1Prognostic values of the integrated model incorporating the volume of metastatic regional cervical lymph node and pretreatment serum Epstein-Barr virus DNA copy number in predicting distant metastasis in patients with N1 nasopharyngeal carcinoma显示文摘Background: According to the 7 th edition of the American Joint Committee on Cancer(AJCC) staging system, over50% of patients with nasopharyngeal carcinoma(NPC) have N1 disease at initial diagnosis. However, patients with N1 NPC are relatively under-researched, and the metastasis risk of this group is not well-stratified. This study aimed to evaluate the prognostic values of gross tumor volume of metastatic regional lymph node(GTVnd) and pretreatment serum copy number of Epstein-Barr virus(EBV) DNA in predicting distant metastasis of patients with N1 NPC, and to develop an integrated prognostic model that incorporates GTVnd and EBV DNA copy number for this group of patients.Methods: The medical records of 787 newly diagnosed patients with nonmetastatic, histologically proven N1 NPC who were treated at Sun Yat-sen University Cancer Center between November 2009 and February 2012 were analyzed. Computed tomography-derived GTVnd was measured using the summation-of-area technique. Blood samples were collected before treatment to quantify plasma EBV DNA. The receiver operating characteristic(ROC) curve analysis was used to evaluate the cut-off point for GTVnd, and the area under the ROC curve was used to assess the predicted validity of GTVnd. The survival rates were assessed by Kaplan-Meier analysis, and the survival curves were compared using a log-rank test. Multivariate analysis was conducted using the Cox proportional hazard regression model.Results: The 5-year distant metastasis-free survival(DMFS) rates for patients with GTVnd > 18.9 vs.≤ 18.9 mL were82.2% vs. 93.2%(P < 0.001), and for patients with EBV DNA copy number > 4000 vs. < 4000 copies/mL were 83.5% vs.93.9%(P < 0.001). After adjusting for GTVnd, EBV DNA copy number, and T category in the Cox regression model, both GTVnd > 18.9 mL and EBV DNA copy number > 4000 copies/mL were significantly associated with poor prognosis(both P < 0.05). According to combination of GTVnd and EBV DNA copy number, all patients were divided into low-,moderate-, and high-risk groups, with the 5-year DMFS rates of 96.1,87.4, and 73.8%, respectively(P < 0.001). Multivariate analysis confirmed the prognostic value of this model for distant metastatic risk stratification(hazard ratio [HR],4.17; 95% confidence interval [CI] 2.34-7.59; P < 0.001).Conclusions: GTVnd and serum EBV DNA copy number are independent prognostic factors for predicting distant metastasis in NPC patients with N1 disease. 2UAV navigation in high dynamic environments:A deep reinforcement learning approach显示文摘Unmanned Aerial Vehicle(UAV) navigation is aimed at guiding a UAV to the desired destinations along a collision-free and efficient path without human interventions, and it plays a crucial role in autonomous missions in harsh environments. The recently emerging Deep Reinforcement Learning(DRL) methods have shown promise for addressing the UAV navigation problem,but most of these methods cannot converge due to the massive amounts of interactive data when a UAV is navigating in high dynamic environments, where there are numerous obstacles moving fast.In this work, we propose an improved DRL-based method to tackle these fundamental limitations.To be specific, we develop a distributed DRL framework to decompose the UAV navigation task into two simpler sub-tasks, each of which is solved through the designed Long Short-Term Memory(LSTM) based DRL network by using only part of the interactive data. Furthermore, a clipped DRL loss function is proposed to closely stack the two sub-solutions into one integral for the UAV navigation problem. 