| 2 | Characterization of drug responses of mini patient-derived xenografts in mice for predicting cancer patient clinical therapeutic response显示文摘Background:Patient-derived organoids and xenografts(PDXs)have emerged as powerful models in functional diag-nostics with high predictive power for anticancer drug response.However,limitations such as engraftment failure and time-consuming for establishing and expanding PDX models followed by testing drug efficacy,and inability to subject to systemic drug administration for ex vivo organoid culture hinder realistic and fast decision-making in selecting the right therapeutics in the clinic.The present study aimed to develop an advanced PDX model,namely MiniPDX,for rapidly testing drug efficacy to strengthen its value in personalized cancer treatment.Methods:We developed a rapid in vivo drug sensitivity assay,OncoVee®MiniPDX,for screening clinically relevant regimens for cancer.In this model,patient-derived tumor cells were arrayed within hollow fiber capsules,implanted subcutaneously into mice and cultured for 7 days.The cellular activity morphology and pharmacokinetics were systematically evaluated.MiniPDX performance(sensitivity,specificity,positive and negative predictive values)was examined using PDX as the reference.Drug responses were examined by tumor cell growth inhibition rate and tumor growth inhibition rate in PDX models and MiniPDX assays respectively.The results from MiniPDX were also used to evaluate its predictive power for clinical outcomes.Results:Morphological and histopathological features of tumor cells within the MiniPDX capsules matched those both in PDX models and in original tumors.Drug responses in the PDX tumor graft assays correlated well with those in the corresponding MiniPDX assays using 26 PDX models generated from patients,including 14 gastric cancer,10 lung cancer and 2 pancreatic cancer.The positive predictive value of MiniPDX was 92%,and the negative predictive value was 81%with a sensitivity of 80%and a specificity of 93%.Through expanding to clinical tumor samples,Min-iPDX assay showed potential of wide clinical application.Conclusions:Fast in vivo MiniPDX assay based on capsule implantation was developed-to assess drug responses of both PDX tumor grafts and clinical cancer specimens.The high correlation between drug responses of paired MiniPDX and PDX tumor graft assay,as well as translational data suggest that MiniPDX assay is an advanced tool for personalized cancer treatment. | Feifei Zhang Wenjie Wang Yuan Long Hui Liu Jijun Cheng Lin Guo Rongyu Li Chao Meng Shan Yu Qingchuan Zhao Shun Lu Lili Wang Haitao Wang Danyi Wen | 2018 | Cancer Communications2018,38,1: | 20 |

| 4 | SHP2 inhibition triggers anti-tumor immunity and synergizes with PD-1 blockade显示文摘Tyrosine phosphatase SHP2 is a promising drug target in cancer immunotherapy due to its bidirectional role in both tumor growth promotion and T-cell inactivation. Its allosteric inhibitor SHP099 is known to inhibit cancer cell growth both in vitro and in vivo. However, whether SHP099-mediated SHP2 inhibition retards tumor growth in vivo via anti-tumor immunity remains elusive. To address this, a CT-26 colon cancer xenograft model was established in mice since this cell line is insensitive to SHP099.Consequently, SHP099 minimally affected CT-26 tumor growth in immuno-deficient nude mice, but significantly decreased the tumor burden in CT-26 tumor-bearing mice with intact immune system.SHP099 augmented anti-tumor immunity, as shown by the elevated proportion of CD8tIFN-γtT cells and the upregulation of cytotoxic T-cell related genes including Granzyme B andPerforin, which decreased the tumor load. In addition, tumor growth in mice with SHP2-deficient T-cells was markedly slowed down because of enhanced anti-tumor responses. Finally, the combination of SHP099 and antiPD-1 antibody showed a higher therapeutic efficacy than either monotherapy in controlling tumor growthin two colon cancer xenograft models, indicating that these agents complement each other. | 9 | Effect of Silicon on Growth, Physiology, and Cadmium Translocation of Tobacco (Nicotiana tabacum L.) in Cadmium-Contaminated Soil显示文摘Silicon(Si) offers beneficial effect on plants under cadmium(Cd) stress such as promoting plant growth and increasing resistance to heavy metal toxicity. In this study, a pot experiment was performed to study the role of Si in alleviating Cd toxicity in tobacco(Nicotiana tabacum L.) plants on a yellow soil taken from Guiyang, China. Nine treatments consisting of three concentrations of Cd(0, 1, and 5 mg kg^(-1)) together with three Si levels(0, 1, and 4 g kg^(-1)) were established. Plant growth parameters, Cd concentration,and the malondialdehyde(MDA), chlorophyll, and carotenoid contents were determined 100 d after transplanting of tobacco seedlings.Application of exogenous Si enhanced the growth of tobacco plants under Cd stress. When 5 mg kg^(-1) Cd was added, Si addition at 1 and 4 g kg^(-1) increased root, stem, and leaf biomass by 26.1%–43.3%, 33.7%–43.8%, and 50.8%–69.9%, respectively, compared to Si addition at 0 g kg^(-1). With Si application, the transfer factor of Cd in tobacco from root to shoot under both 1 and 5 mg kg^(-1) Cd treatments decreased by 21%. The MDA contents in the Si-treated tobacco plants declined by 5.5%–17.1% compared to those in the non-Si-treated plants, indicating a higher Cd tolerance. Silicon