Efficacy and safety of a novel anti-HER2 therapeutic antibody RC48 in patients with HER2-overexpressing,locally advanced or metastatic gastric or gastroesophageal junction cancer:a single-arm phase II study显示文摘Background:Current treatment options for human epidermal growth factor receptor 2(HER2)-overexpressing gastric cancer at third-line have shown limited clinical benefit.Further, there is no specific treatment for HER2 immunohistochemistry(IHC)2+and fluorescence in-situ hybridization-negative patients. Here,we report the efficacy and safety of a novel anti-HER2 antibody RC48 for patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer. Methods:Patients with HER2-overexpressing(IHC 2+or 3+),locally advanced or metastatic gastric or gastroesophageal junction cancer who were under at least second-line therapy were eligible and received RC482.5 mg/kg alone every 2 weeks. The primary endpoint was the objective response rate(ORR)assessed by an independent review committee.Secondary endpoints included progressionfree survival(PFS),overall survival(OS),duration of response, time to progression, disease control rate, and safety. Results:Of 179 patients screened,125 were eligible and received RC48 treatment. The ORR was 24.8%(95%confidence interval[CI]:17.5%-33.3%).The median PFS and OS were 4.1 months(95%CI:3.7-4.9 months)and 7.9 months(95%CI:6.7-9.9 months),respectively. The most frequently reported adverse events were decreased white blood cell count(53.6%),asthenia(53.6%),hair loss(53.6%),decreased neutrophil count(52.0%),anemia(49.6%),and increased aspartate aminotransferase level(43.2%).Serious adverse events(SAEs)occurred in 45(36.0%)patients,and RC48-related SAEs were mainly decreased neutrophil count(3.2%).Seven patients had adverse events that led to death were not RC48-related.Conclusions:RC48 showed promising activity with manageable safety, suggesting potential application in patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer who have previously received at least two lines of chemotherapy. | Zhi Peng Tianshu Liu Jia Wei Airong Wang Yifu He Liuzhong Yang Xizhi Zhang Nanfeng Fan Suxia Luo Zhen Li Kangsheng Gu Jianwei Lu Jianming Xu Qingxia Fan Ruihua Xu Liangming Zhang Enxiao Li Yuping Sun Guohua Yu Chunmei Bai Yong Liu Jiangzheng Zeng Jieer Ying Xinjun Liang Nong Xu Chao Gao Yongqian Shu Dong Ma Guanghai Dai Shengmian Li Ting Deng Yuehong Cui Jianmin Fang Yi Ba Lin Shen | 2021 | Cancer Communications2021,41,11: | 29
Reflective chiral meta-holography: multiplexing holograms for circularly polarized waves显示文摘By allowing almost arbitrary distributions of amplitude and phase of electromagnetic waves to be generated by a layer of sub-wavelength-size unit cells, metasurfaces have given rise to the field of meta-holography.However,holography with circularly polarized waves remains complicated as the achiral building blocks of existing meta-holograms inevitably contribute to holographic images generated by both left-handed and right-handed waves. Here we demonstrate how planar chirality enables the fully independent realization of high-efficiency meta-holograms for one circular polarization or the other.Such circular-polarization-selective meta-holograms are based on chiral building blocks that reflect either left-handed or right-handed circularly polarized waves with an orientation-dependent phase.Using terahertz waves,we experimentally demonstrate that this allows the straightforward design of reflective phase meta-holograms,where the use of alternating structures of opposite handedness yields independent holographic images for circularly polarized waves of opposite handedness with negligible polarization cross-talk. | Qiu Wang Eric Plum Quanlong Yang Xueqian Zhang Quan Xu Yuehong Xu Jiaguang Han Weili Zhang | 2018 | Light(Science & Applications)2018,7,1: | 15
Association of three single nucleotide polymorphisms of ESR1 with breast cancer susceptibility:a meta-analysis显示文摘Expression of estrogen receptors is correlated with breast cancer risk, but inconsistent results have been reported. To clarify potential estrogen receptor(ESR)-related breast cancer risk,we analyzed genetic variants of ESR1 in association with breast cancer susceptibility. We performed a meta-analysis to investigate the association between rs2234693,rs1801132,and rs2046210(single nucleotide polymorphisms of ESR1),and breast cancer risk. Our analysis included 44 case-control studies. For rs2234693,the CC genotype had a higher risk of breast cancer compared to the TT or CT genotype. For rs2046210,the AA,GA,or GA + GG genotype had a much higher risk compared to the GG genotype. No significant association was found for the rs 1801132 polymorphism with breast cancer risk. This meta-analysis demonstrates association between the rs2234693 and rs2046210 polymorphisms of ESR1 and breast cancer risk. The correlation strength between rs2234693 and breast cancer susceptibility differs in subgroup assessment by ethnicity. | Xu Hu Linfei Jiang Chenhui Tang Yuehong Ju Li Jiu Yongyue Wei Li Guo Yang Zhao | 2017 | The Journal of Biomedical Research2017,31,3: | 4
