| 1 | Sirtuin 3 deficiency exacerbates diabetic cardiomyopathy via necroptosis enhancement and NLRP3 activation显示文摘Sirtuin 3(SIRT3)is a potential therapeutic target for cardiovascular,metabolic,and other aging-related diseases.In this study,we investigated the role of SIRT3 in diabetic cardiomyopathy(DCM).Mice were injected with streptozotocin(STZ,60 mg/kg,ip)to induce diabetes mellitus.Our proteomics analysis revealed that SIRT3 expression in the myocardium of diabetic mice was lower than that of control mice,as subsequently confirmed by real-time PCR and Western blotting.To explore the role of SIRT3 in DCM,SIRT3-knockout mice and 129S1/SvImJ wild-type mice were injected with STZ.We found that diabetic mice with SIRT3 deficiency exhibited aggravated cardiac dysfunction,increased lactate dehydrogenase(LDH)level in the serum,decreased adenosine triphosphate(ATP)level in the myocardium,exacerbated myocardial injury,and promoted myocardial reactive oxygen species(ROS)accumulation.Neonatal rat cardiomyocytes were transfected with SIRT3 siRNA,then exposed to high glucose(HG,25.5 mM).We found that downregulation of SIRT3 further increased LDH release,decreased ATP level,suppressed the mitochondrial membrane potential,and elevated oxidative stress in HG-treated cardiomyocytes.SIRT3 deficiency further raised expression of necroptosis-related proteins including receptor-interacting protein kinase 1(RIPK1),RIPK3,and cleaved caspase 3,and upregulated the expression of inflammation-related proteins including NLR family pyrin domain-containing protein 3(NLRP3),caspase 1 p20,and interleukin-1βboth in vitro and in vivo.Collectively,SIRT3 deficiency aggravated hyperglycemia-induced mitochondrial damage,increased ROS accumulation,promoted necroptosis,possibly activated the NLRP3 inflammasome,and ultimately exacerbated DCM in the mice.These results suggest that SIRT3 can be a molecular intervention target for the prevention and treatment of DCM. | Shu Song Yue Ding Guo-liang Dai Yue Zhang Meng-ting Xu Jie-ru Shen Ting-ting Chen Yun Chen Guo-liang Meng | 2021 | Acta Pharmacologica Sinica2021,42,2: | 14 |

| 3 | Insight-HXMT observations of the first binary neutron star merger GW170817显示文摘Finding the electromagnetic(EM) counterpart of binary compact star merger, especially the binary neutron star(BNS) merger,is critically important for gravitational wave(GW) astronomy, cosmology and fundamental physics. On Aug. 17, 2017,Advanced LIGO and Fermi/GBM independently triggered the first BNS merger, GW170817, and its high energy EM counterpart,GRB 170817 A, respectively, resulting in a global observation campaign covering gamma-ray, X-ray, UV, optical, IR, radio as well as neutrinos. The High Energy X-ray telescope(HE) onboard Insight-HXMT(Hard X-ray Modulation Telescope) is the unique high-energy gamma-ray telescope that monitored the entire GW localization area and especially the optical counterpart(SSS17 a/AT2017 gfo) with very large collection area(~1000 cm^2) and microsecond time resolution in 0.2-5 MeV. In addition,Insight-HXMT quickly implemented a Target of Opportunity(ToO) observation to scan the GW localization area for potential X-ray emission from the GW source. Although Insight-HXMT did not detect any significant high energy(0.2-5 MeV) radiation from GW170817, its observation helped to confirm the unexpected weak and soft nature of GRB 170817 A. Meanwhile,Insight-HXMT/HE provides one of the most stringent constraints(~10^(-7) to 10^(-6) erg/cm^2/s) for both GRB170817 A and any other possible precursor or extended emissions in 0.2-5 MeV, which help us to better understand the properties of EM radiation from this BNS merger. Although metal-organic framework (MOF) scaffolds are promisingstrategies for bone tissue regeneration, their potential use for exosome loading remains unexplored. In this study,motivated by the potential