1晚期非小细胞肺癌免疫检查点抑制剂新型生物标志物:血液PD-L1动态检测显示文摘背景免疫检查点抑制剂(immune checkpoint inhibitors,ICIs)具有良好疗效已成为恶性肿瘤治疗领域备受瞩目的翘楚。然而,仅有小部分患者获得长期临床获益。如何选择生物标志物来筛查ICIs治疗优势人群仍是一大热点。方法收集51例晚期恶性肿瘤患者的肿瘤组织和配对血样本,进行PD-L1表达的相关性分析。40例晚期非小细胞肺癌(non-small cell lung cancer,NSCLC)患者接受ICIs治疗,2个月时检测血液PD-L1(bloodPD-L1,bPD-L1)表达的动态变化,包括PD-L1 mRNA、外泌体PD-L1(exosomalPD-L1,exoPD-L1)蛋白和可溶性PD-L1(solublePD-L1,sPD-L1)。应用R软件计算最佳截断值,用于预测最佳客观缓解(best objective response,b OR)、无进展生存期(progression-free survival,PFS)和总生存期(overall survival,OS)。结果51例不同恶性肿瘤患者中,组织PD-L1(tissuePD-L1,tPD-L1)表达阳性患者的PD-L1mRNA表达水平显著升高。40例接受ICIs治疗的晚期NSCLC患者中,bPD-L1 mRNA上调≥2.04的患者b OR、PFS和OS更佳;21例接受ICIs治疗的晚期NSCLC患者中,exoPD-L1上调≥1.86的患者临床获益更大。sPD-L1动态变化与ICIs疗效无显著相关性。PD-L1 mRNA和exoPD-L1联合检测更有利于预测ICIs治疗疗效。结论bPD-L1表达与tPD-L1表达存在正相关;晚期NSCLC患者ICIs治疗初期PD-L1 mRNA和(或)exoPD-L1表达升高,可作为评估临床获益的新型生物标志物。Qiao YANG Mingjing CHEN Jiaoyang GU Kai NIU Xianlan ZHAO Linpeng ZHENG Zihan XU Yongxin YU Feng LI Lingxin MENG Zhengtang CHEN Wenlei ZHUO Luping ZHANG Jianguo SUN 杨峤(译) 孙建国(译) 2021中国肺癌杂志2021,24,12:10
2Effect of blade tip geometry on tip leakage vortex dynamics and cavitation pattern in axial-flow pump显示文摘A series of blade tip geometries, including original plain tip, rounded tip on the pressure side and diverging tip towards the suction side, were adopted to investigate the effect of blade geometry on tip leakage vortex dynamics and cavitation pattern in an axial-flow pump. On the basis of the computation, it clearly shows the flow structure in the clearance for different tip configurations by the detailed data of axial velocity and turbulent kinetic energy. The in-plain trajectory, in aspects of the angle between the blade suction side and vortex core and the initial point of tip leakage vortex, was presented using the maximum swirling strength method. The most striking feature is that the inception location of tip leakage vortex is delayed for chamfered tip due to the change of blade loading on suction side. Some significant non-dimensional parameters, such as pressure, swirling strength and turbulent kinetic energy, were used to depict the characteristics of tip vortex core. By the distribution of circumferential vorticity which dominates the vortical flows near the tip region, it is observed that the endwall detachment as the leakage flow meets the mainstream varies considerably for tested cases. The present study also indicates that the shear layer feeds the turbulence into tip leakage vortex core, but the way is different. For the chamfered tip, high turbulence level in vortex core is mainly from the tip clearance where large turbulent kinetic energy emerges, while it is almost from a layer extending from the suction side corner for rounded tip. 3A Review on Binderless Tungsten Carbide: Development and Application显示文摘WC-Co alloys have enjoyed great practical significance owing to their excellent properties during the past decades.Despite the advantages,however,recently there have been concerns about the challenges associated with the use of Co,i.e.price instability,toxicity and properties degeneration,which necessitates the fabrication of binderless tungsten carbide(BTC).On the other hand,BTC or BTC composites,none of them,to date has been commercialized and produced on an industrial scale,but only used to a limited extent for specialized applications,such as mechanical seals undergoing high burthen as well as high temperature electrical contacts.There are two challenges in developing BTC:fully densifying the sintered body together with achieving a high toughness.Thus,this review applies towards comprehensively summarize the current knowledge of sintering behavior,microstructure,and mechanical properties of BTC,highlighting the densification improving strategies as well as toughening methods,so as to provide reference for those who would like to enhance the performance of BTC with better reliability advancing them to further wide applications and prepare the material in a way that is environment friendly,harmless to human health and low in production cost.This paper shows that the fabrication of highly dense and high-performance BTC is economically and technically feasible.The properties of BTC can be tailored by judiciously selecting the chemical composition coupled with taking into careful account the effects of processing techniques and parameters.Jialin Sun Jun Zhao Zhifu Huang Ke Yan Xuehui Shen Jiandong Xing Yimin Gao Yongxin Jian Hejie Yang Bo Li 2020Nano-Micro Letters2020,12,1:8

