1Phase 1 clinical trial demonstrated that MUC1 positive metastatic seminal vesicle cancer can be effectively eradicated by modified Anti-MUC1 chimeric antigen receptor transduced T cells显示文摘Recent progress in chimeric antigen receptor-modified T-cell(CAR-T cell) technology in cancer therapy is extremely promising, especially in the treatment of patients with B-cell acute lymphoblastic leukemia. In contrast, due to the hostile immunosuppressive microenvironment of a solid tumor, CAR T-cell accessibility and survival continue to pose a considerable challenge, which leads to their limited therapeutic efficacy. In this study, we constructed two anti-MUC1 CAR-T cell lines. One set of CAR-T cells contained SM3 single chain variable fragment(sc Fv) sequence specifically targeting the MUC1 antigen and co-expressing interleukin(IL) 12(named SM3-CAR). The other CAR-T cell line carried the SM3 sc Fv sequence modified to improve its binding to MUC1 antigen(named p SM3-CAR) but did not co-express IL-12. When those two types of CAR-T cells were injected intratumorally into two independent metastatic lesions of the same MUC1+ seminal vesicle cancer patient as part of an interventional treatment strategy, the initial results indicated no side-effects of the MUC1 targeting CAR-T cell approach, and patient serum cytokines responses were positive. 2Advances and prospects of rare earth metal-organic frameworks in catalytic applications显示文摘In recent years,rare earth metal-organic frameworks(RE-MOFs)have attracted increasing attention because of abundant coordination behaviors,adjustable channels and stable networks.The various electron structures of rare earth ions contribute to the catalytic applications of RE-MOFs and their derivates.In this review,we systematically summarize the research progress that using RE-MOFs and their derivates as catalysts for organic reaction,photocatalytic reaction,and CO oxidation reaction.The major parts include synthetic strategies and catalytic applications of the RE-MOFs.Finally,impressive achievements of RE-MOF catalysts are emerging and a prospect is provided for the development of RE-MOF catalysts at the end of the review.Xiaochen Sun Kun Yuan Yawen Zhang 2020Journal of Rare Earths2020,38,8:16

5MXene-composited highly stretchable,sensitive and durable hydrogel for flexible strain sensors显示文摘The flourishing development in flexible electronics has provoked intensive research in flexible strain sensors to realize accurate perception acquisition under different external stimuli.However,building hydrogel-based strain sensors with high stretchability and sensitivity remains a great challenge.Herein,MXene nanosheets were composited into polyacrylamide-sodium alginate matrix to construct mechanical robust and sensitive double networked hydrogel strain sensor.The hydrophilic MXene nanosheets formed strong interactions with the polymer matrix and endowed the hydrogel with excellent tensile properties(3150%),compliant mechanical strength(2.03 kPa^(-1)in Young’s Module)and long-lasting stability and fatigue resistance(1000 dynamic cycles under 1,600%strain).Due to the highly oriented MXene-based three dimensional conductive networks,the hydrogel sensor achieved extremely high tensile sensitivity(18.15 in gauge factor)and compression sensitivity(0.38 kPa^(-1)below 3 kPa).MXene hydrogel-based strain sensors also displayed negligible hysteresis in electromechanical performance,typical frequent-independent feature and rapid response time to external stimuli.Moreover,the sensor exhibited accurate response to different scales of human movements,providing potential application in speech recognition,expression recognition and handwriting verification.Wei Yuan Xinyu Qu Yao Lu Wen Zhao Yanfang Ren Qian Wang Wenjun Wang Xiaochen Dong 2021Chinese Chemical Letters2021,32,6:3
6Tet2 Regulates Osteoclast Differentiation by Interacting with Runx1 and Maintaining Genomic 5-Hydroxymethylcytosine(5hmC)显示文摘As a dioxygenase, Ten-Eleven Translocation 2(TET2) catalyzes subsequent steps of 5-methylcytosine(5 mC) oxidation. TET2 plays a critical role in the self-renewal, proliferation,and differentiation of hematopoietic stem cells, but its impact on mature hematopoietic cells is not well-characterized. Here we show that Tet2 plays an essential role in osteoclastogenesis. Deletion of Tet2 impairs the differentiation of osteoclast precursor cells(macrophages) and their maturation into bone-resorbing osteoclasts in vitro. Furthermore, Tet2^(-/-) mice exhibit mild osteopetrosis, accompanied by decreased number of osteoclasts in vivo. Tet2 loss in macrophages results in the altered expression of a set of genes implicated in osteoclast differentiation, such as Cebpa, Mafb, and Nfkbiz. Tet2 deletion also leads to a genome-wide alteration in the level of 5-hydroxymethylcytosine(5 hmC) and altered expression of a specific subset of macrophage genes associated with osteoclast differentiation. Furthermore, Tet2 interacts with Runx1 and