1ZmMs30 Encoding a Novel GDSL Lipase Is Essential for Male Fertility and Valuable for Hybrid Breeding in Maize显示文摘Genic male sterility (GMS) is very useful for hybrid vigor utilization and hybrid seed production. Although a large number of GMS genes have been identified in plants, little is known about the roles of GDSL lipase members in anther and pollen development. Here, we report a maize GMS gene, ZmMs30, which encodes a novel type of GDSL lipase with diverged catalytic residues. Enzyme kinetics and activity assays show that ZmMs30 has lipase activity and prefers to substrates with a short carbon chain. ZmMs30 is specifically expressed in maize anthers during stages 7-9. Loss of ZmMs30 function resulted in defective anther cuticle, irregular foot layer of pollen exine, and complete male sterility. Cytological and lipidomics analyses demonstrate that ZmMs30 is crucial for the aliphatic metabolic pathway required for pollen exine formation and anther cuticle development. Furthermore, we found that male sterility caused by loss of ZmMs30 function was stable in various inbred lines with different genetic background, and that it didn't show any negative effect on maize heterosis and production, suggesting that ZmMs30 is valuable for crossbreeding and hybrid seed production. We then developed a new multi-control sterility system using ZmMs30 and its mutant line, and demonstrated it is feasible for generating desirable GMS lines and valu. able for hybrid maize seed production. 3Design of Eco-friendly Ultra-high Performance Concrete with Supplementary Cementitious Materials and Coarse Aggregate显示文摘Aiming to investigate the mix design of eco-friendly UHPC with supplementary cementitious materials and coarser aggregates, we comprehensively studied the workability, microstructure, porosity, compressive strength, flexural strength, and Young’s modulus of UHPC. Relationship between compressive strength and Young’s modulus was obtained eventually. It is found that the compressive strength, flexural strength, and Young’s modulus of UHPC increase by 19.01%, 10.81%, and 5.99%, respectively, when 40 wt% cement is replaced with supplementary cementitious materials. 4A specific aggregation-induced emission-conjugated polymer enables visual monitoring of osteogenic differentiation显示文摘Osteogenic differentiation is the basis of bone growth and repair related to many diseases,in which evaluating the degree and ability of osteogenic transformation is quite important and highly desirable.However,fixing or stopping the growth of cells is required for conventional methods to monitor osteogenic differentiation,which cannot realize the full investigation of the dynamic process.Herein,a new anion conjugated polymer featuring aggregation-induced emission(AIE)characteristics is developed with excellent solubility for in-situ monitoring the process of osteogenic differentiation.This novel polymer can bind with osteogenic differentiated cells,and the intracellular fluorescence increases gradually with the enhancement of osteogenic differentiation.Moreover,it possesses good biosafety with negligible effect on cell activity and osteogenic differentiation,which cannot be realized by the typical method of Alizarin Red S staining.Further study shows that the polymer crosses the cell membrane through endocytosis and enriches in lysosomes,whereas no obvious fluorescence is detected with other cells,including non-differentiated osteoblast cells,under the same conditions,demonstrating the high selectivity.This is the first fluorescent probe with excellent specificity to realize real-time observation of the process of osteogenic differentiation.Therefore,PTB-EDTA shows great promise in the study of osteogenic differentiation and related applications.Zhenyu Zheng Taotao Zhou Rong Hu Minjun Huang Xiang Ao Jun Chu Tao Jiang Anjun Qin Zhongmin Zhang 2020Bioactive Materials2020,5,4:3
5Lab-in-cell based on spontaneous amino-yne click polymerization显示文摘Unnatural reaction in the living cells is a powerful tool for biological research. However, the polymerization inside cells is rarely reported. In this work, a lab-in-cell is illustrated based on our developed spontaneous amino-yne click polymerization. Carbonyl group activated terminal diyne can spontaneously polymerize with tetraphenylethene(TPE)-containing primary diamine inside cells, and polymer with weight-average molecular weight of 7,300 was yielded. By utilizing this in vivo amino-yne click polymerization and taking advantage of the aggregation-induced emission feature of TPE, a 'turn-on' cell imaging was realized,and in-situ killing of cells was also acquired by destroying the structures of actin and tubulin, which cannot be realized by preprepared polymer. 