1A two-step lineage reprogramming strategy to generate functionally competent human hepatocytes from fibroblasts显示文摘Terminally differentiated cells can be generated by lineage reprogramming,which is,however,hindered by incomplete conversion with residual initial cell identity and partial functionality. Here, we demonstrate a new reprogramming strategy by mimicking the natural regeneration route, which permits generating expandable hepatic progenitor cells and functionally competent human hepatocytes. Fibroblasts were first induced into human hepatic progenitor-like cells (hHPLCs), which could robustly expand in vitro and efficiently engraft in vivo. Moreover, hHPLCs could be efficiently induced into mature human hepatocytes (hiHeps) in vitro, whose molecular identity highly resembles primary human hepatocytes (PHHs). Most importantly, hiHeps could be generated in large quantity and were functionally competent to replace PHHs for drug-metabolism estimation, toxicity prediction and hepatitis B virus infection modeling. Our results highlight the advantages of the progenitor stage for successful lineage reprogramming. 2Mulching mode and planting density affect canopy interception loss of rainfall and water use efficiency of dryland maize on the Loess Plateau of China显示文摘High and efficient use of limited rainwater resources is of crucial importance for the crop production in arid and semi-arid areas. To investigate the effects of different soil and crop management practices(i.e., mulching mode treatments: flat cultivation with non-mulching, flat cultivation with straw mulching, plastic-covered ridge with bare furrow and plastic-covered ridge with straw-covered furrow; and planting density treatments: low planting density of 45,000 plants/hm^2, medium planting density of 67,500 plants/hm^2 and high planting density of 90,000 plants/hm^2) on rainfall partitioning by dryland maize canopy, especially the resulted net rainfall input beneath the maize canopy, we measured the gross rainfall, throughfall and stemflow at different growth stages of dryland maize in 2015 and 2016 on the Loess Plateau of China. The canopy interception loss was estimated by the water balance method. Soil water storage, leaf area index, grain yield(as well as it components) and water use efficiency of dryland maize were measured or calculated. Results showed that the cumulative throughfall, cumulative stemflow and cumulative canopy interception loss during the whole growing season accounted for 42.3%–77.5%, 15.1%–36.3% and 7.4%–21.4% of the total gross rainfall under different treatments, respectively. Soil mulching could promote the growth and development of dryland maize and enhance the capability of stemflow production and canopy interception loss, thereby increasing the relative stemflow and relative canopy interception loss and reducing the relative throughfall. The relative stemflow and relative canopy interception loss generally increased with increasing planting density, while the relative throughfall decreased with increasing planting density. During the two experimental years, mulching mode had no significant influence on net rainfall due to the compensation between throughfall and stemflow, whereas planting density significantly affected net rainfall. The highest grain yield and water use efficiency of dryland maize were obtained under the combination of medium planting density of 67,500 plants/hm^2 and mulching mode of plastic-covered ridge with straw-covered furrow. Soil mulching can reduce soil evaporation and retain more soil water for dryland maize without reducing the net rainfall input beneath the maize canopy, which may alleviate the contradiction between high soil water consumption and insufficient rainfall input of the soil. 3Chloroplast genomic diversity in Bulbophyllum section Macrocaulia(Orchidaceae,Epidendroideae,Malaxideae):Insights into species divergence and adaptive evolution显示文摘Bulbophyllum is the largest genus in Orchidaceae with a pan tropical distribution.Due to highly significant diversifications,it is considered to be one of the most taxonomically and phylogenetically complex taxa.The diversification pattern and evolutionary adaptation of chloroplast genomes are poorly understood in this species-rich genus,and suitable molecular markers are necessary for species determination and phylogenetic analysis.A natural Asian section Macrocaulia was selected to estimate the interspecific divergence of chloroplast genomes in this study.Here,we