| 1 | Emerging Trends for Microbiome Analysis: From Single-Cell Functional Imaging to Microbiome Big Data显示文摘Method development has always been and will continue to be a core driving force of microbiome science.In this perspective, we argue that in the next decade, method development in microbiome analysis will be driven by three key changes in both ways of thinking and technological platforms:(1) a shift from dissecting microbiota structure by sequencing to tracking microbiota state, function, and intercellular interaction via imaging;(2) a shift from interrogating a consortium or population of cells to probing individual cells;and(3) a shift from microbiome data analysis to microbiome data science. Some of the recent methoddevelopment efforts by Chinese microbiome scientists and their international collaborators that underlie these technological trends are highlighted here. | 2 | Underwater explosion in centrifuge Part Ⅱ:Dynamic responses of defensive steel plate显示文摘Part II of this paper is focused on the dynamic response of a steel plate of the centrifuge model which is fixed on the container. This plate is 5 cm thick, allowing for non-destructive elastic response to all the 19 blasts. Three accelerators and 10 sets of strain gauges are installed on the plate to monitor the acceleration and deformation histories induced by shock waves and bubble oscillations.This article presents the characteristic values measured for shock waves and bubble oscillations response. The detailed information can be found in a database available on the web. In order to obtain the correct acceleration time history, the cut-off frequency of low-pass filtering by Fourier transformation was applied. The results indicated that the acceleration and strain induced by shock wave was nearly unaffected by the increasing gravity, while the effect of gravity has remarkable influence on those induced by bubble oscillation. A preliminary study of comparing the measured dynamic response with numerical simulations has been carried out using ANSYS LS-DYNA. The results captured the dynamic response in terms of characteristics and trends, but deviated in magnitude due the theoretical and technological limitations involved in both the physical and numerical models. | 3 | The Oral Microbiome Bank of China显示文摘The human microbiome project(HMP) promoted further understanding of human oral microbes. However, research on the human oral microbiota has not made as much progress as research on the gut microbiota. Currently, the causal relationship between the oral microbiota and oral diseases remains unclear, and little is known about the link between the oral microbiota and human systemic diseases. To further understand the contribution of the oral microbiota in oral diseases and systemic diseases, a Human Oral Microbiome Database(HOMD) was established in the US. The HOMD includes 619 taxa in 13 phyla, and most of the microorganisms are from American populations. Due to individual differences in the microbiome, the HOMD does not reflect the Chinese oral microbial status. Herein, we established a new oral microbiome database—the Oral Microbiome Bank of China(OMBC,http://www.sklod.org/ombc). Currently, the OMBC includes information on 289 bacterial strains and 720 clinical samples from the Chinese population, along with lab and clinical information. | 6 | Berberine mediates root remodeling in an immature tooth with apical periodontitis by regulating stem cells from apical papilla differentiation显示文摘Once pulp necrosis or apical periodontitis occurs on immature teeth,the weak root and open root apex are challenging to clinicians.Berberine(BBR)is a potential medicine for bone disorders,therefore,we proposed to apply BBR in root canals to enhance root repair in immature teeth.An in vivo model of immature teeth with apical periodontitis was established in rats,and root canals were filled with BBR,calcium hydroxide or sterilized saline for 3 weeks.The shape of the roots was analyzed by micro-computed tomography and histological staining.In vitro,BBR was introduced into stem cells from apical papilla(SCAPs).Osteogenic differentiation of stem cells from apical papilla was investigated by alkaline phosphatase activity,mineralization ability,and gene expression of osteogenic makers.The signaling pathway,which regulated the osteogenesis of SCAPs was evaluated by quantitative real time PCR,Western blot analysis,and immunofluorescence.In rats treated with BBR,more tissue was formed,with longer roots,thicker root walls,and smaller apex diameters.In addition,we found that BBR promoted SCAPs osteogenesis in a time-dependent and concentration-dependent manner.BBR induced the expression ofβ-catenin and enhancedβ-catenin entering into the nucleus,to up-regulate more runt-related nuclear factor 2 downstream.BBR enhanced root repair in immature teeth with apical periodontitis by activating the canonical Wnt/β-catenin pathway in SCAPs. | Yujia Cui Jing Xie Yujie Fu Chuwen Li Liwei Zheng Dingming Huang Changchun Zhou Jianxun Sun Xuedong Zhou | 2020 | International Journal of Oral Science2020,12,2: | 5 |