application also increased the chlorophyll and carotenoid contents by 33.9%–41% and 25.8%–47.3% compared to the Cd only treatments. | 10 | Genome-wide identification and co-expression network analysis of the Os NF-Y gene family in rice显示文摘Nuclear factor Y(NF-Y) is a ubiquitous transcription factor that regulates important physiological and developmental processes. In this study, we identified 34 Os NF-Y genes in rice, including 6 newly identified genes. Expression profile analysis covering the whole life cycle revealed that transcripts of Os NF-Y differentially accumulated in a tissue-specific,preferential or constitutive manner. In addition, gene duplication studies and expression analyses were performed to determine the evolutionary origins of the Os NF-Y gene family.Nine Os NF-Y genes were differentially expressed after treatment of seedlings with one or more abiotic stresses such as drought, salt and cold. Analysis of expression correlation and Gene Ontology annotation suggested that Os NF-Y genes were co-expressed with genes that participated in stress, accumulation of seed storage reserves, and plant development.Co-expression analysis also revealed that Os NF-Y genes might interact with each other,suggesting that NF-Y subunits formed complexes that take part in transcriptional regulation. | 14 | A draft sequence of the rice (Oryza sativa ssp. indica) genome显示文摘The sequence of the rice genome holds fundamental information for its biology, including physiology, genetics, development, and evolution, as well as information on many beneficial phenotypes of economic significance. Using a 'whole genome shotgun' approach, we have pro-duced a draft rice genome sequence of Oryza sativa ssp. in-dica, the major crop rice subspecies in China and many other regions of Asia. The draft genome sequence is constructed from over 4.3 million successful sequencing traces with an accumulative total length of 2214.9 Mb. The initial assembly of the non-redundant sequences reached 409.76 Mb in length, based on 3.30 million successful sequencing traces with a total length of 1797.4 Mb from an indica variant cultivar 93-11, giving an estimated coverage of 95.29% of the rice genome with an average base accuracy of higher than 99%. | 17 | Tuning F-doped degree of rGO:Restraining corrosion-promotion activity of EP/rGO nanocomposite coating显示文摘Given that graphene features high electrical conductivity,it is a kind of material with corrosion-promotion activity.This study aimed to inhibit the corrosion-promotion activity of graphene in coatings.Here,we report an exciting application of epoxy matrix(EP)/F-doped reduced graphene oxide(rGO)coatings for the long-term corrosion protection of steel.The synthesized F-doped rGO(FG)did not reduce the utilization of rGO by a wide margin and possessed distinctive electrically insulating nature.The electrical conductivity of rGO was approximately 1500 S/m,whereas those of FG-1,FG-2 and FG-3 were 1.17,5.217×10^−2 and 3.643×10^-11 S/m,respectively.FG and rGO were then dispersed into epoxy coatings.The chemical structures of rGO and FG were investigated by transmission electron microscopy(TEM),scanning probe microscopy(SPM),X-ray photoelectron spectroscopy(XPS),Fourier-transform infrared spectroscopy(FTIR),and X-ray diffraction(XRD).EP/FG coatings exhibited outstanding corrosion protection in comparison with blank EP and EP/rGO coatings mainly because the corrosion-promotion effect of rGO was eliminated.The anticorrosion ability of EP/FG coatingswasimproved with increased F-doped degree of FG.In addition,electrochemical impendance spectroscopy(EIS)results indicated that the Rc values of EP/FG-2 and EP/FG-3 were four orders of magnitude higher than those of EP/rGO in diluent NaCl solution(3.5 wt.%)after immersion for 90 days. | Lu Shen Yong Li Wenjie Zhao Kui Wang Xiaojing Ci Yangmin Wu Gang Liu Chao Liu Zhiwen Fang | 2020 | Journal of Materials Science & Technology2020,44,9: | 5 |

| 20 | Antidepressant-like effect of active fraction of Polyrhachisvicina Roger in a rat depression model显示文摘OBJECTIVE: To investigate the antidepressant-likeeffect of active fraction of Polyrhachis vicina Roger(AFPR) in a rat depression model, and to elucidate the underlying mechanism.METHODS: AFPR was extracted with ethanol followed by petroleum ether. Its antidepressant-like effect was investigated in mice by tail suspension test(TST), forced swimming test(FST) and open field test(OPT). A repeated dose of reserpine(0.5 mg/kg, daily for 14 d) was used to establish a rat depression model. Fluoxetine was used as positive control agent. The effect of AFPR on reserpine-induced ptosis, hypothermia and akinesia, the levels of monoamines and their metabolites, and the activity of monoamine oxidase(MAO) in hippocampus and prefrontal cortex were determined.RESULTS: Administration of AFPR by gavage at 160 and 320 mg/kg significantly reduced the duration of immobility in the FST and TST, and did not affect locomotor activity in the OPT. In the reserpine-induced depression model, AFPR attenuated anhedonia, demonstrated by reversing hypothermia, akinesia and sucrose consumption. AFPR significantly increased the concentration of monoamines, including dopamine, serotonin, noradrenaline and acetylcholine.CONCLUSION: AFPR normalized the metabolism rates of noradrenaline, serotonin and dopamine,and the activity of MAO, which were altered by chronic reserpine exposure.
