| 1 | Celastrus orbiculatus extract induces mitochondrial-mediated apoptosis in human hepatocellular carcinoma cells显示文摘OBJECTIVE:To investigate the apoptotic effects and underlying molecular mechanisms of Celastrus orbiculatus(C.orbiculatus) extract in human hepatocellular carcinoma cells.METHODS:Human hepatocellular carcinoma cells(HCCLM6) were treated with C.orbiculatus extract(COE) at different nontoxic concentrations(10,20,40,80,and 160 μg/mL).The effect of COE on HC-CLM6 viability was examined using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide(MTT) assays.Cellular apoptosis following COE treatment was assessed by flow cytometry and western blot analysis.RESULTS:COE significantly inhibited cell viability and induced apoptosis of HCCLM6 cells in a dose-dependent manner.Apoptosis was accompanied by increased Bax expression and decreased Bcl-2 expression.In addition,COE treatment led to the release of cytochrome c,activation of caspase-3,and cleavage of poly(ADP-ribose) polymerase(PARP).Furthermore,activation of extracellular signal-regulated kinase(ERK),p38 kinase,and c-Jun N-terminal kinase(JNK) phosphorylation,and down-regulation of Akt phosphorylation was observed.CONCLUSION:COE induces mitochondrial-mediated,caspase-dependent apoptosis in HCCLM6 cells,which might be attributed to the activation of mitogen-activated protein kinase(MAPK) and inhibition of Akt signaling pathways.These data suggest that COE may be a potential treatment for human hepatocellular carcinoma. | Hua Zhang Yayun Qian Yanqing Liu Guoqing Li Pingfang Cui Yaodong Zhu Hui Ma Xue Ji Shiyu Guo Hisamits Tadashi | 2012 | Journal of Traditional Chinese Medicine2012,32,4: | 18 |
| 2 | 2D MOF Nanoflake-Assembled Spherical Microstructures for Enhanced Supercapacitor and Electrocatalysis Performances显示文摘Metal–organic frameworks(MOFs) are of great interest as potential electrochemically active materials.However, few studies have been conducted into understanding whether control of the shape and components of MOFs can optimize their electrochemical performances due to the rational realization of their shapes. Component control of MOFs remains a significant challenge. Herein, we demonstrate a solvothermal method to realize nanostructure engineering of 2D nanoflake MOFs. The hollow structures withNi/Co-and Ni-MOF(denoted as Ni/Co-MOF nanoflakes and Ni-MOF nanoflakes) were assembled for their electrochemical performance optimizations in supercapacitors and in the oxygen reduction reaction(ORR). As a result, the Ni/CoMOF nanoflakes exhibited remarkably enhanced performance with a specific capacitance of 530.4 F g^(-1)at 0.5 A g^(-1)in1 M LiO H aqueous solution, much higher than that of NiMOF(306.8 F g^(-1)) and ZIF-67(168.3 F g^(-1)), a good rate capability, and a robust cycling performance with no capacity fading after 2000 cycles. Ni/Co-MOF nanoflakes also showed improved electrocatalytic performance for the ORR compared to Ni-MOF and ZIF-67. | 4 | Targeted mutagenesis of amino acid transporter genes for rice quality improvement using the CRISPR/Cas9 system显示文摘High grain protein content(GPC) reduces rice eating and cooking quality(ECQ). We generated OsAAP6 and OsAAP10 knockout mutants in three high-yielding japonica varieties and one japonica line using the CRISPR/Cas9 system. Mutation efficiency varied with genetic background in the T_0 generation, and GPC in the T_1 generation decreased significantly,owing mainly to a reduction in glutelin content. Amylose content was down-regulated significantly in some Osaap6 and all Osaap10 mutants. The increased taste value of these mutants was supported by Rapid Visco Analysis(RVA) profiles, which showed higher peak viscosity and breakdown viscosity and lower setback viscosity than the wild type. There were no significant deficiencies in agronomic traits of the mutants. This methodology uses a gamma distribution to model the material degradation, and the impact of imperfect maintenance actions on the system reliability is investigated. The state of a degrading system immediately after the imperfect maintenance action is assumed as a random variable and the maintenance time follows a geometric process. Furthermore, the explicit expressions for the long-run average cost and availability per unit time of the system are evaluated, an optimal policy (ξ*) could be determined numerically or analytically according to the optimization model. | 8 | Impending HRT wave precursors to the Wenchuan M_s8.0 earthquake and methods of earthquake impending prediction by using HRT wave显示文摘We deployed four geo-electric monitoring stations in Sichuan and Yunnan provinces from 2004, using the new generation of equipment (PS-100) and technologies to capture the HRT wave earthquake precursor. Before the Wenchuan Ms8.0 earthquake, we recorded the HRT wave precursor at the only operating station in Hongge (HG, Δ=465 km) and found that significant impending signal had been recorded at the station in the early morning ( 0―5 am) of 12th of May, 2008. The precursor for this earthquake is consistent with precursors recorded for other strong earthquakes. The measured physical properties (geo-resistivity and telluric-current) show tidal wave period oscillations from several days to several months before the earthquakes and the amplitude of such HT oscillation increases significantly towards the occurrence of an earthquake. These HT and RT waves from the epicenter have a causal relationship with the earthquakes that happened several days later. The arrival time of two RT waves is proportional to the distance from the station to the epicenter. The estimated natural decay of the amplitude is correlated with the natural period (T0) of the earthquake fault, which is proportional to the fault length. From this relationship, we can predict the earthquake magnitude. For magnitude 6―9 earthquakes, the natural period is about 1―6 hours. Such oscillation comes from the epicenter area and they can propagate several thousand kilometers in the Earth's crust. Before a strong earthquake in the shallow crust, the conductive pore fluid will experience major changes before the fault rapture. Such fluid change will emit an oscillation in