7Experimental study of the settling of twin spherical particles released side by side: The impact of particle size, fluid viscosity, initial spacing, and particle density显示文摘The settling of solid particles in a fluid is an important process that needs to be considered in many fields of research.For example,the interactions among particles and between particles and the surrounding fluid are important topics in studying suspended sediment transport and water clarification.In this paper,the settling processes and interactions of twin spherical particles released side by side were experimentally studied.The Reynolds number varied in the range of 1-300,which is within the transition zone.Particle Tracking Velocimetry(PTV)and Particle Image Velocimetry(PIV)were utilized to capture the settlement trajectory,and provide insight into the flow fields around the particles.The influences of particle size,fluid viscosity,initial spacing,and particle density on the settling process were systematically investigated.The experimental results reveal that the initial spacing between the twin particles(l_(0)*)and the Reynolds number(Re)are the two most important factors affecting particle settling.The interaction between particles comprises only repulsion when the initial spacing is small,while the density of particles has little effect on the final settling state when the initial spacing is not very small.The flow fields around different particles are similar for the same Re,leading to similar final settlement behaviors,except for the case of l_(0)*=0,when the influence of particle rotation cannot be ignored.Except for the case of l_(0)*=0,although the particle density has little effect on the final settling behavior,it affects the repulsive process during settling.The final repulsive distance between the twin particles is highly dependent on Re and l_(0)*.Two critical Re values exist(~10 and~100),where the repulsive distance is negatively correlated with Re when Re<10,but it is positively correlated with Re when Re>100.However,the repulsive distance is always negatively correlated with l_(0)*.These findings can improve understanding of more complex phenomena such as the particle group settling process and sediment transport.Yang Xiao Jieqing Liu Pei Zhang Jian Zhou Dongfang Liang Zhihao Wang Taotao Zhang Saiyu Yuan Hongwu Tang 2023International Journal of Sediment Research2023,38,1:1

11Plasma membrane calcium ATPase 4b inhibits nitric oxide generation through calcium-induced dynamic interaction with neuronal nitric oxide synthase显示文摘The activation and deactivation of Ca^(2+)- and calmodulindependent neuronal nitric oxide synthase (nNOS) in the central nervous system must be tightly controlled to prevent excessive nitric oxide (NO) generation. Considering plasma membrane calcium ATPase (PMCA) is a key deactivator of nNOS, the present investigation aims to determine the key events involved in nNOS deactivation of by PMCA in living cells to maintain its cellular context. Using time-resolved Förster resonance energy transfer (FRET), we determined the occurrence of Ca^(2+)-induced protein-protein interactions between plasma membrane calcium ATPase 4b (PMCA4b) and nNOS in living cells. PMCA activation significantly decreased the intracellular Ca 2+ concentrations ([Ca^(2+)]_(i)), which deactivates nNOS and slowdowns NO synthesis. Under the basal [Ca^(2+)]_(i) caused by PMCA activation, no protein-protein interactions were observed between PMCA4b and nNOS. Furthermore, both the PDZ domain of nNOS and the PDZ-binding motif of PMCA4b were essential for the protein-protein interaction. The involvement of lipid raft microdomains on the activity of PMCA4b and nNOS was also investigated. Unlike other PMCA isoforms, PMCA4 was relatively more concentrated in the raft fractions. Disruption of lipid rafts altered the intracellular localization of PMCA4b and affected the interaction between PMCA4b and nNOS, which suggest that the unique lipid raft distribution of PMCA4 may be responsible for its regulation of nNOS activity. 13Scientific objectives of the Hot Universe Baryon Surveyor(HUBS) mission显示文摘The Hot Universe Baryon Surveyor(HUBS) is a proposed space-based X-ray telescope for detecting X-ray emissions from the hot gas content in our universe. With its unprecedented spatially-resolved high-resolution spectroscopy and large field of view,the HUBS mission will be uniquely qualified to measure the physical and chemical properties of the hot gas in the interstellar medium, the circumgalactic medium, the intergalactic medium, and the intracluster medium. These measurements will be valuable for two key scientific goals of HUBS, namely to unravel the AGN and stellar feedback physics that governs the formation and evolution of galaxies, and to probe the baryon budget and multi-phase states from galactic to cosmological scales. In addition to these two goals, the HUBS mission will also help us solve some problems in the fields of galaxy clusters, AGNs, difuse X-ray backgrounds, supernova remnants, and compact objects. 