1The landscape of gene-CDS-haplotype diversity in rice:Properties,population organization,footprints of domestication and breeding,and implications for genetic improvement显示文摘Polymorphisms within gene coding regions represent the most important part of the overall genetic diversity of rice.We characterized the gene-coding sequence-haplotype(gcHap)diversity of 45963 rice genes in 3010 rice accessions.With an average of 226±390 gcHaps per gene in rice populations,rice genes could be classified into three main categories:12865 conserved genes,10254 subspecific differentiating genes,and 22844 remaining genes.We found that 39218 rice genes carry>255179 major gcHaps of potential functional importance.Most(87.5%)of the detected gcHaps were specific to subspecies or populations.The inferred proto-ancestors of local landrace populations reconstructed from conserved predominant(ancient)gcHaps correlated strongly with wild rice accessions from the same geographic regions,supporting a multiorigin(domestication)model of Oryza sativa.Past breeding efforts generally increased the gcHap diversity of modern varieties and'caused significant frequency shifts in predominant gcHaps of 14266 genes due to independent selection in the two subspecies.Low frequencies of“favorable”gcHaps at most known genes related to rice yield in modern varieties suggest huge potential for rice improvement by mining and pyramiding of favorable gcHaps.The gcHap data were demonstrated to have greater power than SNPs for the detection of causal genes that affect complex traits.The rice gcHap diversity dataset generated in this study would facilitate rice basic research and improvement in the future.Fan Zhang Chunchao Wang Min Li Yanru Cui Yingyao Shi Zhichao Wu Zhiqiang Hu Wensheng Wang Jianlong Xu Zhikang Li 2021Molecular Plant2021,14,5:5
2A Teosinte-derived Allele of a MYB Transcription Repressor Confers Multiple Disease Resistance in Maize.显示文摘Natural alleles controlling multiple disease resistances (MDR) are valuable for crop breeding. However, only one MDR gene have been cloned in maize, and molecular mechanisms of MDR are not clear. By map-based cloning, we have cloned a teosinte-derived allele of a resistance gene, Mexicana lesion mimic 1 (ZmMM1), which has a lesion mimic phenotype and confers resistance to northern leaf blight (NLB), gray leaf spot (GLS) and southern corn rust (SCR). Strong MDR conferred by the teosinte allele is linked with the polymorphisms in the 3' untranslated region of the ZmMM1 gene that cause increased accumulation of ZmMM1 protein. ZmMM1 acts as a transcription repressor and negatively regulates transcription of specific target genes including ZmMM1-target gene 3 (ZmMT3), which functions as a negative regulator of plant immunity and associated cell death. 4Non‐linearly conductive ZnO microvaristors/epoxy resin composite prepared by wet winding with polyester fibre cloth显示文摘ZnO microvaristors/epoxy resin composite has drawn great attention from academia and industry for its adjustable non‐linear conductivity,high mechanical strength,and good ageing resistance.However,the sedimentation of ZnO microvaristors in epoxy resin during preparation is the key problem,which limits its application in engineering.In this study,a novel method of wet winding with polyester fibre cloth is proposed to prepare the ZnO microvaristors/epoxy resin composite.The anti‐settling effect of ZnO micro-varistors in the composite is verified by scanning electron microscopy(SEM)and thermal gravimetric analysis(TGA).The microstructure shows that ZnO microvaristors distribute uniformly in the composite,and the content difference of ZnO microvaristors at the top and bottom part is only 0.4%.The composite shows typical non‐linear conductivity,and the threshold electric field and the non‐linear coefficient decrease with the content of ZnO microvaristors,while the conductivity in the insulating state shows an increasing trend.To verify the field grading effect of the composite with non‐linear conductivity(CNC),a finite element model of a needle‐plate electrode,simulating the condition of a conductive tip in a solid insulated system,is set up.CNC can adaptively grade the electric field,which reduces the surface electric field of the needle tip by 86.6%and the highest electric field in the system by 82.1%.This wet winding method solidifies the industrial application of CNC in high‐voltage power equipment.Zhikang Yuan Jun Hu Zhiwen Huang Gang Sun Ya Sun Jinliang He 2022High Voltage2022,7,1:2