advantages of hADSCs-Exos and Mg-GA MOF, we designed and synthesized anexosome-functionalized cell-free PLGA/Mg-GA MOF (PLGA/Exo-Mg-GA MOF) scaffold, taking using of thebenefits of hADSCs-Exos, Mg2+, and gallic acid (GA) to construct unique nanostructural interfaces to enhanceosteogenic, angiogenic and anti-inflammatory capabilities simultaneously. Our in vitro work demonstrated thebeneficial effects of PLGA/Exo-Mg-GA MOF composite scaffolds on the osteogenic effects in human bonemarrow-derived mesenchymal stem cells (hBMSCs) and angiogenic effects in human umbilical endothelial cells(HUVECs). Slowly released hADSCs-Exos from composite scaffolds were phagocytosed by co-cultured cells,stabilized the bone graft environment, ensured blood supply, promoted osteogenic differentiation, and acceleratedbone reconstruction. Furthermore, our in vivo experiments with rat calvarial defect model showed thatPLGA/Exo-Mg-GA MOF scaffolds promoted new bone formation and satisfactory osseointegration. | 5 | Hyperbaric oxygen improves functional recovery of rats after spinal cord injury via activating stromal cell-derived factor-1/CXC chemokine receptor 4 axis and promoting brain-derived neurothrophic factor expression显示文摘Background: Spinal cord injury (SCI) is a worldwide medical concern. This study aimed to elucidate the mechanism underlying protective effect of hyperbaric oxygen (HBO) against SCI-induced neurologic defects in rats via exploring the stromal cell-derived factor-1 (SDF-1)/CXC chemokine receptor 4 (CXCR4) axis and expression of brain-derived neurotrophic factor (BDNF). Methods: An acute SCI rat model was established in Sprague-Dawley rats using the Allen method. Sixty rats were divided into four groups (w = 15 in each group): sham-operated, SCI, SCI treated with HBO (SCI + HBO), and SCI treated with both HBO and AMD3100 (an antagonist of CXCR4;SCI + HBO + AMD) groups. The rats were treated with HBO twice a day for 3 days and thereafter once a day after the surgery for up to 28 days. Following the surgery, neurologic assessments were performed with the Basso-Bettie-Bresnahan (BBB) scoring system on postoperative day (POD) 7, 14, 21, and 28. Spinal cord tissues were harvested to assess the expression of SDF-1, CXCR4, and BDNF at mRNA and protein levels, using quantitative real-time polymerase chain reaction, Western blot analysis, and histopathologic analysis. Results: HBO treatment recovered SCI-induced descent of BBB scores on POD 14,(1.25±0.75 vs. 1.03 ±0.66, P< 0.05), 21 (5.27± 0.89 vs. 2.56± 1.24, P< 0.05), and 28 (11.35±0.56 vs. 4.23± 1.20, P<0.05) compared with the SCI group. Significant differences were found in the mRNA levels of SDF-1 (mRNA: day 21, SCI + HBO vs. SCI + HBO + AMD, 2.89± 1.60 vs. 1.56±0.98, P<0.05), CXCR4 (mRNA: day 7, SCI + HBO vs. SCI, 2.99± 1.60 vs. 1.31 ±0.98, P<0.05;day 14, SCI + HBO vs. SCI + HBO + AMD, 4.18± 1.60 vs. 0.80±0.34, P<0.05;day 21, SCI + HBO vs. SCI, 2.10±1.01 vs.1.15±0.03, P<0.05), and BDNF (mRNA: day 7, SCI + HBO vs. SCI, 3.04±0.41 vs. 2.75±0.31, P<0.05;day 14, SCI + HBO vs. SCI, 3.88± 1.59 vs. 1.11 ±0.40, P<0.05), indicating the involvement of SDF-1/CXCR4 axis in the protective effect of HBO. | 9 | Comparison of Ozone and PM2.5 Concentrations over Urban,Suburban,and Background Sites in China显示文摘Surface ozone(O3)and fine particulate matter(PM2.5)are dominant air pollutants in China.Concentrations of these pollutants can show significant differences between urban and nonurban areas.However,such contrast has never been explored on the country level.This study investigates the spatiotemporal characteristics of urban-to-suburban and urban-tobackground difference for O3(Δ[O3])and PM2.5(Δ[PM2.5])concentrations in China using monitoring data from 1171 urban,110 suburban,and 15 background sites built by the China National Environmental Monitoring Center(CNEMC).On the annual mean