1Secukinumab demonstrates high efficacy and a favorable safety profile over 52 weeks in Chinese patients with moderate to severe plaque psoriasis显示文摘Background:Psoriasis is a chronic inflammatory skin disease,affecting about 0.6%of the Chinese population.Many patients are not well controlled by conventional treatments,thus there is need for new treatment regimens.In this study,we assessed the efficacy and safety of secukinumab in Chinese patients with moderate to severe plaque psoriasis.Methods:This study was a 52-week,multicentre,randomized,double-blind,placebo-controlled,parallel-group,Phase 3 trial.A sub-population of study participants(≥18 years)of Chinese ethnicity were randomized to receive subcutaneous injections of 300 or 150 mg secukinumab,or placebo.The co-primary endpoints were psoriasis area severity index(PASI)75 and Investigator’s Global Assessment(IGA)0/1 at Week 12.Results:A total of 441 Chinese patients were enrolled in this study.Co-primary outcomes were achieved;300 and 150 mg secukinumab were superior to placebo as shown in the proportion of patients that achieved PASI 75(97.7%and 87.2%vs.3.7%,respectively;P<0.001),and IGA 0/1(82.3%and 69.7%vs.2.7%;P<0.001)at Week 12.Treatment efficacy was maintained until Week 52.There was no increase in overall adverse events with secukinumab relative to placebo throughout the 52-week period.Conclusion:Secukinumab is highly effective and well tolerated in Chinese patients with moderate to severe plaque psoriasis.Lin Cai Jian-Zhong Zhang Xu Yao Jun Gu Quan-Zhong Liu Min Zheng Shi-Fa Zhang Jin-Hua Xu Cheng-Xin Li Hao Cheng Qing Guo Wei-Li Pan Shen-Qiu Li Ruo-Yu Li Zai-Pei Guo Zhi-Qi Song Shan-Shan Li Xiu-Qin Dong Linda Wang Rong Fu Pascaline Regnault Pascal Charef Ratal Mazur Manmath Patekar 2020Chinese Medical Journal2020,,22:21
2Outcomes and possible risk factors associated with axis alignment and rotational stability after implantation of the Toric implantable collamer lens for high myopic astigmatism显示文摘·AIM:To assess the visual outcomes and possible risk factors associated with axis alignment and rotational stability after implantation of Toric implantable collamer lens (TICL) for the correction of high myopic astigmatism. ·METHODS:In this prospective, nonrandomized clinical study, 54 consecutive eyes of 29 patients with high myopic astigmatism received TICL implantation. To evaluate postoperative axis deviation from the intended axis, a digital anterior segment photograph was taken. The ultrasound biomicroscopy(UBM) was used to observe footplate-position. ·RESULTS:After mean follow-up of 8.6 months, mean manifest refractive cylinder (MRC) decreased 79.3% from (-1.88±1.49)D preoperatively to (0.39±0.61)D postoperatively. MRC within 1.00D occurred in 68.5% (37/54) of eyes, whereas 48.1% (26/54) had MRC within 0.50D. Mean manifest refraction spherical equivalent (MRSE) changed from (-12.08±4.22)D preoperatively to (-0.41±0.61)D postoperatively. Uncorrected binocular vision of 20/20 or better occurred in 72.2%(39/54) of patients compared with binocular best-corrected visual acuity (BCVA) of 20/20 or better in 44.4% (24/54) preoperatively. The mean difference between intended and achieved TICL axes was (6.96±8.37)°. Footplates of TICLs were in the ciliary sulcus in 22 eyes (46.3%), below the ciliary sulcus in 32 eyes (53.7%). The angle of TICL rotation had significant correlation with the footplates-position (t =2.127; P =0.045) and the postoperative TICL vaulting (r =-0.516; P =0.000). ·CONCLUSION:The results of our study further support the safety, efficacy and predictability of TICL for the correct high myopic astigmatism. 