1ELASTIC MODULUS REDUCTION METHOD FOR LIMIT LOAD EVALUATION OF FRAME STRUCTURES显示文摘A new strategy for elastic modulus adjustment is proposed based on the element bearing ratio (EBR),and the elastic modulus reduction method (EMRM) is proposed for limit load evaluation of frame structures. The EBR is defined employing the generalized yield criterion,and the reference EBR is determined by introducing the extrema and the degree of uniformity of EBR in the structure. The elastic modulus in the element with an EBR greater than the reference one is reduced based on the linear elastic finite element analysis and the equilibrium of strain energy. 3Genomic landscape and genetic manipulation of the black soldier fly Hermetia illucens,a natural waste recycler显示文摘The black soldier fly(BSF),Hermetia illucens(Diptera:Stratiomyidae),is renowned for its bioconversion of organic waste into a sustainable source of animal feed.We report a high-quality genome of 1.1 Gb and a consensus set of 16,770 gene models for this beneficial species.Compared to those of other dipteran species,the BSF genome has undergone a substantial expansion in functional modules related to septic adaptation,including immune system factors,olfactory receptors,and cytochrome P450s.We further profiled midgut transcriptomes and associated microbiomes of BSF larvae fed with representative types of organic waste.We find that the pathways related to digestive system and fighting infection are commonly enriched and that Firmicutes bacteria dominate the microbial community in BSF across all diets.To extend its potential practical applications,we further developed an efficient CRISPR/Cas9-based gene editing approach and implemented this to yield flightless and enhanced feeding capacity phenotypes,both of which could expand BSF production capabilities.Our study provides valuable genomic and technical resources for optimizing BSF lines for industrialization.Shuai Zhan Gangqi Fang Minmin Cai Zongqing Kou Jun Xu Yanghui Cao Liang Bai Yixiang Zhang Yongmao Jiang Xingyu Luo Jian Xu Xia Xu Longyu Zheng Ziniu Yu Hong Yang Zhijian Zhang Sibao Wang Jeffery K.Tomberlin Jibin Zhang Yongping Huang 2020Cell Research2020,30,1:9
4FAT1, a direct transcriptional target of E2F1, suppresses cell proliferation, migration and invasion in esophageal squamous cell carcinoma显示文摘Objective: Growing evidence indicates that FAT atypical cadherin 1(FAT1) has aberrant genetic alterations and exhibits potential tumor suppressive function in esophageal squamous cell carcinoma(ESCC). However, the role of FAT1 in ESCC tumorigenesis remains not well elucidated. The aim of this study was to further investigate genetic alterations and biological functions of FAT1, as well as to explore its transcriptional regulation and downstream targets in ESCC.Methods: The mutations of FAT1 in ESCC were achieved by analyzing a combined study from seven published genomic data, while the copy number variants of FAT1 were obtained from an analysis of our previous data as well as of The Cancer Genome Atlas(TCGA) and Cancer Cell Line Encyclopedia(CCLE) databases using the cBioPortal. The transcriptional regulation of FAT1 expression was investigated by chromatin immunoprecipitation(ChIP) and the luciferase reporter assays. In-cell western, Western blot and reverse transcription-quantitative polymerase chain reaction(RT-qPCR) were used to assess the indicated gene expression. In addition, colony formation and Transwell migration/invasion assays were employed to test cell proliferation, migration and invasion.Finally, RNA sequencing was used to study the transcriptomes.Results: FAT1 was frequently mutated in ESCC and was deleted in multiple cancers. Furthermore, the transcription factor E2 F1 occupied the promoter region of FAT1, and depletion of E2 F1 led to a decrease in transcription activity and mRNA levels of FAT1. Moreover, we found that knockdown of FAT1 promoted KYSE30 and KYSE150 cell proliferation, migration and invasion;while overexpression of FAT1 inhibited KYSE30 and KYSE410 cell proliferation, migration and invasion. 5Analysis and prognosis of tropical cyclone genesis over the western North Pacific on the background of global warming显示文摘As revealed by the observational study, there are more tropical cyclones generated over the western North Pacific from the early 1950s to the early 1970s in the 20th century and less tropical cyclones from the mid-1970s to the present. The decadal change of tropical cyclones activities are closely related to the decadal changes of atmospheric general circulation in the troposphere, which provide favorable or unfavorable conditions for the formation of tropical cyclone. Furthermore, based on the simulation of corresponding atmospheric general circulation from a coupled climate model under the schemes of Intergovernmental Panel on Climate Change (IPCC) special report on emission scenarios (SRES) A2 and B2 emissions scenarios an outlook on the tropical cyclone frequency generated over the western North Pacific in the coming half century is presented. 