1Abrogation of USP7 is an alternative strategy to downregulate PD-L1 and sensitize gastric cancer cells to T cells killing显示文摘Targeting immune checkpoints such as programmed cell death protein 1(PD-1)and programmed death ligand-1(PD-L1)have been approved for treating melanoma,gastric cancer(GC)and bladder cancer with clinical benefit.Nevertheless,many patients failed to respond to anti-PD-1/PD-L1 treatment,so it is necessary to seek an alternative strategy for traditional PD-1/PD-L1 targeting immunotherapy.Here with the data from The Cancer Genome Atlas(TCGA)and our in-house tissue library,PD-L1 expression was found to be positively correlated with the expression of ubiquitin-specific processing protease 7(USP7)in GC.Furthermore,USP7 directly interacted with PD-L1 in order to stabilize it,Gastric cancer;Immunosuppression;Cancer biologywhile abrogation of USP7 attenuated PD-L1/PD-1 interaction and sensitized cancer cells to T cell killing in vitro and in vivo.Besides,USP7 inhibitor suppressed GC cells proliferation by stabilizing P53 in vitro and in vivo.Collectively,our findings indicate that in addition to inhibiting cancer cells proliferation,USP7 inhibitor can also downregulate PD-L1 expression to enhance anti-tumor immune response simultaneously.Hence,these data posit USP7 inhibitor as an anti-proliferation agent as well as a novel therapeutic agent in PD-L1/PD-1 blockade strategy that can promote the immune response of the tumor.Zhiru Wang Wenting Kang Ouwen Li Fengyu Qi Junwei Wang Yinghua You Pengxing He Zhenhe Suo Yichao Zheng Hong-Min Liu 2021Acta Pharmaceutica Sinica B2021,11,3:5
2Melanin nanoparticles alleviate sepsis-induced myocardial injury by suppressing ferroptosis and inflammation显示文摘Myocardial injury as one of the severe complications leads to the increasing morbidity and mortality in patients with sepsis.Recent studies reported that reactive oxygen species(ROS)-mediated ferroptosis plays a critical role in the development of heart diseases.Therefore,we hypothesized that anti-ferroptosis agent might be a novel potential therapeutic strategy for sepsis-induced cardiac injury.Herein,we demonstrated that a small biocompatible and MRI-visible melanin nanoparticles(MMPP)improves myocardial function by inhibiting ROS-related ferroptosis signaling pathway.In LPS-induced murine sepsis model,after a single dose intravenously injection of MMPP treatment,MMPP markedly alleviated the myocardial injury including cardiac function and heart structure disorder through suppressing iron-accumulation induced ferroptosis.In vitro,MMPP inhibited cardiomyocyte death by attenuating oxidative stress,inflammation and maintaining mitochondrial homeostasis.Collectively,our findings demonstrated that MMPP protected heart against sepsis-induced myocardial injury via inhibiting ferroptosis and inflammation,which might be a novel therapeutic approach in future.Chang Liu Quan Zou Huixin Tang Jia Liu Shiqi Zhang Caihong Fan Junwei Zhang Ruiqing Liu Yashan Liu Ruiyan Liu Yan Zhao Qiang Wu Zhi Qi Yanna Shen 2023Bioactive Materials2023,,6:5
3PEPT1-mediated prodrug strategy for oral delivery of peramivir显示文摘Peramivir was a novel and highly potent neuraminidase(NA) inhibitor for the treatment of influenza A and B. However, it exhibited a very low oral bioavailability(only 3%) due to the high polarity(log P of-1.4) and the low membrane permeability across the intestine. To utilize the PEPT1-mediated prodrug strategy to improve the oral absorption and develop the oral alternative, seven amino acid ester prodrugs and seven amino acid amide prodrugs have been synthesized. The permeability of these prodrugs across Caco-2 cells were screened. Peramivr-(CH_2)_2-l-Val and Peramivir-l-Ile were of the highest permeability in ester prodrugs and amide prodrugs, respectively, and then they were selected for further studies. Glycylsarcosine(gly-sar) uptake by Caco-2 could be inbihited by Peramivir-(CH_2)_2-l-Val and Peramivir-l-Ile in a concentration-dependent manner, and the IC 50 was 1.34 ± 0.31 m M and 1.78 ± 0.48 m M, respectively. The direct uptake of Peramivir-(CH_2)_2-l-Val and Peramivirl-Ile in MDCK-PEPT1 cells were significantly higher than in MDCK mock cells, and could be markedly inhibited by gly-sar. The uptake of Peramivir-(CH_2)_2-l-Val and Peramivir-l-Ile(0.01 to 50 m M) in MDCK-hPEPT1 cells conformed to Michaelis–Menten Equation. The oral bioavailability of peramivir was 65.3% and 37.3% after the oral administration of Peramivir-(CH_2)_2-l-Val and Peramivir-l-Ile to rats, respectively. The oral absorption and bioactivation of Peramivir-(CH_2)_2-l-Val was rapid and extensive, and no Peramivir-(CH_2)_2-l-Val was found in plasma. Because the amide bond was relatively stable, Peramivir-l-Ile could not be totally converted to the parent drug in vivo. Peramivir-(CH_2)_2-l-Val with good oral profiles and rapid bioactivation might be a promising prodrug for the further clinic development. 