5Auxin signaling module OsSK41-OsIAA10-OsARF regulates grain yield traits in rice显示文摘Auxin is an important phytohormone in plants, and auxin signaling pathways in rice play key roles in regulating its growth, development, and productivity. To investigate how rice grain yield traits are regulated by auxin signaling pathways and to facilitate their application in rice improvement, we validated the functional relationships among regulatory genes such as OsIAA10, OsSK41, and OsARF21 that are involved in one of the auxin(OsIAA10) signaling pathways. We assessed the phenotypic effects of these genes on several grain yield traits across two environments using knockout and/or overexpression transgenic lines.Based on the results, we constructed a model that showed how grain yield traits were regulated by OsIAA10 and OsTIR1, OsAFB2, and OsSK41 and OsmiR393 in the OsSK41-OsIAA10-OsARF module and by OsARF21 in the transcriptional regulation of downstream auxin response genes in the OsSK41-OsIAA10-OsARF module. The population genomic analyses revealed rich genetic diversity and the presence of major functional alleles at most of these loci in rice populations. The strong differentiation of many major alleles between Xian/indica and Geng/japonica subspecies and/or among modern varieties and landraces suggested that they contributed to improved productivity during evolution and breeding. 6OTUB1 promotes osteoblastic bone formation through stabilizing FGFR2显示文摘Bone homeostasis is maintained by the balance between osteoblastic bone formation and osteoclastic bone resorption.Dysregulation of this process leads to multiple diseases,including osteoporosis.However,the underlying molecular mechanisms are not fully understood.Here,we show that the global and conditional osteoblast knockout of a deubiquitinase Otub1 result in low bone mass and poor bone strength due to defects in osteogenic differentiation and mineralization.Mechanistically,the stability of FGFR2,a crucial regulator of osteogenesis,is maintained by OTUB1.OTUB1 attenuates the E3 ligase SMURF1-mediated FGFR2 ubiquitination by inhibiting SMURF1’s E2 binding.In the absence of OTUB1,FGFR2 is ubiquitinated excessively by SMURF1,followed by lysosomal degradation.Consistently,adeno-associated virus serotype 9(AAV9)-delivered FGFR2 in knee joints rescued the bone mass loss in osteoblast-specific Otub1-deleted mice.Moreover,Otub1 mRNA level was significantly downregulated in bones from osteoporotic mice,and restoring OTUB1 levels through an AAV9-delivered system in ovariectomy-induced osteoporotic mice attenuated osteopenia.Taken together,our results suggest that OTUB1 positively regulates osteogenic differentiation and mineralization in bone homeostasis by controlling FGFR2 stability,which provides an optical therapeutic strategy to alleviate osteoporosis.Qiong Zhu Yesheng Fu Chun-Ping Cui Yi Ding Zhikang Deng Chao Ning Fan Hu Chen Qiu Biyue Yu Xuemei Zhou Guan Yang Jiang Peng Weiguo Zou Cui Hua Liu Lingqiang Zhang 2023Signal Transduction and Targeted Therapy2023,8,5:1

11Design of adaptive bushing based on field grading materials显示文摘Electrical bushings,as important power system components,are critical to the reliability of power systems.The capacitance-graded bushing is a type of high voltage bushing that is most commonly used in current power systems.However,the limitations of and problems with capacitance-graded bushings are exposed when the operating voltage level increases.A new direct current wall bushing with adaptive field grading materials is proposed,based on the core features of the problem that the bushing is intended to solve.The main insulation structure of the proposed DC wall bushing with adaptive field grading materials comprises three parts:a field grading layer,a current limiting layer and an electrode extension layer.The proposed bushing has characteristics that include a simple structure,small size and environmental friendliness,and it also meets the re-quirements of existing power systems.The proposed bushing also conforms to the intended development direction for future power equipment and will aid in the devel-opment of power systems for operation at higher voltages.Xiaolei Zhao Jun Hu Zhikang Yuan Jinliang He 2021High Voltage2021,6,4:0
