1Infective viruses produced from full-length complementary DNA of swine vesicular disease viruses HK/70 strain显示文摘The full-length cDNA clone of swine vesicular disease virus HK/70 strain named pSVOK12 was constructed in order to study the antigenicity, replication, maturation and pathogenicity of swine vesicular disease virus. In vitro transcription RNA from pSVOK12 transfected IBRS-2 cells and the re- covered virus RNA were isolated and sequenced, then indirect hemagglutination test, indirect im- munofluorescence assays, eleectron microscope test, 50% tissue culture infecting dose (TCID50) assays and mouse virulence studies were performed to study the antigenicity and virulence of the recovered virus. The result showed that the infectious clones we ob- tained and the virus derived from pSVOK12 had the same biological properties as the parental strain HK/70. 2Emerging molecular subtypes and therapeutic targets in B-cell precursor acute lymphoblasti显示文摘B-cell precursor acute lymphoblastic leukemia(BCP-ALL)is characterized by genetic alterations with high heterogeneity.Precise subtypes with distinct genomic and/or gene expression patterns have been recently revealed using high-throughput sequencing technology.Most of these profiles are associated with recurrent non-overlapping rearrangements or hotspot point mutations that are analogous to the established subtypes,such as DUX4 rearrangements,MEF2D rearrangements,ZNF384/ZNF362 rearrangements,NUTM1 rearrangements,BCL2/MYC and/or BCL6 rearrangements,ETV6-RUNX1-like gene expression,PAX5alt(diverse PAX5 alterations,including rearrangements,intragenic amplifications,or mutations),and hotspot mutations PAX5(p.Pro80Arg)with biallelic PAX5 alterations,IKZF1(p.Asn159Tyr),and ZEB2(p.His1038Arg).These molecular subtypes could be classified by gene expression patterns with RNA-seq technology.Refined molecular classification greatly improved the treatment strategy.Multiagent therapy regimens,including target inhibitors(e.g.,imatinib),immunomodulators,monoclonal antibodies,and chimeric antigen receptor T-cell(CAR-T)therapy,are transforming the clinical practice from chemotherapy drugs to personalized medicine in the field of risk-directed disease management.We provide an update on our knowledge of emerging molecular subtypes and therapeutic targets in BCP-ALL.Jianfeng Li Yuting Dai Liang Wu Ming Zhang Wen Ouyang Jinyan Huang Saijuan Chen 2021Frontiers of Medicine2021,15,3:6

6Orbital-regulated interfacial electronic coupling endows Ni3N with superior catalytic surface for hydrogen evolution reaction显示文摘The interstitial structure and weak Ni-N interaction of Ni3N lead to high unoccupied d orbital energy and unsuitable orbital orientation,which consequently results in weak orbital coupling with H2O and slow water dissociation kinetics for alkaline hydrogen evolution catalysis.Herein,we successfully lower the unoccupied d orbital energy of Ni3N to strengthen the interfacial electronic coupling by employing the strong electron pulling capability of oxygen dopants.The prepared O-Ni3N catalyst delivers an overpotential of 55 mV at 10 mA cm−2,very close to the commercial Pt/C.Refined structural characterization indicates the oxygen incorporation can decrease the electron densities around the Ni sites.Moreover,density functional theory calculation further proves the oxygen incorporation can create more unoccupied orbitals with lower energy and superior orientation for water adsorption and dissociation.The concept of orbital-regulated interfacial electronic coupling could offer a unique approach for the rational design of hydrogen evolution catalysts and beyond.Yanyan Fang Da Sun Shuwen Niu Jinyan Cai Yipeng Zang Yishang Wu Linqin Zhu Yufang Xie Yun Liu Zixuan Zhu Amirabbas Mosallanezhad Di Niu Zheng Lu Junjie Shi Xiaojing Liu Dewei Rao Gongming Wang Yitai Qian 2020Science China Chemistry2020,63,11:1
7Rapid Detection Co-infections of Classical Swine Fever Virus and Porcine Reproductive and Respiratory Syndrome Virus by One-step Multiplex RT-PCR显示文摘Classical swine fever virus(CSFV) and porcine reproductive and respiratory syndrome virus(PRRSV) have caused immense economic loss in the pig industry and are considered to be the two most important infectious diseases of pigs in the world. A multiplex reverse transcription polymerase chain reaction(multiplex RT-PCR) was developed for CSFV and PRRSV co-infections or infections,respectively. A set of two pairs of primer was designed based on the sequence of nonstructural protein NS54B of CSFV and ORF7 gene of PRRSV. The diagnostic accuracy of multiplex RT-PCR assay was evaluated by using 56 field clinical samples by multiplex RT-PCR,single RT-PCR and sequence analysis;and the specificity of multiplex PCR was verified by using constructed plasmids containing the specific viral target fragments of PRRSV and CSFV,respectively. The results indicated that this assay could reliably differentiate PRRSV and CSFV in co-infection samples. 