1Genome-wide association study of vitamin E in sweet corn kernels显示文摘Vitamin E,consisting of tocopherols and tocotrienols,serves as a lipid-soluble antioxidant in sweet corn kernels,providing nutrients to both plants and humans.Though the key genes involved in the vitamin E biosynthesis pathway have been identified in plants,the genetic architecture of vitamin E content in sweet corn kernels remains largely unclear.In the present study,an association panel of 204 inbred lines of sweet corn was constructed.Seven compounds of vitamin E were quantified in sweet corn kernels at 28 days after pollination.A total of 119 loci for vitamin E were identified using a genome-wide association study based on genotyping by sequencing,and a genetic network of vitamin E was constructed.Candidate genes identified were involved mainly in RNA regulation and protein metabolism.The known gene ZmVTE4,encodingγ-tocopherol methyltransferase,was significantly associated with four traits(α-tocopherol,α-tocotrienol,theα/γ-tocopherol ratio,and theα/γ-tocotrienol ratio).The effects of two causative markers on ZmVTE4 were validated by haplotype analysis.Finally,two elite cultivars(Yuetian 9 and Yuetian 22)with a 4.5-fold increase in the sum ofα-andγ-tocopherols were developed by marker-assisted selection,demonstrating the successful biofortification of sweet corn.Three genes(DAHPS,ADT2,and cmu2)involved in chorismate and tyrosine synthesis were significantly associated with theα/γ-tocotrienol ratio.These results shed light on the genetic architecture of vitamin E and may accelerate the nutritional improvement of sweet corn.Yingni Xiao Yongtao Yu Gaoke Li Lihua Xie Xinbo Guo Jiansheng Li Yuliang Li Jianguang Hu 2020The Crop Journal2020,8,2:7
2Crust and upper mantle structures beneath Northeast China from receiver function studies显示文摘P-wave and S-wave receiver function analyses have been performed along a profile consisted of 27broadband seismic stations to image the crustal and upper mantle discontinuities across Northeast China.The results show that the average Moho depth varies from about37 km beneath the Daxing'anling orogenic belt in the west to about 33 km beneath the Songliao Basin,and to about35 km beneath the Changbai mountain region in the east.Our results reveal that the Moho is generally flat beneath the Daxing'anling region and a remarkable Moho offset(about 4 km) exists beneath the basin-mountain boundary,the Daxing'anling-Taihang Gravity Line.Beneath the Tanlu faults zone,which seperates the Songliao Basin and Changbai region,the Moho is uplift and the crustal thickness changes rapidly.We interpret this feature as that the Tanlu faults might deeply penetrate into the upper mantle,and facilitate the mantle upwelling along the faults during the Cenozoic era.The average depth of the lithosphereasthenosphere boundary(LAB) is *80 km along the profile which is thinner than an average thickness of a continental lithosphere.The LAB shows an arc-like shape in the basin,with the shallowest part approximately beneath the center of the basin.The uplift LAB beneath the basin might be related to the extensive lithospheric stretching in theMesozoic.In the mantle transition zone,a structurally complicated 660 km discontinuity with a maximum 35 km depression beneath the Changbai region is observed.The35 km depression is roughly coincident with the location of the stagnant western pacific slab on top of the 660 km discontinuity revealed by the recent P wave tomography.Zhen Guo Yuliang Cao Xianguang Wang Y.John Chen Jieyuan Ning Weiguang He Youcai Tang Yongge Feng 2014Earthquake Science2014,27,3:6
3Autonomous live working robot navigation with real-time detection and motion planning system on distribution line显示文摘In this study,an autonomous robot navigation system is designed for live working on distribution line.The developed system features a real-time detection and motion planning system,incorporating a manipulator capable of grasping power components.In order to accurately identify targets,the authors propose an object detection method based on the Larger Scale‘You Only Look Once’Version 4(LS-YOLOv4)algorithm for detecting the insulators and drop fuses.The LS-YOLOv4 extracts features of power components by Convolutional Neural Network(CNN),and then performs feature fusion.Then the authors develop a motion planning method based on the Node Control Optimal Rapidly Exploring Random Trees(NC-RRT*),which can drive the robot to realise the autonomous robot motion planning and obstacle avoidance.On the grasping function,the authors present a reliable Lightweight-based Convolutional Neural Network(L-CNN)grasping point detection method.Finally,the authors evaluate fully autonomous robotic system in both simulated and real-world experiments.The experimental results demonstrate that the proposed system can effectively identify the target and complete the grasping task in an efficient way.Notably,the proposed motion planning method can take into account both planning efficiency and accuracy to manipulation tasks.Haoning Zhao Chaoqun Wang Rui Guo Xuewen Rong Jiamin Guo Qixun Yang Lecheng Yang Yuliang Zhao Yibin Li 2022High Voltage2022,7,6:2
