| 1 | A complete sequence and comparative analysis of a SARS-associated virus(Isolate BJ01)显示文摘The genome sequence of the Severe Acute Respiratory Syndrome (SARS)-associated virus provides essential information for the identification of pathogen(s), exploration of etiology and evolution, interpretation of transmission and pathogenesis, development of diagnostics, prevention by future vaccination, and treatment by developing new drugs. We report the complete genome sequence and comparative analysis of an isolate (BJ01) of the coronavirus that has been recognized as a pathogen for SARS. The genome is 29725 nt in size and has 11 ORFs (Open Reading Frames). It is composed of a stable region encoding an RNA-dependent RNA polymerase (composed of 2 ORFs) and a variable region representing 4 CDSs (coding sequences) for viral structural genes (the S, E, M, N proteins) and 5 PUPs (putative uncharacterized proteins). Its gene order is identical to that of other known coronaviruses. The sequence alignment with all known RNA viruses places this virus as a member in the family of Coronaviridae. Thirty putative substitutions have been identified by comparative analysis of the 5 SARS- associated virus genome sequences in GenBank. Fifteen of them lead to possible amino acid changes (non-synonymous mutations) in the proteins. Three amino acid changes, with predicted alteration of physical and chemical features, have been detected in the S protein that is postulated to beinvolved in the immunoreactions between the virus and its host. Two amino acid changes have been detected in the Mprotein, which could be related to viral envelope formation. Phylogenetic analysis suggests the possibility of non-human origin of the SARS-associated viruses but provides noevidence that they are man-made. | 2 | Andrographolide sulfonate ameliorates lipopolysaccharide-induced acute lung injury in mice by down-regulating MAPK and NF-κB pathways显示文摘Acute lung injury(ALI) or acute respiratory distress syndrome(ARDS) is a severe, lifethreatening medical condition characterized by widespread inflammation in the lungs, and is a significant source of morbidity and mortality in the patient population. New therapies for the treatment of ALI are desperately needed. In the present study, we examined the effect of andrographolide sulfonate, a water-soluble form of andrographolide(trade name: Xi-Yan-Ping Injection), on lipopolysaccharide(LPS)-induced ALI and inflammation. Andrographolide sulfonate was administered by intraperitoneal injection to mice with LPS-induced ALI. LPS-induced airway inflammatory cell recruitment and lung histological alterations were significantly ameliorated by andrographolide sulfonate. Protein levels of pro-inflammatory cytokines in bronchoalveolar lavage fluid(BALF) and serum were reduced by andrographolide sulfonate administration. m RNA levels of pro-inflammatory cytokines in lung tissue were also suppressed. Moreover, andrographolide sulfonate markedly suppressed the activation of mitogen-activated protein kinase(MAPK) as well as p65 subunit of nuclear factor-κB(NF-κB). In summary, these results suggest that andrographolide sulfonate ameliorated LPS-induced ALI in mice by inhibiting NF-κB and MAPK-mediated inflammatory responses. | 3 | An energy-efficient geographic routing based on cooperative transmission in wireless sensor networks显示文摘In this paper, we propose an energy-efficient cooperative geographic routing (ECGR) to reach energy-efficient routing in wireless sensor networks (WSN). ECGR takes full advantage of cooperative diversity and geographic routing. At physical layer, cooperative nodes transmit the same packet in a cooperative way to achieve farther transmission distance of each hop without more energy consumption. While at the network layer, ECGR adaptively selects the appropriate cooperative nodes to form forwarding clusters to forward the packet to the destination when we consider energy consumption and geographic information of the sensor nodes. Meanwhile, in order to fully assess the energy consumption, we take circuit energy consumption of the sensor nodes into consideration. | 5 | Reconstruction of temperature series of China for the last 1000 years显示文摘This paper reports a study on reconstructing temperature series for ten regions of China over the last 1000 years with a time resolution of 10 a. The regions concerned are: Northeast, North, East, South China, Taiwan, Central, Southwest, Northwest China, Xinjiang and Qinghai-Tibet Plateau. A variety of proxy data, such as ice core, tree-rings, stalagmites, peat, lake sediments, pollen and historical