the pore fluid pressure. This is the mechanism for the HRT wave generation. Since the China Earthquake Administration funded the HRT wave short-term earth-quake prediction project in 2003, the first record of HRT precursor wave has been recorded from the 2004-12-26 Sumatra Mw9.0 earthquake with the largest epicentre distance Δ=2900 km. Thereafter, we have captured HRT waves from more than twenty strong earthquakes, which are well-matched and show repeatability, consistency and regularity. All our observation with the HRT waves demonstrate that HRT wave precursors to earthquakes indeed exist. Strong earthquakes can be predicted and short-term and impending earthquake prediction is achievable in the very near future. | 10 | Synergistic and antagonistic interactions between potassium and magnesium in higher plants显示文摘Magnesium(Mg)affects various critical physiological and biochemical processes in higher plants,and its deficiency impedes plant growth and development.Although potassium(K)-induced Mg deficiency in agricultural production is widespread,the specific relationship of K with Mg and especially its competitive nature is poorly understood.This review summarizes current knowledge on the interactions between K and Mg with respect to their root uptake,root-to-shoot translocation and distribution in plants.Their synergistic effects on certain physiological functions are also described.The antagonistic effect of K on Mg is stronger than that of Mg on K in root absorption and transport within plants,indicating that the balanced use of K and Mg fertilizers is necessary for sustaining high plant-available Mg and alleviating K-induced Mg deficiency,especially in plant species with high K demand or in highavailable-K soil.The relationship between Mg and K in plant tissues may be antagonistic or synergistic depending on plant species,cell type,leaf age,source-and sink organs.There are synergistic effects of K and Mg on photosynthesis,carbohydrate transport and allocation,nitrogen metabolism,and turgor regulation.Definition of optimal K/Mg ratios for soils and plant tissues is desirable for maintaining proper nutritional status in plants,leading to a physiological state supporting crop production.Future research should concentrate on identifying the physiological and molecular mechanisms underlying the interactions between K and Mg in a given physiological function. | Kailiu Xie Ismail Cakmak Shiyu Wang Fusuo Zhang Shiwei Guo | 2021 | The Crop Journal2021,9,2: | 5 |

| 16 | The quality attribute of watershed ecosystem is more important than the landscape attribute in controlling erosion of red soil in southern China显示文摘Landscape and quality attributes are major ecosystem characteristics closely associated with soil conservation service(SCS).However,the intrinsic mechanisms by which these two attributes influence SCS are still unclear.Therefore,this study quantitatively analyzed the landscape pattern,ecological quality,and SCS in the Lianshui River watershed(a typical soil and water loss area of red soil in southern China)and its sub-watersheds in 2019.The boosted regression tree model was used to explore the influence of 15 factors(i.e.,landscape and quality attributes)on SCS at the sub-watershed scale.According to the results,compared with the landscape attribute,the quality attribute of the watershed ecosystem could better explain the spatial heterogeneity of SCS across 66 sub-watersheds.The overall degree of influence of five quality factors on SCS reached 57.81%,with the highest being the normalized differential build-up and bare soil index(NDBSI),at 25.11%.Among 10 landscape factors,aggregation had the greatest influence on SCS,at 28.64%.The relationships between key influencing factors and SCS were nonmonotonic and non-linear,with threshold effects.For example,NDBSI values of 0.18e0.41 had a positive influence on SCS,while NDBSI values of 0.41e0.65 had a negative influence on SCS.The findings broaden our understanding of the response of SCS to changes in landscape and quality attributes at the sub-watershed scale,and could offer comprehensive support for soil erosion management in the watershed ecosystem. | Qing Zhu Xi Guo Jiaxin Guo Jun Wu Yingcong Ye Wenbo Cai Shiyu Liu | 2022 | International Soil and Water Conservation Research2022,10,3: | 2 |

| 18 | Crosstalk between adipose-derived stem cells and chondrocytes:when growth factors matter显示文摘Adipose-derived stem cells(ASCs)and mesenchymal stem cells are promising for tissue repair because of their multilineage differentiation capacity.Our previous data confirmed that the implantation of mixed ASCs and chondrocytes into cartilage defects induced desirable in vivo healing outcomes.However,the paracrine action of ASCs on chondrocytes needs to be further elucidated.In this study,we established a co-culture system to achieve cell-to-cell and cell-to-tissue crosstalk and explored the soluble growth factors in both ASCs and chondrocytes supplemented with 1%fetal bovine serum to mimic the physiological microenvironment.In ASCs,we screened for growth factors by semi-quantitative PCR and quantitative real-time PCR and found that the expression of bone morphogenetic protein 2(BMP-2),vascular endothelial growth factor B(VEGFB),hypoxia inducible factor-1α(HIF-1α),fibroblast growth factor-2(FGF-2),and transforming growth factor-β1 significantly increased after co-culture in comparison with mono-culture.In chondrocytes,VEGFA was significantly enhanced after co-culture.Unexpectedly,the expression of collagen II and aggrecan was significantly down-regulated in the co-culture group compared with the mono-culture group.Meanwhile,among all the growth factors screened,we found that the BMP family members BMP-2,BMP-4,and BMP-5 were down-regulated and that VEGFB,HIF-1α,FGF-2,and PDGF were significantly decreased after co-culture.These results suggest that crosstalk between ASCs and chondrocytes is a pathway through the regulated growth factors that might have potential in cartilage repair and regeneration and could be useful for tissue engineering. | Juan Zhong Bin Guo Jing Xie Shuwen Deng Na Fu Shiyu Lin Guo Li Yunfeng Lin Xiaoxiao Cai | 2015 | Bone Research2015,3,4: | 1 |