4Anti-fatigue activity of Brassica rapa L.extract and correlation among biochemical changes in forced swimming mice显示文摘Brassica rapa L.is an edible and medical plant in China used for treating various diseases,especially in the Tibet region.In this study,the chemical constituents of Brassica rapa L.extract(BE)was clarified by UHPLC-MS and anti-fatigue ability of BE was assessed through the behavior test and weight-loaded forced swimming test in mice.Then we determined several biochemical parameters related to fatigue to explore possible reasons.Results indicate that BE could significantly enhance exercise performance in mice,and there was a significant improvement in both grip-strength and the time on rota-rods.Meanwhile,BE showed significant anti-fatigue effects by decreasing the blood lactic acid(BLA),blood urea nitrogen(BUN),and creatine kinase(CK)levels and increasing the lactate dehydrogenase(LDH)and malondialdehyde(MDA)levels.Increases in superoxide dismutase(SOD)and glutathione peroxidase(GSH-Px)were observed and reactive oxygen species(ROS)levels was regulated.Liver and muscle glycogen levels and adenosine triphosphate(ATP)levels were also significantly increased.Pearson correlation analysis indicates that fatigue-related indexes including metabolites and energy-related indexes were significantly correlated with antioxidant levels.In conclusion,BE possesses remarkable anti-fatigue activity,thereby shows great potential for improving exercise performance and alleviating physical fatigue and can be considered as a functional food supplement for further development.Zilin Li Hongkang Zhu Hanyi Hua Chang Liu Yuliang Cheng Yahui Guo Peng Du He Qian 2022Food Bioscience2022,47,3:2
5Nerve growth factor precursor and sortilin effects on perihematomal brain tissue and the relationship to secondary cell apoptosis显示文摘BACKGROUND: Neuronal apoptosis in perihematomal brain tissues following intracerebral hemorrhage is strongly related to the formation of a compound signal pathway between nerve growth factor precursor (proNGF), p75NTR, and sortilin receptor. Sortilin acts as a co-receptor and molecular switch governing the p75NTR-mediated pro-apoptotic signal induced by proNGF. OBJECTIVE: To investigate proNGF and sortilin expressions in perihematomal brain tissues following intracerebral hemorrhage, and to study the effects of proNGF and sortilin on secondary cell apoptosis. DESIGN, TIME AND SETTING: A paired, comparison study was performed at the Laboratory of Histology and Embryology, Xi'an Jiaotong University from October 2007 to September 2008. MATERIALS: Brain tissue samples were obtained from 15 patients with intracerebral hemorrhage, who were treated at the Department of Neurosurgery, First Affiliated Hospital, Medical College of Xi'an Jiaotong University from October 2007 to March 2008. Rabbit anti-proNGF polyclonal antibody was provided by Chemicon, USA; rabbit antisortilin polyclonal antibody by Abcam, UK; and TUNEL kit by Promega, USA. METHODS: Perihematomal brain tissues selected 0.5 cm from the hemorrhage area were considered to be the hemorrhage group, while brain tissues from the middle temporal gyrus served as the control group. MAIN OUTCOME MEASURES: Histopathological changes were detected using hematoxylin-eosin staining, cell apoptosis was determined using the TUNEL method, and proNGF and sortilin ex- pressions were determined using immunohistochemistry. RESULTS: Edema was clearly observed in perihematomal brain tissues, and infiltration of inflammatory cells was visible, with the presence of irregular and necrotic bodies. The apoptotic rate in the hemorrhage group was significantly greater than in the control group (P < 0.01). Moreover, sortilin expression significantly increased (P < 0.01), but proNGF expression remained unchanged (P > 0.05). Correlation analysis suggested that sortilin expression positively correlated with apoptosis (rs = 0.648, P < 0.01). 