records, were validated with instrumental observations made in the last 120 years, and applied in the recon- struction of the temperature series. A temperature series for whole China is then established by aver- aging the ten regional series with a weighting proportional to the area of each region. | 8 | Characteristics of atmospheric single particles during haze periods in a typical urban area of Beijing:A case study in October,2014显示文摘To investigate the composition and possible sources of particles, especially during heavy haze pollution, a single particle aerosol mass spectrometer(SPAMS) was deployed to measure the changes of single particle species and sizes during October of 2014, in Beijing. A total of 2,871,431 particles with both positive and negative spectra were collected and characterized in combination with the adaptive resonance theory neural network algorithm(ART-2a). Eight types of particles were classified: dust particles(dust, 8.1%), elemental carbon(EC, 29.0%), organic carbon(OC, 18.0%), EC and OC combined particles(ECOC, 9.5%),Na-K containing particles(Na K, 7.9%), K-containing particles(K, 21.8%), organic nitrogen and potassium containing particles(KCN, 2.3%), and metal-containing particles(metal,3.6%). Three haze pollution events(P1, P2, P3) and one clean period(clean) were analyzed,based on the mass and number concentration of PM_(2.5)and the back trajectory results from the hybrid single particle Lagrangian integrated trajectory model(Hysplit-4 model). Results showed that EC, OC and K were the major components of single particles during the three haze pollution periods, which showed clearly increased ratios compared with those in the clean period. | 10 | SHP2 inhibition triggers anti-tumor immunity and synergizes with PD-1 blockade显示文摘Tyrosine phosphatase SHP2 is a promising drug target in cancer immunotherapy due to its bidirectional role in both tumor growth promotion and T-cell inactivation. Its allosteric inhibitor SHP099 is known to inhibit cancer cell growth both in vitro and in vivo. However, whether SHP099-mediated SHP2 inhibition retards tumor growth in vivo via anti-tumor immunity remains elusive. To address this, a CT-26 colon cancer xenograft model was established in mice since this cell line is insensitive to SHP099.Consequently, SHP099 minimally affected CT-26 tumor growth in immuno-deficient nude mice, but significantly decreased the tumor burden in CT-26 tumor-bearing mice with intact immune system.SHP099 augmented anti-tumor immunity, as shown by the elevated proportion of CD8tIFN-γtT cells and the upregulation of cytotoxic T-cell related genes including Granzyme B andPerforin, which decreased the tumor load. In addition, tumor growth in mice with SHP2-deficient T-cells was markedly slowed down because of enhanced anti-tumor responses. Finally, the combination of SHP099 and antiPD-1 antibody showed a higher therapeutic efficacy than either monotherapy in controlling tumor growthin two colon cancer xenograft models, indicating that these agents complement each other. | 13 | The chromosome-level quality genome provides insights into the evolution of the biosynthesis genes for aroma compounds of Osmanthus fragrans显示文摘Sweet osmanthus(Osmanthus fragrans)is a very popular ornamental tree species throughout Southeast Asia and USA particularly for its extremely fragrant aroma.We constructed a chromosome-level reference genome of O.fragrans to assist in studies of the evolution,genetic diversity,and molecular mechanism of aroma development.A total of over 118 Gb of polished reads was produced from HiSeq(45.1 Gb)and PacBio Sequel(73.35 Gb),giving 100×depth coverage for long reads.The combination of Illumina-short reads,PacBio-long reads,and Hi-C data produced the final chromosome quality genome of O.fragrans with a genome size of 727 Mb and a heterozygosity of 1.45%.The genome was annotated using de novo and homology comparison and further refined with transcriptome data.The genome of O.fragrans was predicted to have 45,542 genes,of which 95.68%were functionally annotated.Genome annotation found 49.35%as the repetitive sequences,with long terminal repeats(LTR)being the richest(28.94%).Genome evolution analysis indicated the evidence of whole-genome duplication 15 million years ago,which contributed to the current content of 45,242 genes.Metabolic analysis revealed that linalool,a monoterpene is the main aroma compound.Based on the genome and transcriptome,we further demonstrated the direct connection between terpene synthases(TPSs)and the rich aromatic molecules in O.fragrans.We identified three new flower-specific TPS genes,of which the expression coincided with the production of linalool.Our results suggest that the high number of TPS genes and the flower tissue-and stage-specific TPS genes expressions might drive the strong unique aroma production of O.fragrans. | Xiulian Yang Yuanzheng Yue Haiyan Li Wenjie Ding Gongwei Chen Tingting Shi Junhao Chen Min S.Park Fei Chen Lianggui Wang | 2018 | Horticulture Research2018,5,1: | 16 |