sequenced the complete chloroplast genome of four Bulbophyllum species,including three species from section Macrocaulia.The four chloroplast genomes had a typical quadripartite structure with a genome size ranged from 156,182 to 158,524 bp.The chloroplast genomes included 113 unique genes encoding 79 proteins,30 tRNAs and 4 rRNAs.Comparison of the four chloroplast genomes showed that the three species from section Macrocaulia had similar structure and gene contents,and shared a number of indels,which mainly contribute to its monophyly.In addition,interspecific divergence level was also great.Several exclusive indels and polymorphism SSR loci might be used for taxonomical identification and determining interspecific polymorphisms.A total of 20 intergenic regions and three coding genes of the most variable hotspot regions were proposed as candidate effective molecular markers for future phylogenetic relationships at different taxonomical levels and species divergence in Bulbophyllum.All of chloroplast genes in four Bulbophyllum species were under purifying selection,while 13 sites within six genes exhibited sitespecific selection.A whole chloroplast genome phylogenetic analysis based on Maximum Likelihood,Bayesian and Parsimony methods all supported the monophyly of section Macrocaulia and the genus of Bulbophyllum.Our findings provide valuable molecular markers to use in accurately identifying species,clarifying taxonomy,and resolving the phylogeny and evolution of the genus Bulbophyllum.The molecular markers developed in this study will also contribute to further research of conservation of Bulbophyllum species.Hanqing Tang Lu Tang Shicheng Shao Yulan Peng Lu Li Yan Luo 2021Plant Diversity2021,43,5:3
4Reactive Inorganic Vapor Deposition of Perovskite Oxynitride Films for Solar Energy Conversion显示文摘The synthesis of perovskite oxynitrides,which are promising photoanode candidates for solar energy conversion,is normally accomplished by high-temperature ammonolysis of oxides and carbonate precursors,thus making the deposition of their planar films onto conductive substrates challenging.Here,we proposed a facile strategy to prepare a series of perovskite oxynitride films.Taking SrTaO_(2)N as a prototype,we prepared SrTaO_(2)N films on Ta foils under NH_(3) flow by utilizing the vaporized SrCl_(2)/SrCO_(3) eutectic salt.The SrTaO_(2)N films exhibit solar water-splitting photocurrents of 3.0 mA cm^(-2) at 1.23 V vs.RHE(reversible hydrogen electrode),which increases by 270%compared to the highest photocurrent(1.1 mA cm^(-2) at 1.23 V vs.RHE)of SrTaO_(2)N reported in the literature.This strategy may also be applied to directly prepare a series of perovskite oxynitride films on conductive substrates such as ATaO_(2)N(A=Ca,Ba)and ANbO_(2)N(A=Sr,Ba).Tao Fang Huiting Huang Jianyong Feng Yingfei Hu Qinfeng Qian Shicheng Yan Zhentao Yu Zhaosheng Li Zhigang Zou 2019Research2019,,1:2

8Model predictive control system based on direct yaw moment control for 4WID self-steering agriculture vehicle显示文摘A model predictive control(MPC)approach based on direct yaw moment control(DYC)was proposed to realize the self-steering drive for a newly autonomous four-wheel independent-drive(4WID)agricultural electric vehicle.The front axle and rear axle of the vehicle chassis could rotate simultaneously around their respective center points and cut the turning radius in half at most through specific mechanical chassis structure design and four-wheel electrical drive.It had great potential to reduce wheel traffic damage to field crops if two rear electrical drive wheels can be controlled to follow wheel tracks of two front wheels during self-steering operation.Therefore,firstly,a two-degree-freedom dynamics model presenting this agricultural electric vehicle was constructed.Then,an MPC controller combined with DYC was applied to arrange torques from four wheels to match desired turning angles,direct yaw moments and travel speeds.The simulation results existed small steady error of steering angles below 0.22%as they were set at 5°,followed with yaw moment under 0.17%and velocity less than 1%.Finally,according to experiment results,the vehicle successfully made a working turning radius of 9.1 m with maximum error of 0.55%when desired steering angles were 5°at the speed of 1 m/s and a minimum turning radius of 1.51 m with maximum error of 6.6%when steering angles were 30°at the speed of 0.5 m/s.It verified that the 4WID agricultural electric vehicle could drive autonomously and steady with small self-steering angle error under the proposed control system and has a feasibility to reduce wheel traffic damage during driving and operation.Hui Liu Shicheng Yan Yue Shen Chiheng Li Yafei Zhang Fida Hussain 2021International Journal of Agricultural and Biological Engineering2021,14,2:1

