3Genome-Wide Prediction of Highly Specific Guide RNA Spacers for CRISPR-Cas9-Mediated Genome Editing in Model Plants and Major Crops显示文摘Dear Editor, RNA-guided genome editing (RGE) using the Streptococcus pyogenes CRISPR-Cas9 system (Jinek et al.,2012;Cong et al.,2013;Mali et al.,2013b) is emerging as a simple and highly efficient tool for genome editing in many organisms.The Cas9 nuclease can be programmed by dual or single guide RNA (gRNA) to cut target DNA at specific sites,thereby introducing precise mutations by error-prone non-homologousend-joining repairing or by incorporating foreign DNAs via homologous recombination between target site and donor DNA.The gRNA-Cas9 complex recognizes targets based on the complementarity between one strand of targeted DNA (referred as protospacer) and the 5'-end leading sequence of gRNA (referred to as gRNA spacer) that is approximately 20 base pairs (bp) long (Figure 1A).Besides gRNA-DNA pairing,a protospacer-adjacent motif (PAM) following the paired region in the DNA is also required for Cas9 cleavage.Recent studies reveal that Cas9 could cut the PAM-containing DNA sites that imperfectly match gRNA spacer sequences,resulting in genome editing at undesired positions.Kabin Xie Jianwei Zhang Yinong Yang 2014Molecular Plant2014,7,5:35

7Insight-HXMT observations of the first binary neutron star merger GW170817显示文摘Finding the electromagnetic(EM) counterpart of binary compact star merger, especially the binary neutron star(BNS) merger,is critically important for gravitational wave(GW) astronomy, cosmology and fundamental physics. On Aug. 17, 2017,Advanced LIGO and Fermi/GBM independently triggered the first BNS merger, GW170817, and its high energy EM counterpart,GRB 170817 A, respectively, resulting in a global observation campaign covering gamma-ray, X-ray, UV, optical, IR, radio as well as neutrinos. The High Energy X-ray telescope(HE) onboard Insight-HXMT(Hard X-ray Modulation Telescope) is the unique high-energy gamma-ray telescope that monitored the entire GW localization area and especially the optical counterpart(SSS17 a/AT2017 gfo) with very large collection area(~1000 cm^2) and microsecond time resolution in 0.2-5 MeV. In addition,Insight-HXMT quickly implemented a Target of Opportunity(ToO) observation to scan the GW localization area for potential X-ray emission from the GW source. Although Insight-HXMT did not detect any significant high energy(0.2-5 MeV) radiation from GW170817, its observation helped to confirm the unexpected weak and soft nature of GRB 170817 A. Meanwhile,Insight-HXMT/HE provides one of the most stringent constraints(~10^(-7) to 10^(-6) erg/cm^2/s) for both GRB170817 A and any other possible precursor or extended emissions in 0.2-5 MeV, which help us to better understand the properties of EM radiation from this BNS merger. 8A novel HfNbTaTiV high-entropy alloy of superior mechanical properties designed on the principle of maximum lattice distortion显示文摘This paper reports a synergistic design of high-performance BCC high-entropy alloy based on the combined consideration of the principles of intrinsic ductility of elements,maximum atomic size difference for solid solution strengthening and the valence electron concentration criterion for ductility.The single-phase BCC HfNbTaTiV alloy thus designed exhibited a high compressive yield strength of 1350 MPa and a high compressive ductility of>45%at the room temperature.This represents a 50%increase in yield strength relative to a HfNbTaTiZr alloy.This is attributed to the maximized solid solution strengthening effect caused by lattice distortion,which is estimated to be 1094 MPa.The alloy was also able to retain 53%of its yield strength and 77%of its ductility at 700℃.These properties are superior to those of most refractory BCC high-entropy alloys reported in the literature.Zibing An Shengcheng Mao Yinong Liu Li Wang Hao Zhou Bin Gan Ze Zhang Xiaodong Han 2021Journal of Materials Science & Technology2021,,20:9