| 16 | Concentrations and source identification of organochlorine pesticides (OCPs) in soils from Wolong Natural Reserve显示文摘In order to understand the distribution profile, composition characteristics and sources of OCPs, soil samples collected from Wolong Natural Reserve (WNR) in spring and autumn in 2005 were analyzed using gas chromatography coupled with electron capture detector (GC-ECD). Results showed that all OCPs concentrations were in the range of 10?2 ng/g―10?1 ng/g, representing the background level in those remote sites. Generally, their concentrations were slightly higher in spring than in autumn, which could be explained by OCPs: 1) may be released to soil from snow-melting water in spring; 2) partially evaporate into air from soil due to high temperature and intense radiation in summer and 3) can be relatively high degradation rate in summer. There were notable differences of OCPs seasonal variations between samples from alpine meadow and those from under-treeline. This phenomenon was related to physical-chemical properties of OCPs and vegetation coverage. For alpine meadow soils, levels of OCPs were increased with increasing altitudinal gradient, which was associated with enhanced scav-enging efficiency of wet precipitation. EG-ZⅥ had a high specific surface area and contained sub-micron sized particles of zero-valent iron. Batch experiments were employed to evaluate the Cr(Ⅵ) removal performance. The results showed that the Cr(Ⅵ) removal rate was 98.80% for EG-ZⅥ,which was higher than that for both EG(10.00%) and ZⅥ(29.80%). Furthermore, the removal rate of Cr(Ⅵ) by EG-ZⅥ showed little dependence on solution p H within a p H range of 1–9.Even at pH 11, a Cr(Ⅵ) removal rate of 62.44% was obtained after reaction for 1 hr. EG-ZⅥ could enhance the removal of Cr(Ⅵ) via chemical reduction and physical adsorption,respectively. | 19 | Future changes in coverage of 1.5℃ and 2℃warming thresholds显示文摘The areas covered by 1.5 ℃ and 2.0 ℃ warming thresholds under RCP2.6, RCP4.5, and RCP8.5 were analyzed based on 22 CMIP5 models. More than 90% of the model runs are in agreement that by the end of the 21 st century, near-surface air temperature changes over ~5%(~2%), ~40%(~18%), and ~92%(~86%)of the globe will cross the 1.5 ℃(2.0 ℃) threshold under RCP2.6, RCP4.5, and RCP8.5, respectively. Under RCP8.5, nearly the whole of North America, Europe + Russia, Africa, and Asia–Russia will cross the 1.5 ℃(2.0 ℃) threshold in ~2050(~2060), while the coverage rates over South America and Oceania are ~80%(~75%) and ~50%(~30%), respectively. The threshold-onset time(TOT) for 2 ℃ warming is earliest over Europe + Russia and North America, followed by Africa, Asia–Russia, South America, and finally Oceania under the RCP4.5 and RCP8.5 scenarios. | 20 | Analyses of Extreme Climate Events over China Based on CMIP5 Historical and Future Simulations显示文摘Based on observations and 12 simulations from Coupled Model Intercomparison Project Phase 5(CMIP5) models, climatic extremes and their changes over China in the past and under the future scenarios of three Representative Concentration Pathways(RCPs) are analyzed. In observations, frost days(FD) and low-temperature threshold days(TN10P) show a decreasing trend, and summer days(SU), high-temperature threshold days(TX90P), heavy precipitation days(R20), and the contribution of heavy precipitation days(P95T) show an increasing trend. Most models are able to simulate the main characteristics of most extreme indices. In particular, the mean FD and TX90 P are reproduced the best, and the basic trends of FD, TN10 P, SU and TX90 P are represented. For the FD and SU indexes, most models show good ability in capturing the spatial differences between the mean state of the periods 1986–2005 and 1961–80; however, for other indices, the simulation abilities for spatial disparity are less satisfactory and need to be improved. Under the high emissions scenario of RCP8.5,the century-scale linear changes of the multi-model ensemble(MME) for FD, SU, TN10 P, TX90 P, R20 and P95 T are-46.9,46.0,-27.1, 175.4, and 2.9 days, and 9.9%, respectively; and the spatial change scope for each index is consistent with the emissions intensity.