13Electrochemical Biorefinery toward Chemicals Synthesis and Bio-Oil Upgrading from Lignin显示文摘Recalcitrance and the inherent heterogeneity of lignin structure are the major bottlenecks to impede the popularization of lignin-based chemicals production processes.Recent works suggested a promising pathway for lignin depolymerization and lignin-derived bio-oil upgrading via an electrochemical biorefinery(a process in which lignin valorization is performed via electrochemical oxidation or reduction).This review presents the progress on chemicals synthesis and bio-oil upgrading from lignin by an electrochemical biorefinery,relating to the lignin biosynthesis pathway,reaction pathway of lignin electrochemical conversion,inner-sphere and outer-sphere electron transfer mechanism,basic kinetics and thermodynamics in electrochemistry,and the recent embodiments analysis with the emphasis on the respective feature and limitation for lignin electrochemical oxidative and reductive conversion.Lastly,the challenge and perspective associated with lignin electrochemical biorefinery are discussed.Present-day results indicate that more work should be performed to promote efficiency,selectivity,and stability in pursuing a lignin electrochemical biorefinery.One of the most promising developing directions appears to be integrating various types of lignin electrochemical conversion strategies and other existing or evolving lignin valorization technologies.This review aims to provide more references and discussion on the development for lignin electrochemical biorefinery.Rui Hu Yuying Zhao Chen Tang Yan Shi Gang Luo Jiajun Fan James H.Clark Shicheng Zhang 2023Engineering2023,,8:0
14Genetic Diversities of 23 Y‑Chromosome Short Tandem Repeat Loci in a Han Population in the Beijing Region显示文摘We investigated the polymorphisms of 23 Y‑short tandem repeat(STR)loci in a Han population in the Beijing region.Blood samples were collected from 255 unrelated Han males.DNA templates were amplified using the PowerPlex®Y23 system,and the amplification products were detected with a 3130 genetic analyzer.A total of 254 haplotypes were detected from the 255 unrelated Han males in the Beijing region.The gene diversity of these 23 Y‑STR loci was 0.3952–0.9721.The haplotype diversity was 0.99996 and discrimination capacity(DC)was more than 99.6%.The 23 Y‑STR loci used in this study are highly polymorphic in Han individuals in the Beijing region and are therefore suitable for paternal kinship identification.Studying allelic deletions such as DYS448 and DYS549 are important for examining Y‑STR polymorphisms and forensic testing.Shicheng Hao Xuan Zhang Yan Liu Di Lu 2018Journal of Forensic Science and Medicine2018,4,2:0
15Dynamic Recrystallization Behavior of Q370qE Bridge Steel显示文摘Bridge steel has been widely used in recent years for its excellent performance. Understanding the high-temperature Dynamic Recrystallization (DRX) behavior of high-performance bridge steel plays an important role in guiding the thermomechanical processing process. In the present study, the hot deformation behavior of Q370qE bridge steel was investigated by hot compression tests conducted on a Gleeble 3800-GTC thermal-mechanical physical simulation system at temperatures ranging from 900 ℃ to 1100 ℃ and strain rates ranging from 0.01 s^(−1) to 10 s^(−1). The obtained results were used to plot the true stress-strain and work-hardening rate curves of the experimental steel, with the latter curves used to determine the critical strains for the initiation of DRX. The Zener-Hollomon equation was subsequently applied to establish the correspondence between temperature and strain rate during the high-temperature plastic deformation of bridge steel. In terms of the DRX volume fraction solution, a new method for establishing DRX volume fraction was proposed based on two theoretical models. The good weathering and corrosion resistance of bridge steel lead to difculties in microstructure etching. To solve this, the MTEX technology was used to further develop EBSD data to characterize the original microstructure of Q370qE bridge steel. 