7Sintilimab for relapsed/refractory extranodal NK/T cell lymphoma: a multicenter, single-arm, phase 2 trial (ORIENT-4)显示文摘This study(ORIENT-4)aimed to assess the efficacy and safety of sintilimab,a humanized anti-PD-1 antibody,in patients with relapsed/refractory extranodal NK/T cell lymphoma(r/r ENKTL).ORIENT-4 is a multicenter,single-arm,phase 2 clinical trial(NCT03228836).Patients with r/r ENKTL who failed to at least one asparaginase-based regimen were enrolled to receive sintilimab 200 mg intravenously every 3 weeks for up to 24 months.The primary endpoint was the objective response rate(ORR)based on Lugano 2014 criteria.Twenty-eight patients with r/r ENKTL were enrolled from August 31,2017 to February 7,2018.Twenty-one patients(75.0%,95%CI:55.1–89.3%)achieved an objective response.With a median follow-up of 30.4 months,the median overall survival(OS)was not reached.The 24-month OS rate was 78.6%(95%CI,58.4–89.8%).Most treatment-related adverse events(TRAEs)were grade 1–2(71.4%),and the most common TRAE was decreased lymphocyte count(42.9%).Serious adverse events(SAEs)occurred in 7(25.0%)patients,and no patient died of adverse events.Sintilimab is effective and well tolerated in patients with r/r ENKTL and could be a novel therapeutic approach for the control of ENKTL in patients.Rong Tao Lei Fan Yongping Song Yu Hu Wei Zhang Yafei Wang Wei Xu Jianyong Li 2021Signal Transduction and Targeted Therapy2021,6,11:4

9Genome-wide identification and characterization of the lateral organ boundaries domain gene family in Brassica rapa var.rapa显示文摘The Lateral Organ Boundaries Domain(LBD)genes encode highly conserved plant-specific LOB domain proteins which regulate growth and development in various species.However,members of the LBD gene family have yet to be identified in Brassica rapa var.rapa.In the present study,fifty-nine LBD genes were identified and distributed on 10 chromosomes.The BrrLBD proteins are predicted to encode hydrophobic polypeptides between 118 and 394 amino acids in length and with molecular weights ranging from 13.31 to 44.24 kDa;the theoretical pi for these proteins varies from 4.83 to 9.68.There were 17 paralogous gene pairs in the BrrLBD family,suggesting that the amplification of the BrrLBD gene family involved largescale gene duplication events.Members of the BrrLBD family were divided into 7 subclades(class I a to e,class II a and b).Analysis of gene structure and conserved domains revealed that most BrrLBD genes of the same subclade had similar gene structures and protein motifs.The expression profiles of 59 BrrLBD genes were determined through Quantitative Real-time fluorescent PCR(qRT-PCR).Most BrrLBD genes in the same subclade had similar gene expression profiles.However,the expression patterns of 7 genes differed from their duplicates,indicating that although the gene function of most BrrLBD genes has been conserved,some BrrLBD genes may have undergone evolutionary change.Qin Yu Simin Hu Jiancan Du Yongping Yang Xudong Sun 2020Plant Diversity2020,42,1:3
10H7N9 virus infection triggers lethal cytokine storm by activating gasdermin E-mediated pyroptosis of lung alveolar epithelial cells显示文摘The H7N9 influenza virus emerged in China in 2013,causing more than 1560 human infections,39% of which were fatal.A ‘cytokine storm’ in the lungs of H7N9 patients has been linked to a poor prognosis and death;however,the underlying mechanism that triggers the cytokine storm is unknown.Here,we found that efficient replication of the H7N9 virus in mouse lungs activates gasdermin E(GSDME)-mediated pyroptosis in alveolar epithelial cells,and that the released cytosolic contents then trigger a cytokine storm.Knockout of Gsdme switched the manner of death of A549 and human primary alveolar epithelial cells from pyroptosis to apoptosis upon H7N9 virus infection,and Gsdme knockout mice survived H7N9 virus lethal infection.Our findings reveal that GSDME activation is a key and unique mechanism for the pulmonary cytokine storm and lethal outcome of H7N9 virus infection and thus opens a new door for the development of antivirals against the H7N9 virus.Xiaopeng Wan Jiqing Li Yupeng Wang Xiaofei Yu Xijun He Jianzhong Shi Guohua Deng Xianying Zeng Guobin Tian Yanbing Li Yongping Jiang Yuntao Guan Chengjun Li Feng Shao Hualan Chen 2022National Science Review2022,9,1:3
