2Punicalagin ameliorates collagen-induced arthritis by downregulating M1 macrophage and pyroptosis via NF-κB signaling pathway显示文摘Rheumatoid arthritis(RA)is a chronic inflammatory disease that eventually leads to disability.Inflammatory cell infiltration,severe joint breaking and systemic bone loss are the main clinical symptoms.In this study,we established a collagen-induced arthritis(CIA)model and found a large number of M1 macrophages and pyroptosis,which are important sources of proinflammatory cytokines.Punicalagin(PUN)is an active substance extracted from pomegranate peel.We found that it inhibited joint inflammation,cartilage damage and systemic bone destruction in CIA mice.PUN effectively alleviated the high expression of inflammatory cytokines in synovial tissue in vivo.PUN treatment shifted macrophages from the M1 phenotype to the M2 phenotype after stimulation with lipopolysaccharide(LPS)and interferon(IFN)-γ.The expression of inducible nitric oxide synthase(i NOS)and other proinflammatory cytokines released by M1 macrophages was decreased in the PUN treatment group.However,simultaneously,the expression of markers of anti-inflammatory M2 macrophages,such as arginase(Arg)-1 and interleukin(IL)-10,was increased.In addition,PUN treatment attenuated pyroptosis by downregulating the expression of NLRP3 and caspase-1,thereby preventing inflammatory cell death resulting from the release of IL-1βand IL-18.Mechanistically,PUN inhibited the activation of receptor activators of the nuclear factor-κB(NF-κB)signaling pathway,which contributes to M1 polarization and pyroptosis of macrophages.We concluded that PUN ameliorated pathological inflammation by inhibiting M1 phenotype polarization and pyroptosis and has great potential as a therapeutic treatment for human RA.Gaoran Ge Jiaxiang Bai Qing Wang Xiaolong Liang Huaqiang Tao Hao Chen Minggang Wei Junjie Niu Huilin Yang Yaozeng Xu Yuefeng Hao Yi Xue Dechun Geng 2022Science China(Life Sciences)2022,65,3:14
3Therapeutic effect of nerve growth factor on cerebral infarction in dogs using the hemisphere anomalous volume ratio of diffusion-weighted magnetic resonance imaging显示文摘A model of focal cerebral ischemic infarction was established in dogs through middle cerebral artery occlusion of the right side.Thirty minutes after occlusion,models were injected with nerve growth factor adjacent to the infarct locus.The therapeutic effect of nerve growth factor against cerebral infarction was assessed using the hemisphere anomalous volume ratio,a quantitative index of diffusion-weighted MRI.At 6 hours,24 hours,7 days and 3 months after modeling,the hemisphere anomalous volume ratio was significantly reduced after treatment with nerve growth factor. 4Distribution characteristics of poly-and perfluoroalkyl substances in the Yangtze River Delta显示文摘In this work, a method was developed and optimized for the analysis of polyfluoroalkyl and/or perfluoroalkyl substances(PFASs) content in surface water and sediment samples with high instrumental response and good separation. Surface water and sediment samples were collected from the Yangtze River Delta(YRD) to analyze the distribution characteristics of perfluoroalkyl carboxylic acids(PFCAs), perfluoroalkane sulfonic acids(PFSAs), perfluoroalkyl phosphonic acids(PFPAs), perfluoroalkyl phosphinic acids(PFPiAs), and polyfluoroalkyl phosphoric acid diesters(di PAPs). The results showed that the total concentrations of PFCAs and PFSAs in YRD varied from 31 to 902 ng/L. PFCAs(≥ 11 carbons) and PFSAs(≥ 10 carbons atoms) were not detected in any surface water samples. The mean concentrations of all PFCAs and PFSAs in surface water from the sampling areas decreased in the following order:Yangtze river(191 ng/L) ≈ Taihu lake(189 ng/L) > Huangpu river(122 ng/L) ≈ Qiantang river(120 ng/L) > Jiaxing urban river(100 ng/L). Strong significant(p < 0.05) correlations between