5Pilot Domain NOMA for Grant-Free Massive Random Access in Massive MIMO Marine Communication System显示文摘We propose a pilot domain non-orthogonal multiple access(NOMA)for uplink massive devices grant-free random access scenarios in massive multiple-input multiple-output(MIMO)maritime communication systems.These scenarios are characterized by numerous devices with sporadic access behavior,and therefore only a subset of them are active.Due to massive potential devices in the network,it is infeasible to assign a unique orthogonal pilot to each device in advance.In such scenarios,pilot decontamination is a crucial problem.In this paper,the devices are randomly assigned non-orthogonal pilots which are constructed by a linear combination of some orthogonal pilots.We show that a bipartite graph can conveniently describe the interference cancellation(IC)processes of pilot decontamination.High spectrum efficiency(SE)and low outage probability can be obtained by selecting the numbers of orthogonal pilots according to the given probability distribution.Numerical evaluatioDs show that the proposed pilot domain NOMA decreases the outage probability from 20%to 2 e-12 at the SE of 4 bits/s/Hz for a single device,compared to the conventional method of slotted ALOHA with 1024 antennas at the BS,or increases the spectrum efficiency from 1.2 bits/s/Hz to 4 bit/s/Hz at the outage probability of2 e-12 in contrast with the Welch bound equality(WBE)non-orthogonal pilots.Yongxin Liu Ming Zhao Limin Xiao Shidong Zhou 2020China Communications2020,17,6:3
6Partial anhysteretic remanent magnetization (pARM) of synthetic single-and multidomain magnetites and its paleoenvironmental significance显示文摘In order to isolate magnetic signals carried by single-domain (SD) ferrimagnetic (FM) minerals from multi-domain (MD) FM minerals, we developed a few pa- rameters using partial anhysteretic remanent magnetization (pARM). Because MD fraction contains only soft (easy to be demagnetized) coercivity spectrum, pARM(>20 mT) was sensitive in eliminating MD contributions. Ratio of pARM(5 mT, 10 mT)/pARM(0, 5 mT) is useful in quantifying a rela- tive abundance of mass ratio between SD and MD fractions. 8Rett syndrome-causing mutations compromise MeCP2- mediated liquid—liquid phase separation of chromatin显示文摘Rett syndrome(RTT),a severe postnatal neurodevelopmental disorder,is caused by mutations in the X-linked gene encoding methyl-CpG-binding protein 2(MeCP2).MeCP2 is a chromatin organizer regulating gene expression.RTT-causing mutations have been shown to affect this function.However,the mechanism by which MeCP2 organizes chromatin is unclear.In this study,we found that MeCP2 can induce compaction and liquid-liquid phase separation of nudeosomal arrays in vitro,and DNA methylation further enhances formation of chromatin condensates by MeCP2.Interestingly,RTT-causing mutations compromise MeCP2-mediated chromatin phase separation,while benign variants have little effect on this process.Moreover,MeCP2 competes with linker histone H1 to form mutually exclusive chromatin condensates in vitro and distinct heterochromatin foci in vivo.RTT-causing mutations reduce or even abolish the ability of MeCP2 to compete with histone H1 and to form chromatin condensates.Together,our results identify a novel mechanism by which phase separation underlies MeCP2-mediated heterochromatin formation and reveal the potential link between this process and the pathology of RTT.Liang Wang Mingli Hu Mei-Qing Zuo Jicheng Zhao Di Wu Li Huang Yongxin Wen Yunfan Li Ping Chen Xinhua Bao Meng-Qiu Dong Guohong Li Pilong Li 2020Cell Research2020,30,5:3
9The tribological behavior of a surface-nanocrystallized magnesium alloy AZ31 sheet after ultrasonic shot peening treatment显示文摘A magnesium alloy AZ31 sheet was processed by ultrasonic shot peening treatment to fabricate a surface nanocrystalline,and a ball-on-disk dry sliding wear test was performed to evaluate the tribological behavior after treatment.The microstructure observation indicated a gradient nanocrystalline structure was formed after USSP treatment.The microhardness at the top surface was improved from 60 HV to 145 HV after treatment.The formed nanocrystalline resulted in an easy formation of MgO patches on the surface and reduced the coefficient of friction.Moreover,the formed nanocrystalline leaded to a retard of delamination with increasing the sliding speed and applied load,which was due to its stronger sub-surface.Under high sliding speed(0.5 m/s)and high applied load(50 N),it was firstly found that the formed nanocrystalline prevented the happening of thermal softening and melting.The possible reasons accounting for the prevention of thennal softening and melting were discussed accordingly.Jianyue Zhang Yongxin Jian Xuzhe Zhao Dean Meng Fusheng Pan Qingyou Han 2021Journal of Magnesium and Alloys2021,9,4:3

