| 3 | Discrimination of False Negative Results in RT-PCR Detection of SARS-CoV-2 RNAs in Clinical Specimens by Using an Internal Reference显示文摘Reverse transcription-polymerase chain reaction(RT-PCR)is an essential method for specific diagnosis of SARS-CoV-2 infection.Unfortunately,false negative test results are often reported.In this study,we attempted to determine the principal causes leading to false negative results of RT-PCR detection of SARS-CoV-2 RNAs in respiratory tract specimens.Multiple sputum and throat swab specimens from 161 confirmed COVID-19 patients were tested with a commercialfluorescent RT-PCR kit targeting the ORF1 ab and N regions of SARS-CoV-2 genome.The RNA level of a cellular housekeeping gene ribonuclease P/MRP subunit p30(RPP30)in these specimens was also assessed by RT-PCR.Data for a total of 1052 samples were retrospectively re-analyzed and a strong association between positive results in SARS-CoV-2 RNA tests and high level of RPP30 RNA in respiratory tract specimens was revealed.By using the ROC-AUC analysis,we identified Ct cutoff values for RPP30 RT-PCR which predicted false negative results for SARS-CoV-2 RT-PCR with high sensitivity(95.03%–95.26%)and specificity(83.72%–98.55%)for respective combination of specimen type and ampli-fication reaction.Using these Ct cutoff values,false negative results could be reliably identified.Therefore,the presence of cellular materials,likely infected host cells,are essential for correct SARS-CoV-2 RNA detection by RT-PCR in patient specimens.RPP30 could serve as an indicator for cellular content,or a surrogate indicator for specimen quality.In addition,our results demonstrated that false negativity accounted for a vast majority of contradicting results in SARS-CoV-2 RNA test by RT-PCR. | Yafei Zhang Changtai Wang Mingfeng Han Jun Ye Yong Gao Zhongping Liu Tengfei He Tuantuan Li Mengyuan Xu Luping Zhou Guizhou Zou Mengji Lu Zhenhua Zhang | 2020 | Virologica Sinica2020,35,6: | 5 |

| 7 | Evaluation of Ecological Environment Quality of Typical Forest Parks显示文摘Forest tourist areas have an excellent ecological environment,which is conducive to people's physical and mental health.In this study,Fuzhou National Forest Park and Wuying National Forest Park were selected as typical representatives of the southern and northern forest parks,respectively,which were compared by qualitative and quantitative research methods from such five aspects as air quality,climate quality,water environment quality,acoustic environment quality and biological population in some forest areas.The results showed that Fuzhou National Forest Park has a concentration of negative oxygen ions much higher than Wuying National Forest Park,and is thus more conducive to people's health;and it has the characteristics of warm and comfortable climate throughout the year and high biodiversity,but the noise is 4.35 db higher than Wuying National Forest Park,and the water quality is slightly poorer than that of Wuying National Forest Park.And its DO value is about 3.89 smaller in autumn than in summer.In Wuying National Forest Park,the concentration of negative oxygen ions is 53.6% lower than Fuzhou National Forest Park,but it is also above the national standard.The climate is only comfortable in June-August,and is colder or cooler in other months.The water quality is better than that of Fuzhou National Forest Park.In particular,the PH value is about 0.38 lower than that of Fuzhou National Forest Park.It has its own unique biological characteristics.The number of tourists in summer is higher,resulting in higher noise in summer than in autumn.The two forest parks in the north and the south are both good places for people to relax and enjoy. | Dongmei ZHANG Shuzhen PENG Weijun ZHAO Mengyuan GAO Junqing HAN | 2019 | Agricultural Biotechnology2019,8,5: | 1 |