the concentrations of many of the compounds were found in the sampling areas, suggesting a common source for these compounds. Only perfluorooctanoic acid(PFOA) was observed in all sediment samples, at concentrations varying from 0.02 to 1.35 ng/g. 6Lipoprotein(a) as a Marker for Predicting the Presence and Severity of Coronary Artery Disease in Untreated Chinese Patients Undergoing Coronary Angiography显示文摘Objective Low-density lipoprotein cholesterol(LDL-C) has been well known as the risk factor of coronary artery disease(CAD). However, the role of lipoprotein(a) [Lp(a)] in the development of CAD is of great interest but still controversial. Thus, we aim to explore the effect of Lp(a) on predicting the presence and severity of CAD in Chinese untreated patients, especially in combination with LDL-C. Methods We consecutively recruited 1,980 non-treated patients undergoing coronary angiography, among which 1,162 patients were diagnosed with CAD. Gensini score(GS) was used to assess the severity of CAD. Lp(a) was measured by immunoturbidimetric method. Results Patients with CAD had higher level of LDL-C and Lp(a) compared with non-CAD(P < 0.05). Multivariable logistic regression revealed that Lp(a) > 205 mg/L(highest tertile) predicted 1.437-fold risk for CAD(95% CI: 1.108-1.865, P = 0.006) and 1.480-fold risk for high GS(95% CI: 1.090-2.009, P = 0.012) respectively. Interestingly, concomitant elevated level of Lp(a) and LDL-C conferred the highest risk for both presence [OR = 1.845, 95% CI: 1.339-2.541, P < 0.001] and severity [OR = 1.736, 95% CI: 1.188-2.538, P = 0.004] of CAD. 7New risk factors and new tendency for central nervous system relapse in patients with diffuse large B-cell lymphoma:a retrospective study显示文摘Background: In patients with difuse large B?cell lymphoma(DLBCL), central nervous system(CNS) relapse is uncom?mon but is nearly always fatal. This study aimed to determine the risk factors for CNS relapse in DLBCL patients and to evaluate the eicacy of rituximab and intrathecal chemotherapy prophylaxis for CNS relapse reduction.Methods: A total of 511 patients with newly diagnosed DLBCL treated at the Sun Yat?sen University Cancer Center between January 2003 and December 2012 were included in the study. Among these patients, 376 received R?CHOP regimen(rituximab, cyclophosphamide, doxorubicin, vincristine, and prednisone) as primary treatment, and 135 received CHOP regimen(cyclophosphamide, doxorubicin, vincristine, and prednisone) as primary treatment. Intrathe?cal chemotherapy prophylaxis(methotrexate plus cytarabine) was administered to those who were deemed at high risk for CNS relapse. In the entire cohort and in the R?CHOP set in particular, the Kaplan–Meier method coupled with the log?rank test was used for univariate analysis, and the Cox proportional hazards model was used for multivariate analysis. Diferences were evaluated using a two?tailed test, and P < 0.05 was considered signiicant.Results: At a median follow?up of 46 months, 25(4.9%) patients experienced CNS relapse. There was a trend of reduced occurrence of CNS relapse in patients treated with rituximab; the 3?year cumulative CNS relapse rates were 7.1% in CHOP group and 2.7% in R?CHOP group(P = 0.045). Intrathecal chemotherapy prophylaxis did not confer much beneit in terms of preventing CNS relapse. Bone involvement [hazard ratio(HR) = 4.21, 95% conidence interval(CI) 1.38–12.77], renal involvement(HR = 3.85, 95% CI 1.05–14.19), alkaline phosphatase(ALP) >110 U/L(HR = 3.59, 95% CI 1.25–10.34), serum albumin(ALB) <35 g/L(HR = 3.63, 95% CI 1.25–10.51), treatment with rituxi?mab(HR = 0.34, 95% CI 0.12–0.96), and a time to complete remission ≤ 108 days(HR = 0.22, 95% CI 0.06–0.78) were independent predictive factors for CNS relapse in the entire cohort. Bone involvement(HR = 4.44, 95% CI 1.08–18.35), bone marrow involvement(HR = 11.70, 95% CI 2.24–60.99), and renal involvement(HR = 10.83, 95% CI 2.27–51.65) were independent risk factors for CNS relapse in the R?CHOP set.Conclusions: In the present study, rituximab decreased the CNS relapse rate of DLBCL, whereas intrathecal chemo?therapy prophylaxis alone was not suicient for preventing CNS relapse. Serum levels of ALB and ALP, and the time to complete remission were new independent predictive factors for CNS relapse in the patients with DLBCL. 