| 4 | Preparation, characterization, pharmacokinetics and anticancer effects of PEGylated β-elemene liposomes显示文摘Objective:This study aimed to develop a new polyethylene glycol(PEG)ylatedβ-elemene liposome(PEG-Lipo-β-E)and evaluate its characterization,pharmacokinetics,antitumor effects and safety in vitro and in vivo.Methods:The liposomes were prepared by ethanol injection and high-pressure micro-jet homogenization.Characterization of the liposomes was conducted,and drug content,entrapment efficiency(EE),in vitro release and stability were studied by ultra-fast liquid chromatography(UFLC)and a liquid surface method.Blood was drawn from rats to establish the pharmacokinetic parameters.The anticancer effect was evaluated in a KU-19-19 bladder cancer xenograft model.Histological analyses were performed to evaluate safety.Results:The PEG-Lipo-β-E showed good stability and was characterized as 83.31±0.181 nm in size,0.279±0.004 in polydispersity index(PDI),-21.4±1.06 mV in zeta potential,6.65±0.02 in pH,5.024±0.107 mg/mL inβ-elemene(β-E)content,and 95.53±1.712%in average EE.The Fourier transform infrared spectroscopy(FTIR)and differential scanning calorimetry(DSC)indicated the formation of PEG-Lipo-β-E.Compared to elemene injection,PEG-Lipo-β-E demonstrated a 1.75-fold decrease in clearance,a 1.62-fold increase in half-life,and a 1.76-fold increase in area under the concentration-time curves(AUCs)from 0 hour to 1.5 hours(P<0.05).PEG-Lipo-β-E also showed an enhanced anticancer effect in vivo.Histological analyses showed that there was no evidence of toxicity to the heart,kidney,liver,lung or spleen.Conclusions:The present study demonstrates PEG-Lipo-β-E as a new formulation with ease of preparation,high EE,good stability,improved bioavailability and antitumor effects. | Bingtao Zhai Qibiao Wu Wengang Wang Mingming Zhang Xuemeng Han Qiujie Li Peng Chen Xiaying Chen Xingxing Huang Guohua Li Qin Zhang Ruonan Zhang Yu Xiang Shuiping Liu Ting Duan Jianshu Lou Tian Xie Xinbing Sui | 2020 | Cancer Biology & Medicine2020,17,1: | 6 |

This study took the Qingshitan Reservoir in the Lijiang River as a case, and developed an optimization model to explore a trade-off solution between social-economic interests and nature flow maintenance on a monthly base. The objective function considered irrigation, cruise navigation and water supply aspects. An index of flow alteration degree was proposed to measure the difference between the regulated discharge and the natural flow. The index was then used as an additional constraint in the model besides the conventional constraints on reservoir safety. During model solving, different criteria were applied to the index, representing various degrees of alteration of the natural flow regime in the river. Through the model, a relationship between social-economic interests and flow alteration degree was established. | 8 | The Effect of CD3-Specific Monoclonal Antibody on Treating Experimental Autoimmune Myasthenia Gravis显示文摘CD3-specific monoclonal antibody was the first one used for clinical practice in field of transplantation. Recently,renewed interests have elicited in its capacity to prevent autoimmune diabetes by inducing immune tolerance. In this study, we tested whether this antibody can also be used to treat another kind of autoimmune disease myasthenia gravis (MG) and explored the possible mechanisms. MG is caused by an autoimmune damage mediated by antibody- and complement-mediated destruction of AChR at the neuromuscular junction. We found that administration of CD3-specific antibody (Fab)2 to an animal model with experimental autoimmune myasthenia gravis (EAMG) (B6 mice received 3 times of AChR/CFA immunization) could not significantly improve the clinical signs and clinical score. When the possible mechanisms were tested, we found that CD3 antibody treatment slightly down-regulated the T-cell response to AChR, modestly up-regulation the muscle strength. And no significant difference in the titers of IgG2b was found between CD3 antibody treated and control groups. These data indicated that CD3-specific antibody was not suitable for treating MG, an antibody- and complementmediated autoimmune