4Integrative lipidomic and transcriptomic study unravels the therapeutic effects of saikosaponins A and D on non-alcoholic fatty liver disease显示文摘Nonalcoholic fatty liver disease(NAFLD)has become one of the most prominent causes of chronic liver diseases and malignancies.However,few therapy has been approved.Radix Bupleuri(RB)is the most frequently used herbal medicine for the treatment of liver diseases.In the current study,we aim to systemically evaluate the therapeutic effects of saikosaponin A(SSa)and saikosaponin D(SSd),the major bioactive monomers in RB,against NAFLD and to investigate the underlying mechanisms.Our results demonstrated that both SSa and SSd improved diet-induced NAFLD.Integrative lipidomic and transcriptomic analysis revealed that SSa and SSd modulated glycerolipid metabolism by regulating related genes,like Lipe and Lipg.SSd profoundly suppressed the fatty acid biosynthesis by downregulating Fasn and Acaca expression and promoted fatty acid degradation by inducing Acox1 and Cpt1 a expression.Bioinformatic analysis further predicted the implication of master transcription factors,including peroxisome proliferator-activated receptor alpha(PPARα),in the protective effects of SSa and SSd.These results were further confirmed in vitro in mouse primary hepatocytes.In summary,our study uncoded the complicated mechanisms underlying the promising anti-steatosis activities of saikosaponins(SSs),and provided critical evidence inspiring the discovery of innovative therapies based on SSa and SSd for the treatment of NAFLD and related complications.Xiaojiaoyang Li Junde Ge Yajing Li Yajie Cai Qi Zheng Nana Huang Yiqing Gu Qi Han Yunqian Li Rong Sun Runping Liu 2021Acta Pharmaceutica Sinica B2021,11,11:15
5Complications of high intensity focused ultrasound in patients with recurrent and metastatic abdominal tumors显示文摘AIM: To analyze the local and systemic complications of high intensity focused ultrasound (HIFU) for patients with recurrent and metastatic abdominal tumors.METHODS: From Aug 2001 to Aug 2004, 17 patients with recurrent and metastatic abdominal tumors were enrolled in this study. Real-time sonography was taken, and vital signs, liver and kidney function, skin burns, local reactions, and systemic effects were observed and recored before, during, and after HIFU. CT and MRI were also taken before and after HIFU.RESULTS: All 17 patients had skin burns and pain in the treatment region; the next common complication was neurapraxia of the stomach and intestines to variable degrees. The other local and systemic complications were relatively rare. Severe complications were present in two patients; one developed a superior mesenteric artery infarction resulting in necrosis of the entire small intestines, and the other one suffered from a perforation in terminal ileum due to HIFU treatment. 6Effect of acupuncture at Neiguan (PC 6) on cardiac function using echocardiography in myocardial ischemia rats induced by isoproterenol显示文摘OBJECTIVE: To investigate the effect of acupuncture at Neiguan(PC 6) on cardiac function using echocardiography in rat models of myocardial ischemia(MI) induced by isoproterenol(ISO).METHODS: Twenty-seven Sprague-Dawley rats were randomly assigned to normal, model, and acupuncture groups. The model and acupuncture groups were given injections of ISO(85 mg/kg) to establish the MI model. After model establishment,the acupuncture group was treated with acupuncture at Neiguan(PC 6) for 30 min. Echocardiography was used to monitor diastolic and systolic function for 30 min starting from the time after the acupuncture needles were removed. Changes in the length of left ventricular internal diameter at end-diastole(LVIDd), length of left ventricular internal diameter at end-systole(LVIDs), the ratio of mitral peak velocity at early diastole and atrial contraction(E/A), ejection fraction(EF), fractional shortening(FS), and stroke volume(SV) were measured.RESULTS: Compared with the model group at 0and 15 min after needles were removed, the means of LVIDd and LVIDs were significantly lower(P <0.01) and E/A, EF, FS, and SV significantly higher(P < 0.01) in the acupuncture group. In the acupuncture group, the means of LVIDd and LVIDs15 min after the needles were removed were significantly higher(P < 0.01) than those at 0 min. 8Psychological status and diabetes-related distress of Chinese type 1 diabetes patients in Jiangsu province, China显示文摘We sought to assess the psychological status and diabetes-related distress of Chinese type 1 diabetes