1Characterization of drug responses of mini patient-derived xenografts in mice for predicting cancer patient clinical therapeutic response显示文摘Background:Patient-derived organoids and xenografts(PDXs)have emerged as powerful models in functional diag-nostics with high predictive power for anticancer drug response.However,limitations such as engraftment failure and time-consuming for establishing and expanding PDX models followed by testing drug efficacy,and inability to subject to systemic drug administration for ex vivo organoid culture hinder realistic and fast decision-making in selecting the right therapeutics in the clinic.The present study aimed to develop an advanced PDX model,namely MiniPDX,for rapidly testing drug efficacy to strengthen its value in personalized cancer treatment.Methods:We developed a rapid in vivo drug sensitivity assay,OncoVee®MiniPDX,for screening clinically relevant regimens for cancer.In this model,patient-derived tumor cells were arrayed within hollow fiber capsules,implanted subcutaneously into mice and cultured for 7 days.The cellular activity morphology and pharmacokinetics were systematically evaluated.MiniPDX performance(sensitivity,specificity,positive and negative predictive values)was examined using PDX as the reference.Drug responses were examined by tumor cell growth inhibition rate and tumor growth inhibition rate in PDX models and MiniPDX assays respectively.The results from MiniPDX were also used to evaluate its predictive power for clinical outcomes.Results:Morphological and histopathological features of tumor cells within the MiniPDX capsules matched those both in PDX models and in original tumors.Drug responses in the PDX tumor graft assays correlated well with those in the corresponding MiniPDX assays using 26 PDX models generated from patients,including 14 gastric cancer,10 lung cancer and 2 pancreatic cancer.The positive predictive value of MiniPDX was 92%,and the negative predictive value was 81%with a sensitivity of 80%and a specificity of 93%.Through expanding to clinical tumor samples,Min-iPDX assay showed potential of wide clinical application.Conclusions:Fast in vivo MiniPDX assay based on capsule implantation was developed-to assess drug responses of both PDX tumor grafts and clinical cancer specimens.The high correlation between drug responses of paired MiniPDX and PDX tumor graft assay,as well as translational data suggest that MiniPDX assay is an advanced tool for personalized cancer treatment.Feifei Zhang Wenjie Wang Yuan Long Hui Liu Jijun Cheng Lin Guo Rongyu Li Chao Meng Shan Yu Qingchuan Zhao Shun Lu Lili Wang Haitao Wang Danyi Wen 2018Cancer Communications2018,38,1:20
2Effects of microstructure on the torsional properties of biodegradable WE43 Mg alloy显示文摘Torsional properties are important performance parameters for bone screw applications,but they are seldom studied,especially for newly developed biodegradable Mg alloys.In this study,WE43 Mg alloy with different microstructures was achieved by equal channel angular pressing(ECAP)and heat treatment,and their torsional properties were studied.In addition,tensile properties were also tested as a comparison.The results indicated that grain refinement led to higher torsional strength and ductility,while the second phases improved the torsional strength but reduced the ductility.The texture was strengthened after ECAP,as a result the tensile strength increased,but the torsional strength did not increase and even decreased,especially for 2-pass ECAP sample with a typical basal fiber texture.The basal plane orientation deviation from the extrusion direction after 4-pass ECAP resulted in higher torsional strength and lower torsional ductility,but lower tensile strength and higher tensile ductility were obtained.This implied that a strong fiber texture would reduce the torsional strength but improve the torsional ductility,which was different from its effect on tensile properties.Yi Zhang Lili Tan Qingchuan Wang Ming Gao Iniobong P.Etim Ke Yang 2020Journal of Materials Science & Technology2020,47,16:4