11A novel lignite pre-drying system with low-grade heat integration for modern lignite power plants显示文摘The lignite pre-drying process plays an important role in modern lignite power plants and the fluidized bed dryer with internal heat utilization is a promising drying method which has both high efficiency and cost-effectiveness. After conducting an in-depth analysis of a typical lignite pre-drying power plant, this work proposed a novel lignite pre-drying system with low-grade heat integration. Through system integration, the low-temperature evaporation of the lignite was recovered to heat the combustion air, while the residual heat from the flue gases was used to heat the feed or condensed water, thereby saving a portion of heat from the steam bleeds of the high and intermediate pressure turbines. The results for a1,000 MW lignite-fired power plant showed that, the proposed pre-drying system could yield an increase in net plant efficiency of approximately 3.6 % points and a reduction in the cost of electricity of $1.83/(MW h). 13Pre-deformation for Assembly Performance of Machine Centers显示文摘The current research of machine center accuracy in workspace mainly focuses on the poor geometric error subjected to thermal and gravity load while in operation,however,there are little researches focusing on the effect of machine center elastic deformations on workspace volume.Therefore,a method called pre-deformation for assembly performance is presented.This method is technically based on the characteristics of machine tool assembly and collaborative computer-aided engineering(CAE)analysis.The research goal is to enhance assembly performance,including straightness,positioning,and angular errors,to realize the precision of the machine tool design.A vertical machine center is taken as an example to illustrate the proposed method.The concept of travel error is defined to obtain the law of the guide surface.The machine center assembly performance is analyzed under cold condition and thermal balance condition to establish the function of pre-deformation.Then,the guide surface in normal direction is processed with the pre-deformation function,and the machine tool assembly performance is measured using a laser interferometer.The measuring results show that the straightness deviation of the Z component in the Y-direction is 158.9%of the allowable value primarily because of the gravity of the spindle head,and the straightness of the X and Y components is minimal.When the machine tool is processed in pre-deformation,the straightness of the Z axis moving component is reduced to 91.2%.This research proposes a pre-deformation machine center assembly method which has sufficient capacity to improving assembly accuracy of machine centers.SUN Yongping WANG Delun DONG Huimin XUE Runiu YU Shudong 2014Chinese Journal of Mechanical Engineering2014,27,3:2
14Triangulation of red sprites observed above a mesoscale convective system in North China显示文摘The triangulation of red sprites was obtained, based on concurrent observations over a mesoscale convective system(MCS) in North China from two stations separated by about 450 km. In addition, broadband sferics from the sprite-producing lightning were measured at five ground stations, making it possible to locate and identify the individual causative lightning discharges for different elements in this dancing sprite event. The results of our analyses indicate that the sprites were produced above the trailing stratiform region of the MCS, and their parent strokes were located mainly in the peripheral area of the stratiform. The lateral offset between sprites and causative strokes ranges from a few km to more than 50 km. 19An Experimental Study on Homoharringtonine Liposome and Glaucoma Filtration Surgery显示文摘Purpose: To observe the inhibitory action of homoharringtonine liposome during the healing process of wounds in the filtering sitesMethods: posterior sclerectomies were performed in 14 rabbits. Postoperatively one eye of each rabbit received subconjunctival injections of HH liposome and fellow eye received saline injection in a randomized masked fashion.Results; Fourteen days after operation the IOP of experimental eyes reduced significantly (P < 0.01) as compared with the controlled eyes, and the number of remaining filtering blebs increased noticeably (P< 0.05). Pathohistological examination revealed that the number of fibroblasts per square micron in the filtering sites and the thickness of the scars in the center of the filtering sites of the experimental eyes were less than those of the controlled eyes.