11RNA Interference-based Suppression of Phosphoenolpyruvate Carboxylase Results in Susceptibility of Rapeseed to Osmotic Stress显示文摘The diverse functions of phosphoenolpyruvate carboxylase(PEPCase;EC 4.1.1.31) in C3 plants are not as well understood as in C4 plants.To investigate the functions of PEPCase in C3 plants,rapeseed(Brassica napus L.) PEPCase gene(referred to as BNPE15) was silenced by the RNA interference(RNAi) technique.Under normal growth conditions,no significant difference in lipid content and fatty acid composition were found between wild-type(WT) and transgenic rapeseed plants.However,when these plants were subjected to osmotic stress induced by osmoticum polyethylene glycol(PEG-6000),membrane permeability and membrane lipid peroxidization in roots and leaves of transgenic plants were higher than those of WT plants.It suggested that transgenic plants are more susceptible to osmotic stress than WT plants.Taken together,the results showed that the suppression of PEPCase by RNAi leads to susceptibility to osmotic stress in rapeseed,and PEPCase is involved in the response of C3 plants to environmental stress.Mei Chen Yunlai Tang Jingmei Zhang Mingfeng Yang Yinong Xu 2010Journal of Integrative Plant Biology2010,52,6:4

13Simulation and Analysis on Overtaking Safety Assistance System Based on Vehicle-to-Vehicle Communication显示文摘Because of the complexity and variability of an intelligent vehicle’s driving environment,it is difficult for the application of the vehicular sensors to meet the needs of the surrounding environment information entirely.Vehicle-to-vehicle(V2V)communication technology is used by target vehicles to exchange information,and obtain the driving condition and driving intention of the front driver.To obtain environmental information outside the range of vehicular sensors in advance,in this paper,a vehicle overtaking assistance system is proposed based on V2V communication.The data,including the speed,position,direction angle and steering angle obtained using V2V communication,were preliminarily processed.Then,combined with an overtaking safety distance model,the vehicle parameters,driver’s driving intention and vehicle status information were entered into an overtaking security assistance system to determine the overtaking conditions.Fuzzy theory was used to control the parameters of the overtaking safety distance model.Finally,the overtaking safety assistance system was established and the proposed algorithm was tested using PreScan/MATLAB cooperative simulation software.The results showed that the proposed overtaking safety algorithm effectively provided a warning according to environmental change and the driver’s intention,which assisted the driver to overtake and avoid the occurrence of accidents,which improved the safety performance of the vehicle.Chunmei Mo Yinong Li Ling Zheng 2018Automotive Innovation2018,1,2:4
14Significance of neuroglobin in serum of acute atherosclerotic cerebral infarction patients显示文摘This study sought to examine neuroglobin (NGB) in the serum of acute cerebral infarction patients with double-antibody sandwich enzyme-linked immunosorbent assay to identify all risk factors, calculate infarct size, assess neurological impairment, and analyze the relation between NGB and each of these factors. The double-antibody sandwich assay indicated that levels of NGB in serum were unaltered within 6 hours following acute cerebral infarction compared with normal levels. NGB levels then underwent a distinct change, peaking at 24 hours then returning to normal levels in 72 hours. The results suggest that the level of NGB might be related to infarct size and low-density lipoprotein at 24 hours after acute cerebral infarction. There were no significant differences in neurological impairment scores and infarct size at different periods following infarction. The convergence (divergence) induced by the secondary circulation is the most important factor for frontogenesis (frontolysis) at the top and bottom boundaries. 19Positional distribution of fatty acids on the glycerol backbone during the biosyn-thesis of glycerolipids in Ectocarpus fasciculatus显示文摘The biosynthesis of glycolipids in E.fascicula-tus was studied by 14C label and chase. The fatty acids in sulphoquinovosyl diacylglycerol (SQDG) were almost 16-car-bon and 18-carbon ones. In addition to the two fatty acids, monogalactosyl diacylglycerol (MGDG) and digalactosyl diacylglycerol (DGDG) contained 8.5 mol% and 31.0 mol% of eicosapentaenoic acid (20 : 5), respectively, and this fatty acid was usually distributed in the sn-1 position of the glyc-erol backbone. When plants were incubated with [2-14C] acetate, differences existed in the positional distribution of the labeled fatty acids in sn-1 and sn-2 among the three glycerolipids. In SQDG, 14C-labeled fatty acids were distributed uniformly in the sn-1 and sn-2 positions. In DGDG, 14C-labeled fatty acids were mainly distributed in the sn-2 position.
