1A nano-cocrystal strategy to improve the dissolution rate and oral bioavailability of baicalein显示文摘Baicalein(BE) is one of the main active flavonoids representing the variety of pharmacological effects including anticancer, anti-inflammatory and cardiovascular protective activities, but it's very low solubility, dissolution rate and poor oral absorption limit the therapeutic applications. In this work, a nano-cocrystal strategy was successfully applied to improve the dissolution rate and bioavailability of BE. Baicalein-nicotinamide(BE-NCT) nanococrystals were prepared by high pressure homogenization and evaluated both in vitro and in vivo. Physical characterization results including scanning electron microscopy, dynamic light scattering, powder X-ray diffraction and differential scanning calorimetry demonstrated that BE-NCT nano-cocrystals were changed into amorphous state with mean particle size of 251.53 nm. In the dissolution test, the BE-NCT nano-cocrystals performed 2.17-fold and 2.54-fold enhancement than BE coarse powder in FaSSIF-V2 and FaSSGF. Upon oral administration, the integrated AUC0-t of BE-NCT nano-cocrystals(6.02-fold) was significantly higher than BE coarse powder(1-fold), BE-NCT cocrystals(2.87-fold) and BE nanocrystals(3.32-fold). 8Enhanced photon communication through Bayesian estimation with an SNSPD array显示文摘Laser communication using photons should consider not only the transmission environment’s effects,but also the performance of the single-photon detector used and the photon number distribution.Photon communication based on the superconducting nanowire single-photon detector(SNSPD)is a new technology that addresses the current sensitivity limitations at the level of single photons in deep space communication.The communication’s bit error rate(BER)is limited by dark noise in the space environment and the photon number distribution with a traditional single-pixel SNSPD,which is unable to resolve the photon number distribution.In this work,an enhanced photon communication method was proposed based on the photon number resolving function of four-pixel array SNSPDs.A simulated picture transmission was carried out,and the error rate in this counting mode can be reduced by 2 orders of magnitude when compared with classical optical communication.However,in the communication mode using photon-enhanced counting,the four-pixel response amplitude for counting was found to restrain the communication rate,and this counting mode is extremely dependent on the incident light intensity through experiments,which limits the sensitivity and speed of the SNSPD array’s performance advantage.Therefore,a BER theoretical calculation model for laser communication was presented using the Bayesian estimation algorithm in order to analyze the selection of counting methods for information acquisition under different light intensities and to make better use of the SNSPD array’s high sensitivity and speed and thus to obtain a lower BER.The counting method and theoretical model proposed in this work refer to array SNSPDs in the deep space field.XIANG LI JINGROU TAN KAIMIN ZHENG LABAO ZHANG LIJIAN ZHANG WEIJI HE PENGWEI HUANG HAOCHEN LI BIAO ZHANG QI CHEN RUI GE SHUYA GUO TAO HUANG XIAOQING JIA QINGYUAN ZHAO XUECOU TU LIN KANG JIAN CHEN PEIHENG WU 2020Photonics Research2020,8,5:1

10Analysis of COVID-19 Guideline Quality and Change of Recommendations:A Systematic Review显示文摘Background.Hundreds of coronavirus disease 2019(COVID-19)clinical practice guidelines(CPGs)and expert consensus statements have been developed and published since the outbreak of the epidemic.However,these CPGs are of widely variable quality.So,this review is aimed at systematically evaluating the methodological and reporting qualities of COVID-19 CPGs,exploring factors that may influence their quality,and analyzing the change of recommendations in CPGs with evidence published.Methods.We searched five electronic databases and five websites from 1 January to 31 December 2020 to retrieve all COVID-19 CPGs.The assessment of the methodological and reporting qualities of CPGs was performed using the AGREE II instrument and RIGHT checklist.Recommendations and evidence used to make recommendations in the CPGs regarding some treatments for COVID-19(remdesivir,glucocorticoids,hydroxychloroquine/chloroquine,interferon,and lopinavir-ritonavir)were also systematically assessed.And the statistical inference was performed to identify factors associated with the quality of CPGs.Results.We included a total of 92 COVID-19 CPGs developed by 19 countries.Overall,the