9Esophageal reconstruction with remnant stomach:A case report and review of literature显示文摘The number of patients developing esophageal cancer after gastrectomy has increased.However,gastric remnant is very rarely used for reconstruction in esophageal cancer surgery because of the risk of anastomotic leakage resulting from insufficient blood flow.We present a case of esophageal cancer using gastric remnant for esophageal substitution after distal gastrectomy in a 57-year-old man who presented with a 1-month history of mild dysphagia and a background history of alcohol abuse.Gastroscopy showed a 1.2 cm × 1.0 cm bulge tumor of the lower third esophagus with the upper margin located 39 cm from the dental arcade.Computed tomography of the chest showed lower third esophageal wall thickening.The patient underwent en bloc radical esophagectomy with a two-field lymph node dissection of the upper abdomen and mediastinum via a left-sided posterolateral thoracotomy through the seventh intercostal space.The upper end of the esophagus was resected 5 cm above the tumor.The gastric remnant was used for reconstruction of the esophago-gastrostomy and placed in the left thoracic cavity.The patient started a liquid diet on postoperative day 8 and was discharged on the 10 th postoperative day without complications.In this report,we demonstrate that the gastric remnant may be used for reconstruction in patients with esophageal cancer as a substitute organ after distal gastrectomy.Song-Ping Xie Guo-Hua Fan Gan-Jun Kang Qing Geng Jie Huang Bang-Chang Cheng 2013World Journal of Gastroenterology2013,19,20:4
10Prognostic value of clinicopathological characteristics in patients with pancreatic cancer显示文摘Background:The purpose of this study was to analyze me effects of all clinical characteristics on the overall survival time,in order to provide a basis for determining the prognostic factor of patients with pancreatic cancer.Methods:A total of 103 pancreatic cancer patients were admitted to the Department of Radiotherapy and Chemotherapy of the Ruijin Hospital,Shanghai Jiaotong University School of Medicine,between January 2002 and December 2012.There were 68 men and 35 women;the median age was 62 years.Diagnoses of pancreatic cancer in all patients were confirmed by histopathology,cytology,or clinical diagnosis.The Kaplan-Meier method was performed to calculate the overall survival rate.The log-rank method was used to examine the univariate analysis.The Cox regression model was performed for multivariate analysis.Results:The median survival time was 293 days,the 1-,2-,and 3-year survival rates were 27.18%,5.83%,and 1.94%,respectively.Cox regression analysis revealed that age(P=0.015),Karnofsky performance status(PS)(P=0.002),surgical types(P<0.001),and platelet counts(P<0.001) were independent prognostic factors affecting the overall survival of patients with pancreatic cancer.Conclusions:Pancreatic cancer had a poor prognosis,the general physical condition,age,the availability of radical surgery,and platelet counts were factors influencing the overall survival of patients with pancreatic cancer.Mei Geng Haoping Xu Ruobing Ren Qing Qu Chengfang Shangguan Junwei Wu Jinsong Jiang Hao Li Weiguo Cao 2015Chinese Journal of Cancer Research2015,27,5:4
