| 1 | Assessment of the immune responses to Treponema pallidum Gpd DNA vaccine adjuvanted with IL-2 and chitosan nanoparticles before and after Treponema pallidum challenge in rabbits显示文摘Syphilis is a multistage,sexually transmitted disease caused by the spirochete,Treponema pallidum(Tp).A significantly high incidence of syphilis has been reported in several countries,including China,and there is an urgent need for the development of efficacious vaccines against syphilis.DNA vaccines are a major breakthrough in the field of vaccination with several advantages over traditional vaccines.Animal model studies of Tp DNA vaccines have not been reported elsewhere but our previous reports describe the development of a single-gene Tp DNA vaccine and preclinical immunization study.In this study,chitosan(CS) nanoparticles were used as a vector and an interleukin-2 expression plasmid(pIL-2) as an adjuvant to enhance a TpGpd DNA vaccine candidate(pTpGpd) in a rabbit Tp skin challenge model.At week 8 after the first immunization,three rabbits from each group were used to determine cytokine measurements and spleen lymphocyte proliferation assay.pTpGpd in combination with pIL-2 wrapped with CS led to the greatest enhancement of anti-TpGpd antibodies and T-cell proliferation.During infection,levels of anti-TpGpd antibodies and T-cell proliferation were measured.Both the serum special IgG and IL-2,interferon-γ were significantly increased by the co-injection of the IL-2 plasmid compared with the injection of TpGpd DNA alone(P<0.05).Furthermore,IL-2 plasmid coinjection efficiently enhanced the antigen-specific lymphocyte proliferation response.Additionally,the ratios of positive skin lesions and ulcer lesions in groups immunized with pTpGpd were significantly lower than those of the pIL-2,CS or pIL-2 mixed with CS control groups(P<0.001).CS vectored and pIL-2 adjuvanted pTpGpd immunized animals exhibited the lowest rates of positive skin tests(8.33%) and ulcer lesions(4.17%) and the fastest recovery(42 d).These experiments indicate that co-injection of a pIL-2 plasmid with pTpGpd DNA vaccine wrapped with CS can significantly strengthen the long-term stability of immune response during infection,efficiently improve the protective effect against T.pallidum spirochetes infection and attenuate syphilitic lesion development. | ZHAO FeiJun ZHANG XiaoHong LIU ShuangQuan ZENG TieBing YU Jian GU WeiMing ZHANG YueJun CHEN Xi WU YiMou | 2013 | Science China(Life Sciences)2013,56,2: | 8 |
| 2 | Association between dietary habits and recurrent respiratory infection in children: A case-control study显示文摘Objective:To explore the association between dietary habits and recurrent respiratory infection(RRI)in children aged 0e14 years.Methods:This caseecontrol study compared dietary data of children with(cases)and without RRI(controls)collected via structured questionnaire.Participants were recruited from Chinese medicine clinics,hospitals,and children’s learning institutions in Beijing.A logistic regression analysis and odds ratio(OR)calculations were conducted using SPSS 17.0 software.Results:A total of 241 questionnaires were collected(case:control ratio:approximately 2:1).Frequent consumption of processed foods(OR Z 2.988,95%confidence intervals 1.375e6.491)and high-sugar foods(OR Z 2.268,95%confidence intervals 1.163e4.424),frequent picky eating(OR Z 2.614,95%confidence intervals 1.363e5.014),and a meat-heavy diet with fewer vegetables(OR Z 1.830,95%confidence intervals 1.358e2.467)correlated positively correlated with RRI.Additionally,57.80%of the children with RRI were addicted to high-sugar foods,compared with 41.57%of the children without RRI(P Z.015).Furthermore,63.16%of the children with RRI were picky eaters,compared with 48.31%of the children without RRI(P Z.024).Finally,30.92%of the children with RRI frequently consumed processed foods,compared with only 17.98%of the children without RRI(P Z.027Conclusion:Although RRI correlates positively with several dietary habits,in the future,prospective cohort studies with larger samples are needed to generalize these findings. | Wang Zhang He Yu Jinbang Shang Tiegang Liu Jiaju Ma Xiaohong Gu | 2015 | Journal of Traditional Chinese Medical Sciences2015,2,2: | 5 |
