2Humoral immune response to circulating SARS-CoV-2 variants elicited by inactivated and RBD-subunit vaccines显示文摘SARS-CoV-2 variants could induce immune escape by mutations on the receptor-binding domain(RBD)and N-terminal domain(NTD).Here we report the humoral immune response to circulating SARS-CoV-2 variants,such as 501Y.V2(B.1.351),of the plasma and neutralizing antibodies(NAbs)elicited by CoronaVac(inactivated vaccine),ZF2001(RBD-subunit vaccine)and natural infection.Among 86 potent NAbs identified by high-throughput single-cell VDJ sequencing of peripheral blood mononuclear cells from vaccinees and convalescents,near half anti-RBD NAbs showed major neutralization reductions against the K417N/E484K/N501Y mutation combination,with E484K being the dominant cause.VH3-53/VH3-66 recurrent antibodies respond differently to RBD variants,and K417N compromises the majority of neutralizing activity through reduced polar contacts with complementarity determining regions.In contrast,the 242–244 deletion(242–244Δ)would abolish most neutralization activity of anti-NTD NAbs by interrupting the conformation of NTD antigenic supersite,indicating a much less diversity of anti-NTD NAbs than anti-RBD NAbs.Plasma of convalescents and CoronaVac vaccinees displayed comparable neutralization reductions against pseudo-and authentic 501Y.V2 variants,mainly caused by E484K/N501Y and 242–244Δ,with the effects being additive.Importantly,RBD-subunit vaccinees exhibit markedly higher tolerance to 501Y.V2 than convalescents,since the elicited anti-RBD NAbs display a high diversity and are unaffected by NTD mutations.Moreover,an extended gap between the third and second doses of ZF2001 leads to better neutralizing activity and tolerance to 501Y.V2 than the standard three-dose administration.Together,these results suggest that the deployment of RBD-vaccines,through a third-dose boost,may be ideal for combating SARS-CoV-2 variants when necessary,especially for those carrying mutations that disrupt the NTD supersite.Yunlong Cao Ayijiang Yisimayi Yali Bai Weijin Huang Xiaofeng Li Zhiying Zhang Tianjiao Yuan Ran An Jing Wang Tianhe Xiao Shuo Du Wenping Ma Liyang Song Yongzheng Li Xiang Li Weiliang Song Jiajing Wu Shuo Liu Xuemei Li Yonghong Zhang Bin Su Xianghua Guo Yangyang Wei Chuanping Gao Nana Zhang Yifei Zhang Yang Dou Xiaoyu Xu Rui Shi Bai Lu Ronghua Jin Yingmin Ma Chengfeng Qin Youchun Wang Yingmei Feng Junyu Xiao Xiaoliang Sunney Xie 2021Cell Research2021,31,7:12

5Carbonated Big Mantle Wedge Extending to the NE Edge of the Stagnant Pacific Slab:Constraints from Late Mesozoic-Cenozoic Basalts from Far Eastern Russia显示文摘It has been suggested that the carbonated mantle reflected by Mg-Zn isotopic anomalies of Cenozoic intraplate basalts from East Asia coincides with the stagnant West Pacific slab in the mantle transition zone.However,the northern boundary of such carbonated domain beneath East Asia is uncertain.Late Mesozoic-Cenozoic intraplate basalts are widespread in far eastern Russia and thus provide an opportunity to examine this issue.Here we report major-trace element contents and Sr-NdMg-Zn isotopic compositions for 9 Late Mesozoic-Cenozoic basaltic samples from the Khanka Block and Sikhote-Alin accretionary complex.They are characterized by large variations in SiO_(2)contents(41 wt.%to 50 wt.%)and CaO/Al_(2)O_(3)(0.50 to 0.97),enrichments of large-ion lithophile elements(LILE),positive Nb-Ta anomalies and strongly negative K,Pb,Zr,Hf,Ti,Y anomalies in primitive mantle-normalized trace element spider diagram.Furthermore,the rocks show good correlations of Ti/Ti^(*)with Hf/Hf^(*),La/Yb,Fe/Mn and trace element contents(e.g.,Nb).In addition,they have lighter Mg and heavier Zn isotope compositions than the BSE estimates,coupled with depleted Sr-Nd isotope compositions.These elemental and isotopic characteristics cannot be explained by alteration,magma differentiation or diffusion,but are consistent with the partial melting of carbonated peridotite.By and large,the Late Mesozoic-Cenozoic basalts from far eastern Russia bear very similar geochemical characteristics as those Na-series Cenozoic basalts from eastern China.The extended region of Mg-Zn isotopic anomalies is roughly coincident with the stagnant West Pacific slab beneath East Asia,and all of these alkali basalts can be generated from mantle sources hybridized by recycled Mg-carbonates from the Pacific slab stagnant in the mantle transition zone.We infer that(1)the carbonated big mantle wedge extends to the NE edge of the West Pacific slab and may have also appeared in the Late Mesozoic due to the effect of the Paleo-Pacific slab beneath this region,and(2)decarbonation of stagnant slabs in the mantle transition zone is a key mechanism for carbon outgassing from deep mantle to surface via intraplate alkali melts.Ronghua Cai Shan Xu Dmitri AIonov Jian Huang Sheng-Ao Liu Shuguang Li Jingao Liu 2022Journal of Earth Science2022,33,1:2