| 8 | Properties of Reactive Magnetron Sputtered ITO Films without in-situ Substrate Heating and Post-deposition Annealing显示文摘Indium tin oxide (ITO) films were prepared on polyester, Si and glass substrate with relatively high deposition rate of above 0.9 nm/s by DC reactive magnetron sputtering technique at the sputtering pressure of 0.06 Pa system, respectively. The dependence of resistivity on deposition parameters, such as deposition rate, target-to-substrate distance (TSD), oxygen flow rate and sputtering time (thickness), has been investigated, together with the structural and the optical properties. It was revealed that all ITO films exhibited lattice expansion. The resistivity of ITO thin films shows significant substrate effect: much lower resistivity and broader process window have been reproducibly achieved for the deposition of ITO films onto polyester rather than those prepared on both Si and glass substrates. | 9 | In-situ TEM investigation of MoS2 upon alkali metal intercalation显示文摘Phase transition in two dimensional molybdenum disulfide (MoS_2) can be induced by several methods and has been investigated for decades. Alkali metal insertion of MoS_2 had been proved an effective method to cause phase transition early in 1970s, and has been gaining renewed interest recently, due to the possible application of MoS_2 in energy storage. The alkali metal intercalation of MoS_2 has been studied by various techniques, among which in-situ transmission electron microscopy (TEM) provides unique capability of real time resolving the structural evolution of the materials at high spatial resolutions. Here by in-situ TEM technique we investigated the structural evolution of MoS_2 upon lithium and sodium intercalation, along with transformation of the nanosheet and variation of the electron diffraction patterns. The intercalation process is accompanied by emergence of superstructures, which exist in several forms. The ion intercalation results in phase transition of MoS_2 from 2H to 1T, and the driving mechanism of the phase transition are discussed. | 10 | Electrochemical solid-state amorphization in the immiscible Cu-Li system显示文摘As a typical immiscible binary system, copper (Cu) and lithium (Li) show no alloying and chemical intermixing under normal circumstances. Here we show that, when decreasing Cu nanoparticle sizes into ultrasmall range, the nanoscale size effect can play a subtle yet critical role in mediating the chemical activity of Cu and therefore its miscibility with Li, such that the electrochemical alloying and solidstate amorphization will occur in such an immiscible system. This unusual observation was accomplished by performing in-situ studies of the electrochemical lithiation processes of individual CuO nanowires inside a transmission electron microscopy (TEM). Upon lithiation, CuO nanowires are first electrochemically reduced to form discrete ultrasmall Cu nanocrystals that, unexpectedly, can in turn undergo further electrochemical lithiation to form amorphous Cu Lixnanoalloys. Real-time TEM imaging unveils that there is a critical grain size (ca. 6 nm), below which the nanocrystalline Cu particles can be continuously lithiated and amorphized. | 14 | One-step hydrothermal synthesis of S-defect-controlled ZnIn_(2)S_(4) microflowers with improved kinetics process of charge-carriers for photocatalytic H_(2) evolution显示文摘Engineering lattice defects in two-dimensional(2 D) sulfide semiconductors has been accepted as an effective strategy to enhance the efficiency of the solar-to-fuels conversion.Although many researches have proven the lattice defect-mediated photocatalytic activity of ZnIn_(2)S_(4),the artificial control of Sdefects for optimizing the charge-carrier kinetics process in ZnIn_(2)S_(4) has long been a challenging task.Herein,we report a facile one-step method to modulate the lattice S-content of ZnIn_(2)S_(4) microflowers(MFs) only through adjusting the used amount of S-precursor in the hydrothermal solution that contains the metal precursors with a fixed Zn/In stoichiometric ratio at 1:2.We also demonstrated that the Svacancies at the In facets were the main type of lattice defects in the formed ZnIn_(2)S_(4) MFs,which could enhance both the separation