4Study on the Influence of Airgun Excitation Conditions on Airgun Signals and Travel Time Variation Measurements显示文摘Because of the different excitation conditions of the airgun source,there will be subtle differences in airgun signals.Travel time variation of airgun signals often mix into source information which can’t fully reflect the evolution of the medium.This article uses the airgun signals from the Binchuan Transmitting Seismic Station to analyze the airgun signal′s characteristics of phase and correlation.We conducted a comparative analysis of the effects of the pair difference method and the deconvolution method on eliminating the influence of the excitation conditions in travel time variation.The results show that:(1)The pressure pulse and its subsequent wave of airgun source wavelet are less affected by excitation conditions that we can use it to obtain high-precision excitation moments.(2)Deconvolution can improve the correlation of the airgun signal.(3)The pair difference method can’t eliminate the influence of excitation conditions in travel time variation.Deconvolution can reduce excitation condition interference and the influence of the excitation condition in the travel time variation after deconvolution of the vertical signal is significantly reduced.XIANG Ya YANG Runhai WANG Bin ZHOU Yunyao LU Yongqing WU Huan QI Junwei PENG Jing 2019Earthquake Research in China2019,33,2:3

6Study on an automatic multi-pin insertion system for preparing Z-pin composite laminates显示文摘As a three-dimensional layer enhancement technology, the Z-pin technology is suitable for manufacturing composite prepreg laminates and can also effectively improve their interlaminar properties. In order to manufacture Z-pin composite parts efficiently, this paper proposes a transitional insertion system, and according to the technical requirements of the transitional insertion process and the functional requirements of the transitional insertion platform, proposes a transition insertion plan which can insert multiple Z-pins into a carrier. A prototype machine has been designed and made, and a series of function debugging and verification experiments have been carried out on the machine. Some problems show up during the experiments, but based on the analysis of the reasons, this paper provides lots of solutions, and finally demonstrates the feasibility and practical value of the system. The results show that the system meets all technical requirements of the Z-pin insertion process. It is capable to insert multiple Z-pins into a foam carrier; as a result, the system is as much as 5 times more efficient than existing equipment, and the deviation for the insertion distance can be controlled within 0.3 mm. 10Space Charge Suppression of Polyethylene Induced by Blending with Ethylene-butyl Acrylate Copolymer显示文摘High-voltage direct current(HVDC)power cables have been paid more attention due to the big issue concerning the main insulation materials.The residues in the industrial production process will cause space charge generation and accumulation in the materials,which has become one bottleneck to limit the development of power cables up to higher voltage levels.In this paper,the LDPE matrix was modified by three types of ethylene-butyl acrylate(EBA)copolymers(1.0 wt%)with different polarities(BA content)via melt blending to optimize the space charge behavior of the LDPE insulation.The micromorphology and structure of the blends are examined by polarized light microscope and differential scanning calorimetry.The space charge distribution is tested by the pulsed electro-acoustic method.The trap level is studied by the thermally stimulated current.The results show that EBA introduces more deep traps and decreases the shallow traps.The medium-polar EBA(16%BA content)can effectively suppress charge accumula-tion,and have the same suppressed effect on XLPE/EBA blends.This study will provide important insights into the design and development of advanced HVDC power cable applications.Jintao Zhai Weikang Li Junwei Zha Qi Cheng Xingming Bian Zhimin Dang 2020CSEE Journal of Power and Energy Systems2020,6,1:1

14BoMyrosinase plays an essential role in sulforaphane accumulation in response to selenite treatment in broccoli显示文摘Sulforaphane, a naturally specialized metabolite, plays significant roles in human disease prevention and plant defense. Myrosinase(MY) is a key gene responsible for the catalysis of sulforaphane formation, but the molecular mechanisms through which MY regulates sulforaphane biosynthesis in plants remains largely unknown. Here, we discovered that the change of sulforaphane content in broccoli sprouts caused by exogenous selenite treatments is positively related to BoMY expression. BoMY overexpression in the Arabidopsis thaliana tgg1 mutants could dramatically increase myrosinase activity and sulforaphane content in the rosette leaves of 35S::BoMY/tgg1 and rescue its phenotypes.Moreover, an obvious increase of myrosinase activity and