| 1 | The discovery of Palaeoproterozoic volcanic rocks in the Bulunkuoler Group from the Tianshuihai Massif in Xinjiang of Northwest China and its geological significance显示文摘In the Dabudaer region of the Tianshuihai Massif (Xinjiang, Northwest China), metavolcanic rocks within the mainly meta-sedimentary Bulunkuoler Group are basalt, basaltic andesite, and rhyolite. In situ zircon U-Pb LA-ICP-MS dating on rhyolite magmatic zircons yielded an age of 2481±14 Ma, interpreted as the eruption age of the Bulunkuoler Group volcanic rocks. The basalt and basaltic andesite are low-Al tholeiite series and the rhyolite is low-Al calc-alkaline series. All of rocks are charac-terized by enrichment of LREE and LILE (Rb, Th and Ba), depletion of P, Nb, Ta and Ti, and absence of Eu anomalies. The εNd(t) of basalts ranges from 3.14 to 4.88 indicating a depleted mantle source. The trace element signatures show that these magmas experienced intense crustal contamination during their ascent. Direct evidence for crustal contamination is the xenocrystic zircons in the ryholite, with ages back to ca. 3300 Ma. The primitive-mantle normalized and Zr/Y-Zr diagrams for the basic volcanic rocks show that they formed in an intra-continental tectonic setting. | 2 | Celastrol induces ferroptosis in activated HSCs to ameliorate hepatic fibrosis via targeting peroxiredoxins and HO-1显示文摘Ferroptosis is a form of regulated cell death, characterized by excessive membrane lipid peroxidation in an iron-and ROS-dependent manner. Celastrol, a natural bioactive triterpenoid extracted from Tripterygium wilfordii, shows effective anti-fibrotic and anti-inflammatory activities in multiple hepatic diseases. However, the exact molecular mechanisms of action and the direct protein targets of celastrol in the treatment of liver fibrosis remain largely elusive. Here, we discover that celastrol exerts anti-fibrotic effects via promoting the production of reactive oxygen species(ROS) and inducing ferroptosis in activated hepatic stellate cells(HSCs). By using activity-based protein profiling(ABPP) in combination with bio-orthogonal click chemistry reaction and cellular thermal shift assay(CETSA), we show that celastrol directly binds to peroxiredoxins(PRDXs), including PRDX1, PRDX2, PRDX4 and PRDX6,through the active cysteine sites, and inhibits their anti-oxidant activities. Celastrol also targets to heme oxygenase 1(HO-1) and upregulates its expression in activated-HSCs. Knockdown of PRDX1, PRDX2,PRDX4, PRDX6 or HO-1 in HSCs, to varying extent, elevated cellular ROS levels and induced ferroptosis. | 3 | Deformation mechanism and stability control of roadway along a fault subjected to mining显示文摘It is difficult to maintain the roadway around a fault because of the fractured surroundings,complex stress environment,and large and intense deformation in the mining process.Based on a tailgate of panel S2205 in Tunliu colliery,in Shanxi province,China,we investigated the evolution of stress and displacement of rocks surrounding the roadway during the drivage and mining period using theoretical analysis,numerical simulation and field trial methods.We analyzed the deformation and failure mechanisms of the tailgate near a fault.The deformation of surrounding rock caused by unloading in the drivage period is large and asymmetric,the roadway convergence increases with activation of the fault and secondary fracture develops in the mining period.Therefore,we proposed a specific control technique of the roadway along a fault as follows:(1) High-strength yielding bolt not only supports the shallow rock to loadbearing structures,but also releases primary deformation energy by use of a pressure release device in order to achieve high resistance to the pressure retained;(2) Grouting of near-fault ribside after initial stabilization of the rock deformation is used to reinforce the broken rock,and to improve the integral load-bearing capacity of the roadway.The research results were successfully applied to a field trial. | Yan Shuai Bai Jianbiao Li Wenfeng Chen Jigang Li Lei | 2012 | International Journal of Mining Science and Technology2012,22,4: | 11 |