3Association of Overlapped and Un-overlapped Comorbidities with COVID-19 Severity and Treatment Outcomes: A Retrospective Cohort Study from Nine Provinces in China显示文摘Objective Several COVID-19 patients have overlapping comorbidities. The independent role of each component contributing to the risk of COVID-19 is unknown, and how some non-cardiometabolic comorbidities affect the risk of COVID-19 remains unclear.Methods A retrospective follow-up design was adopted. A total of 1,160 laboratory-confirmed patients were enrolled from nine provinces in China. Data on comorbidities were obtained from the patients’ medical records. Multivariable logistic regression models were used to estimate the odds ratio(OR) and 95% confidence interval(95% CI) of the associations between comorbidities(cardiometabolic or non-cardiometabolic diseases), clinical severity, and treatment outcomes of COVID-19.Results Overall, 158(13.6%) patients were diagnosed with severe illness and 32(2.7%) had unfavorable outcomes. Hypertension(2.87, 1.30–6.32), type 2 diabetes(T2 DM)(3.57, 2.32–5.49),cardiovascular disease(CVD)(3.78, 1.81–7.89), fatty liver disease(7.53, 1.96–28.96), hyperlipidemia(2.15, 1.26–3.67), other lung diseases(6.00, 3.01–11.96), and electrolyte imbalance(10.40, 3.00–26.10)were independently linked to increased odds of being severely ill. T2 DM(6.07, 2.89–12.75), CVD(8.47,6.03–11.89), and electrolyte imbalance(19.44, 11.47–32.96) were also strong predictors of unfavorable outcomes. Women with comorbidities were more likely to have severe disease on admission(5.46,3.25–9.19), while men with comorbidities were more likely to have unfavorable treatment outcomes(6.58, 1.46–29.64) within two weeks.Conclusion Besides hypertension, diabetes, and CVD, fatty liver disease, hyperlipidemia, other lung diseases, and electrolyte imbalance were independent risk factors for COVID-19 severity and poor treatment outcome. 6β-hydroxy-β-methylbutyrate Improves Chronic Renal Failure-Induced Sarcopenia in SD Rats by Regulating Autophagy显示文摘Background Chronic renal failure-induced sarcopenia(CRF-S)seriously affects public health by increasing morbidity and mortality.This study evaluated the therapeutic effects ofβ-hydroxy-β-methylbutyrate acid(HMB)and the underlying mechanism in an adenine-induced CRF-S rat model.Methods Sprague Dawley rats were divided into control,model(CRF-S),and HMB treatment(CRF-S+HMB)groups.The rats in the CRF-S group received 250 mg/kg/d adenine by oral gavage every day for 14 days,followed by administration every other day until the 28th day.In the CRF-S+HMB group,each rat was given 320 mg/kg/d HMB by oral gavage starting from 7 days prior to adenine treatment,and this treatment was maintained for a total of 35 days.The body weight,right anterior tibial muscle weight,blood serum parameters,gastrocnemius muscle and renal tissue histology,and expression of autophagy-related transcripts and proteins were evaluated.Results A rat model of CRF-S was successfully established via adenine gavage,as indicated by reduced body weight and muscle weights,and markers of sarcopenia.There were increased mRNA levels of LC3B,Beclin1,Ulk1,Atg5,Atg7,Atg13 and vps-34,and increased protein expression levels of LC3B and P62.In the rats with HMB intervention,the body weight,right anterior tibial weight and gastrocnemius muscle histology were not significantly different from those in the control group,and the expression levels of autophagy-related proteins(LC3B and P62)were consistent with those in the control group.Conclusion We successfully established a rat model of CRF-S using adenine administered via oral gavage,and observed that HMB treatment prevented CRF-S.Zun Yue Zhang Liu Qing Yang Ya Nan Yang Yun Lin Wang Long Mei Zhang Kun Hua Wang Min Weng 2021Journal of Nutritional Oncology2021,6,4:4
7Bone Loss Induced by Simulated Microgravity,Ionizing Radiation and/or Ultradian Rhythms in the Hindlimbs of Rats显示文摘Objective To better understand the pathological causes of bone loss in a space environment, including microgravity, ionizing radiation, and ultradian rhythms. Methods Sprague Dawley(SD) rats were randomly divided into a baseline group, a control group, a hindlimb suspension group, a radiation group, a ultradian rhythms group and a combined-three-factor group. After four weeks of hindlimb suspension followed by X-ray exposure and/or ultradian rhythms, biomechanical properties, bone mineral density, histological analysis, microstructure parameters, and bone turnover markers were detected to evaluate bone loss in hindlimbs of rats. Results Simulated microgravity or combined-three factors