11A phase field study for influence of elastic energy on L1_0→L1_2 transient ordering in Ni_(75)Al_(17)Zn_8显示文摘The influence of the elastic energy on L10→L12 transient ordering transformation was investigated by microscopic phase field method. It is found that there are three stages experienced in atomic ordering: solute clustering+L10 short range ordering → L10 long range ordering → L12 long range ordering. Before the formation of the high ordered L12 phase, it has firstly taken place the transformation from matrix to L10 phase, and then held the L10→L12 secondary transformation. Elastic energy is proved to take little effect on the stage of short range ordering, but as the elastic energy is multiplied, it obviously shortened the course of the solute clustering, and speeded up the proceeding of the L10 long range ordering transition. Accordingly, the increased elastic energy also strengthens the single crystalline directionality of L10 phase projecting on 2D plane and makes the ordered degree of Al and Zn atoms enhanced. 12Enrichment factors of movable hydrocarbons in lacustrine shale oil and exploration potential of shale oil in Gulong Sag,Songliao Basin,NE China显示文摘The geological characteristics and production practices of the major middle-and high-maturity shale oil exploration areas in China are analyzed.Combined with laboratory results,it is clear that three essential conditions,i.e.economic initial production,commercial cumulative oil production of single well,and large-scale recoverable reserves confirmed by the testing production,determine whether the continental shale oil can be put into large-scale commercial development.The quantity and quality of movable hydrocarbons are confirmed to be crucial to economic development of shale oil,and focuses in evaluation of shale oil enrichment area/interval.The evaluation indexes of movable hydrocarbon enrichment include:(1)the material basis for forming retained hydrocarbon,including TOC>2%(preferentially 3%-4%),and typeⅠ-Ⅱkerogens;(2)the mobility of retained hydrocarbon,which is closely related to the hydrocarbon composition and flow behaviors of light/heavy components,and can be evaluated from the perspectives of thermal maturity(Ro),gas-oil ratio(GOR),crude oil density,quality of hydrocarbon components,preservation conditions;and(3)the reservoir characteristics associated with the engineering reconstruction,including the main pore throat distribution zone,reservoir physical properties(including fractures),lamellation feature and diagenetic stage,etc.Accordingly,13 evaluation indexes in three categories and their reference values are established.The evaluation indicates that the light shale oil zones in the Gulong Sag of Songliao Basin have the most favorable enrichment conditions of movable hydrocarbons,followed by light oil and black oil zones,containing 20.8×10^(8) t light oil resources in reservoirs with R_(0)>1.2%,pressure coefficient greater than 1.4,effective porosity greater than 6%,crude oil density less than 0.82 g/cm^(3),and GOR>100 m/m^(3).The shale oil in the Gulong Sag can be explored and developed separately by the categories(resource sweet spot,engineering sweet spot,and tight oil sweet spot)depending on shale oil flowability.The Gulong Sag is the most promising area to achieve large-scale breakthrough and production of continental shale oil in China.ZHAO Wenzhi BIAN Congsheng LI Yongxin ZHANG Jinyou HE Kun LIU Wei ZHANG Bin LEI Zhengdong LIU Chang ZHANG Jingya GUAN Ming LIU Shijul 2023Petroleum Exploration and Development2023,50,3:2
13Atomic Scale Computer Simulation for Early Precipitation Process of Ni75Al6Vi9 Alloy显示文摘The atomic scale computer simulation for initial precipitation mechanism of Ni75Al6V19 alloy was carried out for the first time by employing the microscopic diffusion equation. The initial precipitation process was invest igated throughsimulating the atomic pictures and calculating the order parameters of the two kinds of ordered phases. Simulationresults show that the γ′ordered phase precipitated earlier than θ ordered phase by congruent ordering+spinodal decomposition mechanism and thus produced a nonstoicheometric γ′ single ordered phase. 14Corilagin inhibits SARS-CoV-2 replication by targeting viral RNA-dependent RNA polymerase显示文摘Severe acute respiratory syndrome coronavirus-2(SARS-CoV-2) has become one major threat to human population health.The RNA-dependent RNA polymerase(RdRp) presents an ideal target of antivirals,whereas nucleoside analogs inhibitor is hindered by the proofreading activity of coronavirus.Herein,we report that corilagin(RAI-S-37) as a non-nucleoside inhibitor of SARS-CoV-2 RdRp,binds directly to RdRp,effectively inhibits the polymerase activity in both cell-free and cell-based assays,fully resists the proofreading activity and potently inhibits SARS-CoV-2 infection with a low 50% effective concentration(EC50) value of 0.13 μmol/L.Computation modeling predicts that RAI-S-37 lands at the palm domain of RdRp and prevents conformational changes required for nucleotide incorporation by RdRp.In addition,combination of RAI-S-37 with remdesivir exhibits additive activity against antiSARS-CoV-2 RdRp.Together with the current data available on the safety and pharmacokinetics of corilagin as a medicinal herbal agent,these results demonstrate the potential of being developed into one of the much-needed SARS-CoV-2 therapeutics.Quanjie Li Dongrong Yi Xiaobo Lei Jianyuan Zhao Yongxin Zhang Xiangling Cui Xia Xiao Tao Jiao Xiaojing Dong Xuesen Zhao Hui Zeng Chen Liang Lili Ren Fei Guo Xiaoyu Li Jianwei Wang Shan Cen 2021Acta Pharmaceutica Sinica B2021,11,6:2