3Mandatory IFRS adoption and executive compensation:Evidence from China显示文摘This study investigates how the mandatory adoption of International Financial Reporting Standards(IFRS) affects the contractual benefits of using accounting information to determine executive compensation in China. After controlling for firm and corporate governance characteristics, we find strong evidence supporting the positive role of mandatory IFRS adoption on the accounting-based performance sensitivity of executive compensation. Subsample analysis suggests that improvements in accounting-based performance sensitivity after IFRS adoption differ across regions with various levels of institutional quality and across firms that are affected to a different extent by the adoption. 4Bi/BiOCl Nanosheets Enriched with Oxygen Vacancies to Enhance Photocatalytic CO_(2)Reduction显示文摘BiOCl has been used in the photoreduction of CO_(2),but exhibits limited photocatalytic activity.In this study,Bi was in situ reduced and deposited on the surface of(001)-dominated BiOCl nanosheets by NaBH 4 to form Bi/BiOCl nanosheets enriched with oxygen vacancies.The as-prepared Bi/BiOCl nanosheets having low thickness(ca.10 nm)showed much higher concentration of oxygen vacancies compared to Bi/BiOCl nanoplates having high thickness(ca.100 nm).Subsequently,the photocatalytic activity of the Bi/BiOCl nanosheets enriched with oxygen vacancies for CO_(2)reduction was dramatically enhanced and much higher than that of BiOCl nanoplates,nanosheets,and Bi/BiOCl nanoplates.It showed that the improved photocatalytic activity in the reduction of CO_(2)can be attributed to the enhanced separation effi ciency of photogenerated electron–hole pairs of the oxygen vacancies on BiOCl nanosheets and Bi metals.This work demonstrated that the in situ reduction of non-noble metals on the surface of BiOCl nanosheets that are enriched with oxygen vacancies is favorable for increasing photocatalytic CO_(2)reduction.Shuqi Wu Junbu Wang Qingchuan Li Zeai Huang Zhiqiang Rao Ying Zhou 2021Transactions of Tianjin University2021,27,2:3
5A global alfalfa diversity panel reveals genomic selection signatures in Chinese varieties and genomic associations with root development显示文摘Alfalfa(Medicago sativa L.) is an important forage crop worldwide. However, little is known about the effects of breeding status and different geographical populations on alfalfa improvement. Here, we sequenced 220 alfalfa core germplasms and determined that Chinese alfalfa cultivars form an independent group, as evidenced by comparisons of FSTvalues between different subgroups, suggesting that geographical origin plays an important role in group differentiation. By tracing the influence of geographical regions on the genetic diversity of alfalfa varieties in China, we identified 350 common candidate genetic regions and 548 genes under selection. We also defined 165 loci associated with 24 important traits from genome-wide association studies. Of those, 17 genomic regions closely associated with a given phenotype were under selection, with the underlying haplotypes showing significant differences between subgroups of distinct geographical origins. Based on results from expression analysis and association mapping,we propose that 6-phosphogluconolactonase(MsPGL) and a gene encoding a protein with NHL domains(MsNHL) are critical candidate genes for root growth. 6Quantifying 3D cell-matrix interactions during mitosis and the effect of anticancer drugs on the interactions显示文摘The mechanical force between cells and the extracellular microenvironment is crucial to many physiological processes such as cancer metastasis and stem cell differentiation. Mitosis plays an essential role in all these processes and thus an in-depth understanding of forces during mitosis gains insight into disease diagnosis and disease treatment. Here, we develop a traction force microscope method based on monolayer fluorescent beads for measuring the weak traction force (tens of Pa) of mitotic cells in three dimensions. We quantify traction forces of human ovarian granulosa (KGN) cells exerted on the extracellular matrix throughout the entire cell cycle in three dimensions. Our measurements reveal how forces vary during the cell cycle, especially during cell division. Furthermore, we study the effect of paclitaxel (PTX) and nocodazole (NDZ) on mitotic KGN cells through the measurement of traction forces. Our results show that mitotic cells with high concentrations of PTX exert a larger force than those with high concentrations of NDZ, which proved to be caused by changes in the structure and number of microtubules. These findings reveal the key functions of microtubule in generating traction forces during cell mitosis and explain how dividing cells regulate themselves in response to anti-mitosis drugs. 