18Textual Content Prediction via Fuzzy Attention Neural Network Model without Predefined Knowledge显示文摘Text analysis is a popular technique for finding the most significant information from texts including semantic,emotional,and other hidden features,which became a research hotspot in the last few years.Specially,there are some text analysis tasks with judgment reports,such as analyzing the criminal process and predicting prison terms.Traditional researches on text analysis are generally based on special feature selection and ontology model generation or require legal experts to provide external knowledge.All these methods require a lot of time and labor costs.Therefore,in this paper,we use textual data such as judgment reports creatively to perform prison term prediction without external legal knowledge.We propose a framework that combines value-based rules and a fuzzy text to predict the target prison term.The procedure in our framework includes information extraction,term fuzzification,and document vector regression.We carry out experiments with real-world judgment reports and compare our model’s performance with those of ten traditional classification and regression models and two deep learning models.The results show that our model achieves competitive results compared with other models as evaluated by the RMSE and R-squared metrics.Finally,we implement a prototype system with a user-friendly GUI that can be used to predict prison terms according to the legal text inputted by the user.Canghong Jin Guangjie Zhang Minghui Wu Shengli Zhou Taotao Fu 2020China Communications2020,17,6:0

20miRNA-141-5p Affects the Levels of Neutrophil Elastase in Preeclampsia by Regulating MAPK1显示文摘Objective:The objective of this study was to investigate the expression levels of microRNA-141-5p(miRNA-141-5p),MAPK1 and neutrophil elastase in patients with and without preeclampsia(PE),and the relationship between miRNA-141-5p and MAPK1 with respect to the secretion of elastase by neutrophils in patients with PE.Methods:Thirty patients with PE and 30 healthy pregnant(HP)women were recruited from The Second Hospital of Shanxi Medical University,Taiyuan,China,between February 2017 and July 2018.Neutrophils were isolated from 8 mL peripheral blood samples and cultured.We recorded neutrophil count and morphology during culture.Apoptosis was detected by flow cytometry in different groups at 0,24,and 48 h.The expression levels of elastase were detected in neutrophils by enzyme-linked immunosorbent assay,whereas the expression levels of miRNA-141-5p in peripheral blood neutrophils were detected by real-time polymerase chain reaction.We used TargetScanHuman Release 7.2 to analyze the target genes of miRNA-141-5p.The expression of MAPK1 in peripheral blood neutrophils was detected by western blotting.Data were analyzed by SPSS version 21.0 software,and comparisons between groups were carried out with the Studentt test.Results:There was no significant difference between the PE and HP groups(P>0.050)with regard to age or body mass index.The weight of newborns in the PE group(2846.00±600.00 g)was significantly lower than that in the HP group(3055.00±230.68 g).The number of neutrophilic granulocytes(NGs)in blood samples from the PE group was significantly higher than that in the HP group(P=0.003).There was no significant difference between the groups with regard to morphology.Apoptosis in the PE group was delayed when compared with the HP group at different time points.TheP value of apoptosis in the PE and HP groups were respectively 0.790,<0.001 and 0.030 at 0 h,24 h and 48 h.The expression levels of miRNA-141-5p in the PE group were significantly lower than those in the HP group(P<0.050).The expression levels of MAPK1 in neutrophils from the PE group were significantly higher than those in the HP group(P<0.050)by western blot.The expression levels of elastase in neutrophils from the PE group were significantly higher than those in the HP group(P<0.050).Furthermore,the number of NGs in peripheral blood from the PE group was higher than that of the HP group;however,the levels of apoptosis were lower.The expression levels of miRNA-141-5p in NGs decreased,the expression of MAPK1 increased,and the secretion of neutrophil elastase in the NG medium increased in the PE group than those in the HP group.Conclusion:Collectively,our analysis suggested that miRNA-141-5p may be involved in the pathogenesis of PE by regulating the MAPK1 signaling pathway to activate neutrophils and increase the secretion of elastase.Keyan Cheng Jialei Cui Wenli Zhou Huiqiang Liu Taotao Yang Yonghong Wang 2022Maternal-Fetal Medicine2022,4,4:0