basis,the urban-to-suburbanΔ[O3]is−3.7 ppbv in Beijing-Tianjin-Hebei,1.0 ppbv in the Yangtze River Delta,−3.5 ppbv in the Pearl River Delta,and−3.8 ppbv in the Sichuan Basin.On the contrary,the urban-to-suburbanΔ[PM2.5]is 15.8,−0.3,3.5 and 2.4μg m^−3 in those areas,respectively.The urban-to-suburban contrast is more significant in winter for bothΔ[O3]andΔ[PM2.5].In eastern China,urban-to-background differences are also moderate during summer,with−5.1 to 6.8 ppbv forΔ[O3]and−0.1 to 22.5μg m^−3 forΔ[PM2.5].However,such contrasts are much larger in winter,with−22.2 to 5.5 ppbv forΔ[O3]and 3.1 to 82.3μg m^−3 forΔ[PM2.5].Since the urban region accounts for only 2%of the whole country’s area,the urban-dominant air quality data from the CNEMC network may overestimate winter[PM2.5]but underestimate winter[O3]over the vast domain of China.The study suggests that the CNEMC monitoring data should be used with caution for evaluating chemical models and assessing ecosystem health,which require more data outside urban areas. | Lan GAO Xu YUE Xiaoyan MENG Li DU Yadong LEI Chenguang TIAN Liang QIU | 2020 | Advances in Atmospheric Sciences2020,37,12: | 5 |
| 10 | Effect of peroral esophageal myotomy for achalasia treatment: A Chinese study显示文摘AIM: To assess the safety and feasibility of peroral esophageal myotomy(POEM) in patients with achalasia.METHODS: From January 2012 to March 2014, 50 patients(28 men, 22 women; mean age: 42.8 years, range: 14-70 years) underwent POEM. Pre- and postoperative symptoms were quantified using the Eckardt scoring system. Barium swallow and esophagogastroscopy were performed before and after POEM, respectively. Esophageal motility was evaluated in all patients, both preoperatively and one month after POEM treatment, using a high-resolution manometry system. Manometry data, Eckardt scores, lower esophageal sphincter pressure and barium swallow results were used to evaluate the effect of the procedure.RESULTS: POEM was successfully completed for all patients. The mean procedure time was 55.4 ± 17.3 min and the mean total length of myotomy of the circular esophagus was 10.5 ± 2.6 cm. No specific complications occurred, with the exception of two patients that developed as ymptomatic pneumomediastinum and subcutaneous emphysema. Clinical improvement in symptoms was achieved in all patients. Approximately 77.5% of patients experienced weight gain 6 mo after POEM, with an average of 4.78 kg(range: 2-15 kg). The lower esophageal sphincter resting pressure, four second integrated relaxation pressure and Eckardt scores were all significantly reduced after POEM(Ps < 0.05). A small segment of proximal esophageal peristalsis appeared postoperatively in two patients, but without normal esophageal peristalsis. The average diameter of the esophageal lumen decreased significantly from 4.39 to 3.09 cm(P < 0.01). | 13 | Impact of Enterprise Ownership and Size on Registered Hematotoxicity in Benzene-exposed Workers显示文摘Benzene is a classified as a human carcinogen that can cause myelodysplastic syndrome and acute myelogenous leukemia[1]. This compound is also a potent toxin suppressing the bone marrow. Accordingly, benzene causes serious hematological adverse effects, such as pancytopenia and aplastic anemia. In the 1920s, the carcinogenicity of benzene was identified in humans, in which it was shown to cause acute myelocytic leukemia (AML)[2]. The reports highlighting the first cases of hematopoietic disease associated with benzene appeared in China in the late 1950s[3]. | 14 | Genetic structure of needle morphological and anatomical traits of Pinus yunnanensis显示文摘Pinus yunnanensis Franch. is an particular conifer tree species in Yunnan–Guizhou plateau in southwest China. The morphological and anatomical traits of needles are important to evaluate geographic variation and population dynamics of conifer species. Seedlings