patients in Jiangsu province,China.According to the World Health Organization criteria,42 patients with type 1 diabetes were enrolled in the study and assessed with the Self-Rating Anxiety Scale(SAS),Self-Rating Depression Scale(SDS)and Diabetes Distress Scale(DDS).All data were tabulated and statistical analyses were performed.The study summarized cases of 42 patients with type 1 diabetes,including 17 males and 25 females with a mean age of 23±12 years and a mean duration of diabetes of 7±6 years.Compared to the Chinese normative data,the SAS standard score was significantly higher,whereas SDS standard score had no statistical significance.The SAS standard score was most highly correlated with diabetes duration(γ=0.547,P=0.011).Additionally,19.5%of the patients had moderate or even severe diabetes-related distress and 21.4%had moderate or even severe emotional burden while 26.2%had regimen-related distress.Multiple stepwise regression analysis showed that the mean correlation between DDS and the four domains was high,particularly the emotional burden domain(estimatedβ=0.363,P<0.001)and regimen-related distress domain(estimatedβ=0.356,P<0.001).The correlation between SAS and DDS was positive(estimatedβ=0.039,P=0.027).In conclusion,the results showed the importance of psychological aspects in Chinese individuals with type 1 diabetes.Screening and treatment of psychological aspects may result in better adherence and increased quality of life for patients with diabetes.Lulu Kong Yun Cai Gang Mei Rong Gu Xiaojiao Zhang Yao Qin Ya Cai Yan Li Hongwen Zhou Mei Zhang Tao Yang 2013The Journal of Biomedical Research2013,27,5:9

10Receptome profiling identifies KREMEN1 and ASGR1 as alternative functional receptors of SARS-CoV-2显示文摘Host cellular receptors play key roles in the determination of virus tropism and pathogenesis.However,little is known about SARS-CoV-2 host receptors with the exception of ACE2.Furthermore,ACE2 alone cannot explain the multi-organ tropism of SARS-CoV-2 nor the clinical differences between SARS-CoV-2 and SARS-CoV,suggesting the involvement of other receptor(s).Here,we performed genomic receptor profiling to screen 5054 human membrane proteins individually for interaction with the SARS-CoV-2 capsid spike(S)protein.Twelve proteins,including ACE2,ASGR1,and KREMEN1,were identified with diverse S-binding affinities and patterns.ASGR1 or KREMEN1 is sufficient for the entry of SARS-CoV-2 but not SARS-CoV in vitro and in vivo.SARS-CoV-2 utilizes distinct ACE2/ASGR1/KREMEN1(ASK)receptor combinations to enter different cell types,and the expression of ASK together displays a markedly stronger correlation with virus susceptibility than that of any individual receptor at both the cell and tissue levels.The cocktail of ASK-related neutralizing antibodies provides the most substantial blockage of SARS-CoV-2 infection in human lung organoids when compared to individual antibodies.Our study revealed an interacting host receptome of SARS-CoV-2,and identified ASGR1 and KREMEN1 as alternative functional receptors that play essential roles in ACE2-independent virus entry,providing insight into SARS-CoV-2 tropism and pathogenesis,as well as a community resource and potential therapeutic strategies for further COVID-19 investigations.Yunqing Gu Jun Cao Xinyu Zhang Hai Gao Yuyan Wang Jia Wang Juan He Xiaoyi Jiang Jinlan Zhang Guanghui Shen Jie Yang Xichen Zheng Gaowei Hu Yuanfei Zhu Shujuan Du Yunkai Zhu Rong Zhang Jianqing Xu Fei Lan Di Qu Guoliang Xu Yun Zhao Dong Gao Youhua Xie Min Luo Zhigang Lu 2022Cell Research2022,32,1:8
