1Efficacy of gefitinib as a first-line single agent treatment in patients with advanced non-small cell lung cancer显示文摘Objective: To assess the efficacy and toxicity of gefitinib as a single agent treatment in Chinese patients with advanced non-small cell lung cancer (NSCLC). Methods: Forty-five patients with advanced NSCLC were treated with gefitinib at 250 mg daily until the disease progressed or the patient could not tolerate the toxicity. Results: None of the patients achieved a complete response (CR), while 15 patients achieved a partial remission (PR) and 17 experienced a stable disease (SD). Thirteen patients continued to have a progressive disease (PD). The response rate and the disease control rate were 33.3% and 71.1%, respectively. The symptom remission rate was 72.5%, and the median remission time was 8 days. The median survival time was 15.3 months. The median progression-free survival time was 6.0 months. The most common toxicities included rash (53.3%) and diarrhea (33.3%). Dehydration and pruritus of the skin developed in 26.7% and 22.2% of the patients, respectively. Hepatic toxicity occurred in 6.7% of patients and oral ulceration occurred in 4.4% of patients. 7Baseline radiologic features as predictors of efficacy in patients with pancreatic neuroendocrine tumors with liver metastases receiving surufatinib显示文摘Objective:Currently,pre-treatment prediction of patients with pancreatic neuroendocrine tumors with liver metastases(PNELM)receiving surufatinib treatment was unsatisfying.Our objective was to examine the association between radiological characteristics and efficacy/prognosis.Methods:We enrolled patients with liver metastases in the phase III,SANET-p trial(NCT02589821)and obtained contrast-enhanced computed tomography(CECT)images.Qualitative and quantitative parameters including hepatic tumor margins,lesion volumes,enhancement pattern,localization types,and enhancement ratios were evaluated.The progression-free survival(PFS)and hazard ratio(HR)were calculated using Cox’s proportional hazard model.Efficacy was analyzed by logistic-regression models.Results:Among 152 patients who had baseline CECT assessments and were included in this analysis,the surufatinib group showed statistically superior efficacy in terms of median PFS compared to placebo across various qualitative and quantitative parameters.In the multivariable analysis of patients receiving surufatinib(N=100),those with higher arterial phase standardized enhancement ratio-peri-lesion(ASER-peri)exhibited longer PFS[HR=0.039;95%confidence interval(95%CI):0.003−0.483;P=0.012].Furthermore,patients with a high enhancement pattern experienced an improvement in the objective response ratio[31.3%vs.14.7%,odds ratio(OR)=3.488;95%CI:1.024−11.875;P=0.046],and well-defined tumor margins were associated with a higher disease control rate(DCR)(89.3%vs.68.2%,OR=4.535;95%CI:1.285−16.011;P=0.019)compared to poorlydefined margins.Conclusions:These pre-treatment radiological features,namely high ASER-peri,high enhancement pattern,and well-defined tumor margins,have the potential to serve as predictive markers of efficacy in patients with PNELM receiving surufatinib.Jianwei Zhang Haibin Zhu Lin Shen Jie Li Xiaoyan Zhang Chunmei Bai Zhiwei Zhou Xianrui Yu Zhiping Li Enxiao Li Xianglin Yuan Wenhui Lou Yihebali Chi Nong Xu Yongmei Yin Yuxian Bai Tao Zhang Dianrong Xiu Jia Chen Shukui Qin Xiuwen Wang Yujie Yang Haoyun Shi Xian Luo Songhua Fan Weiguo Su Ming Lu Jianming Xu 2023Chinese Journal of Cancer Research2023,35,5:0
8TNF-alpha-induced metastasis gene changes in MCF-7 cells显示文摘Objective: Studies have shown that TNF-αsecreted by tumor cells and macrophages infiltrated into the tumor microenvironment might promote the metastasis of a variety of malignant cancers, including breast cancer. The present study was designed to detect global metastasis-related gene expression changes of MCF-7 cells treated by low dose TNF-α and to further explore the mechanisms by which TNF-αcontributes to metastasis. Methods: MCF-7 cells were cultured and treated with low dose TNF-α(20 ng/ml). cDNA array analysis was applied to detect the metastasis related gene expressions. Results: A total of 36 gene expressions were significantly regulated by TNF-α. Functional analysis indicates that the altered genes belong to different functional group. Most of the genes changed may promote the metastasis of MCF-7 cells while the others may inhibit metastasis. The changes observed in gene expression following TNF-αwere somewhat time dependent. Conclusion: TNF-αcan enhance the invasive ability of MCF-7 cells, partly by regulating a series of metastasis related genes, and these genes may take part in every step of metastasis. 10Mapping of genetic modifiers of Plcd1 in scant hair mice(snthr^(1Bao))显示文摘The scant hair mutant mouse(locus symbol:snthr1Bao) is a recessive mutation that originated in an ethylnitrosourea chemical carcinogenesis study using the DBA/2J inbred strain.The gene responsible for the mutation was previously determined to be phospholipase C,delta 1(Plcd1;mutant allele symbol Plcd1snthr1Bao).To map the modifiers of Plcd1,an intercross(DBA/2J-snthr1Bao/snthr1Bao × C57BL/6J+/+) was conducted.The F2 mutant progeny exhibited a variety of alopecia phenotypes;all F2 mutants(n=507) were classified into 3 groups(mild,moderate,and severe alopecia) and genotyped based on 96 microsatellites.A major QTL was identified on mouse chromosome(mChr) 15 at 12 cM with an LOD score greater than 7(P < 0.0001).Three minor QTLs were detected on mChr 2,5,and 7 at 40,84 and 48 cM,respectively.The QTLs on mChr 7 and 15 were associated with minor alopecia while the QTLs on mChr 2 and 5 were associated with moderate to severe alopecia.No antagonistic or synergistic effects among or between the 4 QTLs were found.Integrating the functions of the 4 potential regulatory QTLs and mutant Plcd1snthr1Bao,we found that these QTLs might contribute to variations of scant hair severity by altering the Ca2+ signal pathways in mouse skin.WU BaoJin ZENG YongMei MAO HuiHua YIN LiJing ZHU Jie YANG WeiWei YIN XiaoShu WU PeiLin ZHANG WeiDong 2010Chinese Science Bulletin2010,55,35:0