7Interface Enhancement of Glass Fiber/Unsaturated Polyester Resin Composites with Nano-Silica Treated Using Silane Coupling Agent显示文摘Nano-silica treated with different kinds of coupling agent(KH550, A-143, A-151) was used to modify the surface condition of glass fiber, and then, the modified glass fiber/ unsaturated polyester resin(UPR) composites materials were prepared. Scanning electron microscopy(SEM), dynamic mechanical analysis(DMA), and impact test were used to characterize the composite materials' structure and properties. The morphology of composite materials shows that the adhesion between nano-silica and glass fiber is improved when silane coupling agent is added in. The DMA and impact test results show that silane coupling agent(especially KH550 and A-151) could effectively improve the composite's mechanical properties. 8Interleukin-13 inhibits cytokines synthesis by blocking nuclear factor-κB and c-Jun N-terminal kinase in human mesangial cells显示文摘Objective:Monocytes/macrophages,proinflammatory cytokines and chemokines are important in the pathogenesis of glomerulonephritis.Interleukin(IL)-13 has been shown to exert potent anti-inflammatory properties. This study was designed to investigate the effect of IL-13 on the expression of proinflammatory cytokines,chemokines and profibrogenic cytokines and the involved molecular mechanism in cultured human mesangial cells (HMCs).Methods:The expressions of proinflammatory cytokines,chemokines and profibrogenic cytokines were determined by ribonuclease protection assay(RPA).Activity of nuclear factor-kappa B(NF-κB)and activa- tor protein-1(AP-1)was examined by electrophoretic mobility shift assay(EMSA).NF-κB subunit p65 nuclear transportation and c-Jun N-terminal kinase(JNK)activity were assayed by immunoblot.Results:Recombinant IL-13 inhibited tumor necrosis factor-α(TNF-α),IL-1α,IL-1β,monocyte chemoattractant protein-1(MCP-1), IL-8,and transforming growth factor-β1(TGF-β1)mRNA expressions in a dose-dependent manner.Lipopoly-sacchorides(LPS)dramatically increased NF-κB DNA binding activity of HMCs,which was inhibited by IL-13 in a dose-dependent manner.LPS-activated NF-κB contained p50 and p65 dimers,but not c-Rel subunit.IL-13 blocked LPS-induced NF-κB subunit p65.LPS stimulated JNK/AP-1 activation,which was inhibited by IL-13 in a dose-dependent manner.Conclusion:IL-13 inhibits proinflammatory cytokines,chemokines,and profibrogenic cytokines synthesis by blocking NF-κB and JNK/AP-1 activation.These observations point to the importance of IL-13 in the modulation of inflammatory processes in the renal glomerulus.Chunhua Zhu Aihua Zhang Songming Huang GuixiaDing Xiaoqin Pan Ronghua Chen 2010The Journal of Biomedical Research2010,24,4:2