8Association of systemic inflammation and body mass index with survival in patients with resectable gastric or gastroesophageal junction adenocarcinomas显示文摘Objective:The systemic inflammation index and body mass index(BMI)are easily accessible markers that can predict mortality.However,the prognostic value of the combined use of these two markers remains unclear.The goal of this study was therefore to evaluate the association of these markers with outcomes based on a large cohort of patients with gastric cancer.Methods:A total of 2,542 consecutive patients undergoing radical surgery for gastric or gastroesophageal junction adenocarcinoma between 2009 and 2014 were included.Systemic inflammation was quantified by the preoperative neutrophil-to-lymphocyte ratio(NLR).High systemic inflammation was defined as NLR≥3,and underweight was defined as BMI<18.5 kg/m2.Results:Among 2,542 patients,NLR≥3 and underweight were common[627(25%)and 349(14%),respectively].In the entire cohort,NLR≥3 or underweight independently predicted overall survival(OS)[hazard ratio(HR):1.236,95%confidence interval(95%CI):1.069–1.430;and HR:1.600,95%CI:1.350–1.897,respectively]and recurrence-free survival(RFS)(HR:1.230,95%CI:1.054–1.434;and HR:1.658,95%CI:1.389–1.979,respectively).Patients with both NLR≥3 and underweight(vs.neither)had much worse OS(HR:2.445,95%CI:1.853–3.225)and RFS(HR:2.405,95%CI:1.802–3.209).Furthermore,we observed similar results in subgroup analyses according to pathological stage,age,and postoperative chemotherapy.Conclusions:Our results showed that preoperative elevated NLR and decreased BMI had a significant negative effect on survival.Underweight combined with severe inflammation could enhance prognostication.Taking active therapeutic measures to reduce inflammation and increase nutrition may help improve outcomes.Xianchun Gao Yanan Pan Weili Han Caie Hu Chenchen Wang Ling Chen Yong Guo Yupeng Shi Yan Pan Huahong Xie Liping Yao Jianjun Yang Jianyong Zheng Xiaohua Li Xiaonan Liu Liu Hong Jipeng Li Mengbin Li Gang Ji Zengshan Li Jielai Xia Qingchuan Zhao Daiming Fan Kaichun Wu Yongzhan Nie 2021Cancer Biology & Medicine2021,18,1:2

10High strength and ductility achieved in friction stir processed Ni-Co based superalloy with fine grains and nanotwins显示文摘The trade-off between strength and ductility has been an enormous difficulty in the field of materials for an extended time due to their inverse correlation. In this work, friction stir processing(FSP) was for the first time performed to high-strength and high-melting-point Ni-Co based superalloy(GH4068),and enhanced strength and ductility were achieved in FSP samples. At room temperature, the FSP sample demonstrated significantly higher yield strength and ultimate tensile strength(1290 and 1670 MPa)than that of the base material(BM, 758 and 904 MPa) and advanced wrought GH4068 alloy(982 and 1291 MPa), concurrent with high tensile ductility(~24%). Compared with the BM, 70% higher yield strength of the FSP sample results from the remarkable contribution of grain-boundary and nanotwin strengthening, which has been confirmed by the multimechanistic model studied in this work. More importantly, with increasing temperature, an excellent strength-ductility synergy was obtained at 400 ℃,i.e., the yield strength of the FSP sample was increased by more than 50% compared with the BM(from789 to 1219 MPa);more interestingly, the elongation was also significantly increased from 17.9% in the BM to 28.5% in the FSP sample. Meanwhile, the Portevin-Le Chatelier effect was observed in the engineering stress-strain curve. The occurrence of this effect may be attributed to the interaction between solutes and defects like twins and mobile dislocations. 11Quantitative trait locus mapping of yield and plant height in autotetraploid alfalfa(Medicago sativa L.)显示文摘Alfalfa(Medicago sativa L.)is the most widely grown forage legume crop worldwide.Yield and plant height are important agronomic traits influenced by genetic and environmental factors.The objective of this study was to identify quantitative trait loci(QTL)and molecular markers associated with alfalfa yield and plant height.To understand the genetic basis of these traits,a full-sib F1 population composed of 392 individuals was developed by crossing a low-yielding precocious alfalfa genotype(male parent)with a high-yielding latematuring alfalfa cultivar(female parent).The linkage maps were constructed with 3818 single-nucleotide polymorphism(SNP)markers on 64 linkage groups.QTL for yield and plant height were mapped using phenotypic data for three years.Sixteen QTL associated with yield and plant