treatment led to a significant decrease in the biomechanical properties of bones, reduction in bone mineral density, and deterioration of trabecular parameters. Ionizing radiation exposure also showed adverse impact while ultradian rhythms had no significant effect on these outcomes. Decrease in the concentration of the turnover markers bone alkaline phosphatase(b ALP), osteocalcin(OCN), and tartrate-resistant acid phosphatase-5 b(TRAP-5 b) in serum was in line with the changes in trabecular parameters. Conclusion Simulated microgravity is the main contributor of bone loss. Radiation also results in deleterious effects but ultradian rhythms has no significant effect. 10Association between Coronary Atherosclerotic Plaque Composition and Cardiovascular Disease Risk显示文摘Objective The objective of this study is to determine whether coronary atherosclerotic plaque composition is associated with cardiovascular disease(CVD) risk in Chinese adults. Methods We performed a cross-sectional analysis in 549 subjects without previous diagnosis or clinical symptoms of CVD in a community cohort of middle-aged Chinese adults. The participants underwent coronary computed tomography(CT) angiography for the evaluation of the presence and composition of coronary plaques. CVD risk was evaluated by the Framingham risk score(FRS) and the 10-year atherosclerotic cardiovascular disease(ASCVD) risk score. Results Among the 549 participants, 267(48.6%) had no coronary plaques, 201(36.6%) had noncalcified coronary plaques, and 81(14.8%) had calcified or mixed coronary plaques. The measures of CVD risk including FRS and ASCVD risk score and the likelihood of having elevated FRS significantly increased across the groups of participants without coronary plaques, with noncalcified coronary plaques, and with calcified or mixed coronary plaques. However, only calcified or mixed coronary plaques were significantly associated with an elevated ASCVD risk score [odds ratio(OR) 2.41; 95% confidence interval(CI) 1.09-5.32] compared with no coronary plaques, whereas no significant association was found for noncalcified coronary plaques and elevated ASCVD risk score(OR 1.25; 95% CI 0.71-2.21) after multivariable adjustment. 11Low Phase Angle Predicts Poor Survival in Patients with Hepatocellular Carcinoma:A Retrospective Study显示文摘Background The phase angle(PA)is an indicator of nutritional status derived from bioelectrical impedance analysis(BIA).Studies have shown that the PA is one of several factors related to the survival of cancer patients.However,the correlation between the PA and prognosis in patients with hepatocellular carcinoma(HCC)has not been reported.This study aimed to explore the relationship between the PA,nutrition-related indicators,and patient prognosis.Methods The data from 248 HCC patients who underwent BIA between January 2015 and December 2019 at the First Hospital of Jilin University were retrospectively analyzed.Patients were divided into two groups according to the median PA value,then the nutrition-related indicators and overall survival(OS)were compared between the two groups.The correlations between the PA and laboratory test nutrition-related indicators and the results of a body composition analysis were determined using Pearson correlation analysis.Independent predictors of the patient prognosis were identified using univariate and multivariate analyses.Results The median OS in the high and low PA groups were 70 and 54.6 months,respectively(P=0.004).The Pearson correlation analysis revealed that the PA was associated with both laboratory test nutrition-related indicators and the results of a body composition analysis.The multivariable Cox regression analysis identified the PA as an independent risk factor affecting the prognosis of patients with Barcelona Clinic Liver Cancer stage A or B.Conclusion The PA is a good biomarker for the nutritional status.We found that the PA was associated with the prognosis in patients with HCC,with a low PA indicating a poor nutritional status and worse prognosis.Jin Yu Shi Guang Yang Bo Liu Xiao Shang Guo Zhen Cui Jia Xin Huang Wan Ting Wang Ke Yu Chen Nan Ya Wang 2022Journal of Nutritional Oncology2022,7,2:2