RIGHT checklist reporting rate of COVID-19 CPGs was 33.0%,and the AGREE II domain score was 30.4%.The overall methodological and reporting qualities of COVID-19 CPGs gradually improved during the year 2020.Factors associated with high methodological and reporting qualities included the evidence-based development process,management of conflicts of interest,and use of established rating systems to assess the quality of evidence and strength of recommendations.The recommendations of only seven(7.6%)CPGs were informed by a systematic review of evidence,and these seven CPGs have relatively high methodological and reporting qualities,in which six of them fully meet the Institute of Medicine(IOM)criteria of guidelines.Besides,a rapid advice CPG developed by the World Health Organization(WHO)of the seven CPGs got the highest overall scores in methodological(72.8%)and reporting qualities(83.8%).Many CPGs covered the same clinical questions(it refers to the clinical questions on the effectiveness of treatments of remdesivir,glucocorticoids,hydroxychloroquine/chloroquine,interferon,and lopinavirritonavir in COVID-19 patients)and were published by different countries or organizations.Although randomized controlled trials and systematic reviews on the effectiveness of treatments of remdesivir,glucocorticoids,hydroxychloroquine/chloroquine,interferon,and lopinavir-ritonavir for patients with COVID-19 have been published,the recommendations on those treatments still varied greatly across COVID-19 CPGs published in different countries or regions,which may suggest that the CPGs do not make sufficient use of the latest evidence.Conclusions.Both the methodological and reporting qualities of COVID-19 CPGs increased over time,but there is still room for further improvement.The lack of effective use of available evidence and management of conflicts of interest were the main reasons for the low quality of the CPGs.The use of formal rating systems for the quality of evidence and strength of recommendations may help to improve the quality of CPGs in the context of the COVID-19 pandemic.During the pandemic,we suggest developing a living guideline of which recommendations are supported by a systematic review for it can facilitate the timely translation of the latest research findings to clinical practice.We also suggest that CPG developers should register the guidelines in a registration platform at the beginning for it can reduce duplication development of guidelines on the same clinical question,increase the transparency of the development process,and promote cooperation among guideline developers all over the world.Since the International Practice Guideline Registry Platform has been created,developers could register guidelines prospectively and internationally on this platform.Siya Zhao Shuya Lu Shouyuan Wu Zijun Wang Qiangqiang Guo Qianling Shi Hairong Zhang Juanjuan Zhang Hui Liu Yunlan Liu Xianzhuo Zhang Ling Wang Mengjuan Ren Ping Wang Hui Lan Qi Zhou Yajia Sun Jin Cao Qinyuan Li Janne Estill Joseph LMathew Hyeong Sik Ahn Myeong Soo Lee Xiaohui Wang Chenyan Zhou Yaolong Chen 2021Health Data Science2021,,1:1

12High-timing-precision detection of single X-ray photons by superconducting nanowires显示文摘Precisely acquiring the timing information of individual X-ray photons is important in both fundamental research and practical applications. The timing precision of commonly used X-ray single-photon detectors remains in the range of one hundred picoseconds to microseconds. In this work, we report on high-timing-precision detection of single X-ray photons through the fast transition to the normal state from the superconductive state of superconducting nanowires. We successfully demonstrate a free-running X-ray single-photon detector with a timing resolution of 20.1 ps made of 100-nm-thick niobium nitride film with an active area of 50 μm by 50 μm. By using a repeated differential timing measurement on two adjacent X-ray single-photon detectors, we demonstrate a precision of 0.87 ps in the arrival-time difference of X-ray photon measurements. 16Saturation efficiency for detecting 1550 nm photons with a 2×2 array of Mo0.8Si0.2 nanowires at 2.2 K显示文摘Amorphous materials are attractive candidates for fabricating the superconducting nanowire single-photon detectors(SNSPDs) due to their superior tolerance and scalability over crystalline niobium nitride. However, the reduced superconducting transition temperature degenerates both operating temperature and saturation efficiency. Herein, the SNSPD(6.5 nm thickness and 50 nm width) based on the amorphous Mo0.8Si0.2 film with a high optical absorption coefficient demonstrates close-to-unity intrinsic detection efficiency for 1550 nm photons from 75 m K to 2.2 K. Further, a high-performance array SNSPD with optimized 90 nm-width wires is also demonstrated. 