8The machine protection system for CSNS显示文摘Background The China Spallation Neutron Source(CSNS)accelerator consists of an 80 MeV H-LINAC,a 1.6 GeV rapid cycling synchrotron(RCS)and two beam transport lines.The uncontrolled beam may permanently damage the components or lead to very high residual radiation dose along the beam line.So the equipment protection must be deliberately designed and implemented.Purpose The machine protection system(MPS)protects components from being damaged by the beam.The response time requirement for the CSNS MPS is less than 20 ms,so the PLC(programmable logic controller)was adopted to implement the interlock logic.Methods The MPS was implemented as a two-tier architecture system,and developed through utilizing PLC and Experimental Physics and Industrial Control System(EPICS)software toolkits.The application logic was taken into careful consideration during the implementation stage.An embedded CPU module can function as an IOC accessing PLC I/O modules through the sequence CPU,with an embedded Linux operation system.Results The interlock logic and heartbeat functions were tested with all functions ok.Time consumption has been measured thoroughly since the important requirement,which is around 15 ms to stop the beam.Conclusions MPS was completed in Sep.2017 and then put into operation.It has been operating smoothly for more than 3 years.MPS has played an important role in every stage of CSNS’s commissioning and operation and achieved high reliability during the user’s experiment operation.The accelerator recently runs stably with low equipment failure.Kang Mingtao Zhang Yuliang Jin Dapeng He Yongcheng Zhan Mingchuan Zhu Peng Wu Xuan Guo Fengqin Wang Lin 2021Radiation Detection Technology and Methods2021,5,2:1
9Role of the nerve growth factor precursor-neurotrophin receptor p75 and sortilin pathway on apoptosis in the brain of patients with intracerebral hemorrhage显示文摘This study demonstrated that brain areas surrounding the site of hematoma following intracerebral hemorrhage are characterized by significantly increased apoptosis and expression of neurotrophin receptor p75 and sortilin. However, as detected by terminal deoxynucleotidyl transferase dUTP nick end labeling and immunohistochemical staining, there was no significant change in nerve growth factor precursor expression levels. The appearance of neurotrophin receptor p75 expressing cells was positively correlated with cells that were detected by terminal deoxynucleotidyl transferase dUTP nick end labeling. 12Trans-cinnamaldehyde inhibits Penicillium italicum by damaging mitochondria and inducing apoptosis mechanisms显示文摘Plant-derived essential oils have excellent antifungal effects and can be used for the preservation of fresh foods such as fruits and vegetables, but the detailed mechanism has not been fully elucidated. In this study, we investigated the inhibitory effects of trans-cinnamaldehyde on Penicillium italicum, a common pollution fungus in citrus, and explored the antifungal mechanism of trans-cinnamaldehyde by detecting fungal oxidative damage, mitochondrial metabolism, and cell apoptosis. These results showed that transcinnamaldehyde made the carboxylic acid cycle deregulated by altering the related enzyme activities(succinate dehydrogenase, malate dehydrogenase) and mid product. Moreover, the level of reactive oxygen species rose sharply while the redox level was out of regulation. The mitochondrial membrane potential collapsed, leading to the leakage of cytochrome c, and then triggering the activation of apoptotic protease, which was further confirmed by the significant increase in caspase-3 activity from(3.6 ± 0.6) U to(8.8 ± 1.1) U(P < 0.05). The cytochrome c in mitochondria was detected by confocal Raman microspectroscopy, the characteristic intensity index(I750/I2944) was decreased, indicating that the cytochrome c in mitochondria was reduced and leakage. Besides, the strong negative correlation between Raman intensity and the amount of cytochrome c leakage was established with the correlation coefficient of-0.981 7. 13Supplementation with yak (Bos grunniens) bone collagen hydrolysate altered the structure of gut microbiota and elevated short-chain fatty acid production in mice显示文摘In this study, yak bone collagen hydrolysate(YBCH)was produced by mixed proteases and provided to standard-diet mice at a different dose(low dose(LD), medium dose(MD), and high dose(HD))to investigate its effects on the composition of gut microbiota and short-chain fatty acids(SCFA)production. It was found that YBCH was mainly composed of small molecular peptides whose molecular weight below 2 000 Da. Notably, supplementation with different doses of YBCH could significantly downregulate the ratio of Firmicutes to Bacteroidetes in the fecal microbiota. At the family level, the Lachnospiraceae abundance was significantly reduced in the YBCH gavage groups(mean reduction ratio 41.7 %, 35.2%, and 36.4% for LD, MD, and HD group, respectively). The predicted functions of gut microbes in the MD group were significantly increased at “lipid metabolism” and “glycan biosynthesis and metabolism”. Moreover, the SCFA production in the YBCH groups was elevated. Especially, the concentration of acetic acid, propionic acid, and butyric acid in the MD group was separately increased 79.7%, 89.2%, and 78.8% than that in the NC group. 