| 1 | The FGF metabolic axis显示文摘Members of the fibroblast growth factor (FGF) family play pleiotropic roles in cellular and metabolic homeostasis. During evolution, the ancestor FGF expands into multiple members by acquiring divergent structural elements that enable functional divergence and specification. Heparan sulfate-binding FGFs, which play critical roles in embryonic development and adult tissue remodeling homeostasis, adapt to an autocrine/paracrine mode of action to promote cell proliferation and population growth. By contrast, FGF19, 21, and 23 coevolve through losing binding affinity for extracellular matrix heparan sulfate while acquiring affinity for transmembrane α-Klotho (KL) or β-KL as a coreceptor, thereby adapting to an endocrine mode of action to drive interorgan crosstalk that regulates a broad spectrum of metabolic homeostasis. FGF19 metabolic axis from the ileum to liver negatively controls diurnal bile acid biosynthesis. FGF21 metabolic axes play multifaceted roles in controlling the homeostasis of lipid, glucose, and energy metabolism. FGF23 axes from the bone to kidney and parathyroid regulate metabolic homeostasis of phosphate, calcium, vitamin D, and parathyroid hormone that are important for bone health and systemic mineral balance. The significant divergence in structural elements and multiple functional specifications of FGF19, 21, and 23 in cellular and organismal metabolism instead of cell proliferation and growth sufficiently necessitate a new unified and specific term for these three endocrine FGFs. | 2 | Crop rotation-dependent yield responses to fertilization in winter oilseed rape(Brassica napus L.)显示文摘Differences in soil physical, chemical and biological properties between paddy–upland and continuous upland rotations will influence nutrient relations and crop growth. With the aim of estimating rapeseed yield performance in response to fertilization in rice–rapeseed(RR) and cotton–rapeseed(CR) rotations, on-farm experiments were conducted at 70 sites across Hubei province, central China. The economically optimal fertilizer rates of winter oilseed rape in different rotations were determined. Field experiments showed that previous crops significantly influenced seed yields. Without N fertilization,seed yields were significantly lower for the RR rotation than for the CR rotation. The average yield increase ratio and agronomic efficiency associated with nitrogen(N)fertilization in the RR rotation were 96.6% and 6.56 kg kg- 1, significantly higher than those in the CR rotation. No seed yield differences were detected between the two rotations under phosphorus(P) and potassium(K) fertilization. In contrast to the CR rotation, N fertilizer played a more vital role in maintaining high seed yields in the RR rotation owing to the lower indigenous soil N supply. Compared with local N fertilizer recommendation rates for the RR rotation, on average an additional 18 kg N ha- 1was recommended according to the economically optimal N fertilizer rate(EONFR). In contrast, the EONFR was 14 kg N ha- 1lower than the locally recommended N fertilizer rate for the CR rotation. There were no differences between the two rotations for the average economically optimal P and K fertilization rates. | 6 | A more consistent intraluminal rhesus monkey model of ischemic stroke显示文摘Endovascular surgery is advantageous in experimentally induced ischemic stroke because it causes fewer cranial traumatic lesions than invasive surgery and can closely mimic the pathophysiology in stroke patients. However, the outcomes are highly variable, which limits the accuracy of evaluations of ischemic stroke studies. In this study, eight healthy adult rhesus monkeys were randomized into two groups with four monkeys in each group: middle cerebral artery occlusion at origin segment(M1) and middle cerebral artery occlusion at M2 segment. The blood flow in the middle cerebral artery was blocked completely for 2 hours using the endovascular microcoil placement technique(1 mm × 10 cm)(undetachable), to establish a model of cerebral ischemia. The microcoil was withdrawn and the middle cerebral artery blood flow was restored. A reversible middle cerebral artery occlusion model was identified by hematoxylin-eosin staining, digital subtraction angiography, magnetic resonance angiography, magnetic resonance imaging, and neurological evaluation. The results showed that the middle cerebral artery occlusion model was successfully established in eight adult healthy rhesus monkeys, and ischemic lesions were apparent in the brain tissue of rhesus monkeys at 24 hours after occlusion. The rhesus monkeys had symptoms of neurological deficits. Compared with the M1 occlusion group, the M2 occlusion group had lower infarction volume and higher neurological scores. These experimental findings indicate that reversible middle cerebral artery occlusion can be produced with the endovascular microcoil technique in rhesus monkeys. | 8 | Integrin-interacting protein Kindlin-2 induces mammary tumors in transgenic mice显示文摘Kindlin-2, an integrin-interacting protein, regulates breast cancer progression. However, currently, no animal model to study the role of Kindlin-2 in the carcinogenesis of mammary gland is available. We established a Kindlin-2 transgenic mouse model using a mammary gland-specific promoter, mammary tumor virus(MMTV) long terminal repeat(LTR). Kindlin-2 was overexpressed in the epithelial cells of the transgenic mice. The mammary gland ductal trees were found to grow faster in MMTV-Kindlin-2 transgenic mice than in control mice during puberty. Kindlin-2 promoted mammary gland growth as indicated by more numerous duct branches and larger lumens, and more alveoli were formed in the mammary glands during pregnancy under Kindlin-2 overexpression. Importantly, mammary gland-specific expression of Kindlin-2 induced tumor formation at the age