and migration processes of the photoinduced charge-carriers due to the existence of discrete defect energy-levels(DELs) and the reduced effective mass of electrons,as evidenced by the first-principles calculations and the electron spectra analyses.The ZnIn_(2)S_(4) MFs with the optimal content of S-vacancy obtained by a hydrothermal treatment of the precursors with the Zn/In/S stoichiometric ratio of 1:2:8 possessed the long-lived photoinduced electron(~94.64 ns) for contributing to the photo-physical and-chemical processes.Thus,upon visible light irradiation,the H_(2)-evolution rate of this sample reached ~2.40 mmol h^(-1) g^(-1) with an apparent quantum efficiency of ~0.16% at 420 nm even though only using 5 mg of photocatalysts without any cocatalysts. | Xuedong Jing Na Lu Jindou Huang Peng Zhang Zhenyi Zhang | 2021 | Journal of Energy Chemistry2021,30,7: | 1 |

| 17 | Genome Mining and Biosynthesis Study of a Type B Linaridin Reveals a Highly Versatileα-N-Methyltransferase显示文摘Linaridins are a small but growing family of natural products belonging to the ribosomally synthesized and post-translationally modified peptide(RiPP)superfamily.In this study,a genome mining approach led to the identification of a novel linaridin,mononaridin(MON),from Streptomyces monomycini.In-frame deletion genetic knockout studies showed that,in addition to many genes essential for MON biosynthesis,monM encodes an S-adenosyl methionine(SAM)-dependentα-N-methyltransferase that is responsible for installing two methyl groups in the MON N-terminus.Besides SAM,MonM also accepts ethyl-SAM and allyl-SAM,in which the methyl of SAM is replaced by an ethyl and an allyl,respectively.We showed that ethyl-SAM and allyl-SAM have distinct reactivities in MonM catalysis,and this observation was further investigated in detail by density functional theory(DFT)calculations.Remarkably,MonM acts efficiently on nisin,a prototypic lantibiotic that is structurally very different from the native substrate,and the ability of MonM to transfer an allyl group to the nisin N-terminus allowed production of a fluorescently labeled nisin,which can be further used in microscopic cell analysis.Our studies provide new insights into linaridin biosynthesis and demonstrate the potential of linaridin methyltransferases in bioengineering applications. | Fangting Wang Wanqing Wei Junfeng Zhao Tianlu Mo Xin Wang Xuedong Huang Suze Ma Shu Wang Zixin Deng Wei Ding Yong Liang Qi Zhang | 2021 | CCS Chemistry2021,3,3: | 1 |

| 20 | LncRNA HIF1A-AS2 accelerates malignant phenotypes of renal carcinoma by modulating miR-30a-5p/SOX4 axis as a ceRNA显示文摘Objective:Several reports have proposed that lnc RNAs,as potential biomarkers,participate in the progression and growth of malignant tumors.HIF1 A-AS2 is a novel lnc RNA and potential biomarker,involved in the genesis and development of carcinomas.However,the molecular mechanism of HIF1 A-AS2 in renal carcinoma is unclear.Methods:The relative expression levels of HIF1 A-AS2 and miR-30 a-5 p were detected using RT-qPCR in renal carcinoma tissues and cell lines.Using loss-of-function and overexpression,the biological effects of HIF1 A-AS2 and miR-30 a-5 p in kidney carcinoma progression were characterized.Dual luciferase reporter gene analysis and Western blot were used to detect the potential mechanism of HIF1 A-AS2 in renal carcinomas.Results:HIF1 A-AS2 was upregulated in kidney carcinoma tissues when compared with para-carcinoma tissues(P<0.05).In addition,tumor size,tumor node mestastasis stage and differentiation were identified as being closely associated with HIF1 A-AS2 expression(P<0.05).Knockdown or overexpression of HIF1 A-AS2 either restrained or promoted the malignant phenotype and WNT/β-catenin signaling in renal carcinoma cells(P<0.05).Mi R-30 a-5 p was downregulated in renal cancers and partially reversed HIF1 A-AS2 functions in malignant renal tumor cells.HIF1 A-AS2 acted as a micro RNA sponge that actively regulated the relative expression of SOX4 in sponging miR-30 a-5 p and subsequently increased the malignant phenotypes of renal carcinomas.HIF1 A-AS2 showed a carcinogenic effect and miR-30 a-5 p acted as an antagonist of the anti-oncogene effects in the pathogenesis of renal carcinomas.Conclusions:The HIF1 A-AS2-miR-30 a-5 p-SOX4 axis was associated with the malignant progression and development of renal carcinoma.The relative expression of HIF1 A-AS2 was negatively correlated with the expression of miR-30 a-5 p,and was closely correlated with SOX4 mRNA levels in renal cancers. | Mingwei Chen Xuedong Wei Xiu Shi Le Lu Guangbo Zhang Yuhua Huang Jianquan Hou | 2021 | Cancer Biology & Medicine2021,18,2: | 1 |