from seven populations of P. yunnanensis were analyzed, looking at 22 morphological and anatomical needle traits. The results showed that variations among and within populations were significantly different for all traits and the variance components within populations were generally higher than that among populations in the most tested needle traits. The proportions of three-needle fascicle were significantly different among populations. The traits related to needle size in both morphology and anatomy were positive with latitude and negative with annual temperature and precipitation. Ratio indices, including mesophyllarea/vascular bundle area, mesophyll area/resin canals area, vascular bundle area/resin canals area and mesophyll area/(resin canals area and vascular bundle area), were negatively correlated with elevation and positively correlated with the annual mean temperature, showing some fitness feature for the populations. Needle traits were more significantly correlated with longitude than with other four environmental factors. Needle length was significantly correlated with almost all environmental factors. First four principal components accounted for 81.596 % of the variation with eigenvalues [1; the differences among populations were mainly dependent on needle width,stomatal density, section areas of vascular bundle, total resin canals, and mesophyll, as well as area ratio traits.Seven populations were divided into three categories by Euclidean distance. Variations in needle traits among the populations have shown systematic microevolution in terms of geographic impact on P. yunnanensis. | 17 | Ecological risk assessment of the Gannan Plateau,northeastern Tibetan Plateau显示文摘Desertification, soil salinization and grassland degradation are the major environmental hazards faced by the Gannan Plateau, northeastern Tibetan Plateau. Ecological risk assessment plays an important role in formulating environmental management strategies yet little attention to this region. In this study, we established an ecological risk assessment index system based on 30 evaluation indices in the categories of hydrometeorology, ecological environment, ground surface disturbance, and society and economy for the Gannan Plateau. An entropy method was used to calculate an index weight,and subsequently the matter-element method was used together with extension theory to establish a matter-element extension model of ecological risk. We assessed the ecological risk in this region by calculating the degree of association between index layer, system layer and target layer, and the cumulative ecological risk index. The degrees of ecological risk for the counties of the region were determined by using Arc GIS which would represent a spatial heterogeneity of the risk grade in production. Our results showed that the areas of high ecological risk were in Zhouqu County and Zhuoni County, and others were of low risk(Hezuo City, Diebu County, Xiahe County and Lintan County) or intermediate risk(Maqu County). The results of the assessment were in accord with the actual observed situation. Thus, our ecological risk assessment index system is appropriate for this region and suggests that high risk counties need a priori ecological protection. | 19 | Macrophage exosomes transfer angiotensin Ⅱ type 1 receptor to lung fibroblasts mediating bleomycin-induced pulmonary fibrosis显示文摘Background:Macrophages are involved in the pathogenesis of idiopathic pulmonary fibrosis,partially by activating lung fibroblasts.However,how macrophages communicate with lung fibroblasts is largely unexplored.Exosomes can mediate intercellular communication,whereas its role in lung fibrogenesis is unclear.Here we aim to investigate whether exosomes can mediate the crosstalk between macrophages and lung fibroblasts and subsequently induce fibrosis.Methods:In vivo,bleomycin(BLM)-induced