of 55 weeks on average. Additionally, the levels of estrogen receptor and progesterone receptor were decreased, whereas human epidermal growth factor receptor 2 and β-catenin were upregulated in the Kindlin-2-induced mammary tumors. | 10 | Differential expression analysis and regulatory network reconstruction for genes associated with muscle growth and adipose deposition in obese and lean pigs显示文摘During the growth and development of skeletal muscle cells and adipose cells, the regulatory mechanism of micro-effect polygenes determines porcine meat quality, carcass characteristics and other relative quantitative traits. Obese and lean type pig breeds show obvi- ous differences in muscle growth and adipose deposition; however, the molecular mechanism underlying this phenotypic variation remains unknown. We used pathway-focused oligo microarray studies to examine the expression changes of 140 genes associated with muscle growth and adipose deposition in longissimus dorsi muscle at six growth stages (birth, 1, 2, 3, 4 and 5 months) of Landrace (a leaner, Western breed) and Taihu pigs (a fatty, indigenous, Chinese breed). Variance analysis (ANOVA) revealed that differences in the expression of 18 genes in Landrace pigs and three genes in Taihu pigs were very significant (FDR-adjusted permutation, P < 0.01) and di?erences for 22 genes in Landrace pigs and seven genes in Taihu pigs were significant (FDR-adjusted permutation, P < 0.05) among six growth stages. Clustering analysis revealed a high level of signi?cance (FDR-adjusted, P < 0.01) for four gene expression patterns, in which genes that strongly up-regulated were mainly associated with the positive regulation of myofiber formation and fatty acid biogenesis and genes that strongly down-regulated were mainly associated with the inhibition of cell proliferation and positive regulation of fatty acid b-oxidation. Based on a dynamic Bayesian network (DBN) model, gene regulatory networks (GRNs) were reconstructed from time-series data for each pig breed. These two GRNs initially revealed the distinct differences in physiological and biochemical aspects of muscle growth and adipose deposition between the two pig breeds; from these results, some potential key genes could be identified. Quantitative real-time RT-PCR (QRT-PCR) was used to verify the microarray data for five modulated genes, and a good correlation between the two measures of expression was observed for both two pig breeds at different growth stages (r = 0.876 ± 0.095). | 13 | Atmospheric fine particles in a typical coastal port of Yangtze River Delta显示文摘In recent decades, coastal ports have experienced rapid development and become an important economic and ecological hub in China. Atmospheric particle is a research hotspot in atmospheric environmental sciences in inland regions. However, few studies on the atmospheric particle were conducted in coastal port areas in China, which indeed suffers atmospheric particle pollution. Lack of the physicochemical characteristics of fine particles serves as an obstacle toward the accurate control for air pollution in the coastal port area in China. Here, a field observation was conducted in an important coastal port city in Yangtze River Delta from March 6 to March 19, 2019. The average PM2.5 concentration was 63.7 ±27.8 μg/m^3 and NO3^-, SO4^2-, NH4^+, and organic matter accounted for ?60% of PM 2.5. Fe was the most abundant trace metal element and V as the ship emission indicator was detected. Transmission electron microscopy images showed that SK-rich, soot, Fe, SK-soot and SK-Fe were the major individual particles in the coastal port. V and soluble Fe were detected in sulfate coating of SK-Fe particles. We found that anthropogenic emissions, marine sea salt, and secondary atmosphere process were the major sources of fine particles. Backward trajectory analysis indicated that the dominant air masses were marine air mass, inland air mass from northern Zhejiang and inland-marine mixed air mass from Shandong and Shanghai during the sampling period. | 20 | Fertilization regimes affect the soil biological characteristics of a sudangrass and ryegrass rotation system显示文摘The sudangrass (Sorghum sudanense) and ryegrass (Lolium multiflorum L.) rotation is an intensive and new cropping system in Central China.Nutrient management practices in this rotation system may influence soil fertility,the important aspects of which are soil biological properties and quality.As sensitive soil biological properties and quality indicators,soil microbial community activity,microbial biomass,enzyme activities,soil organic matter (SOM) and total N resulting from different fertilization regimes in this rotation system were studied through a four-year field experiment from April 2005 to May 2009.Treatments included control (CK),fertilizer phosphorus and potassium (PK),fertilizer nitrogen and potassium (NK),fertilizer nitrogen and phosphorus (NP) and a fertilizer nitrogen,phosphorus and potassium combination (NPK).Soil microbial community activities in the NK,NP and NPK treatments were significantly lower than those in the CK and PK treatments after the sudangrass and ryegrass trial.The highest microbial biomass C,microbial biomass N,SOM,total N,sucrase and urease activities were found in the NPK treatment,and these soil quality indicators were significantly higher in the NK,NP and NPK treatments than in the PK and CK treatments.Soil microbial biomass and enzyme activities were positively associated with SOM in the sudangrass and ryegrass rotation system,indicating that fertilization regimes,especially N application,reduced microbial community activity in the soil.Proper fertilization regimes will increase microbial biomass,enzyme activity and SOM and improve soil fertility. | LI WenXi LU JianWei LI FangBai WANG Yan LU JunMing LI XiaoKun | 2011 | Science China(Life Sciences)2011,54,6: | 2 |