15Shale Gas Reservoir Evaluation by Geophysical Measurements:A Case Study of the Upper Ordovician–Lower Silurian in the Fenggang Block,Northern Guizhou Province显示文摘With the aid of geophysical measurements,including seventeen two-dimensional(2 D)seismic lines and the well logging curves of well FGY1,the structure and reservoir characteristics of the Upper Ordovician–Lower Silurian strata in the Fenggang block,northern Guizhou Province,were analyzed thoroughly to identify desert areas and favorable intervals.The results show that Longmaxi-Wufeng is the most prospect-rich formation,consisting of a thick succession of overmature black shale,this formation remaining partially in the Suiyang,Fenggang and Jianchaxi synclines.The Longmaxi-Wufeng shale,especially the lower member,was deposited in a reducing low-energy environment with relatively high U content and a low Th/U value.In this shale,the organic matter type(sapropelic and humic-sapropelic),total organic carbon(TOC)content,gas content,gas adsorption capacity,vitrinite reflectance and brittle mineral content are profitable for shale gas preservation and development.The fractures of this shale were closed because of its high overburden pressure.The gas adsorption capacity of this shale increases with increasing TOC content and Ro.In the Longmaxi-Wufeng Formation at well FGY1,the most favorable intervals are in the depth ranges of 2312.4–2325.1 m and 2325.8–2331.1 m.XIA Peng FU Yong GUO Chuan YANG Zhen HUANG Jinqiang MOU Yuliang 2021Acta Geologica Sinica(English Edition)2021,95,4:0
16Specific detection and effective inhibition of a single bacterial species in situ using peptide mineralized Au cluster probes显示文摘Increasingly serious microbial infections call for the development of new simpler methods for the precise diagnosis and specific inhibition of such pathogens. In this work, a peptide mineralized Au cluster probe was applied as a new simplified strategy to both recognize and inhibit a single bacteria species of Staphylococcus aureus(S. aureus) simultaneously. The probes are composed of peptides and Au clusters. Moreover, the peptides specifically target S. aureus cells and the Au clusters provide fluorescent imaging and have an antibacterial effect. 17Intratree Variation in Viscoelastic Properties of Cell Walls of Masson Pine (Pinus Massoniana Lamb)显示文摘In this study,Pinus massoniana Lamb at different heights,across the annual rings,and between earlywood and latewood was measured by X-ray diffraction and the chemical composition was analyzed by chemical treatment.Results indicated that the microfibril angle(MFA)decreased and the chemical composition changed little with the increase in height from 1 m to 9 m.In the radial direction,the MFA decreased and the chemical composition changed little with an increase in annual rings.The cellulose content of latewood was higher than that of early-wood.The viscoelastic changes of wood cell walls at different heights,across the annual rings by the method of quasi-static nanoindentation and dynamic modulus mapping techniques.Results indicated that the wood cell walls’elastic modulus increased,and the creep rate and creep compliance decreased with the increase in height;The elastic modulus and hardness increased with the annual rings.The cell walls’storage modulus increased and the loss modulus gradually decreased with an increase in height;the storage modulus gradually increased and the loss modulus decreased with the annual rings.Shaoxiang Cai Yuliang Guo Yanjun Li 2022Journal of Renewable Materials2022,10,1:0
18Spontaneous imbibition characteristics of shale oil reservoir under the influence of osmosis显示文摘The spontaneous imbibition(SI)process in shale oil reservoirs is not only infuenced by capillary force,but also by the osmotic pressure between the fracturing fuid and formation water in the nanopores media.In this study,experimental methods are used to investigate the mechanisms of osmosis in the SI,taking into account the presence of initial formation water in shale oil reservoirs.To investigate the efect of osmosis,SI experiments were performed on the fne-grained felsic shale of the Qikou sag of Dagang oilfeld.Low-feld NMR testers and high-precision electronic balances are utilized for the measuring of oil–water migration.The results show that,when Sw≠0,high-salinity fuid SI can be divided into four stages:initial imbibition stage,drainage stage,secondary imbibition stage and stationary stage;when Sw=0,there is no drainage stage of high-salinity fuid SI;when Sw≠0 or Sw=0,low-salinity fuid SI can be called the“osmosis-enhanced SI”;and we have found that“newly formed pores or microfractures”as well as reducing salinity can promote SI.This article presents a systematic study of SI of shale oil reservoirs under the infuence of osmosis,which provide useful information for reservoir numerical simulation and development program design.Yuliang Su Qinghao Sun Wendong Wang Xincheng Guo Jilong Xu Guanqun Li Xiugang Pu Wenzhong Han Zhannan Shi 2022International Journal of Coal Science & Technology2022,9,5:0