negatively modulates its transcriptional activity. Our studies demonstrate a novel molecular mechanism controlling osteoclast differentiation and function by Tet2, that is, through interactions with Runx1 and the maintenance of genomic 5 hmC. 7Exosomes secreted from cardiomyocytes suppress the sensitivity of tumor ferroptosis in ischemic heart failure显示文摘Heart failure(HF)patients in general have a higher risk of developing cancer.Several animal studies have indicated that cardiac remodeling and HF remarkably accelerate tumor progression,highlighting a cause-and-effect relationship between these two disease entities.Targeting ferroptosis,a prevailing form of non-apoptotic cell death,has been considered a promising therapeutic strategy for human cancers.Exosomes critically contribute to proximal and distant organ-organ communications and play crucial roles in regulating diseases in a paracrine manner.However,whether exosomes control the sensitivity of cancer to ferroptosis via regulating the cardiomyocyte-tumor cell crosstalk in ischemic HF has not yet been explored.Here,we demonstrate that myocardial infarction(MI)decreased the sensitivity of cancer cells to the canonical ferroptosis activator erastin or imidazole ketone erastin in a mouse model of xenograft tumor.Post-MI plasma exosomes potently blunted the sensitivity of tumor cells to ferroptosis inducers both in vitro in mouse Lewis lung carcinoma cell line LLC and osteosarcoma cell line K7M2 and in vivo with xenograft tumorigenesis model.The expression of miR-22-3p in cardiomyocytes and plasma-exosomes was significantly upregulated in the failing hearts of mice with chronic MI and of HF patients as well.Incubation of tumor cells with the exosomes isolated from post-MI mouse plasma or overexpression of miR-22-3p alone abrogated erastin-induced ferroptotic cell death in vitro.Cardiomyocyte-enriched miR-22-3p was packaged in exosomes and transferred into tumor cells.Inhibition of cardiomyocyte-specific miR-22-3p by AAV9 sponge increased the sensitivity of cancer cells to ferroptosis.ACSL4,a pro-ferroptotic gene,was experimentally established as a target of miR-22-3p in tumor cells.Taken together,our findings uncovered for the first time that MI suppresses erastin-induced ferroptosis through releasing miR-22-3p-enriched exosomes derived from cardiomyocytes.Therefore,targeting exosome-mediated cardiomyocyte/tumor pathological communication may offer a novel approach for the ferroptosis-based antitumor therapy.Ye Yuan Zhongting Mei Zhezhe Qu Guanghui Li Shuting Yu Yingqi Liu Kuiwu Liu Zhihua Shen Jiaying Pu Yanquan Wang Changhao Wang Zhiyong Sun Qian Liu Xiaochen Pang Ao Wang Zijing Ren Tong Wang Ying Liu Jinhuan Hong Jiajie Xie Xin Li Zhonghua Wang Weijie Du Baofeng Yang 2023Signal Transduction and Targeted Therapy2023,8,4:2

10A Yeast BiFC-seq Method for Genome-wide Interactome Mapping显示文摘Genome-wide physical protein±protein interaction(PPI)mapping remains a major challenge for current technologies.Here,we reported a high-efficiency BiFC-seq method,yeastenhanced green fluorescent protein-based bimolecular fluorescence complementation(y EGFPBiFC)coupled with next-generation DNA sequencing,for interactome mapping.We first applied y EGFP-BiFC method to systematically investigate an intraviral network of the Ebola virus.Two-thirds(9/14)of known interactions of EBOV were recaptured,and five novel interactions were discovered.Next,we used the BiFC-seq method to map the interactome of the tumor protein p53.We identified 97 interactors of p53,more than three-quarters of which were novel.Furthermore,in a more complex background,we screened potential interactors by pooling two BiFC libraries together and revealed a network of 229 interactions among 205 proteins.These results show that BiFC-seq is a highly sensitive,rapid,and economical method for genome-wide interactome mapping.Limin Shang Yuehui Zhang Yuchen Liu Chaozhi Jin Yanzhi Yuan Chunyan Tian Ming Ni Xiaochen Bo Li Zhang Dong Li Fuchu He Jian Wang 2022Genomics, Proteomics & Bioinformatics2022,20,4:1

15Burch colposuspension for stress urinary incontinence: a 14-year prospective follow-up显示文摘We aimed to evaluate the long-term effectiveness and safety of Burch colposuspension(BC) for stress urinary incontinence(SUI). In this prospective cohort study, 84 patients with SUI undergoing BC were enrolled from February 2004 to January 2010.Data on long-term subjective success and postoperative complications were collected at clinic visits and by telephone follow-up.During a mean follow-up period of 14.2 years, 68%(57/84) patients completed the follow-up. A total of 68.4% of patients(39/57) reported absence of SUI symptoms, 73.6%(42/57) were subjectively satisfied according to the Patient Global Impression of Improvement, and 68.4%(39/57) reported subjective success regarding urinary symptoms via the Urinary Distress Inventory Short Form. However, 28.1%(16/57) suffered at least one long-term postoperative complication and incident. Specifically, 1 in25(4.0%) sexually active patients reported dyspareunia, 3 patients(5.3%) had de novo overactive bladder, and 6 patients(10.5%)reported voiding dysfunction. Four patients(7.0%) reported new onset prolapse symptoms, and 3 patients(5.3%) underwent secondary urinary incontinence surgery. Our study indicated that Burch colposuspension is an effective procedure for SUI, and the cure effect was largely maintained for the 14-year follow-up period, with relatively low complication rates. 