lung fibrosis model was established and macrophages infiltration was examined.The effects of GW4869,an exosomes inhibitor,on lung fibrosis were assessed.Moreover,macrophage exosomes were injected into mice to observe its pro-fibrotic effects.In vitro,exosomes derived from angiotensin Ⅱ(Ang Ⅱ)-stimulated macrophages were collected.Then,lung fibroblasts were treated with the exosomes.Twenty-four hours later,protein levels ofα-collagen I,angiotensin Ⅱ type 1 receptor(AT1R),transforming growth factor-β(TGF-β),and phospho-Smad2/3(p-Smad2/3)in lung fibroblasts were examined.The Student's t test or analysis of variance were used for statistical analysis.Results:In vivo,BLM-treated mice showed enhanced infiltration of macrophages,increased fibrotic alterations,and higher levels of Ang Ⅱ and AT1R.GW4869 attenuated BLM-induced pulmonary fibrosis.Mice with exosomes injection showed fibrotic features with higher levels of Ang Ⅱ and AT1R,which was reversed by irbesartan.In vitro,we found that macrophages secreted a great number of exosomes.The exosomes were taken by fibroblasts and resulted in higher levels of AT1R(0.22±0.02 vs.0.07±0.02,t=8.66,P=0.001),TGF-β(0.54±0.05 vs.0.09±0.06,t=10.00,P<0.001),p-Smad2/3(0.58±0.06 vs.0.07±0.03,t=12.86,P<0.001)andα-collagen I(0.27±0.02 vs.0.16±0.01,t=7.01,P=0.002),and increased Ang Ⅱ secretion(62.27±7.32 vs.9.56±1.68,t=12.16,P<0.001).Interestingly,Ang Ⅱ increased the number of macrophage exosomes,and the protein levels of Alix(1.45±0.15 vs.1.00±0.10,t=4.32,P=0.012),AT1R(4.05±0.64 vs.1.00±0.09,t=8.17,P=0.001),and glyceraldehyde-3-phosphate dehydrogenase(2.13±0.36 vs.1.00±0.10,t=5.28,P=0.006)were increased in exosomes secreted by the same number of macrophages,indicating a positive loop between Ang Ⅱ and exosomes production.Conclusions:Exosomes mediate intercellular communication between macrophages and fibroblasts plays an important role in BLM-induced pulmonary fibrosis. | Na-Na Sun Yue Zhang Wen-Hui Huang Bo-Jun Zheng Si-Yi Jin Xu Li Ying Meng | 2021 | Chinese Medical Journal2021,,18: | 1 |
| 20 | Structural Topology and Dynamic Response Analysis of an Electric Torque Vectoring Drive-Axle for Electric Vehicles显示文摘In-wheel motor-drive electric vehicles have the advantage of independently controllable wheel torque and the disadvantages of unsprung mass rise and power restriction.To address the disadvantages,a centralized layout electric torque vectoring drive-axle system(E-TVDS)with dual motors is proposed,which can realize arbitrary distribution of driving torque between the left and right wheels.First,the speed and torque distribution principle of E-TVDS based on velocity diagram are analyzed,and a virtual prototype of the whole vehicle with basic gear ratio relation model of the E-TVDS is built for simulation to verify the theoretical results and the basic effect of E-TVDS on the steering performance of the vehicle.Second,the charac-teristics of 36 types of the novel E-TVDS topology structure are compared and analyzed,and the optimal structure scheme is selected.Third,the accurate multiple degrees of freedom dynamic model for the optimal structure is established by using the bond graph method,and its dynamic response characteristics are analyzed.The results show that the vehicle equipped with the proposed E-TVDS can distribute the driving torque with the almost identical amount but opposite sign between the left and right wheels in any direction,and varying amount according to different chassis dynamics control requirements,and the torque response performance is great with little delay and overshoot.The function and dynamic response of the proposed E-TVDS show that it has potential application value for various performance improvements of electric vehicles. | Junnian Wang Shoulin Gao Yue Qiang Meng Xu Changyang Guan Zidong Zhou | 2022 | Automotive Innovation2022,5,2: | 1 |