19Romidepsin(FK228) improves the survival of allogeneic skin grafts through downregulating the production of donor-specific antibody via suppressing the IRE1α-XBP1 pathway显示文摘Antibody-mediated rejection(AMR)is one of the major causes of graft loss after transplantation.Recently,the regulation of B cell differentiation and the prevention of donor-specific antibody(DSA)production have gained increased attention in transplant research.Herein,we established a secondary allogeneic in vivo skin transplant model to study the effects of romidepsin(FK228)on DSA.The survival of grafted skins was monitored daily.The serum levels of DSA and the number of relevant immunocytes in the recipient spleens were evaluated by flow cytometry.Then,we isolated and purified B cells from B6mouse spleens in vitro by magnetic bead sorting.The B cells were cultured with interleukin-4(IL-4)and anti-clusters of differentiation 40(CD40)antibody with or without FK228 treatment.The immunoglobulin G1(IgG1)and IgM levels in the supernatant were evaluated by enzyme-linked immunosorbent assay(ELISA).Quantitative reverse transcription-polymerase chain reaction(RT-q PCR)and western blotting were conducted to determine the corresponding levels of messenger RNA(mRNA)and protein expression in cultured cells and the recipient spleens.The results showed that FK228 significantly improved the survival of allogeneic skin grafts.Moreover,FK228 inhibited DSA production in the serum along with the suppression of histone deacetylase 1(HADC1)and HDAC2 and the upregulation of the acetylation of histones H2A and H3.It also inhibited the differentiation of B cells to plasma cells,decreased the transcription of positive regulatory domain-containing 1(Prdm1)and X-box-binding protein 1(Xbp1),and decreased the expression of phosphorylated inositol-requiring enzyme 1α(p-IRE1α),XBP1,and B lymphocyte-induced maturation protein-1(Blimp-1).In conclusion,FK228 could decrease the production of antibodies by B cells via inhibition of the IRE1α-XBP1 signaling pathway.Thus,FK228 is considered as a promising therapeutic agent for the clinical treatment of AMR.Yuliang GUO Siyu SONG Xiaoxiao DU Li TIAN Man ZHANG Hongmin ZHOU Zhonghua Klaus CHEN Sheng CHANG 2022Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2022,23,5:0
20Effects of selective cyclooxygenase-2 inhibitor on C6 glioma cell proliferation and apoptosis显示文摘BACKGROUND:Studies have shown that cyclooxygenase-2 is associated with proliferation and apoptosis of glioma cells. OBJECTIVE:To investigate the effects of selective cyclooxygenase-2 inhibitor celecoxib on proliferation and apoptosis of C6 glioma cells in vitro. DESIGN,TIME AND SETTING:A cellular,molecular,controlled study was performed at the Central Laboratory and Room of Electron Microscope,Medical School,Xi'an Jiaotong University,China from March 2007 to March 2008. MATERIALS:C6 glioma cells during in vitro log phase were assigned to control and experimental groups.Celecoxib(Pfizer,USA),dimethyl sulfoxide(Sigma,USA),and MTT(Sigma,USA) were used for this study. METHODS:The control group was subdivided into blank control and dimethyl sulfoxide control groups.C6 glioma cells in the blank control and dimethyl sulfoxide control groups were incubated in Dulbecco's modified Eagle's medium supplemented with 10%calf serum and 0.3%dimethyl sulfoxide respectively.C6 glioma cells in the experimental group were separately treated with 60,80, and 100μmol/L celecoxib. MAIN OUTCOME MEASURES:Activity of C6 glioma cells was examined by MTT assay.C6 glioma cell cycle and apoptosis were determined by annexin V-fluorescein isothiocyanate/propidium iodide double-staining,followed by flow cytometry.Morphology and ultrastructure of C6 glioma cells were observed with an inverted microscope and a transmission electron microscope,respectively. RESULTS:Compared with the blank control group,cell density was reduced,adherence ability weakened,and irregular nuclei were visible,with the presence of chromatin condensation, margination,and some apoptotic bodies in the experimental group.Activity of C6 glioma cells was significantly decreased(P<0.05),cell number was reduced during S phase,cell number was significantly increased during G_2/M phase(P<0.01),and the apoptotic rate was significantly increased(P<0.05) in the experimental group.These results were displayed in a dose- and time-dependent fashion.The outcomes were obvious in the 100μmol/L celecoxib group following 72 hours of treatment.