Emerging roles of NudC family: from molecular regulation to clinical implications显示文摘Nuclear distribution gene C(Nud C) was first found in Aspergillus nidulans as an upstream regulator of Nud F, whose mammalian homolog is Lissencephaly 1(Lis1). Nud C is conserved from fungi to mammals. Vertebrate Nud C has three homologs: Nud C, Nud C-like protein(Nud CL), and Nud C-like protein 2(Nud CL2). All members of the Nud C family share a conserved p23 domain, which possesses chaperone activity both in conjunction with and independently of heat shock protein 90(Hsp90). Our group and the others found that Nud C homologs were involved in cell cycle regulation by stabilizing the components of the LIS1/dynein complex. Additionally, Nud C plays important roles in cell migration, ciliogenesis, thrombopoiesis, and the inflammatory response. It has been reported that Nud CL is essential for the stability of the dynein intermediate chain and ciliogenesis via its interaction with the dynein 2 complex. Our data showed that Nud CL2 regulates the LIS1/dynein pathway by stabilizing LIS1 with Hsp90 chaperone. The fourth distantly related member of the Nud C family, CML66, a tumor-associated antigen in human leukemia, contains a p23 domain and appears to promote oncogenesis by regulating the IGF-1R-MAPK signaling pathway. In this review, we summarize our current knowledge of the Nud C family and highlight its potential clinical relevance. | Qiqin Fu Wei Wang Tianhua Zhou Yuehong Yang | 2016 | Science China(Life Sciences)2016,59,5: | 4
Intense single-band red upconversion emission in BiOCl:Er3+ layered semiconductor via co-doping Ho3+显示文摘Exploring a new tuning way to facilely realize single-band red emission in trivalent rare-earth ions(RE3+) doped upconversion(UC) materials is still desirable. In this work, the intense single-band red emission is achieved by co-doping only Ho3+in the BiOCl:Er3+ under 1550 nm excitation. In the BiOCl layered host,co-doping Ho3+can further enhance the red emission and simultaneously suppress the green emission of Er3+,and thus obviously improve the red-to-green(R/G) ratio. It is found that Ho3+does not se rve as ene rgy trapping through the 5 I6 state as in traditional UC materials but acts as ET bridge(4 S3/2,2 H11/2(Er3+)→5 F4,5 S2(Ho3+)→4 F9/2(Er3+)).The tuning mechanism of Ho3+is discussed in detail and further confirms through a comparative experiment. Our research gives an unusual perspective to tune the UC behavior of Er3+through co-doping Ho3+,which might be inspiring for achievement of single-band red UC emission. | Yuehong Peng Jincai Peng Jiajun Han Tianhui Wang Zhaoyi Yin Jianbei Qiu Qi W ang Zhengwen Yang Zhiguo Song | 2020 | Journal of Rare Earths2020,38,6: | 2

Path Analysis of Agricultural Project Design Based on Function Expansion: A Case Study of Naya Mountain Villa显示文摘The expansion of agricultural function is one of the important means for modern agricultural projects to promote agricultural economic benefits. Successful modern agricultural projects require good creative ideas and design programs. While developing high-efficient modern agricultural production activities,we should fully explore the intangible value of agricultural production activities, combine agriculture with agricultural products, natural conditions, cultural conception and other effective resources,to expand agricultural functions, and promote comprehensive benefits. In order to build a sustainable modern agricultural project operation system,Naya Mountain Villa project planning is taken as an example for analysis. Naya Mountain Villa began construction in 2011; the creative planning based on the agricultural expansion function was carried out in 2013; it had successful access to the capital market in 2015. The project realizes the effective integration of agricultural production system and agricultural function expansion, constructs a set of long-term stable profiting models, and lays an important foundation for entering the capital market. The project is a representative example of the function-expanding modern agricultural project. Through the analysis of the design ideas of the project, this paper discusses the function expansion elements of basic