11A long-form α-neurotoxin from cobra venom produces potent opioid-independent analgesia显示文摘Aim:In light of the antinociceptive activity of the short-chain neurotoxin,cobrotoxin,and other acetylcholine antagonists,the antinociceptive activity andmechanisms of cobratoxin(CTX),a long-chain postsynaptic α-neurotoxin,wasinvestigated in rodent pain models.Methods:CTX was administered intraperito-neally(30,45,68 μg/kg),intra-cerebral ventricularly(4.5 μg/kg)or microinjectedinto periaqueductal gray(PAG;4.5 μg/kg).The antinociceptive action was testedusing the hot-plate and acetic acid writhing tests m mice and rats.The involve-ment of the cholinergic system and opioid system in CTX-induced analgesia wasexamined by pretreatment of animals with atropine(0.5 mg/kg,im;or 10 mg/kg,ip)or naloxone(1 and 5 mg/kg,ip).The effect of CTX on motor activity was testedusing the Animex test.Results:CTX exhibited a dose-dependent analgesic ac-tion in mice as determined by both the hot-plate and acetic acid writhing tests.The peak effect of analgesia was seen 3 h after administration.In the mouse aceticacid writhing test,the intra-cerebral ventricular administration of CTX at 4.5 μg/kg(1/12th of a systemic dose)produced marked analgesic effects.Microinjection ofCTX(4.5 μgkg)into the PAG region did not elicit an analgesic action in rats in thehot-plate test.Atropine at 0.5 mg/kg(im)and naloxone at 1 and 5 mg/kg(ip)bothfailed to block the analgesic effects of CTX,but atropine at 10 mg/kg(ip)didantagonize the analgesia mediated by CTX in the mouse acetic acid writhing test.Acetylsalicylic acid(300 mg/kg)did not enhance the analgesic effects of CTX.Atthe highest effective dose of 68 μg/kg the neurotoxin did not change the sponta-neous mobility of mice.Conclusion:CTX has analgesic effects,which are medi-ated in the central nervous system though not through the PAG.The centralcholinergic system but not opioid system appears to be involved in theantinociceptive action of CTX.Zhi-xin CHEN Hui-ling ZHANG Zhen-lun GU Bo-wen CHEN Rong HAN Paul F REID Laurence N RAYMOND Zheng-hong QIN 2006Acta Pharmacologica Sinica2006,27,4:7
12Exosomal piRNA profiling revealed unique circulating piRNA signatures of cholangiocarcinoma and gallbladder carcinoma显示文摘Cholangiocarcinoma(CCA)and gallbladder carcinoma(GBC)are biliary tract cancers with poor five-year survival and high recurrence rates.Both CCA and GBC patients suffer from lack of circulating diagnostic biomarkers at the early stage.Extracellular vesicles,especially exosomes,have been emerged as promising diagnostic sources for cancers due to easy and quick accessibility.Hence,identification of exosomal biomarkers provides a novel strategy for CCA and GBC diagnosis.Here,five CCA patients and four GBC patients were enrolled for exosomal small RNA sequencing.Our data showed that exosomal piwi-interacting RNA(piRNA)populations were altered in the plasma of CCA and GBC patients.In comparison to healthy individuals,694 and 323 piRNAs were upregulated in CCA and GBC,respectively,while 36 and 191 piRNAs were down regulated.Interestingly,sequencing results predicted that piR-2660989,piR-10506469,piR-20548188,piR-10822895,piR-hsa-23209,and piR-18044111 were upregulated in both CCA and GBC plasma.Importantly,we further included blood samples from 50 health individuals,40 CCA patients,and 25 GBC patients and found that piR-10506469 were significantly increased in the exosomes of plasma from both CCA and GBC patients.Moreover,we analyzed the expression levels of differentially expressed exosomal piRNAs in the plasma of CCA and GBC patient before and after surgeries and found that piR-10506469 and piR-20548188 were significantly decreased in patients underwent surgeries.Taken together,our data revealed that exosomal piRNAs those are differentially expressed in CCA and GBC plasma may serve as potential biomarkers for the diagnosis of CCA and GBC.Xinjin Gu Chen Wang Hui Deng Chong Qing Rong Liu Sanhong Liu Xinying Xue 2020Acta Biochimica et Biophysica Sinica2020,52,5:7

