| 1 | 7,8-Dihydroxyflavone suppresses proliferation and induces apoptosis of human osteosarcoma cells显示文摘Recent studies suggest that 7,8-dihydroxyflavone(7,8-DHF)inhibits the development of several tumors.However,its role in osteosarcoma(OS)remains unknown.This study was designed to investigate the effects and underlying mechanisms of 7,8-DHF that may influence OS development.Human OS cell lines(U2OS and 143B)were treated with 7,8-DHF;cell viability and cell migration were assessed by methylthiazolyldiphenyl-tetrazolium bromide(MTT)assay and wound-healing assay,respectively;and cell death and apoptosis were evaluated by LIVE/DEAD staining and terminal deoxynucleotidyl transferase(TdT)dUTP nick-end labeling(TUNEL)assay,respectively.Reactive oxygen species production was measured using 2,7-dichlorodihydrofluorescein diacetate probe.Akt,Bcl-xL/Bcl-2 asociated death promoter(Bad),p38 mitogen-activated protein kinase(MAPK),extracellular regulated protein kinase(ERK),and c-Jun N-terminal kinase(JNK)expression and their respective phosphorylation levels were detected by western blot analysis.We found that 7,8-DHF reduced cell viability in a dose-dependent manner and also promoted apoptosis,inhibited migration,and induced oxidative stress in OS cells.Moreover,7,8-DHF inhibited Akt,Bad,and p38MAPK,but activated ERK and JNK signals.In summary,our results suggest that 7,8-DHF inhibits OS progression,possibly by regulating Akt/Bad and MAPK signaling.These findings provide new evidence for the pharmacological effects of 7,8-DHF that may improve drug therapy for OS patients. | Jing Zhao Peifeng Li Hua Zhu Fengqin Ge Jie Liu Jingjun Xia Pengzhou Hang | 2021 | Acta Biochimica et Biophysica Sinica2021，53，7： | 3 |
| 2 | First experimental constraints on WIMP couplings in the effective field theory framework from CDEX显示文摘We present weakly interacting massive particles(WIMPs) search results performed using two approaches of effective field theory from the China Dark Matter Experiment(CDEX), based on the data from both CDEX-1B and CDEX-10 stages. In the nonrelativistic effective field theory approach, both time-integrated and annual modulation analyses were used to set new limits for the coupling of WIMP-nucleon effective operators at 90% confidence level(C.L.) and improve over the current bounds in the low mχregion. In the chiral effective field theory approach, data from CDEX-10 were used to set an upper limit on WIMP-pion coupling at 90% C.L. | 3 | First results on ^(76)Ge neutrinoless double beta decay from CDEX-1 experiment显示文摘We report the first results on ^(76)Ge neutrinoless double beta decay from stage one of the China dark-matter experiment(CDEX).A p-type point-contact high-purity germanium detector with a mass of 994 g has been installed to detect neutrinoless double beta decay events, as well as to directly detect dark matter particles. An exposure of 304 kg d has been analyzed over a wide spectral band from 500 keV to 3 MeV. The average event rate obtained was about 0.012 counts per keV per kg per day over the 2.039 MeV energy range. The half-life of ^(76)Ge neutrinoless double beta decay derived based on this result is T^(0ν)1/2>6.4×10^(22) yr(90% C.L.). | 8 | Nanodrug enhances post-ablation immunotherapy of hepatocellular carcinoma via promoting dendritic cell maturation and antigen presentation显示文摘Thermal ablation(TA)as an effective method treating hepatocellular carcinoma(HCC)in clinics is facing great challenges of high recurrence and metastasis.Although immune-checkpoint blockade(ICB)-based immuno-therapy has shown potential to inhibit recurrence and metastasis,the combination strategy of ICB and thermal ablation has shown little progress in HCC treatments.The tremendous hurdle for combining ICB with thermal ablation lies with the insufficient antigen internalization and immaturity of tumor-infiltrating dendritic cells(TIDCs)which leads to an inferior immune response to distant tumor growth and metastasis.Herein,an antigen-capturing nanoplatform,whose surface was modified with mannose as a targeting ligand,was constructed for co-delivering tumor-associated antigens(TAAs)and m6A demethylases inhibitor(i.e.,fat mass and obesity asso-ciated gene(FTO)inhibitor)into TIDCs.In vivo results demonstrate that the intratumoral injection of nanodrug followed by HCC thermal ablation promotes dendritic cells(DCs)maturation,improves tumor infiltration of effector T cells and generates immune memory,which synergize with ICB treatment to inhibit the distant tumor growth and lung metastasis.Therefore,the antigen-capturing and FTO-inhibiting nanodrug holds potential to boost the ICB-based immunotherapy against HCC after thermal ablation. | Zecong Xiao Tan Li Xinyao Zheng Liteng Lin Xiaobin Wang Bo Li Jingjun Huang Yong Wang Xintao Shuai Kangshun Zhu | 2023 | Bioactive Materials2023，，3： | 1 |

| 15 | Performances of a prototype point-contact germanium detector immersed in liquid nitrogen for light dark matter search显示文摘The CDEX-10 experiment searches for light weakly interacting massive particles, a form of dark matter, at the China Jinping Underground Laboratory, where approximately 10 kg of germanium detectors are arranged in an array and immersed in liquid nitrogen. Herein, we report on the experimental apparatus, detector characterization, and spectrum analysis of one prototype detector. Owing to the higher rise-time resolution of the CDEX-10 prototype detector as compared with CDEX-1 B, we identified the origin of an observed category of extremely fast events. For data analysis of the CDEX-10 prototype detector, we introduced and applied an improved bulk/surface event discrimination method. The results of the new method were compared to those of the CDEX-1 B spectrum.