12Interleukin-10 inhibits the expression of nuclear factor kappa B in rats with cerebral ischemia显示文摘BACKGROUND: Plenty of studies have demonstrated that inflammatory reaction is involved in ischemic cerebral damage, and the expression of inflammatory cytokines can be observed at the initial sites of cerebral damage at early period, including interleukin-6, interleukin-8, etc., which are all the target gene products of nuclear factor kappa B (NF-κB). The process of ischemic damage can be affected by adjusting and controlling NF-κB activity via multi-links. OBJECTIVE: To investigate the inhibitory effect of interleukin-10 on the expression of NF-κB in the ischemic sites of rats with focal cerebral ischemia in rats and its molecular mechanisms. DESIGN: A randomized and controlled animal trial. SETTING: Department of Neurology, the Affiliated Union Hospital of Fujian Medical University. MATERIALS: Thirty-two adult male Sprague-Dawley rats weighing (250±30) g were used. NF-κB p65 (RelA) rabbit anti-rat monoclonal primary antibody was the product of Neomarkers Company; Immunohistochemical kit of the SP two-step method was purchased from Beijing Zhongshan Biotechnology Co., Ltd. METHODS: The experiment was carried out in the Affiliated Union Hospital of Fujian Medical University from August 2005 to April 2006. The rats were randomly assigned into sham-operated group, middle cerebral artery occlusion (MCAO) group, vehicle-treated group and interleukin-10 treated group, 8 rats in each group. Focal cerebral ischemia was induced by occlusion of the middle cerebral artery as previously described. Rats in the MCAO group were anesthetized intraperitoneally, thyroid was bluntly dissected. Right common, external and internal carotid arteries were isolated, the trunk of external carotid artery was ligated and freed, an artery clamp was placed at the internal carotid artery, then a 'V' shape incision was made at the free section of external carotid artery, filament was inserted for a depth of (18.5±0.5) mm. The rats in the sham-operated group were given the same treatments with the exception of filament insertion. After the successful model establishment for 1 hour, the rats in the interleukin-10 treated group were injected with human recombinant interleukin-10 (1 μg) via lateral ventricle, whereas those in the vehicle-treated group were injected with 5 mol/L NaP (5 μL). The rectal temperature of rats were kept at about 37 ℃ with heating lamps throughout the operation. Twenty-four hours after MCAO, the rats were examined for neurological deficits. Only those animals that scored at 1-3 points were utilized. The rats were decapitated at 24 hours postoperatively. The expression of NF-κB p65 in peri-infarct core was detected immunohistochemically. The percentage of NF-κB p65 subunit positive cells in 1 000 cells was calculated. MAIN OUTCOME MEASURES: Expression of NF-κB p65 in peri-infarct core; Percentage of NF-κB p65 subunit positive cells. RESULTS: All the 32 rats were involved in the analysis of results. NF-κB p65 expressed in cytoplasm and some nuclei. It was expressed all in cytoplasm in the sham-operated group, and partly expressed in the nucleus after cerebral ischemia. Small amounts of NF-κB p65 positive neurocytes were observed in the sham-operated group[(3.7±0.6)%], those were obviously increased in the MCAO group [(15.4±3.7)%, P < 0.01]. NF-κB p65 positive neurocytes were significantly reduced in the interleukin-10 treated group as compared with those in the vehicle-treated groups [(12.1±2.2)%, (15.5±3.6)%, P < 0.05]. 13Engineering of Co_(3O)_(4)@Ni_(2)P heterostructure as trifunctional electrocatalysts for rechargeable zinc-air battery and self-powered overall water splitting显示文摘Rational design of highly efficient,robust and nonprecious electrocatalysts for the oxygen reduction reaction(ORR),oxygen evolution reaction(OER)and hydrogen evolution reaction(HER)is highly demanded and challenging.Here,heterostructural Co_(3O)_(4)@Ni_(2)P arrays with numerous reaction sites,unique interfacial electronic structure and fast charge transfer kinetics are developed as electrocatalysts for rechargeable Zn-air batteries and overall water splitting.Both density functional theory calculation and X-ray absorption fine structure analysis manifest that the synergistic structural and abundant electronic modulations interfaces are formed,thus simultaneously promoting the electrocatalytic kinetics,activities and stabilities.Specifically,it can achieve an ultralow overpotential of 270 m V and 28 m V at 10 m A cm^(-2) for OER and HER,respectively.The water electrolyzer delivers a current density of 10 m A cm^(-2) at 1.563 V;furthermore,rechargeable Zn-air batteries triggered by this heterostructure can achieve excellent cyclic stability of 177 h(2 h per cycle)at 10 m A cm^(-2);both devices are superior to the Pt/C+Ir/C.This work not only designs an efficient trifunctional electrocatalyst but also paves an avenue to understand the heterostructure engineering for catalysts development and disclose the underlying relationship of interfacial electronic structures and catalytic properties.Xiaolin Hu Tongxin Yang Zuguang Yang Zongyang Li Ronghua Wang Meng Li Guangsheng Huang Bin Jiang Chaohe Xu Fusheng Pan 2022Journal of Materials Science & Technology2022,,20:1

18The design of Co_(3)S_(4)@MXene heterostructure as sulfur host to promote the electrochemical kinetics for reversible magnesium-sulfur batteries显示文摘The rechargeable Mg-S batteries are attractive because of their resource abundances of Mg and S,high volumetric energy density,and less dendrite property of Mg anodes.However,the development is barred by the intrinsic low electronic conductivity of S and the discharge products as well as the lack of understanding the hidden electrochemical kinetics.Here,a Co_(3)S_(4)@MXene heterostructure is proposed as effective sulfur host for reversible Mg-S batteries.XPS results and density functional theory(DFT)calculation confirm that the chemical interaction between the decorated Co_(3)S_(4)nanocrystals host and polysulfide intermediates could well absorb and catalyze the polysulfides conversion,thus improve the electrochemical redox kinetics.Meanwhile,the MXene matrix could promote Mg ion diffusion dynamics greatly.As a result,the developed Mg-S batteries using the Co_(3)S_(4)@MXene-S as the cathode material could demonstrate high sulfur utilization with specific capacity of 1220 mAh g^(-1) and retain a capacity of 528 mAh g^(-1) after 100 cycles,together with a satisfactory rate performance even at 2 C.This work shed light on the advanced cathode design for reversible high energy Mg-S batteries.Qiannan Zhao Ronghua Wang Yuxin Zhang Guangsheng Huang Bin Jiang Chaohe Xu Fusheng Pan 2021Journal of Magnesium and Alloys2021,9,1:1

20Scientific Train of Thought and Methodological Innovation in the Intelligent Decision Support System for Earthquake Prediction in China显示文摘The level of present understanding of earthquake prediction of seismologists at home and abroad is very different. This is because China has opened up a special path of earthquake prediction research that has not been explored by other countries, with its own advantages and potentialities.Therefore, we considered that it is the most practical way to use the advantages and potentialities for raising the earthquake prediction level. For this purpose, we have developed a set of intelligent decision support system for earthquake prediction, with the analysis of cluster anomalies process at the core.