resources, public welfare and agricultural function expansion methods, the formation of general ideas, source and construction logic of creative thinking, and summarizes and abstracts some inspiring design methods of agricultural function expansion. Through the analysis of the key points in the design of the specific technical aspects of the project, this paper provides a reference for solving common difficult problems in the practical design. The summary and refinement of the thinking logic, thinking construction and specific design method of the project is inspiring and repeatable to some extent, which can provide reference for the relevant researchers. | Yuehong SUN Yongsheng WU Rui CHEN Xue YANG | 2018 | Asian Agricultural Research2018,10,2: | 2
Broadband terahertz rotator with an all-dielectric metasurface显示文摘Polarization manipulation is essential in developing cutting-edge photonic devices ranging from optical communication displays to solar energy harvesting. Most previous works for efficient polarization control cannot avoid utilizing metallic components that inevitably suffer from large ohmic loss and thus low operational efficiency.Replacing metallic components with Mie resonance-based dielectric resonators will largely suppress the ohmic loss toward high-efficiency metamaterial devices. Here, we propose an efficient approach for broadband, highquality polarization rotation operating in transmission mode with all-dielectric metamaterials in the terahertz regime. By separating the orthogonal polarization components in space, we obtain rotated output waves with a conversion efficiency of 67.5%. The proposed polarization manipulation strategy shows impressive robustness and flexibility in designing metadevices of both linear-and circular-polarization incidences. | QUANLONG YANG XIEYU CHEN QUAN XU CHUNXIU TIAN YUEHONG XU LONGQING CONG XUEQIAN ZHANG YANFENG LI CAIHONG ZHANG XIXIANG ZHANG JIAGUANG HAN WEILI ZHANG | 2018 | Photonics Research2018,,11: | 2
Discovery of potential biomarkers for human atherosclerotic abdominal aortic aneurysm through untargeted metabolomics and transcriptomics显示文摘Abdominal aortic aneurysm(AAA)and atherosclerosis(AS)have considerable similarities in clinical risk factors and molecular pathogenesis. The aim of our study was to investigate the differences between AAA and AS from the perspective of metabolomics, and to explore the potential mechanisms of differential metabolites via integration analysis with transcriptomics.Plasma samples from 32 AAA and 32 AS patients were applied to characterize the metabolite profiles using untargeted liquid chromatography-mass spectrometry(LC-MS).A total of 18 remarkably different metabolites were identified, and a combination of seven metabolites could potentially serve as a biomarker to distinguish AAA and AS,with an area under the curve(AUC)of0.93.Subsequently,we analyzed both the metabolomics and transcriptomics data and found that seven metabolites, especially 2’-deoxy-D-ribose(2 d DR),were significantly correlated with differentially expressed genes. In conclusion, our study presents a comprehensive landscape of plasma metabolites in AAA and AS patients, and provides a research direction for pathogenetic mechanisms in atherosclerotic AAA. | Lei JI Siliang CHEN Guangchao GU Wei WANG Jinrui REN Fang XU Fangda LI Jianqiang WU Dan YANG Yuehong ZHENG | 2021 | Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2021,22,9: | 1
NudCL2 is an autophagy receptor that mediates selective autophagic degradation of CP110 at mother centrioles to promote ciliogenesis显示文摘Primary cilia extending from mother centrioles are essential for vertebrate development and homeostasis maintenance.Centriolar coiled-coil protein 110(CP110)has been reported to suppress ciliogenesis Initiation by capping the distal ends of mother centrioles. However, the mechanism underlying the specific degradation of mother centriole-capping CP110 to promote cilia initiation remains unknown. Here,we find that autophagy is crucial for CP110 degradation at mother centrioles after serum starvation in MEF cells. We further identify NudC-like protein 2(NudCL2)as a novel selective autophagy receptor at mother centrioles, which contains an LC3-interacting region(UR)motif mediating the association of CP110 and the autophagosome marker LC3.Knockout of NudCL2 induces defects in the removal of CP110 from mother centrioles and ciliogenesis, which are rescued by wild-type NudCL2 but not its UR motif mutant.Knockdown of CP110 significantly attenuates ciliogenesis defects in NudCL2-deficient cells. In addition,NudCL2 morphants exhibit ciliation-related phenotypes in zebrafish, which are reversed by wild-type NudCL2,but not its UR motif mutant.Importantly,CP110 depletion significantly reverses these ciliary phenotypes in NudCL2 morphants.Taken together, our data suggest that NudCL2 functions as an autophagy receptor mediating the selective degradation of mother centriole-capping CP110 to promote ciliogenesis, which is indispensable for embryo development in vertebrates. | Min Liu Wen Zhang Min Li Jiaxing Feng Wenjun Kuang Xiying Chen Feng Yang Qiang Sun Zhangqi Xu Jianfeng Hua Chunxia Yang Wei Liu Qiang Shu Yuehong Yang Tianhua Zhou Shanshan Xie | 2021 | Cell Research2021,31,11: | 1
