water bath at 39C.Gas production was recorded at 0,2,6,12,24,36,48 and 72 h of incubation.After 24 h,fermentation was ceased for 4 replicates and samples were collected.Volatile fatty acids(VFA) concentrations were measured using gas chromatography.Microbial populations were quantified using quantitative real-time PCR(q RT-PCR),and microbial community was analyzed using high throughput sequencing technology.The results showed,cassava residues as substrate had the highest gas production,acetate,propionate and total VFA concentrations(P < 0.05),and corn straw silage had the lowest acetate:propionate ratio(P < 0.05).The lowest numbers of fungi,Ruminococcus albus and Fibrobacter succinogenes,and the highest number of protozoa were observed with cassava residues(P < 0.05).The least abundances of bacterial phyla Firmicutes,Bacteroidetes and genus Prevotella,and substantially higher abundance of phylum proteobacteria(56%) and genus Succinivibrio(52%) were observed with cassava residues.The most abundances of Methanobrevibacter gottschalkii and Entodinium were observed with cassava residues.Spearman's correlations analysis showed,Succinivibrio had strong positive correlations with propionate,butyrate,Metadinium and M.gottschalkii,indicating fermentation products were related to microbial community.In conclusion,incubation with cassava residues resulted in lower number of fiber degrading microbes but higher protozoal population because of its low fiber contents.The microbial community was highly altered by in vitro incubation with cassava residues,whereas remained similar for the other 3 high fiber containing substrates. | Muhammad W.Iqbal Qin Zhang Yingbai Yang Caixia Zou Lili Li Xin Liang Shengju Wei Bo Lin | 2018 | Animal Nutrition2018,,1: | 6 |

| 15 | Recent progress in human body energy harvesting for smart bioelectronic system显示文摘From every heartbeat to every footstep, human beings dissipate energy all the time. Researchers are trying to harvest energy from the human body and convert it into electricity, which can be supplied to electronic medical devices closely related to human health. Such an energy recycling form is currently a research hotspot in the fields of energy harvesting and bioelectronics. This review firstly summarizes the distribution and characteristics of three primary energy sources contained in the human body, including thermal energy, chemical energy, and mechanical energy. Afterwards, the applicable energy harvesting technologies and corresponding working mechanisms for different energy sources are introduced. Some typical demos and practical applications of each type of human body energy harvesting technology are also presented. Specifically, the advantages and critical issues of different energy harvesting technologies are summarized, and corresponding promising solutions are also provided. Besides, the interaction strategies between various energy harvesting devices and the human body are summarized from the aspects of wearable and implantable applications. Finally, the concept of a self-powered closed-loop bioelectronic system (SCBS) is put forward for the first time, which organically combines portable electronic devices, implantable electronic medical devices, energy harvesting devices, and the human body. The prospect of symbiosis between the SCBS and the human body is provided. | 18 | Data processing and initial results from the CE-3 Extreme Ultraviolet Camera显示文摘The Extreme Ultraviolet Camera(EUVC) onboard the Chang'e-3(CE-3)lander is used to observe the structure and dynamics of Earth's plasmasphere from the Moon. By detecting the resonance line emission of helium ions(He+) at 30.4 nm, the EUVC images the entire plasmasphere with a time resolution of 10 min and a spatial resolution of about 0.1 Earth radius(RE) in a single frame. We first present details about the data processing from EUVC and the data acquisition in the commissioning phase, and then report some initial results, which reflect the basic features of the plasmasphere well.