15A comparison study of summer-time synoptic-scale waves in South China and the Yangtze River basin using the TRMM Multi-Satellite Precipitation Analysis daily product显示文摘Summer-time synoptic-scale waves in South China and the Yangtze River basin are quantified and compared by means of analyzing the 6-year (1998―2004) TRMM Multi-Satellite Precipitation Analysis (TMPA) daily product. An innovative 3-dimensional spectrum analysis method is applied. The results indicate that synoptic-scale waves appearing in South China prominently propagate westward within a zonal wavenumber range of 9―21 and a frequency range of-0.12―-0.22 cycles day-1, while those in the Yangtze River basin primarily move eastward with the same characteristic wavenumbers of 9―21, but within a frequency range of 0.2―0.29 cycles day?1. Zonal and meridional distributions, and seasonal variations of these waves are further explored and compared. It shows that summer-time synoptic-scale waves in the South China result from the northward migration and oscillation of the Intertropical Convergence Zone (ITCZ) in the western Pacific, whereas the ones in the Yangtze River basin are generally related to the synoptic troughs within the westerly flow, originating from the Qinghai-Tibet Plateau.Fu Rong Hu Liang Gu GuoJun Li YaoDong 2008Science China Earth Sciences2008,51,1:6
16Allele-aware chromosome-scale assembly of the allopolyploid genome of hexaploid Ma bamboo(Dendrocalamus latiflorus Munro)显示文摘Dendrocalamus latiflorus Munro is a woody clumping bamboo with rapid shoot growth.Both genetic transformation and clustered regularly interspaced short palindromic repeats(CRISPR)/CRISPR-associated protein 9(Cas9)gene editing techniques are available for D.latiflorus,enabling reverse genetic approaches.Thus,D.latiflorus has the potential to be a model bamboo species.However,the genome sequence of D.latiflorus has remained unreported due to its polyploidy and large genome size.Here,we sequenced the D.latiflorus genome and assembled it into three allele-aware subgenomes(AABBCC),representingthe largest genome of a major bamboo species.We assembled 70 allelic chromosomes(2,737 Mb)for hexaploid D.latiflorus using both singlemolecule sequencing from the Pacific Biosciences(Pac Bio)Sequel platform and chromosome conformation capture sequencing(Hi-C).Repetitive sequences comprised 52.65%of the D.latiflorus genome.We annotated 135231 protein-coding genes in the genome based on transcriptomes from eight different tissues.Transcriptome sequencing using RNA-Seq and Pac Bio singlemolecule real-time long-read isoform sequencing revealed highly differential alternative splicing(AS)between non-abortive and abortive shoots,suggesting that AS regulates the abortion rate of bamboo shoots.This high-quality hexaploid genome and comprehensive strand-specific transcriptome datasets for this Poaceae family member will pave the way for bamboo research using D.latiflorus as a model species.Yushan Zheng Deming Yang Jundong Rong Liguang Chen Qiang Zhu Tianyou He Lingyan Chen Jing Ye Lili Fan Yubang Gao Hangxiao Zhang Lianfeng Gu 2022Journal of Integrative Plant Biology2022,64,3:6

18Biotransformation of aesculin by human gut bacteria and identification of its metabolites in rat urine显示文摘AIM:To observe the biotransformation process of a Chinese compound,aesculin,by human gut bacteria,and to identify its metabolites in rat urine.METHODS:Representative human gut bacteria were collected from 20 healthy volunteers,and then utilized in vitro to biotransform aesculin under anaerobic conditions.At 0,2,4,8,12,16,24,48 and 72 h post- incubation,10 mL of culture medium was collected.Metabolites of aesculin were extracted 3×from rat urine with methanol and analyzed by HPLC.For in vivo metabolite analysis,aesculetin(100 mg/kg)was administered to rats via stomach gavage,rat urine was collected from 6 to 48 h post-administration,and metabolite analysis was performed by LC/ESI-MS and MS/MS in the positive and negative modes.RESULTS:Human gut bacteria could completely convert aesculin into aesculetin in vitro.The biotransformation process occurred from 8 to 24 h post-incubation,with its highest activity was seen from 8 to 12 h.The in vitro process was much slower than the in vivo process.In contrast to the in vitro model,six aesculetin metabolites were identified in rat urine,including 6-hydroxy-7-gluco- coumarin(M1),6-hydroxy-7-sulf-coumarin(M2),6,7-di- gluco-coumarin(M3),6-glc-7-gluco-coumarin(M4),6-O-methyl-7-gluco-coumarin(M5)and 6-O-methyl-7- sulf-coumarin(M6).Of which,M2 and M6 were novel metabolites.CONCLUSION:Aesculin can be transferred into aesculetin by human gut bacteria and is further modified by the host in vivo.The diverse metabolites of aesculin may explain its pleiotropic pharmaceutical effects.Wei-Jun Ding Yun Deng Hao Feng Wei-Wei Liu Rong Hu Xiang Li Zhe-Ming Gu Xiao-Ping Dong 2009World Journal of Gastroenterology2009,15,12:5