disease, after this disease has been established. | 13 | Suppression of transmembrane sodium currents on the freshly isolated hippocampal neuron cell with continuous infrared light显示文摘Physiotherapeutic effects of infrared lasers have been proved in clinic.These infrared-based regulations of the bioelectrical activities can roughly be classied into enhancement and suppression of action potential(AP),which are described by sodium(Na)and potassium(K)transmembrane current equations,named as Hodgkin and Huxley(HH)-model.The enhancement effect is able to evoke or strengthen the AP when infrared light is applied.Its corresponding mechanism is commonly ascribed to the changes of the cell membrane capacitance,which is transiently increased in response to the infrared radiation.The distinctive feature of the suppression effect is to inhibit or reduce the AP by the designed protocols of infrared radiation.However,its mechanism presents more complexity than that in enhancement cases.HH-model describes how the Na current determines the initial phase of AP.So,the enhancement and suppression of AP can be also ascribed to the regulations of the corresponding Na currents.Here,a continuous infrared light at the wavelength of 980 nm(CIS-980)was employed to stimulate a freshly isolated hippocampal neuron in vitro and a suppression effect on the Na currents of the neuron cell was observed.Both Na and K currents,which are named as whole cell currents,were simultaneously recorded with the cell membrane capacitance current by using a patch clamp combined with infrared irradiation.The results demonstrated that the CIS-980 was able to reversibly increase the capacitance currents,completely suppressed Na currents,but little changed K currents,which forms the steady outward whole cell currents and plays a major role on the AP repolarization.A conrmation experiment was designed and carried out by synchronizing tens of milliseconds of infrared stimulation on the same kinds of hippocampal neuron cells.After the blocked K channel,a reduction of Na current amplitude was still recorded.This proved that infrared suppression of Na current was irrelevant to K channel.A membrane capacitance mediation process was preliminarily proposed to explain the Na channel suppression process. | Fanyi Kong Xinyu Li Ruonan Jiao Kun Liu Xue Han Changkai Sun Changsen Sun | 2023 | Journal of Innovative Optical Health Sciences2023,16,2: | 0 |

| 16 | Pd cluster decorated free standing flexible cathode for high performance Li-oxygen batteries显示文摘As a promising candidate for the next generation energy storage system,rechargeable lithium-oxygen batteries(LOBs)still face substantial challenges caused by insulating discharge products that preclude their practical application.Exploring highly efficient cathode catalysts capable of facilitating formation/decomposition of discharge products is considered as an essential approach towards high performance LOBs.Herein,Pd decorated Te nanowires(Pd@Te NWs)were synthesized as advanced catalyst in LOBs to maximize Pd utilization and achieve synergistic effect,in which Pd clusters were uniformly grown on Te substrate though regulating the Pd:Te ratio.Meanwhile,Pd@Te nanowires assembled into an interpenetrating network-like structure by vacuum filtration and employed as flexible cathode,enabling LOBs achieved an ultralong 190 cycles stability and a superior specific capacity of 3.35 mAh·cm^(-2).Experimental studies and density functional theory(DFT)calculations reveal the excellent catalytic ability of Pd@Te and synergistic catalytic mechanism of Pd and Te,in which uniform electron distribution,extensive electron exchange,and large adsorption distance between Pd cluster and discharge products promote homogeneous adsorption/desorption of discharge products,while the high adsorption energy of Te substrate for Li species reduces the initial dynamical energy barrier during discharging process.The current work provides viable strategy to design composite catalysts for flexible cathode of LOBs with synergistic catalytic effects. | Liang Guo Guoliang Zhang Ruonan Yang Dongmei Zhang Xiuqi Zhang Han Yu Xia Li Feng Dang | 2024 | Nano Research2024,17,4: | 0 |