| 3 | Phosphine production in anaerobic wastewater treatment under tetracycline antibiotic pressure显示文摘The influence of tetracycline(TC) antibiotics on phosphine(PH_3) production in the anaerobic wastewater treatment was studied. A lab-scale anaerobic baffled reactor with three compartments was employed to simulate this process. The reactor was operated in a TC-absence wastewater and 250 μg/L TC-presence wastewater for three months after a start-up period,respectively. The responses of p H, oxidation–reduction potential(ORP), chemical oxygen demand(COD), total phosphorus(TP), enzymes activity(dehydrogenase and acid phosphatase),and microbial community were investigated to reveal the effect of TC on PH_3 production.Results suggested that the dehydrogenase(DH) activity, acid phosphatase(ACP) activity and COD have positive relationship with PH_3 production, while p H, ORP level and the TP in liquid phase have negative relationship with PH_3 production. | 6 | The efficacy of NP11-4-derived immunotoxin scFv-artesunate in reducing hepatic fibrosis inducedby Schistosoma japonicum in mice显示文摘Schistosomiasis is one of the most prevalent parasitic diseases in China,and hepatic fibrosis caused by schistosome infection is the principal cause of death.The aim of this study was to evaluate the efficacy of NP11-4derived immunotoxin scFv-artesunate on Schistosoma japonicum-induced hepatic fibrosis.A single-chain variable fragment (scFv) was generated from the murine anti-Schistosoma japonicum (S.japanicum) monoclonal antibody NP11-4.The scFv was expressed as a soluble protein and purified by Ni-affinity chromatography.After conjugation with artesunate,the binding ability with soluble egg antigens (SEA) was determined by an enzyme-linked immunosorbent assay (ELISA).The biological activity of purified scFv,scFv-artesunate (immunotoxin),and artesunate was detected in vivo.Image-Pro Plus software was used to analyze the size of egg granuloma and the extent of liver fibrosis.The recombinant scFv expession vector was constructed and expressed successfully.After purification by a His-trap Ni-affinity column,the scFv yield was approximately 0.8 mg/L of culture medium.ELISA results showed that chemical conjugation did not affect the binding activity of the immunotoxin.Our animal experiments indicated that the immunotoxin could significantly reduce the size of egg granuloma in the liver and inhibit hepatic fibrosis.The immunotoxin could be used as a promising candidate in the targeted therapy of S.japonicum-induced hepatic fibrosis. | Hong Li Chunyan Gu Yongya Ren Yang Dai Xiaojuan Zhu Jing Xu Yuhua Li Zhenning Qiu Jin Zhu Yinchang Zhu Xiaohong Guan Zhenqing Feng | 2011 | The Journal of Biomedical Research2011,25,2: | 3 |
| 7 | Drug resistance mutation profiles of the drug-nave and first-line regimen-treated HIV-1-infected population of Suzhou, China显示文摘Little is known about the prevalence of drug-resistant mutations in HIV-1-positive individuals in Suzhou, China. To elucidate the transmitted drug resistance(TDR) and acquired drug resistance mutation(ADR) profiles, we collected blood specimens from 127 drug-naive and 117 first-line drugtreated HIV-1-infected individuals sampled from 2014 to 2016 in Suzhou. We successfully amplified po/fragments from 100 drug-naive and 20 drug-treated samples. We then determined the drugresistant mutations to protease(PR) and reverse-transcriptase(RT) inhibitors according to the Stanford drug resistance database. Overall, 11 and 13 individuals had transmitted(drug-naive group) and acquired(treated group) resistance mutations, respectively. Six transmitted drugresistant mutations were found, including two mutations(L33F and L76V) in the protease region and four(K70N/E and V179D/E) in the RT region. Only L76 V was a major mutation, and K70N/E and V179D/E are known to cause low-level resistance to RT inhibitors. All 13 treated participants who had major drug resistance mutations demonstrated intermediate to high resistance to efavirenz and nevirapine, and six had a treatment