12Differential microRNA expression between shoots and rhizomes in Oryza longistaminata using high-throughput RNA sequencing显示文摘Plant microRNAs(miRNAs)play important roles in biological processes such as development and stress responses.Although the diverse functions of miRNAs in model organisms have been well studied,their function in wild rice is poorly understood.In this study,high-throughput small RNA sequencing was performed to characterize tissue-specific transcriptomes in Oryza longistaminata.A total of 603 miRNAs,380 known rice miRNAs,72 conserved plant miRNAs,and151 predicted novel miRNAs were identified as being expressed in aerial shoots and rhizomes.Additionally,99 and 79 miRNAs were expressed exclusively or differentially,respectively,in the two tissues,and 144 potential targets were predicted for the differentially expressed miRNAs in the rhizomes.Functional annotation of these targets suggested that transcription factors,including squamosa promoter binding proteins and auxin response factors,function in rhizome growth and development.The expression levels of several miRNAs and target genes in the rhizomes were quantified by RT-PCR,and the results indicated the existence of complex regulatory mechanisms between the miRNAs and their targets.Eight target cleavage sites were verified by RNA ligase-mediated rapid 5′end amplification.These results provide valuable information on the composition,expression and function of miRNAs in O.longistaminata,and will aid in understanding the molecular mechanisms of rhizome development.Ying Zong Liyu Huang Ting Zhang Qiao Qin Wensheng Wang Xiuqin Zhao Fengyi Hu Binying Fu Zhikang Li 2014The Crop Journal2014,2,Z1:0
13Tissue-specific transcriptomics reveals a central role of CcNST1 in regulating the fruit lignification pattern in Camellia chekiangoleosa,a woody oil-crop显示文摘Fruit lignification is of significant economic importance because it affects the quality of fruit and the production of seed oil.The specified lignification pattern in Camellia chekiangoleosa fruits plays critical roles in its seed oil yield,but little is known about how this lignification process is regulated.Here,we report on a comprehensive tissue-specific transcriptomics analysis conducted for C.chekiangoleosa fruit.By mining the differentially expressed genes,we found that lignin biosynthesis and transcriptional regulation pathways were significantly enriched in the lignified tissues.The homolog of NST-like transcription factor,CcNST1,was highly expressed in lignified seed coat and endocarp tissues;transgenic analyses of CcNST1 in Arabidopsis and hybrid poplar revealed the enhanced lignification levels of various tissues.Gene expression analysis of the transgenic lines uncovered potential downstream genes involved in the regulation of lignin biosynthesis.This work provides a valuable gene expression resource and identified the pivotal role of CcNST1 in regulating the lignin biosynthesis underlying fruit lignification.Chao Yan Ziyan Nie Zhikang Hu Hu Huang Xianjin Ma Sijia Li Jiyuan Li Xiaohua Yao Hengfu Yin 2022Forestry Research2022,2,1:0

15Self‐adaptive electrostatic discharge performance of ZnO microvaristors doped silicone rubber composites显示文摘With the increase in applied voltage and the decrease in volume,integrated electronic modules have become electrostatic sensitive.ZnO microvaristors doped silicone rubber(SiR)composite,with electric field‐dependent conductivity,shows promising prospects for solving the electrostatic problem.In this study,20 vol%to 50 vol%ZnO/SiR composite films,filled with 10–30μm ZnO microvaristors,were prepared and put into the test of electrostatic discharge.Compared with the insulating and conducting materials,ZnO/SiR composites achieve the lowest initial voltage and a short time constant of electrostatic discharge.With the increase of ZnO microvaristors volume fraction,from 20%to 50%,the initial voltage and the time constant of electrostatic discharge drop from 1.78 to 0.72 kV and 3.1 ms to 0.4μs.When the ZnO volume fraction is higher than 30%,there is no residual voltage after 0.1 s.To explore the reason behind different electrostatic discharge performances of the ZnO/SiR composites,the conductivities of the com-posites were measured.It is found that the break‐over voltage of the composite drops with the increase in the ZnO microvaristors volume fraction.When a high voltage im-pulse is applied on the ZnO/SiR film,the composite will turn to the conducting state and release the electrostatic charge adaptively so that the initial voltage can be controlled.This work supplies a novel electrostatic discharge idea for electric and electronic devices.Ya Sun Zhikang Yuan Jun Hu Zhiwen Huang Xuan Liu Gang Sun Qi Li Jinliang He 2023High Voltage2023,8,6:0