11Microvessel density at different levels of normal or injured bile duct in dogs and its surgical implications显示文摘BACKGROUND:Ischemic recurrent stricture after surgical repair for iatrogenic bile duct injury(BDI)remains a challenge in clinical practice.The present study was designed to investigate whether ischemia is universal and of varied severity at different levels of the proximal bile duct after BDI. METHODS:A total of 30 beagle dogs were randomly divided into control,BDI,and BDI-repaired groups.The BDI animal model was established based on the classic pattern of laparoscopic cholecystectomy-related BDI.The animals were sacrificed on postoperative day 15,and bile duct tissue was harvested to assess microvessel density(MVD)at selected levels of the normal,post-BDI and BDI-repaired bile duct with the CD34 immunohistochemistry technique. RESULTS:In the control group,MVD at level H(high level) was remarkably higher than that at level L(low level).No significant difference was found between MVDs at levels H and M(middle level),as well as at levels M and L.However, the tendency was noted that the closer the level to the hilus, the greater the MVD at that level.In both the BDI and BDI-repaired groups,MVDs at level H were generally greater than those at level L,despite the unremarkable differences between MVDs at neighboring levels.In these two groups,a similar tendency of MVD distribution to that in the control group was found;the closer the level to the injury site,the lower was the MVD at that level.Moreover,compared with the MDVs at the levels M and L in the control group,MVDs at the corresponding levels in the BDI and BDI-repaired groups were all remarkably reduced(P<0.05).In addition,MVDs at all three levels in the BDI group significantly declined further after BDI repair.CONCLUSIONS:After BDI,universal ischemic damage in the injured proximal bile duct develops close to the injury site, while close to the hilus,ischemia is relatively slight.High hepaticojejunostomy,rather than low biloenterostomy or end-to-end duct anastomosis,should be recommended for BDI repair.Great care should be taken to protect the peribiliary plexus during repair.Lin Geng,Ding Luo,Hao-Chun Zhang,Liang-Song Zhao and De-Ke Qing Department of Hepatobiliary Surgery,Kunming General Hospital of PLA,Kunming 650032,China 2011Hepatobiliary & Pancreatic Diseases International2011,10,1:4

13Th17 cells are involved in mouse chronic obstructive pulmonary disease complicated with invasive pulmonary aspergillosis显示文摘Background:The incidence of chronic obstructive pulmonary disease(COPD)complicated with invasive pulmonary aspergillosis(IPA)has increased in the last two decades.The mechanism underpinning susceptibility to and high mortality of COPD complicated with IPA is unclear,and the role of T helper cells 17(Th17 cells)in the compound disease remains unknown.Therefore,this study aimed to assess the function of Th17 cells in COPD combined with IPA.Methods:COPD,IPA,and COPD+IPA mouse models were established in male wild type C57/BL6 mice.The amounts of Th17 cells and retinoic acid-related orphan receptorsγt(RORyt)were tested by flow cytometry.Then,serum interleukin(IL)-17 and IL-23.levels were detected by enzyme-linked immunosorbent assay(ELISA)in the control,COPD,IPA and COPD+IPA groups.In addition,COPD+IPA was induced in IL-17 knockout(KO)mice,for determining the role of Th17 cells in COPD+IPA.Results:Compared with the COPD group,the COPD+IPA group showed higher amounts of blood RORyt([35.09±16.12]%vs.[17.92±4.91]%,P=0.02)and serum IL-17(17.96±9.59 pg/mL vs.8.05±4.44 pg/mL,P=0.02),but blood([5.18±1.09]%vs.[4.15±0.87]%,P=0.28)and lung levels of Th17 cells(1.98±0.83]%vs.[2.03±0.98]%,P=0.91),lung levels of RORyt([9.58±6.93]%vs.[9.63±5.98]%,P=0.49)and serum IL-23(51.55±27.82 pg/mL us.68.70±15.20 pg/mL,P=0.15)showed no significant differences.Compared with the IPA group,the COPD+IPA group displayed lower amounts of blood([5.18±1.09]%vs.[9.21±3.56]%,P=0.01)and lung Th17 cells([1.98±0.83]%vs.