| 4 | Finite element simulation and experimental test of the wear behavior for self-lubricating spherical plain bearings显示文摘In this study, based on the classical Archard adhesion wear theory, a three-dimensional finite element model was established, with the aim of simulating the failure process of self-lubricating spherical plain bearings in the swinging wear condition. The results show that the self-lubricating spherical plain bearings go through two different stages during the wear process, namely, initial wear stage and stable wear stage. Because the large contact points wear out during the initial wear stage, the maximum contact pressure decreases as the test period increases. The relatively larger wear depth region shows elliptical distribution, and the maximum distribution appears in the central contact area. The wear depth reaches 0.974 mm after swinging 25,000 times. PTFE fibers, which possess a good friction performance but poor abrasion resistance, abundantly exist on the friction surfaces of the fabric liner. Consequently, the friction torque during the initial wear stage is slightly smaller than the friction torque during the stable wear stage; however, the wear rate during the initial wear stage is high. The reliability and effectiveness of the finite element model are verified by experiment. | 9 | Distribution of dissolved oxygen and causes of maximum concentration in the Bering Sea in July 2010显示文摘According to data obtained in the Bering Sea during the 4th Chinese National Arctic Research Expedition, the distribution of dissolved oxygen(DO) was studied, causes of its maximum concentration were discussed, and the relationships between DO and other parameters, such as salinity, temperature, and chlorophyll a were analyzed. The results showed DO concentration ranged from 0.53 to 12.05 mg/L in the Bering Sea basin. The upper waters contained high concentrations and the maximum occurred at the depth range from 20 to 50 m. The DO concentration decreased rapidly when the depth was deeper than 200 m and reached the minimum at the depth range from 500 to 1 000 m, and then increased slowly with the depth increasing but still kept at a low level. On the shelf, the DO concentration ranged from 6.53 to 16.63 mg/L with a mean value of 10.75 mg/L, and showed a characteristic of decreasing from north to south. The DO concentration was higher in the area between the Bering Sea and Lawrence Island and was lower in the southeast and southwest of Lawrence Island at the latitude of 62°N. The formation of maximum DO concentration was concerned with phytoplankton photosynthesis and formation of the themocline. To the south of Sta. B07 in the Bering Sea basin, the oxygen produced by photosynthesis permeated to the deeper water and the themocline made it difficult to exchange vertically, and to the north of Sta. B07, the maximum DO concentration occurred above the themocline due to phytoplankton activities. On the shelf, the oxygen produced by phytoplankton photosynthesis gathered at the bottom of the thermocline and formed the DO maximum concentration. | 11 | Synergistic effects of autophagy/mitophagy inhibitors and magnolol promote apoptosis and antitumor efficacy显示文摘Mitochondria as a signaling platform play crucial roles in deciding cell fate.Many classic anticancer agents are known to trigger cell death through induction of mitochondrial damage.Mitophagy,one selective autophagy,is the key mitochondrial quality control that effectively removes damaged mitochondria.However,the precise roles of mitophagy in tumorigenesis and anticancer agent treatment remain largely unclear.Here,we examined the functional implication of mitophagy in the anticancer properties of magnolol,a natural product isolated from herbal Magnolia officinalis.First,we found that magnolol induces mitochondrial depolarization,causes excessive mitochondrial fragmentation,and increases mitochondrial reactive oxygen species(mtROS).Second,magnolol induces PTEN-induced