13Amplification and expression of c-MET correlate with poor prognosis of patients with gastric cancer and upregulate the expression of PDL1显示文摘The prognostic significance of c-MET in gastric cancer(GC)remains uncertain.In the present study,we examined the amplification,expression,and the prognostic value of c-MET,human epidermal growth factor receptor 2(HER2),and programmed cell death 1 ligand 1(PDL1),together with the correlations among them in a large cohort of Chinese samples.A total of 444 patients were included.The immunohistochemistry(IHC)and the dual-color silver in situ hybridization(SISH)were performed to examine their expression and amplification.Univariate and multivariate analyses were performed by the Cox proportional hazard regression model,and survival curves were estimated by the Kaplan-Meier method.The positivity determined by IHC of c-MET was 24.8%,and the MET amplification rate was 2.3%.The positivity rates of HER2 and PDL1 were 8%and 34.7%,respectively.PDL1 expression had a significantly positive association with c-MET expression.c-MET positivity played a significant prognostic role in disease-free survival(DFS)(P=0.032).Patients with mesenchymal-epithelial transition(MET)amplification had significantly poorer prognosis on both DFS and overall survival(OS).Subgroup analysis showed that in HER2-negative patients,but not in HER2-positive patients,MET-positive patients had significantly worse DFS(P=0.000)and OS(P=0.006).c-MET regulated the expression of PDL1 through an AKT-dependent pathway.c-MET inhibitor enhanced the T-cell killing ability and increased the efficacy of PD1 antibody.c-MET was found to be an independent prognostic factor for DFS of GC patients.A combination of c-MET inhibitors and PD1 antibodies could enhance the killing capacity of T cells,providing a preliminary basis for the clinical research on the same combination in GC treatment.Ya’nan Yang Chenchen Wang Congqi Dai Xinyang Liu Wenhua Li Mingzhu Huang Xiaoying Zhao Dongmei Ji Jin Li Weijian Guo 2021Acta Biochimica et Biophysica Sinica2021,53,5:2
14The Influence of Diabetes, Hypertension, and Hyperlipidemia on the Onset of Age-Related Macular Degeneration in North China: The Kailuan Eye Study显示文摘Objective To analyze the prevalence of dry and wet age-related macular degeneration(AMD)in patients with diabetes,hypertension and hyperlipidemia,and to analyze the risk factors for AMD.Methods A population-based cross-sectional epidemiologic study was conducted involving 14,440 individuals.We assessed the prevalence of dry and wet AMD in diabetic and non-diabetic subjects and analyzed the risk factors for AMD.Results The prevalence of wet AMD in diabetic and non-diabetic patients was 0.3%and 0.5%,respectively,and the prevalence of dry AMD was 17%and 16.4%,respectively.The prevalence of wet AMD in healthy,hypertensive,hyperlipidemic,and hypertensive/hyperlipidemic populations was 0.5%,0.3%,0.2%,and 0.7%,respectively.The prevalence of dry AMD in healthy,hypertensive,hyperlipidemic,and hypertensive/hyperlipidemic populations was 16.6%,16.2%,15.2%,and 17.2%,respectively.Age,sex,body mass index,and use of hypoglycemic drugs or lowering blood pressure drugs were corrected in the risk factor analysis of AMD.Diabetes,diabetes/hypertension,diabetes/hyperlipidemia,and diabetes/hypertension/hyperlipidemia were analyzed.None of the factors analyzed in the current study increased the risk for the onset of AMD.Conclusion There was no significant difference in the prevalence of wet and dry AMD among diabetic and non-diabetic subjects.Similarly,there was no significant difference in the prevalence of wet and dry AMD among subjects with hypertension and hyperlipidemia.Diabetes co-existing with hypertension and hyperlipidemia were not shown to be risk factors for the onset of dry AMD.ZHANG Yong Peng WANG Ya Xing ZHOU Jin Qiong WANG Qian YAN Yan Ni YANG Xuan YANG Jing Yan ZHOU Wen Jia WANG Ping SHEN Chang YANG Ming LUAN Ya Nan WANG Jin Yuan WU Shou Ling CHEN Shuo Hua WANG Hai Wei FANG Li Jian WAN Qian Qian ZHU Jing Yuan NIE Zi Han CHEN Yu Ning XIE Ying JONAS JB WEI Wen Bin 2022Biomedical and Environmental Sciences2022,35,7:2