19A short-chain α-neurotoxin from Naja naja atra produces potent cholinergic-dependent analgesia显示文摘Objective To investigate the analgesia induced by cobrotoxin (CT) from venom of Naja naja atra, and the effects of atropine and naloxone on the antinociceptive activity of CT in rodent pain models. Methods CT was administered intraperitoneally (33.3,50,7μg/kg) , intra-cerebral venticularly (2.4μg /kg) or microinjected into periaque-ductal gray (PAG,1.2μg /kg). The antinociceptive action was tested using the hot-plate test and the acetic acid writhing test in mice and rats. The involvement of cholinergic system and the opioid system in CT-induced analgesia was examined by pretreatment of animals with atropine (0.5mg/kg, im or 10mg/kg, ip) or naloxone (3mg/kg,ip) . The effect of CT on motor activity was tested using the Animex test. Results CT (33.3,50 and 75μg/kg,ip) exhibited a dose-dependent analgesic action in mice as determined with hot-plate test and acetic acid writhing test. In the mouse acetic acid writhing test, the intra-cerebral ventricle administration of CT 2.4μg/kg (1/23th of a systemic dose) produced marked analgesic effects. Microinjection of CT 1.2μg/kg (1/46th of systemic dose) into the PAG also elicited a robust analgesic action in the hot-plate test in rats. Atropine at 0.5 mg/kg (im) or naloxone at 3 mg/kg (ip) failed to block the analgesic effects of CT, but atropine at 10 mg/kg (ip) did antagonize the analgesia mediated by CT in the mouse acetic acid writhing test. At the highest effective dose of antinociception (75μg/kg) , CT did not change the spontaneous mobility of mice. Conclusion These results suggest that CT from Naja naja atra venom has analgesic effects. Central nervous system may be involved in CT' analgesic effects and the PAG may be the primary central site where CT exerts its effects. 20Analysis of severe human adenovirus infection outbreak in Guangdong Province,southern China in 2019显示文摘During 2018–2019,a severe human adenovirus(HAdV)infection outbreak occurred in southern China.Here,we screened 18 respiratory pathogens in 1704 children(≤14 years old)hospitalized with acute respiratory illness in Guangzhou,China,in 2019.In total,151 patients had positive HAdV test results;34.4%(52/151)of them exhibited severe illness.HAdV infection occurred throughout the year,with a peak in summer.The median patient age was 3.0(interquartile range:1.1–5.0)years.Patients with severe HAdV infection exhibited increases in12 clinical indexes(P≤0.019)and decreases in four indexes(P≤0.007),compared with patients exhibiting nonsevere infection.No significant differences were found in age or sex distribution according to HAdV infection severity(P>0.05);however,the distributions of comorbid disease and HAdV co-infection differed according to HAdV infection severity(P<0.05).The main epidemic types were HAdV-3(47.0%,71/151)and HAdV-7(46.4%,70/151).However,the severe illness rate was significantly higher in patients with HAdV-7(51.4%)than in patients with HAdV-3(19.7%)and other types of HAdV(20%)(P<0.001).Sequencing analysis of genomes/capsid genes of 13 HAdV-7 isolates revealed high similarity to previous Chinese isolates.A representative HAdV-7isolate exhibited a similar proliferation curve to the curve described for the epidemic HAdV-3 strain Guangzhou01(accession no.DQ099432)(P>0.05);the HAdV-7 isolate exhibited stronger virulence and infectivity,compared with HAdV-3(P<0.001).Overall,comorbid disease,HAdV co-infection,and high virulence and infectivity of HAdV-7 were critical risk factors for severe HAdV infection;these data can facilitate treatment,control,and prevention of HAdV infection.Wenkuan Liu Shuyan Qiu Li Zhang Hongkai Wu Xingui Tian Xiao Li Duo Xu Jing Dai Shujun Gu Qian Liu Dehui Chen Rong Zhou 2022Virologica Sinica2022,37,3:5