Vp of muscovite-biotite gneiss up to 950℃ at 400 MPa: Constraints on the origin of abnormal seismic layers in continental crust显示文摘Here we present experimental results of compressional wave velocity (Vp) of muscovite-biotite gneiss from Higher Himalayan Crystallines (HHC) at the temperature up to 950℃ and the pressure of 0.1―400 MPa. At 400 MPa, when the temperature is lower than 600℃, Vp decreases linearly with increasing temperature at the rate of (Vp/T)p -4.43×10-4 km/s ℃. In the temperature range of 600―800℃, Vp drops significantly and the signal is degraded gradually due to the dehydration of muscovite and α-quartz softening. When the temperature rises from 800℃ to 875℃, Vp increases and the signals become clear again as a result of the temperature going through the β-quartz range. The experiments indicate that the duration has great influence on the experimental results when temperature is above the dehydration point of biotite. During the first 30 h at 950℃, the Vp decreases substantially from 5.9 to 5.4 km/s and the signal amplitude is attenuated by more than 80%. After the 30-h transition, the Vp and the amplitude of ultrasonic wave signals become steady. The decrease of Vp and attenuation of the signals at 950℃ are associated with the breakdown reactions of biotite. The experiments suggest that the breakdown of muscovite and/or quartz softening can contribute to the low seismic wave velocity in thickened quartz-rich felsic-crust such as what is beneath southern Tibet. Additionally, α-β quartz transition generates a measurable high seismic velocity zone, which provides a possibility of precisely constraining the temperature in the upper-middle continental crust. Our study also demonstrates that duration is a key factor to obtain credible experimental results. | YANG XiaoSong ZHOU Ping MING YueHong | 2007 | Chinese Science Bulletin2007,52,24: | 1

Impact of preoperative therapy on surgical outcomes of laparoscopic total gastrectomy for gastric/gastroesophageal junction cancer显示文摘Objective: As laparoscopic surgery is widely applied for primarily treated gastric cancer(GC)/gastroesophageal junction cancer(GEJC) and gains many advantages, the feasibility of laparoscopic total gastrectomy(LTG) for GC/GEJC patients who have received preoperative therapy(PT) has come to the fore. This study aims to analyze the safety and feasibility of LTG after PT for GC/GEJC patients. Methods: We retrospectively analyzed the data of 511 patients with GC/GEJC undergoing LTG, of which 405received LTG(LTG group) and 106 received PT+LTG(PT-LTG group) at Nanfang Hospital between June 2018and September 2022. The surgical outcomes were compared between the two groups. Results: The surgical duration was significantly longer in the PT-LTG group(P<0.001), while the incidence of intraoperative complications(P=1.000), postoperative complications(LTG group vs. PT-LTG group: 26.2% vs.23.6%, P=0.587), the classification of complication severity(P=0.271), and postoperative recovery was similar between two groups. Notably, the incidence of anastomotic complications of esophagojejunostomy was also comparable between the two groups(LTG group vs. PT-LTG group: 5.9% vs. 5.7%, P=0.918). The univariate and multivariate analysis confirmed that positive proximal margin [positive vs. negative: odds ratio(OR)=14.094, 95%confidence interval(95% CI): 2.639-75.260, P=0.002], rather than PT, has an impact on anastomotic complications after LTG(OR=0.945, 95% CI: 0.371-2.408, P=0.905).Conclusions: PT did not increase the surgical risk of LTG for GC/GEJC. Therefore, considering the positive effect of PT on long-term survival, the broader application of PT and LTG for GC/GEJC is supported by our findings. | Yuehong Chen Zhijing Yang Mingli Zhao Chuanjin Xu Yuxuan Zhu Huimin Zhang Huilin Huang Yanmei Peng Yanfeng Hu Tian Lin Tao Chen Hao Chen Liying Zhao Hao Liu Guoxin Li Jiang Yu Xinhua Chen | 2023 | Chinese Journal of Cancer Research2023,35,4: | 0