16High-temperature phase relations of ZrN–ZrO_2–Y_2O_3 ternary system显示文摘Zirconium nitride(ZrN) ceramics were prepared via hot pressed sintering(HP) at 1750 ℃ in N_2 atmosphere with ZrO_2–Y_2O_3 as sintering additive. X-ray diffraction was applied to analyze the phase composition of the as-prepared ceramics to study the high temperature phase relation in ZrN –ZrO_2–Y_2O_3 ternary system and establish ZrN –ZrO_2–Y_2O_3 ternary phase diagrams. 17Association between homologous recombination deficiency and outcomes with platinum and platinum-free chemotherapy in patients with triple-negative breast cancer显示文摘Objective:The choice of chemotherapeutic regimen for triple-negative breast cancer(TNBC)remains controversial.Homologous recombination deficiency(HRD)has attracted increasing attention in informing chemotherapy treatment.This study was aimed at investigating the feasibility of HRD as a clinically actionable biomarker for platinum-containing and platinum-free therapy.Methods:Chinese patients with TNBC who received chemotherapy between May 1,2008 and March 31,2020 were retrospectively analyzed with a customized 3D-HRD panel.HRD positivity was defined by an HRD score≥30 or deleterious BRCA1/2 mutation.A total of 386 chemotherapy-treated patients with TNBC were screened from a surgical cohort(NCT01150513)and a metastatic cohort,and 189 patients with available clinical and tumor sequencing data were included.Results:In the entire cohort,49.2%(93/189)of patients were identified as HRD positive(40 with deleterious BRCA1/2 mutations and 53 with BRCA1/2 intact with an HRD score of≥30).In the first-line metastatic setting,platinum therapy was associated with longer median progression-free survival(mPFS)than platinum-free therapy[9.1 vs.3.0 months;hazard ratio(HR),0.43;95%confidence interval 0.22–0.84;P=0.01].Among HRD-positive patients,the mPFS was significantly longer in those treated with platinum rather than platinum-free therapy(13.6 vs.2.0 months;HR,0.11;P=0.001).Among patients administered a platinum-free regimen,HRD-negative patients showed a PFS significantly superior to that of HRD-positive patients(P=0.02;treatment-biomarker P-interaction=0.001).Similar results were observed in the BRCA1/2-intact subset.In the adjuvant setting,HRD-positive patients tended to benefit more from platinum chemotherapy than from platinum-free chemotherapy(P=0.05,P-interaction=0.02).Conclusions:HRD characterization may guide decision-making regarding the use of platinum treatment in patients with TNBC in both adjuvant and metastatic settings.Yimeng Chen Xue Wang Feng Du Jian Yue Yiran Si Xiaochen Zhao Lina Cui Bei Zhang Ting Bei Binghe Xu Peng Yuan 2023Cancer Biology & Medicine2023,20,2:1

20Automatic interval management for aircraft based on dynamic fuzzy speed control considering uncertainty显示文摘A novel real-time autonomous Interval Management System(IMS)is proposed to automate interval management,which considers the effect of wind uncertainty using the Dynamic Fuzzy Velocity Decision(DFVD)algorithm.The membership function can be generated dynamically based on the True Air Speed(TAS)limitation changes in real time and the interval criterion of the adjacent aircraft,and combined with human cognition to formulate fuzzy rules for speed adjusting decision-making.Three groups of experiments were conducted during the en-route descent stage to validate the proposed IMS and DFVD performances,and to analyze the impact factors of the algorithm.The verification experimental results show that compared with actual flight status data under controllers’command,the IMS reduces the descent time by approaching 30%with favorable wind uncertainty suppression performance.Sensitivity analysis shows that the ability improvement of DFVD is mainly affected by the boundary value of the membership function.Additionally,the dynamic generation of the velocity membership function has greater advantages than the static method in terms of safety and stability.Through the analysis of influencing factors,we found that the interval criterion and aircraft category have no significant effect on the capability of IMS.In a higher initial altitude scenario,the initial interval should be appropriately increased to enhance safety and efficiency during the descent process.This prototype system could evolve into a realtime Flight-deck Interval Management(FIM)tool in the future.Jie YUAN Yang PEI Yan XU Xiaochen LI Yuxue GE 2023Chinese Journal of Aeronautics2023,36,11:0