17Rapid and High-throughput Identification of Recombinant Bacteria with Mass Spectrometry Assay显示文摘Objective To construct a rapid and high-throughput assay for identifying recombinant bacteria based on mass spectrometry.Methods Matrix assisted laser desorption ionization time-of-flight mass spectrometry(MALDI-TOF MS) techniques were used to identify 12 recombinant proteins(10 of Yersinia pestis,1 of Campylobacter jejuni and 1 of Helicobacter pylori).A classification model for the various phase of recombinant bacteria was established,optimized and validated,using MALDI-TOF MS-ClinProTools system.The differences in the peptide mass spectra were analyzed by using Biotyper and FlexAnalysis softwares.Results Models of GA,SNN,and QC were established.After optimizing the parameters,the GA recognition model showed good classification capabilities:RC=100%,mean CVA=98.7%(the CVA was96.4%in phase 1,100%in phase 2,98.4%in phase 3,and 100%in phase 4,respectively) and PPV=95%.This model can be used to classify the bacteria and their recombinant,which only requires 3.7xl03 cells for analysis.The total time needed is only 10 min from protein extraction to reporting the result for one sample.Furthermore,this assay can automatically detect and test 96 samples concurrently.A total of 48specific peaks(9,16,9,and 14 for the four stages,respectively) was found in the various phase of recombinant bacteria.Conclusion MALDI-TOF MS can be used as a fast,accurate,and high-throughput method to identify recombinant bacteria,which provide a new ideas not only for recombinant bacteria but also for the identification of mutant strains and bioterrorism pathogens.XIAO Di TAO Xiao Xia WANG Peng LIU Guo Dong GONG Ya Nan ZHANG Hui Fang WANG Hai Bin ZHANG Jian Zhong 2014Biomedical and Environmental Sciences2014,27,4:2
18Performance analysis and optimization model based on dynamics equation for heterogeneous wireless networks显示文摘In this paper, we propose a model based on dynamics equation for performance analysis and optimization for heterogeneous wireless networks(HWNs). First, the channel occupation state with time of HWNs is modelled by dynamics equation, in which users' mobility as an important factor affecting system performance is considered. Then the steady state probability distribution of channel occupation is derived. Based on the results, the expression of the throughput of HWNs is deduced, which includes a factor ρ which is the ratio of service arrival rate accessing one of the networks to the total service arrival rate in the overlapping area. And this paper proposes to maximize the throughput of the HWNs by optimizing the factor ρ to efficiently utilize the resources. 19Flexible double narrowband near-infrared photodetector based on PMMA/core-shell upconversion nanoparticle composites显示文摘Flexible narrowband near infrared(NIR)photodetectors(PDs)are urgently in demand in the fastdeveloping era of flexible electronics,due to their crucial roles in various innovative applications.Hence,we designed and synthesized the core-shell structured NaYF4:Yb^(3+),Er^(3+)@NaYF4:Nd^(3+)upconversion nanoparticles(UCNPs),which can be pumped by the 808 and 980 nm lights.The upconversion luminescence(UCL)are significantly enhanced after being assembled with the opal photonic crystals(OPCs)due to the photonic crystal effect,with 55 and 48 folds of enhancement factors under illuminations of 808 and 980 nm lights,respectively.Based on this hybrid,the flexible narrowband PDs were successfully fabricated on the PET substrate with the structure of OPCs/NaYF4:Yb^(3+),Er^(3+)@Nd^(3+)/MAPbl3,which displays excellent detection performance to double narrowband NIR light(808 and 980 nm)benefiting from the amplified UCL,with responsivity of 8.79 and 7.39 A/W,detectivity of 3.01×10^(11)and2.68×10^(11)cm·Hz^(1/2)/W for 808 and 980 nm lights detection respectively,along with short response time in the range of 120-160 ms.Furthermore,the OPCs/NaYF4:Yb^(3+),Er^(3+)@NaYF4:Nd^(3+)/MAPbI3 double narrowband PDs display low photodetection power threshold(0.05 W/cm2),outstanding flexibility,prominent moisture resistance,and good long-time stability.This work displays a new concept of narrowband NIR PDs,which open a new field for specific NIR light detections.Ya’nan Ji Donglei Zhou Nan Wang Nan Ding Wen Xu Hongwei Song 2022Journal of Rare Earths2022,40,2:2