height were identified on chromosomes 1 to 8.Six QTL explained more than 10%of phenotypic variation,representing major loci controlling yield and plant height.One locus on chromosome 1 controlling yield traits had not been identified in previous studies.Three QTL co-located with other QTL(qyield-1 and qheight-7,qheight-5 and qyield-4,qheight-6,and qyield-6).With further validation,the markers closely linked with these QTL may be used for marker-assisted selection in breeding new alfalfa varieties with high yield.Fei He Ruicai Long Tiejun Zhang Fan Zhang Zhen Wang Xijiang Yang Xueqian Jiang Changfu Yang Xuxin Zhi Mingna Li Longxi Yu Junmei Kang Qingchuan Yang 2020The Crop Journal2020,8,5:1

14Genome Assembly of Alfalfa Cultivar Zhongmu-4 and Identification of SNPs Associated with Agronomic Traits显示文摘Alfalfa(Medicago sativa L.)is the most important legume forage crop worldwide with high nutritional value and yield.For a long time,the breeding of alfalfa was hampered by lacking reliable information on the autotetraploid genome and molecular markers linked to important agronomic traits.We herein reported the de novo assembly of the allele-aware chromosome-level genome of Zhongmu-4,a cultivar widely cultivated in China,and a comprehensive database of genomic variations based on resequencing of 220 germplasms.Approximate 2.74 Gb contigs(N50 of 2.06 Mb),accounting for 88.39%of the estimated genome,were assembled,and 2.56 Gb contigs were anchored to 32 pseudo-chromosomes.A total of 34,922 allelic genes were identified from the allele-aware genome.We observed the expansion of gene families,especially those related to the nitrogen metabolism,and the increase of repetitive elements including transposable elements,which probably resulted in the increase of Zhongmu-4 genome compared with Medicago truncatula.Population structure analysis revealed that the accessions from Asia and South America had relatively lower genetic diversity than those from Europe,suggesting that geography may influence alfalfa genetic divergence during local adaption.Genome-wide association studies identified 101 single nucleotide polymorphisms(SNPs)associated with 27 agronomic traits.Two candidate genes were predicted to be correlated with fall dormancy and salt response.We believe that the alleleaware chromosome-level genome sequence of Zhongmu-4 combined with the resequencing data of the diverse alfalfa germplasms will facilitate genetic research and genomics-assisted breeding in variety improvement of alfalfa.Ruicai Long Fan Zhang Zhiwu Zhang Mingna Li Lin Chen Xue Wang Wenwen Liu Tiejun Zhang Long-Xi Yu Fei He Xueqian Jiang Xijiang Yang Changfu Yang Zhen Wang Junmei Kang Qingchuan Yang 2022Genomics, Proteomics & Bioinformatics2022,20,1:1

20Genotype analysis of rotaviruses isolated from children during a phase III clinical trial with the hexavalent rotavirus vaccine in China显示文摘The oral hexavalent live human-bovine reassortant rotavirus vaccine(RV6)developed by Wuhan Institute of Biological Products Co.,Ltd(WIBP)has finished a randomized,placebo-controlled phase III clinical trial in four provinces of China in 2021.The trail demonstrated that RV6 has a high vaccine efficacy against the prevalent strains and is safe for use in infants.During the phase III clinical trial(2019–2021),200 rotavirus-positive fecal samples from children with RV gastroenteritis(RVGE)were further studied.Using reverse transcription-polymerase chain reaction and high-throughput sequencing,VP7 and VP4 sequences were obtained and their genetic characteristics,as well as the differences in antigenic epitopes of VP7,were analyzed in detail.Seven rotavirus genotypes were identified.The predominant rotavirus genotype was G9P[8](77.0%),followed by prevalent strains G8P[8](8.0%),G3P[8](3.5%),G3P[9](1.5%),G1P[8](1.0%),G2P[4](1.0%),and G4P[6](1.0%).The amino acid sequence identities of G1,G2,G3,G4,G8,and G9 genotypes of isolates compared to the vaccine strains were 98.8%,98.2%–99.7%,88.4%–99.4%,98.2%,94.2%–100%,and 93.9%–100%,respectively.Notably,the vaccine strains exhibited high similarity in amino acid sequence,with only minor differences in antigenic epitopes compared to the Chinese endemic strains.This supports the potential application of the vaccine in preventing diseases caused by rotaviruses.Wenqi Zou Qingchuan Yu Yan Liu Qingliang Li Hong Chen Jiamei Gao Chen Shi Ying Wang Wei Chen Xuan Bai Biao Yang Jiuwei Zhang Ben Dong Bo Ruan Liuyifan Zhou Gelin Xu Zhongyu Hu Xiaoming Yang 2023Virologica Sinica2023,38,6:0