duration of less than three months. No major mutations to RT inhibitors were found, implying that the epidemic of transmitted resistance mutations was not significant in this area. Our results suggest that more frequent virus load and drug resistance mutation tests should be conducted for individuals receiving antiretroviral treatment, especially for newly treated patients. | 9 | Growth and feeding habits of invasive Pseudorasbora parva in the Chabalang Wetland(Lhasa,China) and its trophic impacts on native fish显示文摘In recent years, fish invasion has become one of the main reasons for the decline of native fish stocks. Pseudorasbora parva is considered one of the major invasive species worldwide. The present study investigated the fish resources of the Chabalang Wetland (Lhasa, Tibet) during diff erent seasons in 2009 and 2013. Four hundred and twelve individuals were subsampled to estimate age, growth, and feeding habit of P . parva . Furthermore, food relationships between P . parva and the native Schizothoracinae fish were also examined. The results revealed a significant shift in species composition and community structure characterized by the disappearance of native fish and outbreak of non-native fish. The percentage of nonnative P . parva in the fish collections significantly increased from 33.64% in 2009 to 64.08% in 2013. The standard length (SL) ranged from 22.00 to 78.71 mm, and their age was 1–5 a. The von Bertalanff y function was used to model the observed length-at-age data as L t =112.19(1–e^-0.1495 ( t +0.8012)) for females and as L t =123.12 (1–e^-0.1500 ( t +0.7132)) for males. The results indicated that P . parva in Tibet has lower growth and mortality rates compared with that from the native ranges. Ninety-seven prey taxa belonging to 9 prey categories were identified in the gut of 38 P . parva . P . parva can be considered a generalized and opportunistic predator, competing with the native fish, especially Schizothorax o’connori , Schizopygopsis younghusbandi younghusbandi , and Ptychobarbus dipogon , for Bacillariophyta and Chironomid larvae. | 16 | CTCs Detection and Whole-exome Sequencing Might Be Used to Differentiate Benign and Malignant Pulmonary Nodules显示文摘Background and objective Low-density computed tomography(LDCT)improved early lung cancer diagnosis but introduces an excess of false-positive pulmonary nodules data.Hence,accurate diagnosis of early-stage lung cancer remains challenging.The purpose of the study was to assess the feasibility of using circulating tumour cells(CTCs)to differentiate malignant from benign pulmonary nodules.Materials and methods 122 patients with suspected malignant pulmonary nodules detected on chest CT in preparation for surgery were prospectively recruited.Peripheral blood samples were collected before surgery,and CTCs were identified upon isolation by size of epithelial tumour cells and morphological analysis.Laser capture microdissection,MALBAC amplification,and whole-exome sequencing were performed on 8 samples.The diagnostic efficacy of CTCs counting,and the genomic variation profile of benign and malignant CTCs samples were analysed.Results Using 2.5 cells/5 m L as the cut-off value,the area under the receiver operating characteristic curve was of 0.651(95%confidence interval:0.538-0.764),with a sensitivity and specificity of 0.526 and 0.800,respectively,and positive and negative predictive values of 91.1%and 30.3%,respectively.Distinct sequence variations differences in DNA damage repair-related and driver genes were observed in benign and malignant samples.TP53 mutations were identified in CTCs of four malignant cases;in particular,g.7578115T>C,g.7578645C>T,and g.7579472G>C were exclusively detected in all four malignant samples.Conclusion CTCs play an ancillary role in the diagnosis of pulmonary nodules.TP53 mutations in CTCs might be used to identify benign and malignant pulmonary nodules. | Changdan XU Xiaohong XU Weipeng SHAO Hongliang SUN Xiaohong LIU Hongxiang FENG Xianbo ZUO Jingyang GAO Guohui WANG Xiongtao YANG Runchuan GU Shutong GE Shijie WANG Liwei GAO Guangying ZHU | 2023 | 中国肺癌杂志2023,26,6: | 1 |