13Accelerating water dissociation kinetics of Ni3N by tuning interfacial orbital coupling显示文摘The high unoccupied d band energy of Ni_(3)N basically results in weak orbital coupling with water molecule,consequently leading to slow water dissociation kinetics.Herein,we demonstrate Cr doping can downshift the unoccupied d orbitals and strengthen the interfacial orbital coupling to boost the water dissociation kinetics.The prepared Cr-Ni_(3)N/Ni displays an impressive overpotential of 37 mV at 10 mA·cmgeo-2,close to the benchmark Pt/C in 1.0 M KOH solution.Refined structural analysis reveals the Cr dopant exists as the Cr-N_(6)states and the average d band energy of Ni_(3)N is also lowered.Density functional theory calculation further confirms the downshifted d band energy can strengthen the orbital coupling between the unpaired electrons in O 2p and the unoccupied state of Ni 3d,which thus facilitates the water adsorption and dissociation.The work provides a new concept to achieve on-demand functions for hydrogen evolution catalysis and beyond,by regulating the interfacial orbital coupling.Yishang Wu Yufang Xie Shuwen Niu Yipeng Zang Jinyan Cai Zenan Bian Xuanwei Yin Yanyan Fang Da Sun Di Niu Zheng Lu Amirabbas Mosallanezhad Huijuan Wang Dewei Rao Hongge Pan Gongming Wang 2021Nano Research2021,14,10:1

18USP7 deubiquitinates and stabilizes NOTCH1 in T-cell acute lymphoblastic leukemia显示文摘T-cell acute lymphoblastic leukemia(T-ALL)is a highly aggressive leukemia that is primarily caused by aberrant activation of the NOTCH1 signaling pathway.Recent studies have revealed that posttranslational modifications,such as ubiquitination,regulate NOTCH1 stability,activity,and localization.However,the specific deubiquitinase that affects NOTCH1 protein stability remains unestablished.Here,we report that ubiquitin-specific protease 7(USP7)can stabilize NOTCH1.USP7 deubiquitinated NOTCH1 in vivo and in vitro,whereas knockdown of USP7 increased the ubiquitination of NOTCH1.USP7 interacted with NOTCH1 protein in T-ALL cells,and the MATH and UBL domains of USP7 were responsible for this interaction.Depletion of USP7 significantly suppressed the proliferation of T-ALL cells in vitro and in vivo,accompanied by downregulation of the NOTCH1 protein level.Similarly,pharmacologic inhibition of USP7 led to apoptosis of T-ALL cells.More importantly,we found that USP7 was significantly upregulated in human T-ALL cell lines and patient samples,and a USP7 inhibitor exhibited cell cytotoxicity toward primary T-ALL cells,indicating the clinical relevance of these findings.Overall,our results demonstrate that USP7 is a novel deubiquitinase that stabilizes NOTCH1.Therefore,USP7 may be a promising therapeutic target in the currently incurable T-ALL.Huizhuang Shan Xiangyun Li Xinhua Xiao Yuting Dai Jinyan Huang Junjun Song Meng Liu Li Yang Hu Lei Yin Tong Li Zhou Hanzhang Xu Yingli Wu 2018Signal Transduction and Targeted Therapy2018,3,1:0
19Solving quantified constraint satisfaction problems with value selection rules显示文摘Solving a quantified constraint satisfaction problem(QCSP)is usually a hard task due to its computational complexity.Exact algorithms play an important role in solving this problem,among which backtrack algorithms are effective.In a backtrack algorithm,an important step is assigning a variable by a chosen value when exploiting a branch,and thus a good value selection rule may speed up greatly.In this paper,we propose two value selection rules for existentially and universally quantified variables,respectively,to avoid unnecessary searching.The rule for universally quantified variables is prior to trying failure values in previous branches,and the rule for existentially quantified variables selects the promising values first.Two rules are integrated into the state-of-the-art QCSP solver,i.e.,QCSP-Solve,which is an exact solver based on backtracking.We perform a number of experiments to evaluate improvements brought by our rules.From computational results,we can conclude that the new value selection rules speed up the solver by 5 times on average and 30 times at most.We also show both rules perform well particularly on instances with existentially and universally quantified variables occurring alternatively.Jian Gao Jinyan Wang Kuixian Wu Rong Chen 2020Frontiers of Computer Science2020,14,5:0