18Canopy structure:An intermediate factor regulating grassland diversity-function relationships under human disturbances显示文摘Grasslands are one of the largest coupled human-nature terrestrial ecosystems on Earth,and severe anthropogenic-induced grassland ecosystem function declines have been reported recently.Understanding factors influencing grassland ecosystem functions is critical for making sustainable management policies.Canopy structure is an important factor influencing plant growth through mediating within-canopy microclimate(e.g.,light,water,and wind),and it is found coordinating tightly with plant species diversity to influence forest ecosystem functions.However,the role of canopy structure in regulating grassland ecosystem functions along with plant species diversity has been rarely investigated.Here,we investigated this problem by collecting field data from 170 field plots distributed along an over 2000 km transect across the northern agro-pastoral ecotone of China.Aboveground net primary productivity(ANPP)and resilience,two indicators of grassland ecosystem functions,were measured from field data and satellite remote sensing data.Terrestrial laser scanning data were collected to measure canopy structure(represented by mean height and canopy cover).Our results showed that plant species diversity was positively correlated to canopy structural traits,and negatively correlated to human activity intensity.Canopy structure was a significant indicator for ANPP and resilience,but their correlations were inconsistent under different human activity intensity levels.Compared to plant species diversity,canopy structural traits were better indicators for grassland ecosystem functions,especially for ANPP.Through structure equation modeling analyses,we found that plant species diversity did not have a direct influence on ANPP under human disturbances.Instead,it had a strong indirect effect on ANPP by altering canopy structural traits.As to resilience,plant species diversity had both a direct positive contribution and an indirect contribution through mediating canopy cover.This study highlights that canopy structure is an important intermediate factor regulating grassland diversity-function relationships under human disturbances,which should be included in future grassland monitoring and management.Xiaoxia Zhao Yuhao Feng Kexin Xu Mengqi Cao Shuya Hu Qiuli Yang Xiaoqiang Liu Qin Ma Tianyu Hu Maggi Kelly Qinghua Guo Yanjun Su 2023Fundamental Research2023,3,2:0

20Regulation of cytokinin biosynthesis using PtRD26pro-IPT module improves drought tolerance through PtARR10-PtYUC4/5-mediated reactive oxygen species removal in Populus显示文摘Drought is a critical environmental factor which constrains plant survival and growth.Genetic engineering provides a credible strategy to improve drought tolerance of plants.Here,we generated transgenic poplar lines expressing the isopentenyl transferase gene(IPT)under the driver of Pt RD26 promoter(PtRD26 pro-IPT).Pt RD26 is a senescence and drought-inducible NAC transcription factor.Pt RD26 pro-IPT plants displayed multiple phenotypes,including improved growth and drought tolerance.Transcriptome analysis revealed that auxin biosynthesis pathway was activated in the PtRD26 pro-IPT plants,leading to an increase in auxin contents.Biochemical analysis revealed that ARABIDOPSIS RESPONSE REGULATOR10(PtARR10),one of the type-B ARR transcription factors in the cytokinin pathway,was induced in PtRD26 pro-IPT plants and directly regulated the transcripts of YUCCA4(Pt YUC4)and YUCCA5(PtYUC5),two enzymes in the auxin biosynthesis pathway.Overexpression of PtYUC4 enhanced drought tolerance,while simultaneous silencing of PtYUC4/5 evidently attenuated the drought tolerance of Pt RD26 pro-IPT plants.Intriguingly,Pt YUC4/5 displayed a conserved thioredoxin reductase activity that is required for drought tolerance by deterring reactive oxygen species accumulation.Our work reveals the molecular basis of cytokinin and auxin interactions in response to environmental stresses,and shed light on the improvement of drought tolerance without a growth penalty in trees by molecular breeding.Hou-Ling Wang Qi Yang Shuya Tan Ting Wang Yi Zhang Yanli Yang Weilun Yin Xinli Xia Hongwei Guo Zhonghai Li 2022Journal of Integrative Plant Biology2022,64,3:0