18Identification of Half‑Sisters from Different Mothers by Autosomal and X Chromosomal Short Tandem Repeats:A Case Study显示文摘Complex kinship identification such as half‑sibling identification is a difficult task in forensic biology Here we represented an approach in dealing with half‑sisters from different mothers,with the combination of autosomal and X chromosomal short‑tandem repeats(STRs)data.X chromosomal STRs can offer additional information,especially in some cases where autosomal STRs alone may not provide enough information for an accurate opinion.In this case,half‑sister or unrelated relationship between two women(S_(1)and S_(2))with different mothers were distinguished.23 autosomal and 31 X chromosomal STRs of S_(1),S_(2),S_(1)’s mother(M1),S_(2)’s mother(M2)and S_(1)’s grandmother(G1)were profiled with three different commercial kits.As to X‑chromosome STRs,likelihood ratios(LRs)were calculated by FamLinkX with consideration of linkage,linkage disequilibrium,and mutations.When only the profiles of the two individuals(S_(1)and S_(2))were available,LRs between S_(1)and S_(2)were 1.1110×10^(2)based on 23 autosomal STRs and 3.2257 om107 based on 31 X chromosomal STRs.When the maternal genotypes were taken into consideration,LRs increased to 2.5297×10^(3)and 3.0563×10^(18).Therefore,both the DNA profiles of each mothers and X chromosomal STRs are important in dealing with the identification of half‑sisters from different mothers.Jinpei Zhang Shicheng Hao Yan Liu Li Yuan 2021Journal of Forensic Science and Medicine2021,7,2:0
19Study of Autosomal Short Tandem Repeat Loci Using ITO Method in Full‑Sibling Identification显示文摘This study aimed to investigate the application of autosomal short tandem repeat(STR)loci using the ITO method and discriminant function algorithm for full‑sibling(FS)identification.A total of 342 pairs of full siblings(FSs)and 3900 pairs of unrelated individuals(UIs)were genotyped at 51 STR loci.The groups were in accordance with discrimination power(DP)values and the number of loci,and the values of FS index(FSI)of FSs and UIs were calculated by the ITO method.The discriminant functions of FS–UI were established using the Fisher’s discriminant analysis method with SPSS 19.0 software.All the lgFSI values in the FS and UI groups followed a normal distribution,and there were significant differences between the two pairs.A higher average DP value was associated with a more significant difference,as was a greater number of STR loci detected.Receiver operator characteristic curves showed that the accuracy of FS identification can be affected by both locus polymorphism and the number of loci detected.Comparing the rate of false positives and false negatives of discriminant function between the two groups,a higher average DP value and larger number of loci detected were associated with a lower rate of miscarriage of justice and were more helpful for FS–UI identification.The ITO‑based discriminant analysis method has high applicability in FS–UI tests.Testing of a greater number of STR loci promotes FS identification.Li Yuan Xu Xu He Ren Zhao Zhao Tong Wang Shicheng Hao Jinpei Zhang Yan Liu Yan Xu 2020Journal of Forensic Science and Medicine2020,6,1:0
20Genetic Polymorphism Investigation of 19 X‑STR Loci in the Han Population in Northern China显示文摘To investigate the genetic polymorphisms of 19 X‑STR loci in the Han population in Northern China,samples from 628 unrelated individuals(314 males and 314 females)were collected and 19 X‑STR loci were amplified by AGCU X19 STR System.A total of 270 different alleles were detected in 19 X‑STR loci.All loci were in Hardy−Weinberg equilibrium and there was only one pair of linkage loci(DXS10103‑DXS10101).There was no significant difference in allele frequency between male and female populations.The combined power of discrimination in males was 1–1.8667×10−13,while the combined power of discrimination in females was 1–3.6532×10−22.The combined mean paternity exclusion chance(CMEC)for X-chromosomal markers in father/daughter or mother/son duos Mean paternity exclusion chance(MECDesmarais Duo)was 1–5.1109×10−9.Moreover,the CMEC for X-chromosomal markers in trios involving daughters(MECDesmarais)was 1–2.0292×10−12.The compound amplification system composed of 19 X‑STR in this study showed high polymorphism in the Han population of Northern China,which had a high application value in difficult genetic relationship identification.Shicheng Hao Yan Liu Yan Xu Dong Zhao Gexin Liu Jinpei Zhang Li Yuan 2022Journal of Forensic Science and Medicine2022,8,3:0