[6.29±1.11]%,P=0.01)and serum IL-23(51.55±27.82 pg/mL vs.154.90±64.60 pg/mL,P=0.01)and IL-17(17.96±9.59 pg/mL uUs.39.81±2.37 pg/mL,P=0.02),while comparable blood([35.09±16.12]%Vs.[29.86±15.42]%,P=0.25)and lung levels of RORγt(9.58±6.93]%VUS,[15.10±2.95]%,P=0.18)were found in these two groups.Finally,Aspergillus load in IL-17 KO COPD+IPA mice was almost 2 times that of COPD+IPA mice(1,851,687.69±944,480.43'vs.892,958.10±686,808.80,t=2.32,P=0.02).Conclusion:These findings indicate that Th17 cells might be involved in the pathogenesis of COPD combined with IPA,with L-17 likely playing an antifungal role.Wan-Ru Geng Hang-Yong He Qing Zhang Zhao-Hui Tong 2021Chinese Medical Journal2021,,5:3
14First experimental constraints on WIMP couplings in the effective field theory framework from CDEX显示文摘We present weakly interacting massive particles(WIMPs) search results performed using two approaches of effective field theory from the China Dark Matter Experiment(CDEX), based on the data from both CDEX-1B and CDEX-10 stages. In the nonrelativistic effective field theory approach, both time-integrated and annual modulation analyses were used to set new limits for the coupling of WIMP-nucleon effective operators at 90% confidence level(C.L.) and improve over the current bounds in the low mχregion. In the chiral effective field theory approach, data from CDEX-10 were used to set an upper limit on WIMP-pion coupling at 90% C.L. 15Advances in Conceptual Electronic Nanodevices based on 0D and 1D Nanomaterials显示文摘Nanoelectronic devices are being extensively developed in these years with a large variety of potential applications. In this article, some recent developments in nanoelectronic devices, including their principles, structures and potential applications are reviewed. As nanodevices work in nanometer dimensions, they consume much less power and function much faster than conventional microelectronic devices. 16Plasma apo CⅢ Levels in Relation to Inflammatory Traits and Metabolic Syndrome in Patients not Treated with Lipid-lowering Drugs Undergoing Coronary Angiography显示文摘Objective Assessment of the comprehensive relationship among apolipoprotein CIII(apoCⅢ) levels, inflammation, and metabolic disorders is rare. Methods A total of 1455 consecutive patients not treated with lipid-lowering drugs and undergoing coronary angiography were enrolled in this cross-sectional study. A mediation analysis was used to detect the underlying role of apoCⅢ in the association of inflammation with metabolic syndrome(MetS). Results Patients with MetS showed higher levels of apoCⅢ [95.1(73.1-131.4) vs. 81.7(58.6-112.4) μg/mL, P < 0.001] and inflammatory markers [high sensitivity C-reactive protein, 1.7(0.8-3.4) vs. 1.1(0.5-2.2) mg/L; white blood cell count,(6.48 ± 1.68) vs.(6.11 ± 1.67) × 10~9/L]. The levels of apoCⅢ and inflammatory markers increased with the number of metabolic risk components(all P < 0.001). Furthermore, apoCⅢ levels were associated with virtually all individual MetS risk factors and inflammatory markers(all P < 0.05). Importantly, the prevalence of MetS in each metabolic disorder rose as apoCⅢ levels increased(all P < 0.05). Mediation analysis showed that apoCⅢ partially mediated the effect of inflammation on MetS independently from triglycerides. 19Analysis of Spatial Clustering Optimization显示文摘Spatial clustering is widely used in many fields such as WSN (Wireless Sensor Networks), web clustering, remote sensing and so on for discovery groups and to identify interesting distributions in the underlying database. By discussing the relationships between the optimal clustering and the initial seeds, a clustering validity index and the principle of seeking initial seeds were proposed, and on this principle we recommend an initial seed-seeking strategy: SSPG (Single-Shortest-Path Graph). With SSPG strategy used in clustering algorithms, we find that the result of clustering is optimized with more probability.