Effect of demethyltransferase FTO on tumor progression显示文摘N6-methyladenosine(m6A)modification is the most widespread and conserved internal mRNA modification in mammalian cells. It greatly affects genetic regulation by enhancing the involvement of diverse cellular enzymes and thus, plays a significant role in basic life processes.Numerous studies on m6A modification identified FTO as a crucial demethylase that participates in various biological processes. Not only does FTO play a pivotal role in obesity-related conditions, but it also influences the occurrence, development, and prognosis of several cancers, such as acute myeloid leukemia, breast cancer, liver cancer, and lung cancer. Moreover,FTO also shows a close association with immunity and viral infections. This article summarized the molecular mechanism of FTO in tumorigenesis and tumor progression. | LING SHENG YUEHONG SHEN HONGYU YANG | 2022 | BIOCELL2022,46,11: | 0

Zoning of rural water conservation in China:A case study at Ashihe River Basin显示文摘With the effective control of point source(PS)pollution accomplished, water pollution problems caused by non-point source(NPS)pollution have increased in recent years. The worsening agricultural NPS pollution has drawn the attention of the Chinese Government and researcher scientists and has resulted in the often mentioned“three red lines”on water resources management. One of the red lines is to control water pollution within a rational range. The Agricultural NPS pollution, which includes pollution from housing, and from livestock and crop production,is the main source.Based on the NPS pollution statutes,an index system for integrated evaluation of water quality, and a zoning scheme for rural water conservation were established.Using the method of one-dimensional Euclidean distance, this country is divided into 9 sub-zones at the provincial level, which are the first level zones. The zoning themes include natural resources,socio-economic development, water use efficiency, and pollutants emission intensity.According to pollution types of livestock, agriculture,or both, the first level zones are divided into 25 second level zones. The third class zoning is divided also based on pollution intensity of total nitrogen(TN),total phosphorus(TP),ammonia nitrogen(NH3-N),chemical oxygen demand(COD),and biochemical oxygen demand(BOD).On the basis of the second level zoning, there were formed 70 rural water conservation third level zones. This case study in the Ashihe river watershed indicated that the main pollution sources are consistent with the zoning research result, and this zoning has shown a good way to guide the agricultural NPS pollution control in not only the wide rural area of China but also other parts of the world.&2015 International Research and Training Center on Erosion and Sedimentation and China Water and Power Press.Production and Hosting by Elsevier B.V.This is an open access article under the CC BY-NC-ND license(http://creativecommons.org/licenses/by-nc-nd/4.0/). | Xiaoying Liu Sha Zhou Shi Qi Bo Yang Yuehong Chen Ruan Huang Pengfei Du | 2015 | International Soil and Water Conservation Research2015,3,2: | 0
Effects of Freeze-Thaw Cycles on Physical and Mechanical Properties of Glulam Exposed to Outdoor Environment显示文摘This paper presents an experimental investigation to identify suitable indices to assess durability of glulam when subjected to freeze-thaw cycles in an exposed enviroenment. In this study, two types of glulam specimens were tested for their performance when subjected to different levels of aging due to freezing and thawing.Effect of aging treatment on various parameters including thickness swelling rate, static bending strength, elastic modulus, shear strength, and peeling rate of adhesive layer were studied.Obtained results showed that freeze-thaw aging treatment did not affect the water-resistance of the specimens as measured by thickness swelling rate and had little effect on the dimensional stability of the material. However, the applied aging treatment weakened the bending resistance of the glulam specimens with more pronounced effects on on low-density wood.On the other hand, bond strength of high-density wood was relatively more affected due to the appliedfreeze-thaw cycles. For highdensity wood,it is suggested that the shear strength of the adhesive layer be taken as an important index to determine the durability of freeze-thaw cycles aging. For low-density wood,on the other hand, the static bending strength can be used as an index to determine the durability of glulam under freeze-thaw cycles aging. | Ruyuan Yang Haitao Li Assima Dauletbek Mahmud Ashraf Rodolfo Lorenzo Youfu Sun Yuehong Wu | 2021 | Journal of Renewable Materials2021,9,7: | 0