putative kinase protein 1(PINK1)-Parkin-mediated mitophagy through regulating two positive feedforward amplification loops.Third,magnolol triggers cancer cell death and inhibits neuroblastoma tumor growth via the intrinsic apoptosis pathway.Moreover,magnolol prolongs the survival time of tumor-bearing mice.Finally,inhibition of mitophagy by PINK1/Parkin knockdown or using inhibitors targeting different autophagy/mitophagy stages significantly promotes magnolol-induced cell death and enhances magnolol's anticancer efficacy,both in vitro and in vivo.Altogether,our study demonstrates that magnolol can induce autophagy/mitophagy and apoptosis,whereas blockage of autophagy/mitophagy remarkably enhances the anticancer efficacy of magnolol,suggesting that targeting mitophagy may be a promising strategy to overcome chemoresistance and improve anticancer therapy. | Yancheng Tang Liming Wang Tao Yi Jun Xu Jigang Wang Jiang-Jiang Qin Qilei Chen Ka-Man Yip Yihang Pan Peng Hong Yingying Lu Han-Ming Shen Hu-Biao Chen | 2021 | Acta Pharmaceutica Sinica B2021,11,12: | 2 |
| 12 | Kinectin 1 promotes the growth of triple-negative breast cancer via directly co-activating NF-kappaB/p65 and enhancing its transcriptional activity显示文摘Triple-negative breast cancer(TNBC)is the most challenging subtype of breast cancer.Various endeavor has been made to explore the molecular biology basis of TNBC.Herein,we reported a novel function of factor Kinectin 1(KTN1)as a carcinogenic promoter in TNBC.KTN1 expression in TNBC was increased compared with adjacent tissues or luminal or Her2 subtypes of breast cancer,and TNBC patients with high KTN1 expression have poor prognosis.In functional studies,knockdown of KTN1 inhibited the proliferation and invasiveness of TNBC both in vitro and in vivo,while overexpression of KTN1 promoted cancer cell proliferation and invasiveness.RNA-seq analysis revealed that the interaction of cytokine-cytokine receptor,particularly CXCL8 gene,was upregulated by KTN1,which was supported by the further experiments.CXCL8 depletion inhibited the tumorigenesis and progression of TNBC.Additionally,rescue experiments validated that KTN1-mediated cell growth acceleration in TNBC was dependent on CXCL8 both in vitro and in vivo.Furthermore,it was found that KTN1 enhanced the phosphorylation of NF-κB/p65 protein at Ser536 site,and specifically bound to NF-κB/p65 protein in the nucleus and cytoplasm of cells.Moreover,the transcription of CXCL8 gene was directly upregulated by the complex of KTN1 and NF-κB/p65 protein.Taken together,our results elucidated a novel mechanism of KTN1 gene in TNBC tumorigenesis and progression.KTN1 may be a potential molecular target for the development of TNBC treatment. | Lin Gao Shanze Chen Malin Hong Wenbin Zhou Bilan Wang Junying Qiu Jinquan Xia Pan Zhao Li Fu Jigang Wang Yong Dai Ni Xie Qinhe Yang Hsien-Da Huang Xiang Gao Chang Zou | 2021 | Signal Transduction and Targeted Therapy2021,6,8: | 2 |

| 16 | Differential DNA methylation profiles of human B lymphocytes and Epstein-Barr virus-immortalized B lymphocytes显示文摘Objective: This study aimed to comprehensively assess Epstein-Barr virus(EBV)-induced methylation alterations of B cell across whole genome.Methods: We compared DNA methylation patterns of primary B cells and corresponding lymphoblastoid cell lines(LCLs) from eight participants. The genome-wide DNA methylation profiles were compared at over 850,000 genome-wide methylation sites.Results: DNA methylation analysis revealed 87,732 differentially methylated Cp G sites, representing approximately 12.41% of all sites in LCLs compared to primary B cells. The hypermethylated and hypomethylated Cp G sites were about 22.75% or 77.25%, respectively. Only 0.8% of hypomethylated sites and 4.5% of hypermethylated sites were located in Cp G islands, whereas 8.0% of hypomethylated sites and 16.3% of hypermethylated sites were located in shore(N_shore and S_shore). Using principal component analysis of the DNA methylation profiles, primary B cells and LCLs could be accurately predicted.
