| 1 | Neuro-protective effects of CNTF on hippocampal neurons via an unknown signal transduction pathway显示文摘In our previous study, we proposed that there may be an unknown pathway in the upper stream of the known signal transduction pathway of Ciliary neurotrophic factor (CNTF) that mediates the neuro-protective function of CNTF. In the present experiment, we observed that the neuro-protective function of the non-classic signal transduction path- way in a L-NMDA (a glutamic acid ion type receptor atagonist) induced hippocampal neuron injury model, using primary culture rat hippocampal neurons, con- tinuous photography and gp130 immunohistochemi- cal assay. The results showed that L-NMDA induced injurious reaction of hippocampal neurons, and CNTF was able to inhibit the toxic action of L-NMDA on hippocampal neurons. Additionally, when JAK/STATs in the known classic signal transduction pathway of CNTF were blocked by PTPi-2, the protective effect of CNTF against L-NMDA injury still existed. L-NMDA caused a rapid increase in the concentration of hip- pocampal intracellular free [Ca2+]i. | 2 | Utilization and quality assessment of digestive endoscopy in China:results from 5-year consecutive nationwide surveys显示文摘Background:Worldwide,the volume and availability of digestive endoscopy have undergone dramatic development in recent years,with increasing attention on quality assurance.We investigated the utilization and quality of digestive endoscopy in China from 2015 to 2019 and developed a quantitative quality evaluation tool for medical institutions.Methods:We invited all tertiary/secondary hospitals in Chinese mainland to participate in the survey annually.The questionnaires included the personnel,annual volume,and quality indicators of endoscopy.An endoscopy quality index(EQI)was developed based on recorded quality indicators using principal component analysis to determine the relative weight.Results:From 2015 to 2019,806,1412,2644,2468,and 2541 hospitals were respectively enrolled in this study.The average annual volume of endoscopy increased from 12,445 to 16,206(1.30-fold)and from 2938 to 4255(1.45-fold)in tertiary and secondary hospitals,respectively.The most obvious growth was observed in diagnostic colonoscopy(1.44-fold for all hospitals after standardization).The proportion of early cancer among all esophageal and gastric cancers during diagnostic esophagogastroduodenoscopy increased from 12.3%(55,210/448,861)to 17.7%(85,429/482,647)and from 11.4%(69,411/608,866)to 16.9%(107,192/634,235),respectively.The adenoma detection rate of diagnostic colonoscopy increased from 14.9%(2,118,123/14,215,592)to 19.3%(3,943,203/20,431,104).The EQI model included 12 quality indicators,incorporating 64.9%(7.792/12)of the total variance into one comprehensive index.According to the EQI measurements,the quality of endoscopy was higher in tertiary hospitals and hospitals in developed areas with higher volume or more endoscopists than that in other hospitals.Conclusions:Digestive endoscopy in China has developed considerably in recent years in terms of both volume and quality.The EQI is a promising tool to quantify the quality of endoscopy at different hospitals. | Lei Xin Ye Gao Zhiyuan Cheng Tianjiao Wang Han Lin Yanan Pang Chang Sun Zengjun Fu Zhaoshen Li Xudong Ma Luowei Wang | 2022 | Chinese Medical Journal2022,135,16: | 3 |

| 16 | Effect and safety of anaprazole in the treatment of duodenal ulcers:a randomized,rabeprazole-controlled,phase Ⅲ non-inferiority study显示文摘Background:The pharmacokinetic and clinical behaviors of many proton pump inhibitors(PPIs)in peptic ulcer treatment are altered by CYP2C19 genetic polymorphisms.This non-inferiority study evaluated the efficacy and safety of the novel PPI anaprazole compared with rabeprazole.We also explored the influence of Helicobacter pylori(H.pylori)infection status and CYP2C19 polymorphism on anaprazole.Methods:In this multicenter,randomized,double-blind,double-dummy,positive-drug parallel-controlled,phase Ⅲ study,Chinese patients with duodenal ulcers were randomized 1:1 to receive rabeprazole 10 mg+anaprazole placebo or rabeprazole placebo+anaprazole 20 mg once daily for 4 weeks.The primary efficacy endpoint was the 4-week ulcer healing rate assessed by blinded independent review.Secondary endpoints were the proportion of patients with improved overall and individual duodenal ulcer symptoms at 4 weeks.Furthermore,exploratory subgroup analysis of the primary endpoint by H.pylori status and CYP2C19 polymorphism was conducted.Adverse events were monitored for safety.Non-inferiority analysis was conducted for the primary endpoint.Results:The study enrolled 448 patients(anaprazole,n=225;rabeprazole,n=223).The 4-week healing rates were 90.9%and 93.7%for anaprazole and rabeprazole,respectively(difference,-2.8%[95%confidence interval,-7.7%,2.2%]),demonstrating non-inferiority of anaprazole to rabeprazole.Overall duodenal ulcer symptoms improved in 90.9%and 92.5%of patients,respectively.Improvement rates of individual symptoms were similar between the groups.Healing rates did not significantly differ by H.pylori status or CYP2C19 genotype for either treatment group.The incidence of treatment-emergent adverse events was similar for anaprazole(72/220,32.7%)and rabeprazole(84/219,38.4%).Conclusions:The efficacy of anaprazole is non-inferior to that of rabeprazole in Chinese patients with duodenal ulcers.Registration:ClinicalTrials.gov,NCT04215653. | Huiyun Zhu Xue Pan Li Zhang Hongxin Sun Huizhen Fan Zhongwei Pan Caibin Huang Zhenwang Shi Jin Ding Qi Wang Yiqi Du Nonghua Lyu Zhaoshen Li | 2022 | Chinese Medical Journal2022,135,24: | 1 |

| 20 | Engineered tumor cell-derived vaccines against cancer: The art of combating poison with poison显示文摘Tumor vaccination is a promising approach for tumor immunotherapy because it presents high specificity and few side effects.However,tumor vaccines that contain only a single tumor antigen can allow immune system evasion by tumor variants.Tumor antigens are complex and heterogeneous,and identifying a single antigen that is uniformly expressed by tumor cells is challenging.Whole tumor cells can produce comprehensive antigens that trigger extensive tumor-specific immune responses.Therefore,tumor cells are an ideal source of antigens for tumor vaccines.A better understanding of tumor cell-derived vaccines and their characteristics,along with the development of new technologies for antigen delivery,can help improve vaccine design.In this review,we summarize the recent advances in tumor cell-derived vaccines in cancer immunotherapy and highlight the different types of engineered approaches,mechanisms,administration methods,and future perspectives.We discuss tumor cell-derived vaccines,including whole tumor cell components,extracellular vesicles,and cell membrane-encapsulated nanoparticles.Tumor cell-derived vaccines contain multiple tumor antigens and can induce extensive and potent tumor immune responses.However,they should be engineered to overcome limitations such as insufficient immunogenicity and weak targeting.The genetic and chemical engineering of tumor cell-derived vaccines can greatly enhance their targeting,intelligence,and functionality,thereby realizing stronger tumor immunotherapy effects.Further advances in materials science,biomedicine,and oncology can facilitate the clinical translation of tumor cell-derived vaccines. | Xinyi Zhang Hengqing Cui Wenjun Zhang Zhaoshen Li Jie Gao | 2023 | Bioactive Materials2023,,4: | 0 |