| 15 | 8.30%Efficiency P3HT-based all-polymer solar cells enabled by a miscible polymer acceptor with high energy levels and efficient electron transport显示文摘P3HT stands out from numerous polymer donors owing to the merits of low cost and high scalability of synthesis.However,the photovoltaic performance of P3HT-based blends lags significantly behind the state-of-the-art systems,especially for all-polymer solar cells(APSCs) that generally show efficiency of around 3%–4% due to the lack of matched polymer acceptors.Herein,a polymer acceptor,named IDTBTC8-CN,was designed and synthesized with indacenodithiophene(IDT) and mono-cyano(CN)-substituted benzothiadiazole(BT-CN) as building blocks.Introducing a CN group endowed the polymer with decreased bandgap,and apparent n-type charge transport character despite the relatively high energy levels.Additionally,IDTBTC8-CN showed largely improved miscibility with P3HT,compared with that of BT-based control polymer IDTBTC8.The high miscibility between P3HT and IDTBTC8-CN as well as the amorphous aggregation behavior of IDTBTC8-CN enabled a broad manipulation room for the blend film to acquire favorable morphology.Eventually,a champion efficiency of 8.30% was achieved,in sharp contrast to that of the IDTBTC8-based system(1.21%).Such efficiency is a new record for P3HT-based APSCs reported so far.Moreover,P3HT:IDTBTC8-CN blend film also exhibited excellent mechanical robustness.This study implies the guidance of molecular design of the polymer acceptors and morphology control for P3HT-based APSCs. | Ziqi Liang Jiangting He Bin Zhao Mengyuan Gao Yu Chen Long Ye Miaomiao Li Yanhou Geng | 2023 | Science China Chemistry2023,66,1: | 0 |

| 19 | Anti-apoptotic protein BCL-XL as a therapeutic vulnerability in gastric cancer显示文摘Background: New therapeutic targets are needed to improve the outcomes for gastric cancer(GC) patients with advanced disease. Evasion of programmed cell death(apoptosis) is a hallmark of cancer cells and direct induction of apoptosis by targeting the pro-survival BCL2 family proteins represents a promising therapeutic strategy for cancer treatment. Therefore, understanding the molecular mechanisms underpinning cancer cell survival could provide a molecular basis for potential therapeutic interventions. Method: Here we explored the role of BCL2L1 and the encoded anti-apoptotic BCL-XL in GC. Using Droplet Digital PCR(ddPCR) technology to investigate the DNA amplification of BCL2L1 in GC samples and GC cell lines, the sensitivity of GC cell lines to selective BCL-XL inhibitors A1155463 and A1331852, pan-inhibitor ABT-263, and VHL-based PROTAC-BCL-XL was analyzed using(CellTiter-Glo) CTG assay in vitro. Western Blot(WB) was used to detect the protein expression of BCL2 family members in GC cell lines and the manner in which PROTAC-BCL-XL kills GC cells. Coimmunoprecipitation(Co-IP) was used to investigate the mechanism of A1331852 and ABT-263 kills GC cell lines. DDPCR, WB, and real-time PCR(RTPCR) were used to investigate the correlation between DNA, RNA, protein levels, and drug activity. Results: The functional assay showed that a subset of GC cell lines relies on BCL-XL for survival. In gastric cancer cell lines, BCL-XL inhibitors A1155463 and A1331852 are more sensitive than the pan BCL2 family inhibitor ABT-263, indicating that ABT-263 is not an optimal inhibitor of BCL-XL. VHL-based PROTAC-BCL-XL DT2216 appears to be active in GC cells. DT2216 induces apoptosis of gastric cancer cells in a time-and dose-dependent manner through the proteasome pathway. Statistical analysis showed that the BCL-XL protein level predicts the response of GC cells to BCL-XL targeting therapy and BCL2L1 gene CNVs do not reliably predict BCL-XL expression.Conclusion: We identified BCL-XL as a promising therapeutic target in a subset of GC cases with high levels of BCL-XL protein expression. Functionally, we demonstrated that both selective BCL-XL inhibitors and VHL-based PROTAC BCL-XL can potently kill GC cells that are reliant on BCL-XL for survival. However, we found that BCL2L1 copy number variations(CNVs) cannot reliably predict BCL-XL expression, but the BCL-XL protein level serves as a useful biomarker for predicting the sensitivity of GC cells to BCL-XL-targeting compounds.