sulforaphane content was displayed in transgenic BoMY-overexpressed broccoli lines.In addition, a 2 033 bp promoter fragment of BoMY was isolated. Yeast one-hybrid(Y1H) library screening experiment uncovered that one bHLH transcription factor, BoFAMA, could directly bind to BoMY promoter to activate its expression, which was further evidenced by Y1H assay and dual-luciferase reporter assay. BoFAMA is a selenite-responsive transcription factor that is highly expressed in broccoli leaves;its protein is solely localized to nucleus. Additionally, genetic evidence suggested that the knockdown of FAMA gene in Arabidopsis thaliana could significantly decrease sulforaphane yield by inhibiting the expression of myrosinase genes. Interestingly, exogenous selenite supply could partially restore the low level of sulforaphane content in transgenic Arabidopsis FAMA-silencing plants. 16Development of an indirect immunofluorescence assay for PCV3 antibody detection based on capsid protein显示文摘Porcine circovirus type 3(PCV3)is a novel porcine circovirus associated with porcine dermatitis and nephritis syndrome(PDNS),reproductive failure,and multisystemic inflammation.Capsid protein(Cap)encoded by PCV3 ORF2 gene has been identified as an immunogenic protein.Currently,there is no immunofluorescence assay(IFA)available for serological diagnosis.Here,the N-terminal 33 amino acids of Cap protein were predicted to serve as a PCV3 nuclear localization signal(NLS).Two types of recombinant plasmids were constructed for recombinant protein expression in 5f9 cells by using a baculovirus expression system:plasmid rvBac-Pc for full-length Cap protein expression and rvBac-Sc for Cap protein expression with a honeybee melittin signal peptide in place of the predicted NLS sequence.Expression of the nuclear localization sequences was further analyzed by IFA.Strong and specific fluorescence signals were observed in the nucleus of rvBac-Pc-transfected cells and in the cytoplasm of rvBac-Sc-transfected cells.No cross-reactivity was observed with porcine circovirus type 2,porcine pseudorabies virus,classical swine fever virus,or porcine reproductive and respiratory syndrome virus.In summary,we developed two fluorescence detection modes for Cap protein that can be used to detect PCV3 antibodies.This method is suitable for the diagnosis and epidemiological investigation of PCV3.This study provides a reliable detection method for monitoring PCV3 antibody level in pigs in the future.Lun Yao Chang Li Junwei Wang Yufang Cheng Ahmed H.Ghonaim Qi Sun Xuexiang Yu Weijie Niu Shengxian Fan Qigai He 2021Animal Diseases2021,1,2:0
17In situ growth of phosphorized ZIF-67-derived amorphous CoP/Cu_(2)O@CF electrocatalyst for efficient hydrogen evolution reaction显示文摘Transition metal phosphides have been extensively studied for catalytic applications in water splitting.Herein,we report an in situ phosphorization of zeolitic imidazole frameworks(ZIF-67)to generate amorphous cobalt phosphide/ZIF-67 heterojunction on a self-supporting copper foam(CF)substrate with excellent performance for hydrogen evolution reaction(HER).The needleleaf like copper hydroxide was anchored on CF surface,which acted as implantation to grow ZIF-67.The intermediate product was phosphorized to obtain final electrocatalyst(CoP/Cu_(2)O@CF)with uniform particle size,exhibiting a rhombic dodecahedron structure with wrinkles on the surface.The electrochemical measurement proved that CoP/Cu_(2)O@CF catalyst exhibited excellent HER activity and long-term stability in 1.0 mol·L^(–1)KOH solution.The overpotential was only 62 mV with the Tafel slope of 83 mV·dec^(–1)at a current density of 10 mA·cm^(–2),with a large electrochemical active surface area.It also showed competitive performance at large current which indicated the potential application to industrial water electrolysis to produce hydrogen.First-principle calculations illustrated that benefit from the construction of CoP/ZIF-67 heterojunction,the d-band center of CoP downshifted after bonding with ZIF-67 and the Gibbs free energy(ΔGH*)changed from–0.18 to–0.11 eV,confirming both decrease in overpotential and excellent HER activity.This work illustrates the efficient HER activity of CoP/Cu_(2)O@CF catalyst,which will act as a potential candidate for precious metal electrocatalysts.Ruiwen Qi Xiao Liu Hongkai Bu Xueqing Niu Xiaoyang Ji Junwei Ma Hongtao Gao 2023Frontiers of Chemical Science and Engineering2023,17,10:0

19Comprehensive Evaluation of New Fresh Edible Sweet-glutinous Maize Varieties Based on Entropy Weight Method显示文摘In this study,the entropy weight method was used to comprehensively evaluate the eight sweet-glutinous maize varieties in the regional trials in Shanxi Province in 2016. The results showed that the order of the performance of comprehensive traits of the tested varieties was Heitiannuo 631,Heitiannuo No. 2,Cainuo No. 5,Xinnuo 8601,FYN 1601,Xinnuo 8608,Jindannuo 41,Caitiannuo 1958( from good to poor).
